ABSTRACT

Atherosclerosis is usually the underlying cause of cardiovascular diseases. With the change in diet structure and living environment, it has become an increasingly serious global health problem, posing a huge challenge to public health. Berberine, also known as flavonidol, is an isoquinoline-type quaternary alkaloid with purgative and detoxifying effects. Berberine and its derivatives have antibacterial, antiviral, anti-inflammatory, antioxidant, hypoglycemic, hypolipidemic, and atherosclerosis prevention effects, etc. Recent research results showed that berberine and its derivatives can play an important role in atherosclerosis prevention through a hypolipidemic effect, anti-oxidative stress and anti-inflammatory activity, improvement of vascular endothelial dysfunction, and regulation of intestinal microbiota. In this review paper, the research progress on the mechanism of action of berberine and its derivatives in the prevention of atherosclerosis was reviewed from the perspectives of a lipid-regulating effect, inhibition of oxidative stress and the inflammatory response, improvement of vascular endothelial dysfunction, and regulation of intestinal microbiota. The aim of this paper is to provide a theoretical basis for reducing the occurrence of atherosclerosis, improving the clinical symptoms of patients, and further developing berberine-based drugs.

ABSTRACT

As a cardiovascular complication of diabetes, diabetic cardiomyopathy seriously affects the prognosis of patients with diabetes. The incidence and mortality of diabetic cardiomyopathy increase, and has emerged as a research hotspot in current years. The pathogenesis of diabetic cardiomyopathy is complex, involving a range of signaling pathways in its incidence and development. Nuclear factor NF-E2-related factor 2 (Nrf2), as a powerful antioxidant gene, enhances the capacity of the myocardium to withstand oxidative stress via interplay with different signaling elements and exerts anti-inflammatory response, anti-myocardial fibrosis, and anti-apoptosis effects. Researches have shown that certain ingredients of traditional Chinese medicine can alleviate the myocardial injury by affecting the relationship of Nrf2 with other signaling factors via enhancing the expression of Nrf2. Here we review the role of Nrf2 and therapy of traditional Chinese medicine in diabetic cardiomyopathy in hope of providing referential idea for the treatment of diabetic cardiomyopathy. A reference for the prevention and therapy of diabetic cardiomyopathy.

ABSTRACT

As a common disease worldwide, alcoholic liver injury is caused by long-term or excessive intake of alcohol and triggers cell death due to alcohol metabolism and reactive oxygen species(ROS)-mediated cytotoxicity. Wangshi Baochi(WSBC) Pills have been widely adopted in clinical practice for evacuating stasis, resolving turbidity, clearing heat, tranquilizing mind, invigorating sto-mach, promoting digestion, purging fire and removing toxin. This study aimed to investigate the efficacy of WSBC Pills in dispelling the effect of alcohol and protecting against acute alcoholic liver/stomach injury in mice, and preliminarily investigate its possible mole-cular mechanism. The results found that the preventive treatment with WSBC Pills contributed to elevating the activity of alcohol dehydrogenase(ADH) and its expression in liver and shortening the time required for sobering up of mice with acute alcoholic liver injury. The staining of liver pathological sections as well as the detection of serum aspartate aminotransferase(AST) and alanine aminotransferase(ALT) and liver ROS levels revealed that WSBC Pills protected the liver by reducing serum AST and ALT. It suppressed oxidative stress-induced liver injury by lowering liver ROS and elevating superoxide dismutase(SOD), and the liver-protecting effect was superior to that of silibinin. Western blot assay confirmed that WSBC Pills inhibited the oxidative stress by up-regulating SOD1 and NAD(P)H: quinone oxidoreductase 1(NQO-1). In addition, WSBC Pills lowered the ROS level to protect against the acute alcoholic stomach injury in mice. The findings have suggested that WSBC Pills alleviated the acute alcoholic liver/stomach injury in mice by increasing ADH and resisting oxidative stress.

ABSTRACT

Patchouli Alcohol (PA) is the main bioactive constituent of Pogostemon cablin Pharmacological researches have reported various benefits of PA, including anti-infammatory, anti-gastric ulcer, anticancer, protecting against alung and brain injury, antibacterial, prevention of mastitis, colitis and arteriosclerosis, anti-photoaging, treatment of diarrhea irritable bowel syndrome, antioxidant stress, vasodilation and anti-injury.The preliminary study on the mechanism showed that anti-inflammatory effect was related to the down-regulation of mRNA expression of inflammatory mediators to inhibit the production of proinflammatory cytokines and the reduction of extracellular regulated protein kinases (ERK)-mediated nuclear factor kappa B (NF-κB) activation. The anti-gastric ulcer, protection of lung injury and brain injury and anti-photoaging were related to antioxidant and anti-inflammatory effects. The mechanism of anti-human leukemic cell line MV4-11 migh be associated with the changes of NF-κB, phosphorylated M2 pyruvate kinase (p-PKM2) and cysteine aspartate protease-3 (Caspase-3) protein expressions. The proposed mechanisms of anti-human colorectal cancer cells included the inhibition of histone deacetylase (HDAC)-2 expression and HDAC enzyme activity, and subsequent downregulation of c-myc and activation of NF-κB pathway.Apoptosis of A549 cells was induced by blocking the phosphorylation of epidermal growth factor receptor (EGFR) pathway and activating c-Jun N-terminal kinase (JNK). Anti-prostate cancer was related to the activation of mitochondrial cytochrome C apoptosis pathway, and its antiviral effect may be related to the regulation of RIG-1-like helicases (RLH) signal pathway or the direct binding of virus protein or nucleic acid and the binding site of virus to cells to block the adsorption of virus to cells. The mechanism of its antibacterial, prevention of mastitis, colitis and arteriosclerosis was closely related to anti-inflammation.The pharmacological effects of PA need to be further studied. In order to accelerate the exploration and discovery other molecular mechanism of PA, and expand its application prospects. This review comprehensively summarizes the metabolism, biological activities, and possible mechanism of PA, in order to provide references for the development and application of PA.

ABSTRACT

OBJECTIVE@#Infectious diseases such as typhoid fever lead to the formation of free radicals which can damage the body. Many medicinal plants have antioxidant molecules that neutralize free radicals. The present work evaluated the antioxidant activity and histopathological effects of the dichloromethane fraction of Dichrocephala integrifolia in Salmonella typhi-infected rats.@*METHODS@#The S. typhi-infected rats concurrently received daily doses of D. integrifolia extract at doses of 25, 50 and 100 mg/kg body weight or ciprofloxacin (5 mg/kg body weight) for 15 days. Body temperature was measured daily during infection and treatment periods. At the end of treatment period, the animals were sacrificed and biological responses including hematological parameters, superoxide dismutase and catalase activities, and glutathione, malondialdehyde and nitric oxide concentrations were evaluated.@*RESULTS@#The elevated body temperature induced by infection was significantly decreased in animals treated with 25, 50 or 100 mg/kg of the extract. Platelet levels decreased slightly in infected rats, while treatment with the dichloromethane fraction of D. integrifolia significantly increased platelet levels; this response was greater than that elicited by ciprofloxacin. The doses of 50 and 100 mg/kg of the dichloromethane fraction of D. integrifolia notably decreased monocyte and neutrophil values. Activity of superoxide dismutase and catalase and levels of glutathione in the tissues of treated animals were increased significantly (P < 0.01), while malondialdehyde and nitric oxide levels were significantly decreased (P < 0.01), following treatment with the dichloromethane fraction of D. integrifolia.@*CONCLUSION@#The results of this study show that the dichloromethane fraction of D. integrifolia has protective effects against a series of pathological conditions initiated by oxidation and tissue damage in the course of a S. typhi infection.

ABSTRACT

Sepsis is a critical complication of severe trauma, large area burns, infection, and major surgery etc., which can induce septic shock and multiple organ dysfunction syndrome (MODS), and its incidence and mortality are always high. Nuclear factor-E2 related factor 2 (Nrf 2) is an important transcription factor of antioxidant stress response. It plays a key role in the treatment of sepsis by regulating the expression of antioxidant enzyme and phase Ⅱ detoxification enzyme mediated by antioxidant response element (ARE). In this article we review the changes, related regulation mechanism and reversing measures of Nrf 2 in sepsis, aiming to provide a reference for the effective intervention measures of sepsis.
