ABSTRACT

Background & objectives: WNIN/Ob (obese and euglycaemic) and WNIN/GR-Ob (obesity with impaired glucose tolerance), were isolated and established from Wistar rat stock (WNIN). Both strains showed physical, physiological and biochemical indices related to obesity. We present here haematology, histology and pathophysiological changes between the phenotypes of these strains, lean (+/+), carrier (+/-) and obese (-/-). Methods: A total of 72 animals of equal gender consisting of three phenotypes were used for the study. Haematology, organ weights were measured and histopathology of the tissues studied using standard procedures. In 12 lean and obese rats (equal gender) of WNIN/GR-Ob group morphometry of pancreatic islets was done immunohistochemically (IHC). Results: Obese rats of both the strains showed normal haematology (except low platelet count), but exhibited changes in the organ weights and in histopathology in organs like liver, kidney, brain and testis/ovary. Hyperplasia of adipocytes was seen in obese rats as compared to lean and carrier. IHC of obese rat pancreas showed that both islet density and volume were significantly (P<0.05) increased compared to lean littermates. Interpretation & conclusions: The histological and pathophysiological changes seen in these mutants were in tune with obese phenotype exhibited by these animals.

ABSTRACT

Nevus sebaceous (NS) is a type of classical nevus or congenital malformation that is often present at birth and commonly involves the scalp or face. The lesion usually presents as a linear, yellow, hairless, and verrucous plaque. It has been well-established that several benign and malignant tumors can develop from the NS; however, there have been no reports about ectopic fat cells in the dermis, and cornoid lamella arising from the NS. We report a case of NS on the scalp with accompanying unusual histopathologic findings.

ABSTRACT

Objective To explore the effect of Feier Recuperates Fluid on lipid metablism, fat weight, fatty cell shape. Method Trophism fat rat model was induced by feeding hyperalimentation feedstuff for six weeks in infancy rat. Drugs were administered orally after two week for four weeks. The cholesterol total (TC), triglyceride total (TG) and high-density lipoprotein-cholesterol (HDL-C) contents in the serum were determined. Fat weight was weighted. Number and diameter of fat cells were observed. Result Feier Recuperates Fluid (63.0 g/kg) could reduce fat weight, TC and TG contents, and increase HDL-C. Feier Recuperates Fluid could decrease diameter of fat cells, and increase fat cells number slightly. Conclusion Feier Recuperates Fluid could improve markedly lipid metablism of trophism fat rat.

ABSTRACT

PURPOSE: In the autologous fat injection, the centrifugation is useful for the refinement of harvested fat. As it can be an injury to the fat cell, we studied the fat cell viability with the change of centrifugation velocity and centrifugation time in order to get the limits of centrifugation velocity and centrifugation time. METHODS: We used the Colman System in 8 patients. We handled the control group with no centrifugation, group I with the centrifugation with 1500rpm for 1 minute, group II with 1500 rpm for 3 minutes, group III with 1500rpm for 5 minutes, group IV with 3000rpm for 1 minute, group V with 3000rpm for 3 minutes, group VI with 3000rpm for 5 minutes, group VII with 5000rpm for 1 minute, group VIII with 5000rpm for 3 minutes, group IX with 5000rpm for 5 minutes. We used the collagenase to separate the fat tissue. We had evaluated the fat cell viability by checking survival cell counts. RESULTS: There was no significance in group I, II, IV, V, but there was significant difference in group III, VI, VII, VIII, IX. CONCLUSION: The centrifugation with 3000rpm for 3 minutes is recommendable.

ABSTRACT

Objective To explore genes expression of adiponectin receptors during differentiation of SW872 preadipocytes. Met-(hods) SW872 preadipocytes were cultured in vitro and induced to differentiate by 0.6 mmol/L oleic acid. During the progress of diffe-(rentiation), the morphological changes of SW872 cells were observed and the differentiation rate was assayed by oil-red O staining. Adiponectin receptors mRNA was measured by reverse transcription-polymerase chain reaction during differentiation of SW872 preadipocytes. Results 1.After stimulated by 0.6 mmol/L oleic acid for 72 hours, almost all SW872 cells were differentiated,and there were lots of fat droplets in the cells.2.There were adiponectin receptors genes expressions in SW872 preadipocytes.After 72 hours,and the levels of adiponectin receptor(AdipoR) 1 mRNA and AdipoR 2 mRNA were markedly increased up to 2.54 and 4.09 times,respectively. Conclusion There are AdipoR1 and AdipoR2 genes expressions in fat cells and the expressions are differentiation-dependent.

ABSTRACT

The purpose of this study was to investigate the effects of wax gourd on weight, triglyceride, leptin and fat cell size in rats fed a high-fat diet. Male Sprague Dawley rats were fed an experimental diet containing total dietary fat at 40% of calories with wax gourd 0%, 5%, 10% and 15% (w/w) for 4 weeks. Weight gain and triglyceride level fell significantly in the 15% wax gourd group compared to the control group. Epididymal fat pad, abdominal fat and perirenal fat tended to decrease in the 15% wax gourd group. Leptin and free fatty acid level were not significantly different among the groups. Fat cell size significantly decreased in the 10% and 15% wax gourd groups compared to the control group. Weight gain correlated positively with visceral fat masses and the levels of leptin and triglyceride. Fat cell size significantly correlated with visceral fat and leptin level. Therefore, the 15% wax gourd diet substantially reduced weight, triglyceride and fat cell size.
