ABSTRACT

Objective To study the influence of sequential and contemporaneous revascularizations of portal vein and hepatic artery during liver transplantation with retrograde reperfusion on the ultra-structural changes in intrahepatic cholangiocytes of the graft with liver ischernia reperfusion injury,and the recovery of short-term liver function after operation.Methods The clinical data of 97 patients who underwent orthotopic liver transplantation (OLT) using retrograde reperfusion via the vena cava (IVC) in our hospital from January,2009 to June,2013 were retrospectively analyzed.Using to the different sequences of revascularization,97 digible patients were divided into three groups:group R-IPR:the initial arterial revascularization group (35 patients);group R-IAR:the initial portal revascularization group (30 patients);group R-SAPR:simultaneous reperfusion group (32 patients).Liver function,coagulation function and other related parameters were continuously monitored for the transplant recipients at the following time points:1 day before operation,2 hours after reperfusion and on postoperative Day 1,2,5 and 7.The ultra-structure of intrahepatic bile duct epithelial cells at 2 hours after reperfusion were studied by transmission electron microscopy and morphometric analysis of mitochondria was performed by the computer image analysis system.Results The levels of ALT,GGT and TB in group R-IAR were significantly lower than those in group R-IPR and group R-SAPR (P<0.05).There were no significant differences among the three groups in other parameters that reflected liver function at any time point (P>0.05).In group R-IAR and group R-IPR,mitochondrial swelling of bile duct epithelial cells was more severe than that in group R-SAPR.There were significant differences among the three groups in the mitochondrial average area and the average perimeter (P<0.05).Mild swelling of mitochondria in hepatic cells was observed in group R-IAR and group R-IPR.There was no significant difference in the mitochondrial average area and average perimeter between the two groups (P>0.05).Mitochondrial swelling of hepatic cells was most severe in group R-SAPR.There were significant differences in the average area and average perimeter of hepatic cell mitochondria between group R-SAPR and the other two groups (P<0.05).Conclusion During liver transplantation with retrograde reperfusion,initial arterial revascularization reduced the short-term levels of transaminases,improved early liver function after operation and protected the intrahepatic bile duct against the second ischemia reperfusion injury.

ABSTRACT

Objective To observe the effects ofXiaotan Sanjie Decoction on ultramicrostructure in MKN-45 human gastric cancer orthotopic xenograft;To discuss its mechanism for gastric cancer. Methods MKN-45 human gastric cancer cell line was used to establish subcutaneous tumor model in nude mice as an experimental model which passed three generations of tumor source. Then OB glue was used to establish human gastric cancer orthotopic xenograft model. 40 model nude mice were divided into control group, model group (received gavage with saline),Xiaotan Sanjie Decoction group (received garage withXiaotan Sanjie Decoction) and Tegafur group (received gavage with Tegafur), 10 mice in each group. Mice in the control group were under normal diet, without medicine intervention. After 6 weeks of treatment intervention, tumor weight was detected, inhibitory rate was calculated, the ultrastructural changes of gastric cancer cells were detected through transmission electron microscopy.Results The tumor weight inXiaotan Sanjie Decoction group was significantly lower than that in control group and model group (P<0.05,P<0.01). The tumor inhibition rate was 46.2%. Under the electron microscope, MKN-45 gastric cancer cells strain changed to typical apoptosis.ConclusionXiaotan Sanjie Decoction can lead to apoptosis effects on MKN-45 human gastric cancer orthotopic xenografts. All evidences indicate that inducing apoptosis may be one of the most important mechanisms ofXiaotan Sanjie Decoction in treating gastric cancer.

ABSTRACT

Connarus suberosus is a typical species of the Brazilian Cerrado biome, and its inflorescences and young vegetative branches are densely covered by dendritic trichomes. The objective of this study was to report the occurrence of a previously undescribed glandular trichome of this species. The localization, origin and structure of these trichomes were investigated under light, transmission and scanning electron microscopy. Collections were made throughout the year, from five adult specimens of Connarus suberosus near Botucatu, São Paulo, Brazil, including vegetative and reproductive apices, leaves and fruits in different developmental stages, as well as floral buds and flowers at anthesis. Glandular trichomes (GTs) occurred on vegetative and reproductive organs during their juvenile stages. The GTs consisted of a uniseriate, multicellular peduncle, whose cells contain phenolic compounds, as well as a multicellular glandular portion that accumulates lipids. The glandular cell has thin wall, dense cytoplasm (with many mitochondria, plastids and dictyosomes), and a large nucleus with a visible nucleolus. The starch present in the plastids was hydrolyzed during the synthesis phase, reducing the density of the plastid stroma. Some plastids were fused to vacuoles, and some evidence suggested the conversion of plastids into vacuoles. During the final activity stages of the GTs, a darkening of the protoplasm was observed in some of the glandular cells, as a programmed cell death; afterwards, became caducous. The GTs in C. suberosus had a temporal restriction, being limited to the juvenile phase of the organs. Their presence on the exposed surfaces of developing organs and the chemical nature of the reserve products, suggest that these structures are food bodies. Field observations and detailed studies of plant-environment interactions, as well as chemical analysis of the reserve compounds, are still necessary to confirm the role of these GTs as feeding rewards.


Connarus suberosus es una especie típica del bioma Cerrado brasileño, y sus inflorescencias junto con los ramos vegetativos jóvenes están densamente cubiertos por tricomas dendríticas. El objetivo de este estudio es presentar la ocurrencia de un tricoma glandular sin descripción previa para esta especie. La localización, origen y estructura de estos tricomas se observó mediante microscopia óptica, así como electrónica de transmisión y de barrido. Los tricomas glandulares (TGs) se producen en los órganos vegetativos y reproductivos, en sus fases jóvenes. Los TGs consisten en un pedúnculo uniseriado y multicelular, cuyas células contienen compuestos fenólicos, así como un sitio en el cual las células glandulares acumulan compuestos pécticos, almidón y lípidos. La acumulación de aceite en el citosol es impresionante, pues ocupa una gran parte del volumen celular. La porción glandular presenta células con paredes delgadas, un denso citoplasma y un núcleo grande con nucleolo visible. El citoplasma contiene gran cantidad de mitocondrias, plástidos y dictiosomas. El almidón presente en los plástidos se hidroliza durante la fase de síntesis, de tal modo que reduce la densidad del estroma plastidial. Algunos plástidos se fusionan con las vacuolas, además hay pruebas de la conversión de los plástidos en vacuolas. Durante las etapas finales de la actividad de los TGs, se observó un oscurecimiento del protoplasma en algunas de las células glandulares lo que sugiere la existencia de la muerte celular programada; después de lo anterior, los TGs se convierten en caducas. Por lo tanto la aparición de TGs en las superficies expuestas de los órganos en desarrollo en C. suberosus y la naturaleza química de los productos de reserva sugieren que estas estructuras son cuerpos alimenticios. Hay evidencias sobre el papel de estos tricomas en las interacciones planta-hormiga, especialmente en el bioma Cerrado.

ABSTRACT

Objective To explore the effect of Baogan Jiedu Granule on liver cell ultrastructure of model mice with acute liver injury induced by tetracycline.Methods Totally 60 mice were randomly divided into tetracycline model group,large-dosage,medium-dosage and small-dosage groups of Baogan Jiedu Granule,Ganlixin group and control group.Each group was respectively perfused with corresponding drug for 5 days and tetracycline was perfused once to make the model.The tissues were observed by electron microscope,and the liver cell ultrastructure was measured and analyzed by three-dimensional metrology.Results The amount of mitochondria Vv,heterochromatin Vv,mitochondria ?m,endoplasmic reticulum ?m and Golgi's apparatus ?m all obviously decreased in model group,while that of euchromatin Vv and lipid droplet Vv increased.Baogan Jiedu Granule could remarkably increase the amount of indexes referred above,and decrease the amount of euchromatin Vv and lipid droplet Vv.Conclusion Baogan Jiedu Granule can significantly ameliorate the pathologic change of liver cell ultrastructure,thus having a good hepatoprotective effect.

ABSTRACT

The present study is designed to establish a xenograft model of human promyelocytic leukemia cell mutant (HL-60-AR) deficient in hypoxanthine guanine phosphoribosyltransferase (HGPRT) in nude mice. A solid leukemia sarcoma developed after subcutaneous inoculation with HL-60-AR cells. Comparative studies of HL-60-AR/Nu tumor cells in nude mice and cultured HL-60-AR cells in vitro revealed virtual identity as shown by light microscopic morphology, ultrastructure of cell, cytochemistry, chromosome analysis, LDH isoenzyme pattern, genetic markers and differentiated characters assay. Up to now, twelve generations haw been transmitted in rive by inoculating with the solid tumor Cells developed in nude mice. This nude mice model in which human leukemia cells grew could be considered as a useful model for in rive studies of human leukemic cells proliferation, differentiation and the screening for anti-leukemia drugs.
