ABSTRACT

OBJECTIVE:To provide reference for rational use of Clindamycin hydrochloride injection. METHODS:The utilization of Clindamycin hydrochloride injection in orthopedic inpatients of a third grade class A general hospital dur-ing 2013-2015 was analyzed and investigated retrospectively. The rationality evaluation standard was formulated to evaluate the rationality of drug use in those patients. RESULTS:A total of 189 orthopedic inpatients received Clindamycin hydro-chloride injection,and there were 152 cases of irrational drug use among which there were 16 cases of irrational therapeu-tic drug use and 136 cases of irrational prophylactic drug use. Irrational drug use mainly included irrational usage and dos-age(86.18%),irrational drug selection(44.74%)and irrational drug replacement(15.79%). Two patients suffered from ADE,i.e. rash and nausea. After drug withdrawal and symptomatic treatment,the symptoms were improved,but irrational drug use was found in one case. CONCLUSIONS:The utilization of Clindamycin hydrochloride injection is irrational in or-thopedic department. It is necessary to further standardize medication behavior and guarantee the safety and effectiveness of drug use.

ABSTRACT

OBJECTIVE:To establish the method for simultaneous determination of the content of metronidazole,clindamycin hydrochloride,spironolactone and vitamin B6 in Compound metronidazole clindamycin cream.METHODS:HPLC method was adopted.The determination was performed on Diamonsil C18 column with mobile phase consisted of acetonitrile-[0.68% dipotassium hydrogen phosphate solution (containing 1% triethylamine)] (gradient elution) at the flow rate of 1.0 mL/min.The detection wavelength was set at 210 nm,and column temperature was 25 ℃.The sample size was 20 μL.RESULTS:The linear ranges of metronidazole,clindamycin hydrochloride,spironolactone and vitamin B8 were respectively 20.0-120.0 i.tg/mL(r=0.999 9),10.0-60.0μg/mL(r=0.999 1),4.0-24.0 μg/mL(r=0.999 4),10.0-60.0 μg/mL (r=0.999 4).The quantitative limits were respectively 13.06,7.36,2.43,6.42 μg/mL.The detection limits were 4.18,1.82,0.76,1.79 μg/mL.RSDs of the precision,stability,repeatability tests were all less than 2%.The recovery rates were respectively 98.97%-100.91% (RSD=0.79%,n=9),99.02%-101.80% (RSD=1.03%,n=9),98.18%-101.05% (RSD=1.01%,n=9) and 98.75%-101.36% (RSD=0.87%,n=9).CONCLUSIONS:The method is simple,rapid,accurate and can be used for the determination of metronidazole,clindamycin hydrochloride,spironolactone and vitamin B6 in Compound clindamycin metronidazole cream.

ABSTRACT

Objective To investigate the effect of Zhibaidihuangwan combined with clindamycin hydrochloride on serum IL-8, TNF-α, IL-10 levels in periodontitis and its clinical efficacy.Methods A total of 98 patients with periodontitis from our hospital were collected and randomly divided into the control group and the experimental group, 49 cases each group.Patients in the control group were treated on the basis of the foundation therapy with clindamycin hydrochloride, patients in the experimental group were treated with Zhibaidihuangwan on the basis of control group .The periodontal pocket depth, plaque index, gingival index and positive rate of probe bleeding, serum IL-8, TNF-a, IL-10 levels were measured, and the clinical efficacy were compared.Results The effective rate of the experimental group(93.88%) was higher than the control group(79.59%), with statistical significance (P<0.05).The positive rate of probe bleeding in experimental group (38.78%) was lower than control group of two groups(59.18%), with statistical significance (P<0.05).The periodontal pocket depth, plaque index, gingival index in experimental group were lower than the control group after treatment, the TNF-αand IL-8 levels in experimental group were lower than the control group, IL-10 was higher than the control group after treatment, with statistical significance (P<0.05).Conclusion Zhibaidihuangwan combined with clindamycin hydrochloride could reduce periodontitis serum IL-8 and TNF-αlevels, improve the level of IL-10.

ABSTRACT

Introducción: la presencia de microorganismos residuales post-preparación químico-quirúrgico de los conductos radiculares, sugiere el empleo de medicación intracanal eficaz y biológicamente compatible con los tejidos periapicales. Objetivo: comparar la citotoxicidad de medicaciones de uso intracanal en fibroblastos gingivales humanos. Métodos: estudio de tipo experimental. Con cuatro diferentes grupos: control (G1), Clorhidrato de Clindamicina 2g asociado a fosfato de dexametasona 0,32g (Clindex) vehiculado en solución alcohólica (G2), Clindex Vehiculado en polietilenoglicol 400 (G3), y NDP: paramonoclorofenol asociado a fosfato de dexametasona 0,32g vehiculado en polietilenoglicol 400 y solución salina (G4). Las células fueron estimuladas con las medicaciones por 24, 48 y 72 horas y la viabilidad celular fue evaluada usando la técnica de análisis de MTT y la lectura fue realizada en el espectrofotómetro de ELISA. Resultados: fueron analizados por medio de la actividad mitocondrial y mostraron que el G2 obtuvo los mejores resultados en los tres tiempos estudiados, seguido por el G3 y el G4. Se realizó análisis estadístico (ANOVA y complementado con la prueba de Tukey) mostraron diferencias significante a nivel de 1% G2 y G3 cuando se compararon con el G4. Conclusión: la combinación Clindamicina dexametasona independiente del vehículo presentó mayor viabilidad celular que el paramonoclorofenol asociado a fosfato de dexametasona.


Introduction: the presence of residual microorganisms after chemical-surgical preparation of root canals suggests the use of effective intracanal medication, biologically compatible with periapical tissues. Objective: compare the cytotoxicity of intracanal medications in gingival human fibroblasts. Methods: an experimental study was conducted with four different groups: control (G1), Clindamycin Hydrochloride 2g associated to dexamethasone phosphate 0.32g (Clindex) vehicled in alcoholic solution (G2), Clindex vehicled in polyethylene glycol 400 (G3), and NDP: paramonochlorophenol associated to dexamethasone phosphate 0.32g vehicled in polyethylene glycol 400 and saline solution (G4). The cells were stimulated with the medications for 24, 48 and 72 hours. Cell viability was evaluated with the technique of MTT analysis. Readings were performed in an ELISA spectrophotometer. Results: the groups were analyzed in terms of mitochondrial activity. G2 had the best results in the three times studied, followed by G3 and G4. A statistical analysis was performed (ANOVA and complemented by Tukey's test). Significant differences of 1% were found when comparing G2 and G3 with G4. Conclusion: the Clindamycin dexamethasone combination, irrespective of the vehicle, showed greater cell viability than paramonochlorophenol associated to dexamethasone phosphate.

ABSTRACT

OBJECTIVE:To prepare Xiaodouning cream and establish a method for its quality control. METHODS: The cream was prepared using glycerol monstearate as matrix with clindamycin hydrochloride and metronidazole as principle agents. The contents of the principal agents were determined by HPLC. The stability of the cream within 6 months was investigated. RESULTS: The cream was well-distributed and delicate with suitable viscidity,and it met the criteria of Chinese Pharmacopeia(2005 edition) in test and identification. The linear ranges of clindamycin hydrochloride and metronidazole were 2.342~11.71 ?g (r=0.999 8) and 0.97~4.85 ?g (r=0.999 8),respectively,with average recoveries at 98.3% (RSD=0.7%) and 100.3% (RSD=1.6%),respectively. In the stability test,no obvious change was noted for all indexes. CONCLUSION: The preparation technology of this cream is simple and feasible and its quality is stable and controllable.

ABSTRACT

OBJECTIVE:To establish microbial limit test for Compound clindamycin hydrochloride gel and to carry out the validation of three methods.METHODS:According to the appendices volumeⅡof Chinese Pharmacopeia(edition 2005),microbial limit test which contained routine test,medium dilution method and membrane-filter method was adopted to validate Compound clindamycin hydrochloride gel.Recoveries of three methods were compared to make sure the suitable validation method.RESULTS:In routine test,recovery of trial bacteria was lower than 70%and controlled bacteria couldn't be found out.In medium di-lution method,recovery of trial bacteria was higher than 70%and still lower than the recovery of diluents.In membrane-filter method,recovery of trial bacteria was higher than 90%,and controlled bacteria grew well.Membrane-filter method could eliminate inhibition effect of Compound clindamycin hydrochloride gel on bacteria.CONCLUSION:Membrane-filter method is effective and practical for microbial limit test of Compound clindamycin hydrochloride gel.
