ABSTRACT

Objective To establish mouse models of intracerebral hemorrhage using autologous arterial blood,to study the physiological property of hippocampal neurons,brain edema changes and learning ability in the mouse models after intracerebral hemorrhage.Methods Eighty male C57/BL6 mice were randomly divided into intracerebral hemorrhage group and control group (n=40); 20 μL arterial blood from the tail arteries or normal saline were injected into the caudate nucleus of intracerebral hemorrhage group and control group by stereotactic technique,respectively.One,three,five and seven d after injection,the neurological impairment was scored; the behavioral changes of the mice in the Morris water maze (navigation test and space exploration experiment) were observed; brain edema was measured by wet and dry weight method and electrophysiological differences of hippocampal neurons were recorded by whole-cell patch-clamp technique and computer software.Results As compared with those in the control group,significantly increased neurological deficit scores one,three,five and seven d after injection,statistically decreased residence time in the platform on the fifth d of training,obviously increased water content around the brain edema one,three,five and seven d after injection,and significantly decreased resting membrane potential and input resistance in the hippocampal CA1 pyramidal cells five d after injection of mice in the intracerebral hemorrhage group were noted (P<0.05).Conclusion The hippocampus-dependent spatial leaming ability of intracerebral hemorrhage mice is decreased,and the permeability of potassium channels is enhanced.

ABSTRACT

Objective To explore the clinical, imaging and electrophysiological characteristics of Creutzfeldt-Jakob disease ( CJD) .Methods The clinical data of 28 very probable CJD patients were analyzed retrospectively. Results Twenty-eight patients in this group, acute onset was in 5 cases, subacute onset was in 17 cases, chronic onset was in 6 cases.The initial symptoms were dizziness, gait instability in 11 cases ( 39.3%) , cognitive dysfunction in 10 cases (35.7%), visual symptoms in 3 cases (10.7%), mental symptoms, insomnia, repeated falls and barylalia in 1 case (3.6%) respectively.The clinical symptoms were progressive dementia in 28 cases (100%), myoclonus in 21 cases (75.0%), ataxia in 19 cases (67.9%), abnormal mental behavior in 7 cases (25.0%), pyramidal signs in 20 cases (71.4%), extrapyramidal symptoms in 15 cases (53.6%), visual impairment in 6 cases (21.4%), sleep disorder in 11 cases (39.3%), and aphasia in 9 cases (32.1%).Twelve patients (42.9%) had positive results with CSF 14-3-3 protein analysis.Twenty-four patients (85.7%) had MRI high signal abnormalities in caudate nucleus and putamen or/and at least two cortical regions.Eight cases (28.6%) experienced a typical periodic sharp wave complexes (PSWCs), 12 cases (42.9%) had a diffuse symmetric or asymmetric slow-wave activity, 8 cases (28.6%) showed rhythmic three-phase or delta wave, with the whole guide or forehead synchronization.Conclusion Most CJD patients are subacute or chronic onset.Initial symptoms of CJD are atypical, progressed to classical clinical characteristics presented in middle-late stages.Nearly half patients have CSF 14-3-3 protein positive.MRI DWI of mostly CJD patients have abnormal signal.EEG of nearly a third of CJD patients shows PSWCs.

ABSTRACT

Object To study the influence of main active principles of ZHIGANCAO DECOCTIDN (ZD), glycyrrhizic acid (GA), ginseng total saponin (GTS) and Ophiopogon total saponin (OTS) on electrophysiology of isolated rat myocardium Methods The influence of GA, GTS and OTS on the automaticity, excitability and functional refractory period of isolated rat atrium and papillary muscle were studied in comparison with ZD made free of the above said active ingredients Results The combine use of GA, GTS and OTS significantly decreased the automaticity, inhibited excitaibility and prolonged the functional refractory period of isolated rat atrium; decreased the automaticity and arrhythmia of papillary muscle induced by epinephrine, while ZD made free of GS, GTS and OTS showed much less effect than the intact decoction Conclusion GA, GTS and OTS proved to be the main effective ingredients responsible for the antiarrhythmic activity of ZD
