ABSTRACT

Abstract: This study evaluated the relationships between environmental factors and the spatio-temporal distribution of H. pudibundus, with the hypothesis of differential occupation in coastal areas of southeastern Brazil. The samplings took place monthly in January-December 2000 period, along nine transects from 2 to 40 m of depth, in Ubatuba region, northern coast of São Paulo. We collected 1808 individuals of H. pudibundus. The highest abundance was recorded in winter in the transects 10-25 m deep. Abundance was positively correlated with organic matter content and texture sediment (phi values). With the retreat of the South Atlantic Central Water (SACW) in autumn and winter, the sediment swirls, suspending the detritivore and filter-feeding macrofauna, increasing the food availability. Sites characterized by finer sediment offer higher food availability, besides facilitating H. pudibundus burying behavior. Due to its opportunistic predatory behavior, this species feeds on a variety of organisms, including mollusks, annelids and foraminifera, which are preys more abundant in the studied area and in sediments of finer grain size.


Resumo: Este estudo avaliou as relações dos fatores ambientais e a distribuição espaço-temporal de H. pudibundus, com a hipótese de ocupação diferencial em uma região do litoral sudeste do Brasil. As amostragens ocorreram mensalmente no período de janeiro a dezembro de 2000, ao longo de nove transectos de 2 a 40 m de profundidade, na região de Ubatuba, litoral norte de São Paulo. Foram coletados 1808 indivíduos de H. pudibundus. A maior abundância foi registrada no inverno nos transectos 10 a 25 m de profundidade. A abundância foi positivamente correlacionada com o teor de matéria orgânica e a textura do sedimento (valores de phi). Durante o outono e inverno com a retração da ACAS ocorre o revolvimento do sedimento e a supensão da macrofauna detritívora e filtradora, aumentando a disponibilidade de alimento. Locais caracterizados por sedimentos mais finos oferecem maior disponibilidade de alimentos, além de facilitar o comportamento de H. pudibundus se enterrar. Devido ao comportamento predatório oportunista, esta espécie se alimenta de uma grande variedade de organismos, incluindo moluscos, anelídeos e foraminíferos, que são as presas mais abundantes nas áreas estudadas, principalmente em locais com grãos menores.

ABSTRACT

Abstract This molecular study is the first report, to the best of our knowledge, on identification of norovirus, NoV GII.4 Sydney 2012 variants, from blue mussels collected from UK coastal waters. Blue mussels (three pooled samples from twelve mussels) collected during the 2013 summer months from UK coastal sites were screened by RT-PCR assays. PCR products of RdRP gene for noroviruses were purified, sequenced and subjected to phylogenetic analysis. All the samples tested positive for NoVs. Sequencing revealed that the NoV partial RdRP gene sequences from two pooled samples clustered with the pandemic "GII.4 Sydney variants" whilst the other pooled sample clustered with the NoV GII.2 variants. This molecular study indicated mussel contamination with pathogenic NoVs even during mid-summer in UK coastal waters which posed potential risk of NoV outbreaks irrespective of season. As the detection of Sydney 2012 NoV from our preliminary study of natural coastal mussels interestingly corroborated with NoV outbreaks in nearby areas during the same period, it emphasizes the importance of environmental surveillance work for forecast of high risk zones of NoV outbreaks.

Aims@#Vibrio parahaemolyticus is a marine and estuarine bacterium that has been documented as the causative agent of food-borne outbreak worldwide. The aim of this study was to confirm the identification of presumptive V. parahaemolyticus isolates to the species level by using PCR targeted to the outer membrane protein regulation operon gene (toxR) and to investigate antibiotic resistance, plasmid profile, and the main core virulence genes of thermostable direct hemolysin (tdh) and tdh-related hemolysin (trh). @*Methodology and results@# A total of 56 presumptive isolates of V. parahaemolyticus were isolated from seawater collected during year a 2010 sampling pilot study performed along the Arabian Gulf coast of the Eastern Province of Saudi Arabia. The purpose of this study was to confirm the identification of presumptive V. parahaemolyticus isolates to the species level by using PCR targeted to the toxR gene and to investigate antibiotic resistance, plasmid profile, and the main core virulence genes of tdh and trh. The toxR-specific PCR assay revealed that a total of 30 out of 56 isolates tested positive for V. parahaemolyticus. None of the 30 strains of the toxR gene were tested positive for tdh and trh genes. All (100%) of isolates were highly resistant to amikacin, cefuroxime, ampicillin, ticarcillin, cefaclor (80%), and tetracycline (70%). The multiple antibiotic resistance (MAR) index was measured for all 16 antimicrobial agents, and the high ranged from 0.25 to 0.56. Among the isolated V. parahaemolyticus, 22 out of 30 strains contained plasmid DNA bands ranging in size from 1.5 to 55 kb and no correlation was observed between the plasmid profiles and antibiotic resistance patterns. @*Conclusion, significance and impact of study@#The results obtained in this study indicate that V. parahaemolyticus is present in the coastal environment of the Eastern Province of Saudi Arabia.

ABSTRACT

El agua de mar refinada aparece en la literatura científica reciente como una potencial ayuda terapéutica en el tratamiento de varias enfermedades en humanos y animales por su contenido de minerales y oligoelementos. En Colombia, Nicaragua y España se utiliza de forma natural por recogida de la orilla e ingesta; sin embargo, esto puede representar un riesgo para la salud por los problemas de contaminación microbiológica y química. Los tratamientos de control microbiano del agua de mar permiten mejorar su calidad microbiológica. Objetivo: comparar la eficiencia de tres métodos de control microbiano: microfiltración, exposición solar y cuarentena. Metodología: se recolectaron 30 muestras de agua de mar en recipientes de polietileno de alta densidad con capacidad de 20 litros, en tres lugares diferentes de la costa atlántica colombiana. Resultados: de 30 muestras recolectadas, 15 resultaron con en- terobacterias como E. coli y bacterias halófilas como Vibrio y Aeromonas. La microfiltración a través de cerámica de 0,5 μm produce una desinfección de 100% de las muestras, mientras que la cuarentena por cinco meses y la desinfección solar son efectivas en 66 y 21% respectivamente. Esta última requiere de ciertas condiciones climáticas para alcanzar la desinfección y solo permite el manejo de pequeños volúmenes. Discusión: Respecto de la contaminación química en ciertos lugares, que no es controlable por los métodos de desinfección, se recomienda recoger el agua en altamar, en lugares limpios, y realizar la microfiltración.


Recent scientific literature presents seawater as a potential aid to solve a variety of health diseases in animals and human beings because by means of its mineral and trace elements content. In Colombia, Nicaragua and Spain it is collected in a natural way from de shore and drunk; however, this can represent a health risk because of the problems related to chemical and microbiological contamination. Microbial control of seawater allows the improvement of its microbiological quality. Objective: to compare the efficiency of three microbial control methods: microfiltration, solar exposition and quarantine. Methodology: 30 samples were collected in 20-liter high density polyethylene containers in three different places in the Colombian Atlantic coast. Results: 15 samples out of 30 showed the presence of bacteria such as E. coli and halophiles bacteria like Vibrio and Aeromonas. Microfiltration through ceramic filters of 0.5 μm produces disinfection in 100% of the samples but the quarantine for five months and solar disinfection are effective in 66 and 21% respectively. The latter requires certain weather conditions to achieve disinfection and it only allows managing small quantities of water. Dicussion: Considering chemical contamination in some places which cannot be controlled through disinfection methods, the collection of water offshore in clean places is suggested and then microfiltration treatment should be performed.
