ABSTRACT

OBJECTIVE:To study the effects of Lactobacillus fermentum(Lb-f)on inhibition of cytoxan(CTX) on HCT116 cells of human colorectal cancer in vitro and in vivo. METHODS:HCT116 cells were cultured in vitro. MTT assay was used to in-vestigate the effects of 4,2,1μg/ml CTX and combined with 10μg/ml Lb-f on the survival rate of HCT116 cells. HCT116 cell xe-nograft tumor nude mice model was induced to investigate the effects of 100,50,25 mg/kg CTX and combined with 180 mg/kg Lb-f on tumor volume,tumor weight,relative tumor inhibition rate,the sensitization effect of chemotherapy (by q value),the number of peripheral blood leucocyte and platelet. RESULTS:Compared with CTX alone,CTX combined with Lb-f had no signifi-cant effect on survival rate of HCT116,relative inhibition rate of tumor volume in nude mice, relative inhibition rate of tumor weight and q value (P>0.05),but increased the number of peripheral blood leucocyte and platelet (P<0.05). CONCLUSIONS:No synergistic effects of Lb-f is found on the inhibition of CTX on the growth of HCT116 in vitro and in vivo;Lb-f can inhibit the decrease of peripheral blood leucocyte and platelet of HCT116 cell xenograft tumor nude mice induced by CTX.

ABSTRACT

Objective To explore the regulatory effect of astragalus polysaccharide ( APS) on immune function in immunosuppressed mice, and to establish a set of feasible indexes of immunological regulation and evaluation system of pol-ysaccharides.Methods Both Cytoxan ( CTX)-induced immunosuppressed mice and normal mice were treated with astrag-alus polysaccharide ( APS) .Spleen index, thymus index, phagocytosis rate and phagocytic index of peritoneal macrophages were assessed, and the spleen and thymus tissues were examined by histology.The percentage of CD3 +, CD4 +, and CD8 +cells in peripheral blood were determined by flow cytometry.Results APS significantly increased the thymus and spleen indexes in both immunosuppressed mice and normal mice, promoted the histological development of spleen and thy-mus in normal mice, enhanced the recovery of histological structures in the spleen and thymus in the CTX-induced immuno-suppressed mice.APS also enhanced the phagocytosis rate and phagocytic index of peritoneal macrophages in both immuno-suppressed mice and normal mice.In addition, the percentage of CD3 +and CD4 +cells,the ratio of CD4 +/CD8 +in pe-ripheral blood of both CTX-induced immunosuppressed mice and normal mice were increased while CD8 +cells were de-creased.Conclusions The results of this study suggest that astragalus polysaccharide can improve the non-specific immu-nity and cellular immunity in mice, and indicate that we established comprehensive evaluation indexes of immunoregulatory effects of polysaccharides.

ABSTRACT

Background and purpose:The clear-cell cancer ovarian cancer have worse prognosis than the other ovarian cancer.The patient's survival rate of the clear-cell ovarian cancer has been analyzed in this article in order to study the effect of the treatment with CAPcytoxan(CTX),cisplatinum(DDP),epiadriamycin(E-ADM)plus mitomycin(MMC).Methods:33 cases(group A)with clear-cell ovarian cancer between Jan.1th 1999 and Dec.31th 1999 were compared to 37 cases(B group)with other pathological ovarian cancer.All cases underwent the tumor reductive surgery and been capable of remain the residual tumor size less than 1 cm.Patients in the two groups all underwent CAP based chemotherapy,and patients in group A with additional MMC chemotherapy at the same time.Group A had been compared with the clear-cell ovarian cancer with the CAP protocol(group C,stage Ⅰ/Ⅱ 15 cases,stage Ⅲ/Ⅳ 9 cases).Results:There was significant statistical different value of the CA125 in stage Ⅰ/Ⅱ before operation and no significant statistical difference for stage Ⅲ/Ⅳ between the two groups.There were significant decrease in the CA125 value for the stage Ⅰ/Ⅱ and no significant decrease for stage Ⅲ/Ⅳ between two groups after three and six courses chemotherapy.There were 11(33.33%)cases developed with endometriosis and 7(21.21%)with deep venous thrombosis(DVT),however the DVT had no direct correlation to the survival rate.The average survival time for stages Ⅰ/Ⅱ in group A and B was(38.3?2.4),and(38.3?2.7)months,compared to(20?3)and(34?4)months in stage Ⅲ/Ⅳ,respectively.There was no significant statistical difference(P=0.471)in four-year survival rate between groups A and B with stage Ⅰ/Ⅱ and there was significant statistical difference(P

ABSTRACT

Objective: To investigate the effect of acupuncture or moxibustion at acupoint Geshu (BL 17) on raising leukocyte and promoting marrow proliferation. Methods: Wister rats were selected to establish the leukopenia and immuno-suppression model by intraperitoneal injection of cytoxan (CTX). After they were grouped and treated, leukocyte and marrow karyocyte were counted in different phases to evaluate the effect of acupuncture or moxibustion at acupoint Geshu (BL 17) on raising leukocyte and promoting marrow hematopoiesis. Results: Acupuncture or moxibustion at acupoint Geshu (BL 17) and routine western medicine could all significantly raise leukocyte and increase marrow karyocyte. The curative effect was better in moxibustion group than in acupuncture group and in routine western medicine group (P<0.05). Conclusion: Moxibustion at acupoint Geshu (BL 17) can markedly raise peripheral blood leukocyte and marrow karyocyte of the leucopenia model in rats.

ABSTRACT

Objectives: To evaluate the efficacy, toxicity and side effects of CAP (cytoxan, adriamycin, cisplatin), NP(navelbine, cisplatin), GP(gemcitabine, cisplatin) in the treatment of patients with non small cell Lung cancer(NSCLC). Methods:146 patients with NSCLC diagnosed by pathology or cytology were treated with CAP, GP or GP regimen. Results:The overall response rate of the three groups (CAP, NP, GP)were 33.33%, 46.43% and 47.92% respectively. The major toxicity and side effects was bone marrow inhibition and the gastrointestinal reaction. Grade Ⅲ-Ⅳ side effects of CAP group were significant higher than the others. Conclusions:The regimen of navelbine and cisplatin or gemcitabine and cisplatin was more effective and less toxic than cytoxan, adriamycin, cisplatin and cisplatin.

Objective To investigate the levels of serum tumor necrosis factor-alpha (TNF-?琢) and soluble tumor necrosis factor receptor (sTNF-R) and sTNF-R/TNF-?琢 ratios in patients with severe lupus nephritis (SLN) and the impact of double pulse therapy with methylprednisolone (MP) combined cytoxan (CTX). Methods Serum TNF-?琢, soluble tumor necrosis factor receptor type Ⅰ(sTNF-RⅠ) and soluble tumor necrosis factor receptor type Ⅱ (sTNF-RⅡ) levels were determined in 38 cases of SLN patients and 35 health controls by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) before and after the double pulse therapy with MP and CTX. Anti-dsDNA antibody was detected by ELISA. Antinuclear antibody (ANA) was detected by indirect immunofluorescence experiments. Complement C3 and complement C4 were detected by rate nephelometry. Results The serum levels of TNF-?琢, sTNF-RⅠ, sTNF-RⅡand ratios of TNF-?琢/sTNF-RⅠ, TNF-a/sTNF-RⅡ were significantly higher in SLN patients than those in normal control group (P

ABSTRACT

Objective To study the synergistic antitumor effect of PCF when combine used with CTX.Methods Inoculated 0.2 mL H22 tumor fluid into the right armpit of mouse,and then the animals were divided into seven groups randomly: control group;CTX group(CTX 10 mg?kg-1);three test group(PCF 1000,500 and 250 mg?kg-1) and two combined use groups (PCF 1000,500 mg?kg-1with CTX 10 mg?kg-1).PCF were given by intragastric administration for 10 days,and CTX by intraperitoneal injection for 10 days.Same volume of saline was given to the control group.The mice were killed 24 hours after the last medication and the tumor inhibition rate was calculated.Results Three PCF groups had no effect on tumor inhibition,while two combined treatusement groups showed significant effect on tumor inhibition,while at the same doses the tumor inhibition rates were raised to 55.9% and 52.9% respectively,which were higher than that of CTX when used alone.Conclusion PCF can enhance the antitumor effects of CTX.

ABSTRACT

Aim The effect of extracts of astralus(EA) on learning, memory and immune function of CTX treated mice was observed.Methods The step-down test was adopted to investigate learning and memory in mice; the method of enveloping CRBC was adopted to explore the function of macrophage in abdominal cavity; 3H-TdR marking method was used to observe the prolifernation of T lymphocytes and the method of ELISA was applied to measure the concentration of IFN-? in blood. Results EA (25 mg?kg-1, 50 mg?kg-1,ig, 21 d ) could boost learning and memory , enhance the phagocytic activity of monocytes and the proliferation of T lymphocytes of CTX treated mice, and increase the concentration of IFN-? in blood of CTX treated mice. Conclusion EA not only promote learning and memory, but regulate immune function.

ABSTRACT

Objective To study the efficiency enhancement and toxicity reduction effects of scutellaria barbata polysaccharides(SBPS) on mice treated with cytoxan(CTX).Methods Transplanted H22 hepatoma mice were randomly divided into negative control group,CTX group and combined SBPS and CTX group.After 10 d of consecutive drug administration,the tumor inhibitory rate,blood white cell count,phagocytic index and activity of IL-2 were measured.Results SBPS increased the tumor inhibitory rate and reduced the toxicity of cytoxan,and enhanced the function of mononuclear phagocytes and the activity of IL-2 and TNF-?.Conclusions SBPS has efficiency enhancement and toxicity reduction effects on cytoxan,and the mechanism may be by increasing the immunologic function of the organism.

ABSTRACT

AIM To study antitumor activity of melatonin in mice and its synergetic effects when MT combined with chemotherapeutic agents cytoxan(CTX). METHODS At a tolerable dose level,MT was administrated singlely or in combination with CTX to treat the transplanted murine solid tumor U 14 . The inhibition of tumor weight were observed. The weight of spleen and the hepatic cells' structure were also be measured. RESULTS The high dosed group of MT(80 mg?kg -1 ?d -1 ,sc) had a certain inhibition on the growth of the U 14 with inhibition rate of 27 1%. The low dosed group of MT(40 mg?kg -1 ?d -1 , sc) and the high dosed one combined with the CTX (25 mg?kg -1 ?d -1 ?1,ip) showed much pronounced than that of solely administration of MT. There was no significant influnce on the weight of spleen and the hepatic cells' structure. CONCLUSIONS The antitumor effect was observed by independent administration of MT. There was a synergism when MT combined with CTX but their toxicities were not obviously enhanced.
