ABSTRACT

@#ObjectiveTo observe the effect of stimulating “Zhiyang” (DU 9) with electric acupuncture apparatus on contents of cAMP and cGMP in plasma of experimental acute myocardial ischemic rabbits.Methods40 rabbits were randomly divided into four groups: the normal control group (A), model group (B), model plus electric acupuncture on “Neiguan” (P 6) group (C) and model plus electric acupuncture on DU 9 group (D). The acute myocardial ischemic model was established by pituitrin (2 U/kg) injected into the vein of rabbit. Electrocardiogram (ECG) of Ⅱ leads was observed and contents of cAMP and cGMP in plasma were tested after electric acupuncture by radio immunoassay.ResultsThe cAMP contents of the group C and group D were lower than that of the group B significantly ( P<0.01), but not different from that of the group A ( P>0.05), and there was no difference between the group C and group D. The contents of cGMP were not different among four groups ( P>0.05).ConclusionPuncturing DU 9 can raise the myocardial endurance to the insufficiency of blood and oxygen. The effects of electric acupuncture on DU 9 and P 6 are the same.

ABSTRACT

Objective:To explore the action of the second signal system in the activatied and proliferating T cells and the induction of apoptosis of the hepatoma cells.Methods:The T cells were costimulated by anti CD28 and anti CD80(B7.1)McAb and acted on the hepatoma cells(BEL 7402),then testing the concentration of cAMP?cGMP and Ca 2+ in the T cells and the apoptosis of the hepatoma cells.Results:The concentration of cAMP was increased temporarily at first,then decreased rapidly,and increased 1 2 times again when acted on the hepatoma cells.The concentration of cGMP was increased 6 8 times fast and the concentration of Ca 2+ obviously increased 2 3 times too.The peak of them was at the fourth day and positive related to apoptosis of the hepatoma cells.Conclusion:The level of the second signal system of cAMP?cGMP and Ca 2+ were significant correlated with the T cells activated and porliferating and the cytotoxic effect.

ABSTRACT

Aim To investigate the effects of tamoxifen on the proliferation and DNA synthesis of cultured human pituitary adenoma cells and make a further investigation of the mechanism of the inhibitory effect of tamoxifen on the proliferation of pituitary adenoma cells. Methods The techniques of MTT colorimetry, 3H-TdR, flow cytometry, PKC activity detection and cAMP/ cGMP levels detection were used to detect or observe the effects of tamoxifen on proliferation, DNA synthesis, cell cycle, PKC activity and cAMP/ cGMP levels of cultured human pituitary adenoma cells, respectively.Results ①Tamoxifen (0.1,1 and 10 ?mol?L -1) inhibited the proliferation and DNA synthesis of cultured human pituitary adenoma cells in a dose-dependent manner.② tamoxifen (1,10 and 20 ?mol?L -1) increased the ratio of G_1 phase of pituitary adenoma cells, and decreased the ratio of S and G_2 phase markedly;②compared with control, PMA, a PKC activator, increased the activity of membrane and total PKC in human pituitary adenoma cells. However, after a 15-min treatment with tamoxifen (10 ?mol?L -1),a significant reduction of the activity of cytoplasm, membrane and total PKC in human pituitary adenoma cells was observed;③tamoxifen (1 and 10 ?mol?L -1) increased the amount of cAMP in the cytoplasm of human pituitary adenoma cells, but had no effect on that of cGMP. Conclusion These data provide an important clue to explore the molecular mechanisms of the inhibitory effect of tamoxifen on the proliferation of pituitary adenoma cells, and suggest that the modulating effect of tamoxifen on the proliferation of pituitary adenoma cells results from interactions of several cellular signaling pathways.

ABSTRACT

The concentration of the cyclic nucleotides and its relation with the E-RFC were determined by using ginseng rhizome saponin in human peripharal leukocytes.Result were compared with the ckange of the E-RFC by using cAMP.cGMP and theophylline. Experiments proved that girseng rhizome saponin mainly affected the lymphocytes and significantly increased the concentration of cAMP and cGMP,especially,the cAMP. Lymphocytes were incubated with ginseng rhizome saponin to form Et-RFC. It is different where lymphocytes were incubated in diffierent times. cAMP and theophylline reduced the Ea-RFC and Et-RFC, cGMP increased the Ea— RFC,but no effect to the Et-RFC. Ginseng rhizome or root saponin could promote the Ea-RFC,but inhibited the Et-RFC. It is consistent with the effect of both cAMP and cGMP. Experiments showed that ginseng rhizome saponin increased the concentration of cAMP and cGMP in human lyphocytes followed by that of E-RFC.It was suggested that the change of cyclic nucleotides in lymphocytes may be a mechanism of adjusting the immunity on cell level.
