ABSTRACT

Objective To improve the performance of artificial joint at biological fixed interface by frettingmechanical stimulation. Methods Taking the bonding sample of active rat bone tissues and inactive titanium bead coating as the research object, by using the self-developed fretting mechanical stimulation test device, the effects of mechanical stimulation with different tangential fretting amplitudes on growth promotion and micro-injury of bone tissues at fixed interface were explored, and the mechanism of growth promotion and micro-injury of bonetissues on the surface of titanium bead coating under fretting stimulation was revealed. Results The mechanical stimulation group with 40 μm fretting amplitude showed the maximum interface bonding force between bone tissues and titanium bead coating. The tangential fraction force-displacement curves changed from elliptic shape to linear shape, and the bonding interface was in adhesion area, which was difficult to become loose. The bone tissues had the most complete structure, and the number of tissue cells adhering and growing on the surface of titanium bead coating was the largest, as well as the distribution range was the widest. With the increase of fretting amplitude, the promoting effect of fretting stimulation on bone growth weakened, the bonding strength between bone tissues and titanium bead coating interface gradually decreased, the internal cavity area increased, the number and proliferation activity of tissue cells decreased to varying degrees as well. Conclusions Fretting stimulation with different amplitudes can promote the growth of bone tissue and cause micro-injury at the same time, and the optimal fretting amplitude is 40 μm.

ABSTRACT

Introduction: The growth of skeletal tissues constitutes a secondary, compensatory and mechanically obliged response to change the functional matrixes. When the growth of functional matrixes is affected for any reason, the skeletal tissues also respond with a degree of affectation depending on the moment in which it occurs. Then the mandible, as part of the viscerocranium, must present that relationship with the hyoid bone directly. Objective: To associate the morphological behavior of the hyoid bone with the morphological variables of the mandible and verify if the morphology of the bones belonging to the skeletons studied is determined by the set of soft tissues that surround them and set the pace of the growth remodeling process. Material and Methods: The continuation of the osteological study was carried out in a bone sample of 82 skulls by performing morphometric measurements of the hyoid bone and the mandible. Pearson's linear correlation coefficient matrices in SPSS Version 22 were used to evaluate the relationship between the morphology of the hyoid bone and the bones of the mandible. Results: The relationship between the morphology of the hyoid bone and the growth of the viscerocranium is corroborated by the positive and significant correlation between several morphological variables of the hyoid bone obtained - both at the level of its body and its greater horns -, and the mandible. Conclusions: These findings corroborate the association between the morphology of the hyoid bone and the growth of the mandible(AU)

Introduction: Body movement obeys and produces activity in the skeletal muscle for which there must be a static muscle equilibrium that produces the movement of the anatomic elements involved in it, either as a result of volition or as the unconscious perception of a stimulus. Objective: To associate the morphological behavior of the hyoid bone with some morphological variables of the viscerocranium of skeletons (except the jaw). Material and Methods: An osteological study was carried out in a bone sample of 82 skulls by performing morphometric measurements of the hyoid bone and the bones of the viscerocranium. Pearson's correlation coefficient and SPSS Version 22 were used to evaluate the relationship between the morphology of the hyoid bone and the facial bones. Morphological variables of the viscerocranium include: bizygomatic width (BW), external transverse width of the hard palate (ETWHP), external sagittal width of the hard palate (ESWHP), and the height of the middle third of the face (MTF). Results: A very strong positive correlation between different morphological variables of the hyoid bone, -both at the level of its antlers or greater horns- and the morphological variables of the viscerocranium was obtained. Conclusions: These findings corroborate the association between the hyoid bone and the growth of facial bones(AU)

ABSTRACT

SUMMARY: An increased thickening of the frontal bone by irregular laminar additions on the inner surface just deep to the dura mater have been known in the archaeological and medical record as hyperostosis frontalis interna (HFI). The distribution of this is idiosyncratically restricted to the frontal and has no known etiology. The prevalence among post-menopausal females and rarity in males suggests that it is hormonally driven. Here we report histopathological findings of particularly hormonally active organs (pituitaries, gonads and liver) from a geriatric cadaveric sample in which HFI is assessed. HFI was present in 50 % of males (7/14) and 95 % (21/22) of females. All males with HFI had testicular atrophy or had testes absent. Males with HFI category C or D had moderate to severe testicular atrophy. Decreased numbers of interstitial cells (Leydig cells) were present in 83.3 % of males with HFI. All but one female (21/22) from this study exhibited evidence of HFI, and ovarian pathologies were unevenly distributed (fibromas in two) and most exhibited signs of being healthy and post-menopausal. Liver pathologies had opposite patterns between the sexes, with more liver pathologies occurring among males without HFI (particularly passive congestion and bile stasis). The only exceptions were that the one case of liver neoplasia was found in a male with HFI and steotosis was found in two cases with HFI and one case without HFI. In females all cases of liver pathologies (steotosis, hepatitis, passive congestion, fibrosis, and bile stasis) were associated with HFI. It appears that gonadal pathology is most closely associated with HFI in males but not females, suggesting that the role of estradiol in this unusual growth of bone in geriatric humans may be worth investigating further.

ABSTRACT

BACKGROUND: At present, scholars at home and abroad try to promote the growth of bone graft by changing the types and morphology of implant materials, and improving the thickness and smoothness of induced membrane. OBJECTIVE; To compare the degree of vascularization and the expression of some osteogenic factors in the polymethylmethacrylate bone cement membrane with different surface roughness degrees at different femoral bone defects in rats. METHODS: Forty-eight male Sprague-Dawley rats (purchased from the Experimental Animal Center of Guangzhou University of Chinese Medicine) were used to establish rat models of critical-size femur defect. These rat modes were then randomly divided into groups A, B, C and D. Polymethylmethacrylate bone cement occupiers with surface roughness 0.05). Immunohistochemical staining revealed that the positive expression of bone morphogenetic protein 2, transforming growth factor (31 and vascular endothelial growth factor protein was similar among four groups (P>0.05). These findings suggest that the change of surface roughness of bone cement has no significant effect on the histomorphological structure of induced membrane and the expression of bone morphogenetic protein 2, transforming growth factor pi and vascular endothelial growth factor in induced membrane at 6 weeks after implantation of bone cement spacer.

ABSTRACT

Zoledronic acid (ZA) is an antiresorptive drug used in children with bone diseases like osteogenesis imperfecta, juvenil osteoporosis, fibrous dysplasia and primary bone tumors. The aim of the present study was to evaluate the effects of ZA dose accumulation on growing bone during different periods of treatment in normal rats. Methods: A 4x2 factorial design was used to study the effect of the dose of ZA (D: 0-2.5-12.5-25 µg Z/kg body weight/s.c. weekly) and the length of treatment (T: 15-30 days) in normal female Sprague Dawley rats. Bone morphometric, histomorphometric, densitometric and biomechanical studies were performed. Results: Femoral length and cross-sectional area were affected by both D and T. A significant interaction between D and T was observed in length with a lower value at higher dose and 30 days of treatment. Growth plate of the tibia showed a decrease in total thickness with D and T. Histomorphometric and connectivity parameters of trabecular bone were significantly increased with D and several parameters were also affected by T. Cortical bone strength was increased only with T. Biomechanical parameters of trabecular bone showed significant interaction with greater effect at higher D and T. Conclusion: Even though a mild negative effect of the highest dose of ZA on linear and appositional growth was observed, the other bone parameters evaluated were improved. A careful risk/benefit analysis would lead us to conclude that the mild deleterious effects of ZA during growth are outweighed by the benefit obtained with treatment. (AU)

(AU)

ABSTRACT

ABSTRACT: An experiment was performed to evaluate the effect of decreased levels of vitamin D3 in the premix and 1,25-dyhydroxyvitamin D3-glycoside (1,25(OH)2D3-glycoside) supplementation on performance, carcass yield and bone quality in 42d old broilers. Seven-d-old male chickens Cobb500® were distributed in a randomized design with six treatments: a control diet with inclusion of vitamin D3 in the premix, without supplementation of 1,25(OH)2D3-glycoside, and five diets with decreased levels of vitamin D3 (100%, 75%, 50%, 25% and 0% about the control) plus the addition of 1,25(OH)2D3-glycoside, 50g ton-1 of diet. The main results were to reduce the tenor of Vitamin D3 in the premix when the addition of 1,25(OH)2D3-glycoside did not affect (P>0.05) the performance, carcass yield and bone quality variables. However, performance (feed intake, gain weight, feed conversion), yield (warm carcass weight) and bone quality (dry weight, length, mineral matter and breaking strength) of broilers fed with diets without vitamin D3 in the premix and with addition of 1,25(OH)2D3-glycoside, which was the single source of vitamin D, had as a result very low (P<0.05) values comparing to the control. For the purposes of the present research, it was concluded that is possible the reduction of vitamin D3 tenor in the premix up to 75% when the diet of male broilers is supplemented with 1,25(OH)2D3-glycoside. However, the use of 1,25(OH)2D3-glycoside as a single source of vitamin D, as tested here, is not recommended for broilers diets.

ABSTRACT

ABSTRACT Objective To evaluate, in rat offspring, bone changes induced by excess maternal thyroxin during pregnancy and lactation, and to assess the reversibility of these changes after weaning. Material and methods Twenty Wistar rats were distributed in two groups, hyperthyroid and control, that were treated daily with L-thyroxin (50 mcg/animal) and placebo, respectively. The treatment was initiated seven days before mating and continued throughout pregnancy and lactation. From every female of each of the two groups, two offspring were euthanized after birth, two at 21 days of age (weaning), and two at 42 days of age (21 days after weaning). In newborns, the length of pelvic and thoracic limbs were measured, and in the other animals, the length and width of the femur and humerus were measured. Bones were dissected, decalcified, embedded in paraffin, and analyzed histomorphometrically. Results Excess maternal thyroxin significantly reduced the length of the pelvic limb in neonates. In 21-day-old individuals, excess maternal thyroxine reduced the length and the width of the femur and the humerus. It also increased thickness of the epiphyseal plate and the percentage of trabecular bone tissue. In 42-day-old individuals, there were no significant differences between groups in relation to the parameters evaluated in the previous periods. Conclusion Excess maternal thyroxine reduced growth in suckling rats both at birth and at weaning, and it also increased the percentage of trabecular bone tissue in 21-day-old animals. These changes, however, were reversible at 42 days, i.e., 21 days after weaning. Arch Endocrinol Metab. 2016;60(2):130-7.

ABSTRACT

PURPOSE: The objective of this study was to investigate the effects of a high fructose and fat diet on bone growth and maturation in growing female rats. METHODS: Three-week-old female SD rats were randomly assigned to four experimental groups; the control group (CON: fed control diet based on AIN-93G, n = 8); the high-fructose diet group (HFrc: fed control diet with 30% fructose, n = 8); the high-fat diet group (Hfat: fed control diet with 45 kcal% fat, n = 8); and the high-fat diet plus high fructose group (HFrc + HFat: fed diets 45 kcal% fat with 30% fructose, n = 8). Each group was assigned their respective diets for the remaining eight weeks. Bone-related parameters (bone mineral density (BMD) and structural parameters, osteocalcin (OC), deoxypyridinoline (DPD)) and morphologic changes of kidney were analyzed at the end of the experiment. RESULTS: Final body weights and weight gain were higher in the HFat and HFrc + HFat groups and showed higher tendency in the HFrc group compared with those of the CON group (p < 0.05); however, no significant difference in caloric intake was observed among the four experimental groups. The serum OC levels of the HFrc and HFrc + HFat groups were lower than those of the CON and HFat groups (p < 0.05). Urinary levels of DPD did not differ among the experimental groups. BV/TV and Tb.N of trabecular bone were higher in the HFrc + HFat group and showed a higher tendency in the HFrc group than those of the CON and HFat groups (p < 0.05). Tb.Pf of trabecular bone were lower in the HFrc + HFat group than those in the CON and HFat groups (p < 0.05). However, no difference in trabecular BMD was observed among the experimental groups. Cortical bone volume was higher in the HFat and HFrc + HFat groups than in the CON and HFrc groups (p < 0.05). No morphology change in kidney was observed among the experimental groups. CONCLUSION: Our study suggests that 8 weeks of high-fructose and high fat intake could improve the bone quality (Structural parameters) of trabecular and cortical bone of tibia in growing female rats.

ABSTRACT

BACKGROUND: The chin shape and position is important in determining the general shape of the face, and augmentation genioplasty is performed alone or in combination with other aesthetic procedures. However, augmentation genioplasty using osteotomy is an invasive and complex procedure with the potential to damage mentalis muscle and mental nerve, to affect chin growth, and prolonged recovery. Our aim was to present our experience with a modified augmentation genioplasty procedure for hypoplastic chins using a Gore-Tex implant. METHODS: Two vertical slit incisions were made at the canine level to create a supraperiosteal pocket between the incisions, preserving the periosteum and mentalis muscle. Minimal sub-periosteal dissection was performed lateral to the incisions along the mandibular border. The both wings of implant were inserted under the periosteum to achieve a stable dual plane implantation. RESULTS: In total, 47 patients underwent dual plane chin augmentation using a Gore-Tex implant between January 2008 and May 2013. The mean age at operation was 25.77 years (range, 15-55 years). There were 3 cases of infection; one patient was treated with antibiotics, the others underwent implant removal. Additionally, two patients complained of postoperative parasthesia that spontaneously improved without any additional treatment. Most patients were satisfied with the postoperative outcomes, and no chin growth problems were observed among the younger patients. CONCLUSION: Dual plane Gore-Tex chin augmentation is a minimally-invasive operation that is simple and safe. All implants yielded satisfactory results with no significant complications such as mental nerve injury, lower lip incompetence, or chin growth limitation.

ABSTRACT

The aim of this study was to conduct a literature review of impacts of protein malnutrition and/or early food deprivation on the formation of the growth plate in young animals. A search was performed using the databases PubMed/MEDLINE, LILACS, SciELO and the Cochrane Library, without language restrictions or any limit as to year. Initially, 150 articles were identified, however, after application of the inclusion and exclusion criteria only five remained. In three studies the nutritional stress was due to protein malnutrition, another due to fasting and the fifth was due to food restriction. All the studies had some methodological omissions. The studies included in this review-demonstrated corroboration in the results obtained, regardless of whether the animals had been subjected to protein malnutrition, fasting or food restriction. The findings uncovered were reduction in height of the epiphyseal plate, in the number of proliferative and hypertrophic chondrocytes, in the bone growth rate and in the longitudinal length of the bone in animals subjected to nutritional stress. In this systematic review, it was possible to observe the susceptibility of the epiphyseal plate in the first place, and secondarily, of the long bones to the effects of nutritional stress by means of protein malnutrition or food deprivation applied in young animals.

ABSTRACT

Mechanical signaling plays a critical role in bone growth, remodeling and healing. Recent studies discover that estrogen can modulate proliferation, apoptosis and functional activity of bone cell tissues via estrogen receptor (ER) and affect bone formation and absorption. Thus ER plays an important role in bone growth and remodeling. ER also involves in the adaptive response to mechanical stimulation, which is affected by ER numbers and activity. These findings suggest that mechanical stimulation and estrogen can act together to regulate functions of bone cells via common signaling pathways. In this review, the role and mechanism of ER in bone tissues and its respons to mechanical stimulation are discussed.

ABSTRACT

Large bone defects, congenital or caused by diseases, trauma or surgery, do not heal spontaneously and are usually a clinical challenge in the orthopedic and dental practices. A critical review concerning strategies to substitute lost bone or stimulate osteogenesis was undertaken. Pivotal concepts ranging from traditional bone grafting and use of biomaterials to local application of growth factors and gene therapy were addressed, including critical comments on the efficacy and safety, difficulties, advantages and disadvantages of each method. The most predictable results are still obtained with autogenous bone graft, despite the inconveniences of morbidity and limited availability of graft material. Satisfactory results have been reported for recombinant bone morphogenetic proteins (rhBMPs)-2 and -7, which distinguish for their osteoinductive property, the difficulty being the need for a degradable carrier that allows its continuous release in a rate compatible to that of new bone formation. Other bone growth factors are currently under evaluation in preclinical models of bone defects; however their efficacy is also dependent on the competence of a delivery strategy and on an appropriate delineation of “which one”, “which dose” and “when”. Parameters of efficiency and safety for gene therapy are still being established. In conclusion, given the variety of growth factors involved in the complex cascade of bone repair and the biological interactions between them, it remains a challenge to accomplish the ideal strategy to stimulate reparational bone formation in specific conditions of the medical as in the dental practices.

ABSTRACT

The osteointegration is the structural and functional connection between bone and implant. When an implant is inserted in bone, it creates the so-called bone-implant interphase, a joint zone between implant biomaterial surface and the surrounding bone. The healing of this interphase depends on bone biological conditions, characteristic of implant design and the distribution of loads between bone and implant. The aim of present article is to review of healing process of bone-implant interphase for a dental implant and also to describe the sequence of biological events beginning with lesion caused by implant insertion and leading to the formation of a new bone in the interphase. This description includes a novel classification of mechanical phenomena present in the healing process of tissues affected. This mechanobiological description of dental bone-implant interphase is used to determine the more significant features to be into account in formulation of a mathematical model of the osteointegration of dental implants(AU)
