ABSTRACT

Objetivando conhecer a situação da tuberculose ovina no Maranhão, avaliaram-se 343 ovinos da região sudoeste do estado, 247 fêmeas e 96 machos, com idade acima de 6 meses, por meio do teste cervical comparativo (TCC), com inoculação de tuberculinas na região cervical direita. Cranialmente, injetou-se derivado proteico purificado (PPD) aviário; e caudalmente, PPD bovino, em doses individuais de 0,1 mL. Todos os animais apresentaram resultado negativo para tuberculose, entretanto, 65 ovinos apresentaram reação à tuberculina bovina superior à aviária. Para investigar a possibilidade de baixa sensibilização ou anergia em animais com tuberculose avançada, foi necropsiado um ovino caquético, idade acima de quatro anos, com resultado negativo ao TCC, porém, com reação à tuberculina aviária e linfonodos aumentados com consistência endurecida. Os resultados indicaram infecções por C. pseudotuberculosis, Actinomyces pyogenes, Staphylococcus aureus e Mycobacterium sp. A cultura e o isolamento resultaram em Pseudomonas aeruginosas e Bacillus sp. Não foram visualizadas estruturas bacterianas compatíveis com bacilo álcool ácido resistente (BAAR). Os resultados confirmam ausência de animais reagentes ao M. bovis, pressupondo inexistência da tuberculose em ovinos na região estudada.(AU)

No bacterial structures compatible with acid-fast bacteria were found. The results confirmed the absence of animals reactive to M. bovis, assuming no tuberculosis in sheep in the study area.(AU)

ABSTRACT

To study the comparability of myocardial enzyme spectrum detection results by two different bio-chemical testing systems to provide references for mutual accredit of detection results in one lab. Following NC-CLS document EP9-A2, 8 clinical specimens were selected every day for five days and tested for AST, LDH, CK and CK-MB respectively with Siemens ADVIA1800 and Johnson & Johnson Victro350 systems. Acknowledging Siemens AD-VIA1800 as objective testing system, the results were utilized to make correlation analysis and clinical acceptability esti-mation. There was high precision in Siemens ADVIA1800 and Johnson & Johnson Victro350 (CV≤1/3CLIA′88 allowable error). It's proved by t test that the difference of the myocardial enzyme results tested with Siemens ADVIA1800 and Johnson Victro350 was statistically significant (P0.975). In the comparison with the Siemens ADVIA1800, the standard errors (SE%) of AST, CK and CK-MB tested with Johnson Victro350 were 4.0%-6.6%, 9.6% and 13.3%, which were accepted by clinical standard; while the SE% of LDH tested with Johnson Victro350 were 11.2%-12.7%, which were beyond clinically acceptable range. In case one item is tested by different testing systems in one laboratory, the comparison and bias estimation acceptable eval-uationare necessary to judge the clinical acceptability, which can ensure the accuracy and consistency of the results.

ABSTRACT

Objective To investigate comparability between Sysmex different hematology analyzers.The consistency of all he-matology analyzers with the same fresh sample.Methods Selected Sysmex XE-2100 as reference instrument which attend external quality assessment.Fresh high,medium and low blood were detected in Sysmex XS-800i,Sysmex XT-4000i and Sys-mex XT-1800i,compared results of WBC,RBC,HGB,HCT,MCV and PLT.Results The bias of XS-800i and XE-2100 was WBC 2.85%,RBC 1.44%,HGB 0.75%,HCT 2.11% and PLT 5.53%.The bias of XT-4000i and XE-2100 was WBC 1.26%,RBC 0.95%,HGB 0.68%,HCT 1.35% and PLT 2.68%.The bias of XT-1800i and XE-2100 was WBC 5.21%, RBC 1.96%,HGB 1.60%,HCT 1.96% and PLT 4.95%.It had good compatability.All various parameters was in the al-lowed range.Conclusion Should maintenance and compare the hematology analyzers periodically,found the problem then calibration instruments in time,ensure the consistency of measurement results between different instruments and guarantee accuracy of the results.

ABSTRACT

BACKGROUND: The Korean Red Cross blood laboratory centers have been performing comparative tests for NAT reactive specimens since February 2005. However, five discrepant specimens were found in HCV-diluted specimens between 2007 and 2008 and the reasons for this has been investigated. METHODS: For the five discrepant specimens, the HCV RNA concentration was measured in 5 tubes for each speciment. Subsequently, in order to compare the sensitivity of the low titer specimens measured by RT-PCR and TMA, comparative tests of diluted samples were examined six times per sample. Finally, the genotype was tested in order to determine the characteristics of the discrepant samples. RESULTS: Result of the quantitative tests for HCV RNA demonstrated that aliquots of the plasma bag were homogenous in term of viral load quantitation. As a result of the comparative test, all samples were found to contain over 1.0x10(1) IU/mL as detected by the two analytical systems. In contrast, those less than 1.0x10(1) IU/mL were not entirely detected by the two systems. CONCLUSION: It was impossible to completely detect using the two NAT system and the detection rates for both systems were equivalent for the samples examined. In particular, with respect to HCV, it may be undetectable on the NAT test because viral load decreases rapidly before and after sero-conversion. This result indicates that anti-HCV and NAT should be performed together as an HCV screening test prior to blood donation.
