ABSTRACT

Neuropeptides play an important role in the physiological functions of the human body. The physiological activities such as pain, sleep, mood, learning and memory are affected by neuropeptides. Neuropeptides mainly exist in the nerve tissue of the body, and a small amount of them are distributed in body fluid and organs. At present, analysis of large-scale identification of neuropeptides in whole brain tissue is still challenging. Therefore, high-throughput detection of these neuropeptides is greatly significant to understand the composition and function of neuropeptides. In this study, 1 830 endogenous peptides and 99 novel putative neuropeptides were identified by extraction of endogenous peptides from whole brain tissue of mice by liquid phase tandem mass spectrometry (LC-MS / MS). The identification of these endogenous peptides provides not only a reference value in the treatment and mechanism studies of diseases and the development of drugs, but also the basis for the study of a new neuropeptides and their functions.

ABSTRACT

【Objective】To observe the effect of plasma ?-endogenous peptides (?-EP) on gestational function in endometriosis(EMT) rats.【Methods】 Female SD rats at the third estrous cycle were used to establish EMT model.Then 20 EMT rats were equally randomized into the model group and naltrexone(an opioid receptor antagonist) group,and other 10 rats were in the pseudo-operation group.Naltrexone group was given naltrexone 2?mg?kg~-1?d~-1 by gastric gavage during proestrus and normal saline at other time,and the model group and pseudo-operation group were given normal saline for one month.Serum levels of ?-EP,luteinizing hormone(LH) and prolactin(PRL) were examined during proestrus,and serum progesterone(P) level and gestational function were observed after estrus.【Results】Serum levels of ?-EP and PRL increased and LH decreased during proestrus,and serum P level also decreased after estrus in the model group(P

ABSTRACT

The conformational proclivity of leucine and methionine enkephalinamides in deuterated dimethyl sulphoxide has been investigated using proton magnetic resonance at 500 MHz. The resonances from the spin system of the various amino acid residues have been assigned from the 2-dimensional correlated spectroscopy spectra. The temperature variation of the amide proton shifts indicates that none of the amide proton is intramolecularly hydrogen-bonded or solvent-shielded. The analysis of vicinal coupling constants, 3JHN.C 2 H,along with temperature coefficients and the absence of characteristic nuclear Overhauser effect cross peaks between the NH protons reveal that there is no evidence of the chain folding in these molecules. However, the observation of nuclear Overhauser effect cross peaks between the NH and the CαH of the preceding residue indicates preference for extended backbone conformation with preferred side chain orientations particularly of Tyr and Phe in both [Leu5] - and [Met5]-enkephalinamides.
