ABSTRACT

Abstract Bone regeneration is crucial for repairing bone tissue following various injuries. Research techniques that enable the study of metabolic changes in bone tissue under different conditions are important for understanding bone repair and remodeling. This study used bone scintigraphy to evaluate osteogenesis secondary to osteotomy in a preclinical model of New Zealand rabbits. For this purpose, we conducted a longitudinal, prospective, case-control study in which scintigraphic variables were measured in both the right forearm (case-operated) and the left forearm (control - non-operated). The study sample consisted of 10 rabbits subjected to osteotomy, followed by a 12-week postoperative evaluation period, divided into six imaging stages at 1, 2, 3, 4, 8, and 12 weeks. We observed that the operated forearm showed significantly higher external radiation than the control side, using the pinhole collimator, denoting an increase in the biodistribution and tropism of the radiopharmaceutical to the operated forearm. Among the three evaluated time points, osteoblastic activity was highest in the second week and presented a significant decline in the 8th and 12th weeks, denoting regeneration and resolution of the surgical injury; the control forearm was also influenced by the inactivity imposed by the operated forearm. This fact was notably evidenced by the reduction in the metabolic activity of osteoblasts in the left forearm. Our study suggested that bone scintigraphy was sensitive enough to semi-quantitatively differentiate the metabolic activity of osteoblasts in the operated forearm in the three temporal landmarks evaluated in the study.

ABSTRACT

Resumo Objetivo Avaliar a aplicabilidade e a efetividade de um jogo com elementos de Roleplaying Game (RPG) enquanto ferramenta didático pedagógica para o ensino em saúde. Método A população deste estudo foi de discentes de cursos de graduação e pós-graduação, da área da saúde, da Universidade Federal do Rio Grande do Sul, Brasil. Trata-se de uma pesquisa qualitativa. O estudo foi realizado entre os meses de abril a junho de 2018, em duas etapas. Primeiramente foi realizado o RPG com as turmas envolvidas, e após uma semana, entrevistas de GF para a obtenção de informações, contando com voluntários dessas turmas que aceitaram participar da pesquisa. A análise das informações foi baseada em aspectos da Teoria Fundamentada nos Dados (TFD). Resultados Da análise emergiram as categorias:1) Criação do jogo, 2) Impacto da experiência no ensino aprendizagem, 3) Reflexão acerca do jogo. Os resultados demonstram que o uso do jogo com elementos de RPG tem um enorme potencial a ser explorado no processo de ensino-aprendizagem na área da saúde. Trata-se de um método inovador e ainda pouco utilizado na área da saúde. É importante seguir algumas premissas para que a ferramenta seja utilizada de forma potente. Conclusão Nessa direção entende-se que o jogo auxilia na formação em saúde, enriquecendo conhecimentos e habilidades através de uma experiência desafiadora e conectada com a realidade.


Abstract Objective To assess the applicability and effectiveness of a game incorporating Roleplaying Game (RPG) elements as a pedagogical tool for health education. Method The study population comprised undergraduate and postgraduate students in the health field at the Universidade Federal do Rio Grande do Sul, Brazil. This qualitative research was conducted between April and June 2018 in two stages. Initially, the RPG was implemented with the involved classes, followed by Focus Group interviews conducted a week later to gather information, with volunteers from these classes who willingly participated in the research. Information analysis was grounded in aspects of Grounded Theory (GT). Results The analysis yielded the following categories: 1) Game creation, 2) Impact of the experience on teaching and learning, 3) Reflection on the game. The results demonstrate that the use of RPG-influenced games holds significant untapped potential in the health education process. This innovative method remains underutilized within the health domain, necessitating adherence to specific premises for its potent application. Conclusion In this context, it is understood that the game contributes to health education, enhancing knowledge and skills through a challenging and reality-connected experience.

ABSTRACT

Aim To explore the potential targets and related signaling pathways of Agaricus blazei Murill (AbM ) extract in the treatment of chronic myeloid leukemia (CML) based on liquid chromatography mass spectrometry ( LC-MS ), network pharmacology, molecular docking, and were further verified by experiments in vitro. Methods The active components of AbM extract were retrieved from LC-MS, Swiss Target Prediction database was used to predict related targets, and CML disease target genes were obtained from Gen- eCards and DisGeNET databases. After screening the common targets of drug and CML, the protein-protein interaction network of the common targets was performed by STRING, and GO and KEGG enrichment a- nalysis were done by DAVID database. Cytoscape software was used to construct the network of target protein. Molecular docking was carried out by DockThor, and the Pymol software was used to make a visual picture. The inhibitory effect of AbM extract on leukemia cells K562 was determined by CCK-8 experiment, and the effect of AbM extract on the expression and phosphorylation level of related proteins was verified by Western blot. Results The prediction results showed that 126 active components of AbM extract, and 172 common targets were collected. ABSTRACT

The study was carried out to understand the changes in the ethical cognition status of laboratory animals and the effectiveness of laboratory animal ethics education among medical students in Xiangya School of Medicine of Central South University (CSU), and provide new enlightenment for further strengthening the ethical education of laboratory animals. In the study, the same self-compiled questionnaire was used to investigate the ethical cognition of experimental animals among medical students in Xiangya School of Medicine of CSU in 2011 and 2021, and 359 and 363 questionnaires were collected respectively. Through comparative analysis of the questionnaire results before and after ten years, it was found that medical students’ animal experiment operation and attitudes towards laboratory animals, cognition of experimental animal ethics knowledge and their attitude to animal experiment ethics education were significantly improved. It showed that the state of experimental animal ethics cognition among medical students in Xiangya School of Medicine of CSU had improved significantly in recent 10 years, but the cognition of experimental animal ethics knowledge was higher than the actual behavior of caring for experimental animals, and there was the phenomenon of "separation of knowledge and action". The ethics education of experimental animals needs to pay more attention to the development of students’ behavior of caring for experimental animals.

ABSTRACT

ObjectiveTo investigate the role and mechanism of total saponins of Dioscorea (TSD) in mitigating nonalcoholic steatohepatitis (NASH) in mice. MethodForty-eight C57BL/6J mice were randomized into a normal group and a modeling group. The mice for modeling were fed with a high-fat and high-cholesterol diet + 20% fructose solution for 16 weeks and randomized into model, atorvastatin (4 mg·kg-1·d-1), and high-, medium-, and low-dose (200, 60, and 20 mg·kg-1·d-1) TSD groups. The mice were administrated with corresponding doses of drugs by gavage for 8 weeks. The mouse activity, liver index, levels of total cholesterol (TC), triglycerides (TG), and free fatty acids (FFAs) in the liver, and levels of TC, TG, aspartate aminotransferase (AST), alanine aminotransferase (ALT), γ-glutamyl transferase (GGT), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) in the serum were measured. Hematoxylin-eosin staining, Masson staining, oil red O staining, and transmission electron microscopy were employed to observe the pathological changes, lipid accumulation, and morphological changes of liver ultrastructure. Western blot was employed to determine the protein levels of AMP-activated protein kinase (AMPK), sterol regulatory element-binding protein-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), and phosphorylated ACC (p-ACC) in the liver tissue. ResultCompared with the normal group, the activity of mice in the model group decreased(P<0.05, P<0.01), the levels of TC, TG, FFA and serum TC, TG, ALT, AST, GGT, IL-1β and TNF-α, liver coefficient and liver pathology scores were significantly increased, the expression of p-AMPK/AMPK and p-ACC proteins in liver tissues was significantly reduced, and the expressions of SREBP-1c and ACC proteins were significantly increased (P<0.01). Compared with the model group, atorvastatin increased the mouse activity (P<0.05), while each dose of TSD caused no significant changed in the mouse activity. The levels of TC, TG, FFA in liver and serum TC, TG, ALT, AST, GGT, IL-1β, TNF-α, liver coefficient and liver pathological score in TSD and atorvastatin groups were significantly decreased, and the expressions of p-AMPK/AMPK and p-ACC in liver tissue were significantly increased. The expressions of SREBP-1c and ACC were significantly decreased (P<0.05,P<0.01). ConclusionTSD may alleviate NASH in mice by regulating the AMPK/SREBP-1c/ACC signaling pathway to reduce lipid synthesis.

ABSTRACT

Multiple sclerosis (MS) is a neuroinflammatory demyelinating disease, mediated by pathogenic T helper 17 (Th17) cells. However, the therapeutic effect is accompanied by the fluctuation of the proportion and function of Th17 cells, which prompted us to find the key regulator of Th17 differentiation in MS. Here, we demonstrated that the triggering receptor expressed on myeloid cells 2 (TREM-2), a modulator of pattern recognition receptors on innate immune cells, was highly expressed on pathogenic CD4-positive T lymphocyte (CD4+ T) cells in both patients with MS and experimental autoimmune encephalomyelitis (EAE) mouse models. Conditional knockout of Trem-2 in CD4+ T cells significantly alleviated the disease activity and reduced Th17 cell infiltration, activation, differentiation, and inflammatory cytokine production and secretion in EAE mice. Furthermore, with Trem-2 knockout in vivo experiments and in vitro inhibitor assays, the TREM-2/zeta-chain associated protein kinase 70 (ZAP70)/signal transducer and activator of transcription 3 (STAT3) signal axis was essential for Th17 activation and differentiation in EAE progression. In conclusion, TREM-2 is a key regulator of pathogenic Th17 in EAE mice, and this sheds new light on the potential of this therapeutic target for MS.

ABSTRACT

ObjectiveThe active ingredients, action targets, and signaling pathways of Cuscutae Semen to control premature ovarian failure were initially predicted by network pharmacology and molecular docking techniques, and an animal model of premature ovarian failure was constructed to explore the mechanism of Cuscutae Semen based on lipid and atherosclerosis signaling pathways. MethodThe effective components and corresponding targets of drugs were obtained from Traditional Chinese Medicines Systems Pharmacology Platform (TCMSP), Swiss Target Prediction, Pharmmapper, and other databases. GeneCards database was used to collect disease-related targets. Venny2.1.0 online tool was used to screen out the intersection targets of drugs and diseases, and STRING database and Cytoscape v3.7.2 software were used to construct the network diagram of "drug-component-target" and protein-protein interaction (PPI). The gene ontology (GO) and the Kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses of the intersection targets were performed by running the R language script. The molecular docking technology was utilized to dock drug components with targets and visualize some of the docking results. The mice were randomly divided into a blank group, a model group, a Cuscutae Semen group, and an estradiol valerate group, and the ovarian premature failure model was prepared by chronic stress. The blank group and the model group were gavaged with the same amount of normal saline, and the Cuscutae Semen group was given a Cuscutae Semen decoction of 2.6 g·kg-1·d-1. The estradiol valerate group was given an estradiol valerate solution of 0.13 mg·kg-1·d-1. After four weeks, samples were collected, and hematoxylin-eosin (HE) staining was performed to observe the histopathological changes in the ovary. Serum levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), Muller's tube inhibitor/anti-Muller's tube hormone (AMH), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were determined by enzyme-linked immunosorbent assay (ELISA). The expression levels of extracellular regulatory protein kinase (ERK), nuclear transcription factor-κB p65 (NF-κB p65), nuclear transcription factor-κB suppressor α (IκBα), interleukin-1β (IL-1β), IL-6, and tumor necrosis factor-α (TNF-α) were measured by Western blot. ResultA total of 171 targets of Cuscutae Semen for the prevention and treatment of premature ovarian failure were screened, mainly including tumor protein p53 (TP53), protein kinase B1 (Akt1), sarcoma (SRC), tumor necrosis factor (TNF), epidermal growth factor receptor (EGFR), etc. KEGG pathway enrichment analysis predicts that Cuscutae Semen is mainly involved in lipid and atherosclerosis, TNF signaling pathway, and TP53 signaling pathway to control premature ovarian failure. The animal experiments show that compared with the premature ovarian failure model group, the Cuscutae Semen group can significantly upregulate AMH, E2, and HDL-C (P<0.05, P<0.01), significantly downregulate LH, TC, and LDL-C (P<0.01), greatly reduce IL-1β, IL-6, and TNF-α protein levels, as well as ERK, NF-κB p65, and their phosphorylation levels (P<0.01). ConclusionCuscutae Semen can regulate hormone levels and improve ovarian function through a multi-component, multi-target, and multi-pathway approach, and the mechanism may be related to the regulation of lipid and atherosclerosis signaling pathways.

ABSTRACT

ObjectiveThis study observes the intervention effect of Longmu Piyan prescription on oxidative stress in BALB/c mice with atopic dermatitis (AD) induced by 2,4-dinitrochlorobenzene (DNCB) and explores its mechanism. MethodThe AD model was established using the method of DNCB sensitization on the back skin of BALB/c mice. Forty male BALB/c mice were randomly divided into a blank group, a model group, a vitamin C control group (0.5×10-3 mg·kg-1), and a Longmu Piyan prescription group (26 g·kg-1). Except for the blank group, other groups were sensitized with different concentrations of DNCB on the back to induce AD, and the blank group was treated with matrix coating. The gastric administration was started on the seventh day after sensitization with 2% DNCB and on the 24th day after sensitization with 0.2% DNCB continuously for 21 days. The changes in skin lesions of each group were directly observed after the experiment. Enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of interleukin (IL)-4, tumor necrosis factor (TNF)-α, immunoglobulin E (IgE), and reactive oxygen species (ROS) in the serum of each group. The total antioxidant capacity determination kit-trace method (ABTS method) was used to measure the level of total antioxidant capacity (TAOC) in serum. The Hematoxylin eosin (HE) staining method was used to observe the pathological and morphological changes of the skin lesion site. The immunohistochemical method was used to detect the expression of thymic stromal lymphopoietin (TSLP) in the skin lesion site. Western blot was used to detect the expression of filaggrin (FLG) in the dorsal skin lesions. ResultThe results showed that compared with the blank group, the skin lesion score of the model group mice was significantly increased (P<0.01), and HE staining showed characteristic pathological changes of AD in the skin lesion site. At the same time, the expression of TSLP in the skin lesion was significantly increased, and that of FLG was reduced (P<0.05). The levels of TNF-α, IL-4, IgE, and ROS in serum increased, while the activity of TAOC decreased (P<0.01). Compared with the model group, the Longmu Piyan prescription group showed a significant decrease in skin lesion scores and a significant improvement in skin lesion pathology. At the same time, the expression of TSLP decreased, and the expression of FLG increased in the skin lesions (P<0.05). In addition, compared with the model group, the serum levels of TNF-α, IL-4, IgE, and ROS also decreased to varying degrees (P<0.05,P<0.01), and TAOC activity increased in the Longmu Piyan prescription group (P<0.01). ConclusionThere is a significant correlation among the degree of oxidative stress, the severity of skin lesions in AD, and the levels of inflammatory cytokines. Longmu Piyandu prescription can improve AD-like skin lesions in BALB/c mice by promoting ROS clearance, enhancing TAOC, and inhibiting oxidative stress, thus protecting the skin barrier and reducing inflammation.

ABSTRACT

La deficiencia de zinc puede ser un factor mediador en los trastornos del crecimiento fetal en la descendencia de la gestante diabética. Se persiguió como objetivo determinar la influencia de un suplemento con zinc sobre la morfometría externa corporal y craneofacial en fetos de ratas diabéticas con hiperglucemias moderadas. Durante la gestación, ratas diabéticas y controles fueron suplementadas por vía oral con sulfato de zinc (50 mg/kg-pc) o no recibieron tratamiento. Los fetos descendientes del grupo diabético suplementado presentaron niveles similares a los controles en las variables de crecimiento somático determinadas. La suplementación con zinc a ratas diabéticas favoreció el crecimiento intrauterino en los fetos. Los resultados de esta investigación constituyen aportes para dilucidar los requerimientos de zinc que permitan prevenir los trastornos del crecimiento fetal en la descendencia de gestantes diabéticas.


Zinc deficiency may be a mediating factor in fetal growth disorders in the offspring of diabetic pregnant women. The objective was to determine the influence of a zinc supplement on external body and craniofacial morphometry in diabetic rat fetuses with moderate hyperglycemia. During gestation, diabetic and control rats were orally supplemented with zinc sulphate (50 mg/kg bw) or received no treatment. The fetuses descendants of the supplemented diabetic group had levels similar to the control ones in the determined somatic growth variables. Zinc supplementation to diabetic rats favoured intrauterine growth in fetuses. The results of this research constitute a contribution to elucidate zinc requirements that allow preventing fetal growth disorders in the offspring of diabetic pregnant women.

Introduction. Nutritional profiling systems (NPS) are mainly used in front-end labeling policies in order to make the purchase and consumption choice conscious and healthy. Objetive. This study systematically reviewed evidence from interventions on the effect of NPS in the front-of- package on food purchases. Materials and methods. A bibliographic search was carried out in electronic sources from Medline, Elsevier, Scielo and Lilacs, of experimental studies and intervention between 2012 and 2022. A total of 14 articles were included in the review. They were analyzed according to the intervention modality used: 4 studies analyzed the effect of NPS in a real purchase situation and 10 evaluated purchase perception/intention. Results. According to the modality of intervention, the 6 studies that analyzed the NPS-warning system, all recorded healthier purchases compared to the control groups. While for the NPS-NutriScore, Health Star Rating and Multiple Traffic Lights were effective in the decision to purchase healthier foods, in 5 of 7 studies for the first NPS, in 4 of 7 for the second and in 4 of 8 for the third, compared with control groups. Conclusions. Findings of this study suggest that NPSs may be effective for healthy purchase choices, even so it is necessary to strengthen the system and policies with nutritional food education campaigns(AU)


Introducción. Los sistemas de perfilado nutricional (SPN) son utilizados principalmente en políticas de etiquetado frontal con la finalidad de que la elección de compra y consumo sea consciente y saludable. Objetivo. Este estudio revisó sistemáticamente la evidencia de las intervenciones sobre el efecto de SPN en el frente del paquete en las compras de alimentos. Materiales y métodos. Se realizó una búsqueda bibliográfica en fuentes electrónicas de Medline, Elsevier, Scielo y Lilacs, de estudios experimentales e intervenciones entre 2012 y 2022. Se incluyeron un total de 14 artículos en la revisión. Fueron analizados según la modalidad de intervención utilizada: 4 estudios analizaron el efecto de los SPN en una situación de compra real y 10 evaluaron percepción/intención de compra. Resultados. Según la modalidad de intervención, los 6 estudios que analizaron el SPN-sistema de advertencia, todos registraron compras más saludables en comparación con grupo controles. Mientras que para los SPN-NutriScore, Estrellas de Salud y Semáforo Tricolor Múltiple fueron efectivas en la decisión de compra de alimentos más saludables, en 5 de 7 estudios para el primer SPN, en 4 de 7 para el segundo y en 4 de 8 para el tercero, comparados con grupo controles. Los hallazgos de este estudio sugieren que los SPN pueden ser efectivos para elecciones de compras saludables, aun así, es necesario reforzar el sistema y las políticas con campañas de educación alimentaria nutricional(AU)

ABSTRACT

This is a methodological study that aimed to develop a board game to develop autonomy and social skills in children with autism. The game's development was supported by the toy library of the Hospital Universitário Júlio Bandeira [Julio Bandeira University Hospital]. Models were collected through an integrative review and an interview with mothers of autistic children. The game was based on methods extracted from articles, from the Picture Exchange Communication System, Social Stories, and the difficulties encountered during the interview. It has a board with 32 squares, 22 cards (three different models), one die, and small cars used to advance through the squares on the board. Due to their playful nature, board games have the potential to be used by children with autism to develop autonomy and social skills. It was concluded that the production of care-educational technologies aimed at the autistic public is essential for cognitive and social development.


Trata-se de estudo metodológico que objetivou elaborar um jogo de tabuleiro para desenvolver a autonomia e as habilidades sociais nas crianças com autismo. A produção do jogo contou com o apoio da brinquedoteca do Hospital Universitário Júlio Bandeira. Captaram-se modelos mediante uma revisão integrativa e uma entrevista com mães de crianças autistas. O jogo se baseou nos métodos extraídos dos artigos, no Picture Exchange Communication System, nas Histórias Sociais e nas dificuldades encontradas na entrevista. Ele conta com um tabuleiro de 32 casas, 22 cartas (3 modelos diferentes), 1 dado e carrinhos utilizados para avançar as casas do tabuleiro. Devido ao seu caráter lúdico, os jogos de tabuleiro têm potencial de ser usados por crianças com autismo para desenvolver a autonomia e habilidades sociais. Concluiu-se que a produção de tecnologias cuidativo-educacionais voltadas para o público autista é fundamental para o desenvolvimento cognitivo e social.

ABSTRACT

La investigación fundamental en homeopatía ha avanzado considerablemente en los últimos 20 años: desde estudios exploratorios con animales y plantas hasta la caracterización de los efectos sistémicos de los medicamentos homeopáticos y estudios in vitro con sistemas celulares aislados para evaluar los cambios en los mecanismos de adaptación celular y señalización intracelular frente a tratamientos homeopáticos variables. El número de artículos publicados a lo largo del tiempo ha permitido realizar varias revisiones sistemáticas. Recientemente, la demostración de que los medicamentos homeopáticos podrían modificar las funciones celulares a través de mecanismos epigenéticos (metilación y desmetilación de ADN) preparó el camino para un campo de investigación completamente nuevo. En paralelo, el descubrimiento de las nanopartículas y propiedades físicas específicas de las diluciones homeopáticas ha arrojado luz hacia un campo antes poco conocido, dado que se consideraba que las diluciones homeopáticas no consistían más que de agua. Así las cosas, los retos para el futuro conciernen a la demostración, o no, de la interrelación entre ambos fenómenos.


Fundamental research in homeopathy has much advanced in the past 20 years. From exploratory studies with animals and plants to the characterization of the systemic effects of homeopathic medicines and in vitro studies with isolated cell systems to assess changes in the mechanisms of cell adaptation and intracellular signaling facing variable homeopathic treatments. The amount of articles published over time enabled several systematic reviews. Recently, demonstration that homeopathic medicines might modify cell functions through epigenetic mechanisms (DNA methylation and demethylation) paved the road for a fully new field of research. In parallel, the discovery of nanoparticles and specific physical properties of homeopathic dilutions brought light to a previously poorly known field, as it was believed that homeopathic dilutions consist in nothing but water. Thus being, challenges for the future concern the demonstration, or not, of the interrelationship between both phenomena.

ABSTRACT

RESUMO Objetivo comparar as eficácias da terapia fonológica tradicional e terapia fonológica associada à estratégia de gamificação em sujeitos com Transtorno Fonológico (TF). Método participaram dez sujeitos com TF que apresentavam o processo de substituição de líquidas. Os sujeitos foram randomizados em dois grupos: terapia fonológica tradicional (grupo controle - GC) e terapia fonológica associada a estratégia de gamificação mediada por computador (grupo gamificação - GG). A intervenção fonológica compreendeu, para ambos os grupos, etapas de percepção e produção de fala. As intervenções se diferenciaram na etapa de percepção, na qual o GG foi submetido ao jogo com estratégia de gamificação. Ao final de cada sessão, foram registrados a produção de fala dos sujeitos (% de acerto) para cada etapa terapêutica, a partir de palavras-alvo e palavras-sondagem. Para análise foram considerados: média de acerto dos sujeitos para cada etapa terapêutica; valores de PCC-R (Porcentagem de Consoantes Corretas Revisado) pré e pós terapia; além do número de sessões utilizadas para se atingir 85% de produção correta. Resultados não houve diferença estatisticamente significante entre os tipos de intervenção considerando a média de acertos das produções e o número de sessões. Houve efeito significante para as condições pré e pós terapia na comparação dos valores de PCC-R para ambos os modelos. Os sujeitos do GC tiveram os valores de PCC-R maiores do que as do GG. Conclusão ambos os modelos de intervenção apresentam resultados semelhantes, propiciando melhora no desempenho fonológico do sujeito desde a primeira sessão.


ABSTRACT Purpose to compare the efficacies of traditional phonological therapy and phonology associated with the gamification strategy in children with Phonological Disorder (PD). Methods ten individuals with PD participated who showed the process of replacing liquids. They were randomized into two groups: traditional phonological therapy (control group - CG) and phonological therapy associated with a gamification strategy mediated by computer (gamification group - GG). The phonological intervention comprised, for both groups, stages of speech perception and production. Interventions differed in the perception stage, in which the GG was submitted to the game with gamification strategies. At the end of each session, individuals speech production (% of correct answers) were registered for each therapeutic stage, based on target words and sounding words. For analysis the following were considered: The individuals mean of correct answers for each therapeutic stage; PCC-R value (percentage of correct consonants) pre and post therapy; beyond of the number of sessions used to reach 85% of correct production. Results there was no statistical difference between the types of intervention considering the average of correct answers of the productions and the number of sessions. There was a significant effect for pre- and post-therapy conditions in the comparison PCC-R values ​​for both models. The individuals in the GC had the PCC-R values higher than those of GG. Conclusion both models of intervention present similar results, providing an improvement in the individuals phonological performance from the first session.

Abstract Background Rheumatoid arthritis is an autoimmune inflammatory disease that often leads patients to muscle impairment and physical disability. This study aimed to evaluate changes in the activity of proteasome system in skeletal muscles of mice with collagen-induced arthritis (CIA) and treated with etanercept or methotrexate. Methods Male DBA1/J mice were divided into four groups (n = 8 each): CIA-Vehicle (treated with saline), CIA-ETN (treated with etanercept, 5.5 mg/kg), CIA-MTX (treated with methotrexate, 35 mg/kg) and CO (healthy control group). Mice were treated two times a week for 6 weeks. Clinical score and hind paw edema were measured. Muscles were weighted after euthanasia and used to quantify proteasome activity, gene (MuRF-1, PMSα4, PSMβ5, PMSβ6, PSMβ7, PSMβ8, PSMβ9, and PSMβ10), and protein (PSMβ1, PSMβ5, PSMβ1i, PSMβ5i) expression of proteasome subunits. Results Both treatments slowed disease development, but only CIA-ETN maintained muscle weight compared to CIA-MTX and CIA-Vehicle groups. Etanercept treatment showed caspase-like activity of 26S proteasome similar to CO group, while CIA-Vehicle and CIA-MTX had higher activity compared to CO group (p: 0.0057). MuRF-1 mRNA expression was decreased after etanercept administration compared to CIA-Vehicle and CO groups (p: 0.002, p: 0.007, respectively). PSMβ8 and PSMβ9 mRNA levels were increased in CIA-Vehicle and CIA-MTX compared to CO group, while CIA-ETN presented no difference from CO. PMSβ6 mRNA expression was higher in CIA-Vehicle and CIA-MTX groups than in CO group. Protein levels of the PSMβ5 subunit were increased in CO group compared to CIA-Vehicle; after both etanercept and methotrexate treatments, PSMβ5 expression was higher than in CIA-Vehicle group and did not differ from CO group expression (p: 0.0025, p: 0.001, respectively). The inflammation-induced subunit β1 (LMP2) was enhanced after methotrexate treatment compared to CO group (p: 0.043). Conclusions The results of CIA-Vehicle show that arthritis increases muscle proteasome activation by enhanced caspase-like activity of 26S proteasome and increased PSMβ8 and PSMβ9 mRNA levels. Etanercept treatment was able to maintain the muscle weight and to modulate proteasome so that its activity and gene expression were compared to CO after TNF inhibition. The protein expression of inflammation-induced proteasome subunit was increased in muscle of CIA-MTX group but not following etanercept treatment. Thus, anti-TNF treatment may be an interesting approach to attenuate the arthritis-related muscle wasting.

ABSTRACT Objective: to develop a playful-educational technology for learning Nursing Semiology and Semiotics, as well as to carry out the evaluation of its playability before and after implementing adjustments with undergraduate Nursing students. Method: a methodological study that followed the planning, prototyping and production stages corresponding to the first playable version for development. For the playability evaluation, 62 students were divided into two groups: one for to evaluate the preliminary version and the other for after implementing adjustments; and, for this, the Friedman test was used, which allows detecting differences between medians of more than two paired samples, considering p-value below 5% as statistical significance. Results: the game had three sequential phases: the first addressed vital signs; the second, biosafety measures; and the third, the physical examination. Referring to the playability evaluation, it was possible to observe that the "Concentration" and "Feedback" categories and the general median presented better evaluation scores after implementing adjustments. Conclusion: the current study sought to develop a technology that would allow students attending Bachelor's degree courses in Nursing to sharpen their knowledge in Nursing Semiology and Semiotics. In addition to that, it was sought to evaluate the playability of Semio em Jogo ®; thus, it was observed that the academic community considered the game satisfactory after the modifications as much as before implementing them.


RESUMEN Objetivos: desarrollar una tecnología lúdico-educativa para la enseñanza de Semiología y Semiotécnica en Enfermería; y evaluar la facilidad de juego de una tecnología lúdico-educativa antes y después de implementar ajustes con estudiantes de la carrera de grado en Enfermería. Métodos: estudio metodológico que siguió las etapas de planificación, diseño del prototipo y producción de la primera versión apta para jugar para el desarrollo. A fin de evaluar la facilidad de juego se recurrió a 62 estudiantes divididos en dos grupos: uno para evaluar la versión preliminar y otro para luego de implementar los ajustes; y para ello se utilizó la prueba de Friedman, que permite detectar diferencias entre medianas de más de dos muestras emparejadas, considerando valores de p inferiores al 5% como significancia estadística. Resultados: el juego estuvo compuesto por tres fases secuenciales: en la primera se abordaron los signos vitales; en la segunda, las medidas de bioseguridad; y en la tercera, el examen físico. En cuanto a la evaluación de la facilidad de juego, se pudo observar que las categorías "Concentración" y "Feedback" y la mediana general presentaron una mejor evaluación después de implementar ajustes. Conclusión: el presente estudio pretendió desarrollar una tecnología que permitiese que los alumnos de la Licenciatura en Enfermería agudicen sus conocimientos de Semiología y Semiotécnica en Enfermería. También se buscó evaluar la facilidad de juego de Semio em Jogo ®; de esta manera, se observó que la comunidad académica consideró que el juego alcanzó niveles satisfactorios tanto antes como después de las modificaciones.

Método: estudo metodológico, seguindo as etapas de planejamento, prototipagem e produção da primeira versão jogável para o desenvolvimento. Para a avaliação da jogabilidade, contou-se com 62 estudantes divididos em dois grupos, um para a avaliação da versão preliminar e outro para após a implementação de ajustes, e, para tal, utilizou-se o teste de Friedman que permite detectar diferenças entre medianas de mais de duas amostras emparelhadas, considerando como significância estatística valores de p-valor menor que 5%. Resultados: o jogo contou com três fases sequenciais: na primeira foram abordados os sinais vitais; na segunda, as medidas de biossegurança; e na terceira, o exame físico. Referente à avaliação da jogabilidade, foi possível observar que as categorias "concentração", "feedback" e mediana geral apresentaram melhor avaliação após a implementação de ajustes. Conclusão: o presente estudo buscou desenvolver uma tecnologia que possibilitasse ao aluno de cursos de bacharel em enfermagem aguçar seus conhecimentos em semiologia e semiotécnica em enfermagem. Além disso, buscou-se avaliar a jogabilidade do Semio em Jogo®, assim, observou-se que a comunidade acadêmica considerou o jogo como satisfatório após as modificações tanto quanto antes delas.

ABSTRACT Objective: to develop and analyze content and face validity evidence of a board game to promote healthy lifestyle habits for patients with coronary artery disease. Method: this is a psychometric study of development and validity of a board game for health education. Data collection was carried out between October 2021 and January 2022. Initially, the game rules were described and, sequentially, themes related to cardiovascular prevention were identified, used as a basis for card development. The game was submitted to assessment by 14 experts in terms of clarity, theoretical relevance, practical relevance, image visibility and relationship between image and text, through a Likert scale score. The content validity coefficient was calculated for each round, with a coefficient greater than 0.57 being considered valid. New assessment rounds were performed until the appropriate coefficient was reached. Results: the board game was built containing images of anthropomorphized hearts alluding to healthy lifestyle habits and 45 spaces. A total of 69 cards were developed containing the themes of healthy eating, physical activity and the importance of smoking and alcohol cessation. In the first round of assessment, three cards did not reach the minimum content validity coefficient. They were reformulated and sent to a second round of assessment, and was considered validated in this new round. Conclusion: the game, its rules and its content showed adequate content validity evidence after the second round. Further studies to analyze its effect on lifestyle modification should be carried out.


RESUMEN Objetivo: desarrollar y analizar evidencias de validez de contenido y apariencia de un juego de mesa para promover hábitos de vida saludables en pacientes con enfermedad arterial coronaria. Método: estudio psicométrico de desarrollo y validación de un juego de mesa para educación en salud. Os dados se recopilaron desde octubre de 2021 hasta noviembre de 2022. Inicialmente, se describieron las reglas del juego y, en forma secuencial, se identificaron temas relacionados con la prevención cardiovascular, utilizado como base para el desarrollo de gráficos. El juego fue sometido a la evaluación de 14 especialistas en términos de claridad, pertinencia teórica, pertinencia práctica, nitidez de las imágenes y relación entre la imagen y el texto, a través de la puntuación de la escala Likert. Se calculó el coeficiente de validez de contenido para cada ronda, considerándose válido un coeficiente superior a 0,57. Se realizaron nuevas rondas de evaluación hasta alcanzar el coeficiente adecuado. Resultados: se construyó el juego de mesa que contenía imágenes de corazones antropomorfizados alusivos a hábitos de vida saludables y 45 casas. Se desarrollaron 69 cartas que contenían los temas de alimentación saludable, actividad física y la importancia de dejar de fumar y de beber alcohol. En la primera ronda de evaluación, tres letras no alcanzaron el coeficiente mínimo de validez de contenido. Fueron reformulados y enviados a la segunda ronda de evaluación, considerándose validados en esta nueva ronda. Conclusión: el juego, sus reglas y su contenido mostraron evidencia adecuada de validez de contenido después de la segunda ronda. Se deben realizar más estudios para analizar su efecto en la modificación del estilo de vida.

Inicialmente, foram descritas as regras do jogo e sequencialmente foram identificados temas relacionados à prevenção cardiovascular, usados como base para o desenvolvimento de cartas. O jogo foi submetido à avaliação de 14 especialistas em relação à clareza, relevância teórica, pertinência prática, nitidez das imagens e relação da imagem com o texto, por meio da pontuação em escala de Likert. Foi calculado o coeficiente de validade de conteúdo para cada rodada, sendo considerado válido um coeficiente superior a 0,57. Novas rodadas de avaliação foram realizadas até que o coeficiente adequado fosse atingido. Resultados: o jogo de tabuleiro foi construído contendo imagens de corações antropomorfizados aludindo a hábitos saudáveis de vida e 45 casas. Foram desenvolvidas 69 cartas contendo as temáticas de alimentação saudável, atividade física e a importância da cessação do tabagismo e do álcool. Na primeira rodada de avaliação, três cartas não atingiram o coeficiente de validade de conteúdo mínimo. Foram reformuladas e enviadas para a segunda rodada de avaliação, sendo consideradas validadas nessa nova rodada. Conclusão: o jogo, suas regras e seu conteúdo apresentaram adequadas evidências de validade de conteúdo após a segunda rodada. Estudos posteriores para análise do seu efeito na modificação do estilo de vida devem ser realizados.

Abstract Background Sepsis and septic shock still represent great challenges in critical care medicine. Sildenafil has been largely used in the treatment of pulmonary arterial hypertension, but its effects in sepsis are unknown. The aim of this study was to investigate the hypothesis that sildenafil can attenuate endotoxin-induced pulmonary hypertension in a porcine model of endotoxemia. Methods Twenty pigs were randomly assigned to Control group (n = 10), which received saline solution; or to Sildenafil group (n = 10), which received sildenafil orally (100 mg). After 30 minutes, both groups were submitted to endotoxemia with intravenous bacterial lipopolysaccharide endotoxin (LPS) infusion (4 µg.kg-1.h-1) for 180 minutes. We evaluated hemodynamic and oxygenation functions, and also lung histology and plasma cytokine (TNFα, IL-1β, IL6, and IL10) and troponin I response. Results Significant hemodynamic alterations were observed after 30 minutes of LPS continuous infusion, mainly in pulmonary arterial pressure (from Baseline 19 ± 2 mmHg to LPS30 52 ± 4 mmHg, p< 0.05). There was also a significant decrease in PaO2/FiO2 (from Baseline 411 ± 29 to LPS180 334 ± 49, p< 0.05). Pulmonary arterial pressure was significantly lower in the Sildenafil group (35 ± 4 mmHg at LPS30, p< 0.05). The Sildenafil group also presented lower values of systemic arterial pressure. Sildenafil maintained oxygenation with higher PaO2/FiO2 and lower oxygen extraction rate than Control group but had no effect on intrapulmonary shunt. All cytokines and troponin increased after LPS infusion in both groups similarly. Conclusion Sildenafil attenuated endotoxin-induced pulmonary hypertension preserving the correct heart function without improving lung lesions or inflammation.

Abstract Introduction: Treatments of Inflammatory Bowel Disease (IBD) are able to control symptoms in most cases, however, a fraction of patients do not improve or have a loss of response to treatments, making it important to explore new therapeutic strategies. Hyperbaric oxygen therapy (HBO) may represent one of them. The aim of this study was to evaluate the effects of HBO therapy in an experimental model of IBD. Methods: Sixty male BALBc mice were divided into six groups. Group 1 was colitis-induced with trinitrobenzene sulfonic acid (TNBS) + ethanol, group 2 received TNBS + ethanol plus HBO, group 3 received only ethanol, group 4 received ethanol plus HBO, group 5 received saline solution, and group 6 received saline solution plus HBO. HBO was performed for four days, subsequently, the mice were evaluated daily. At the end of the study, samples from the intestine were collected for histological analysis as well as for measurement of antioxidant enzymes and cytokine levels. Results: HBO significantly improved the clinical and histological status of the animals. Treatment with HBO increased the activity of the antioxidant enzymes superoxide dismutase (SOD) and glutathione peroxidase (GPx) in all of the groups; moreover, the difference was only significant between the TNBS and TNBS + HBO groups and treatments promoted a reduction in the proinflammatory cytokines IFN-γ, IL-12, IL-17 and TNF-α and increased the anti-inflammatory cytokines IL-4 and IL-10, with no changes in IL-13. Conclusion: HBO effectively treats TNBS-induced colitis by increasing the activity of antioxidant enzymes and modulating cytokine profiles.

RESUMO A capacidade tática nos jogos esportivos coletivos se baseia na relação com os conhecimentos que o praticante possui por meio de suas experiências anteriores, e a escola deve oportunizá-las, a fim de promover a formação geral dos alunos. O objetivo do estudo foi avaliar os impactos no conhecimento tático processual (CTP) de escolares em formação esportiva geral após um programa híbrido de ensino. Participaram 40 escolares (20 no grupo experimental-GE e 20 no grupo controle-GC) com idades entre 8 e 10 anos (7,85 ± 0,48), de ambos os sexos. Aplicou-se o Teste de Conhecimento Tático Processual: Orientação Esportiva para avaliação do CTP antes e após 17 sessões de aulas com um programa híbrido de ensino que utilizou conteúdos das propostas metodológicas da Iniciação Esportiva Universal (IEU) e da Escola da Bola (EB). A intervenção no GE provocou melhora significativa no CTP dos escolares para as ações em ataque e defesa com mãos e pés quando comparado ao CG com tamanho de efeito grande. Conclui-se que a utilização desse programa híbrido de ensino (IEU + EB) aplicado nas aulas de educação física (EF) foi capaz de melhorar o CTP de escolares.


ABSTRACT The tactical capacity in team sports is based on the relationship with the knowledge that the practitioner has through his previous experiences, and the school must provide them in order to promote the general formation of the students. The aim of this study was to evaluate the impacts on the procedural tactical knowledge (PTK) of students in general sports formation after a hybrid teaching program. Participants were 40 students (20 in the experimental group-EG and 20 in the control group-CG) aged between 8 and 10 years old (7.85 ± 0.48), of both sexes. The PTK test: Sports Orientation was applied to evaluate the PTK before and after 17 sessions of school classes with a hybrid teaching program that used contents of the methodological proposals of the Universal Sports Initiation (USI) and the Ball School (BS). The intervention in the EG caused a significant improvement in the PTK of the students for the actions in attack and defense with hands and feet when compared to the GC with large effect size. It is concluded that the use of this hybrid teaching program (USI + BS) applied in physical education classes was able to improve the PTK of students.

Resumo Objetivo Avaliar a efetividade do jogo de tabuleiro Enfermeiro Diagnosticador para o ensino do raciocínio diagnóstico em estudantes de enfermagem. Métodos Estudo quase-experimental, desenvolvido em três etapas: construção do jogo de tabuleiro Enfermeiro Diagnosticador; análise de conteúdo dos casos clínicos por especialistas; e aplicação do jogo de tabuleiro Enfermeiro Diagnosticador. Participaram 19 especialistas na análise de conteúdo. O pré-teste e o pós-teste foram realizados com 11 discentes do Curso de Graduação em Enfermagem, cursando a Disciplina de Bases da Semiologia. Resultados Os nove casos clínicos que compuseram o jogo foram organizados em pistas capazes de subsidiar a composição dos diagnósticos de enfermagem. Esses foram avaliados com boa adequação por 19 especialistas em raciocínio diagnóstico e/ou tecnologia educacional. Comparando o desempenho dos discentes antes e após o experimento, a inferência diagnóstica correta apresentou diferença estatística significativa na resolução do primeiro e segundo diagnósticos (p=0,008). Além disso, a escrita correta do diagnóstico foi ponto de destaque no pós-teste e todos os itens avaliados apresentaram melhora no momento pós-aplicação do jogo de tabuleiro. Conclusão Conclui-se que o jogo de tabuleiro Enfermeiro Diagnosticador é efetivo no apoio ao ensino do raciocínio diagnóstico em enfermagem. Desta forma, almeja-se avançar no estado da arte no que concerne a jogos educativos de baixo custo e fácil acesso para dar suporte ao raciocínio diagnóstico em enfermagem.


Resumen Objetivo Evaluar la efectividad del juego Enfermero Diagnosticador para la enseñanza del razonamiento diagnóstico en estudiantes de enfermería. Métodos Estudio cuasi experimental desarrollado en tres etapas: elaboración del juego de mesa Enfermero Diagnosticador, análisis de contenido de los casos clínicos por especialistas y aplicación del juego de mesa Enfermero Diagnosticador. Participaron 19 especialistas en el análisis de contenido. El pretest y el postest se realizó con 11 estudiantes de la carrera de Enfermería que cursaban la materia Bases de la Semiología. Resultados Los nueve casos clínicos que formaban parte del juego fueron organizados en pistas que podían respaldar la composición de los diagnósticos de enfermería. Estos casos fueron evaluados con buena idoneidad por 19 especialistas en razonamiento diagnóstico o tecnología educativa. Al comparar el rendimiento de los estudiantes antes y después del experimento, la inferencia diagnóstica correcta presentó diferencia estadística significativa en la resolución del primer y segundo diagnóstico (p=0,008). Además, la escritura correcta del diagnóstico fue un punto destacado en el postest, y todos los ítems evaluados presentaron una mejora en el momento posterior a la aplicación del juego de mesa. Conclusión Se concluye que el juego de mesa Enfermero Diagnosticador es efectivo para respaldar la enseñanza del razonamiento diagnóstico en enfermería. De esta forma, se pretende avanzar en el estado del diseño en lo que respecta a juegos educativos de bajo costo y fácil acceso para respaldar el razonamiento diagnóstico en enfermería.


Abstract Objective To assess the "Enfermeiro Diagnosticador" board game effectiveness for teaching diagnostic reasoning among nursing students. Methods This is a quasi-experimental study, developed in three stages: "Enfermeiro Diagnosticador" (Diagnostician Nurse) game construction; content analysis of clinical cases by experts; and "Enfermeiro Diagnosticador" game application. Nineteen experts participated in content analysis. Pre- and post-tests were performed with 11 undergraduate nursing students, attending semiology, a baseline subject. Results The nine clinical cases that comprised the game were organized into clues capable of subsidizing the composition of nursing diagnoses. These were assessed with good suitability by 19 experts in diagnostic reasoning and/or educational technology. Comparing students' performance before and after the experiment, the correct diagnostic inference showed a statistically significant difference in the resolution of the first and second diagnoses (p=0.008). Moreover, the correct writing of diagnosis was a highlight in the post-test and all the items assessed showed improvement after the board game application. Conclusion It is concluded that "Enfermeiro Diagnosticador" is effective in supporting the teaching of diagnostic reasoning in nursing. In this way, we aim to advance in the state of the art with regard to low-cost and easily accessible educational games to support diagnostic reasoning in nursing.
