ABSTRACT

Objective To analyze the distribution of human papillomavirus (HPV) genotype in the diseases associated with HPV infection and explore the role of HPV genotype in these diseases. Methods Retrospective analysis was carried out to compare HPV genotypes of 353 outpatients suffering the diseases associated with HPV infection including cervical caner, cervical intraepithelial neoplasia (CIN),genital duc condyloma, and subclinical cervical human papillomavirus infection (SPI) in our department from January to October 2008. HybriMax were used to detect HPV genotype. The distribution data of HPV genotype were statistically analyzed. Results Totally 176 HPV positive females were found with a positive rate of 49.86%. HPV positive rate in cervical cancer was 100%,and all of them were high risk HPV. The positive rate was increased with the severity of CIN,and the higher histological-grade was,the higher HPV positive rate was (P

ABSTRACT

Objective To determine the resistance reversion of mitomycin (MMC) by 3′-Keto-bmt 1-val 2-cyclosporin (SDZ PSC 833) in human cervical cancer in vitro and in vivo. Metheds A xenografted mitomycin resistant mice model of cervical cancer was devolped. The reversion of mitomycin resistance by SDZ PSC 833 (1 or 3 mg/L) was detected from human cervical cancer cell (Hela) and its resistant subline Hela/MMC in vitro and in vivo. Studies in vitro include drug resistance reversion experiment and the changes of morphology. Studies in vivo including tumor volume and tumor related histopathological changes in the autopsied specimen were evaluated by comparing random sections of each group. Results Nontoxic doses of SDZ PSC 833 could result in almost partial reversion of MMC-resistance of Hela/MMC.In vivo studies also showed SDZ PSC 833 augmented the growth inhibitory effect of mitomycin on Hela/MMC xenografted in nude mice. Conclusion SDZ PSC 833 can overcome mitomycin resistance of Hela/MMC in vitro and in vivo ,so SDZ PSC 833 will be a better candidate clinically for reversing multidrug resistance.

ABSTRACT

Objective:To investigate the expressions of cFLIP and FADD in cervical carcinoma and explore their associations with the carcinogenesis and development of cervical carcinoma, and to evaluate their clinical significances. Methods:The expressions of cFLIP and FADD proteins were detected in cervical tissue samples including 64 cervical carcinoma tissues,15CIN,12 chronic cervicitis tissues and 10 normal cervix tissues by immunohistochemistry(SP).And PCNA-LI in 64 cervical carcinoma samples was determined. Results:cFLIP and FADD expressions were significantly different among cervical carcinoma,CIN,chronic cervicitis tissue and normal cervix. The high expression of cFLIP and low expression of FADD were simultaneously found in cervical carcinoma.Statistical analysis showed that there was a positive correlation between the level of cFLIP expression and PCNA-LI(r1=0.7738,P
