ABSTRACT

Objective To analyze the effect of non -xanthine adenosine receptor antagonist (SLV320)on the ventricular remodeling and renin -angiotensin -aldosterone system (RAAS)in animal experimental models of chronic heart failure (CHF).Methods The 40 healthy male New Zealand rabbits were received adriamycin by intra-venous injection to establishing the experimental animal models and were randomly divided into 4 groups,which were high -dosage group (injected with SLV320,10.0μg·kg -1 ·d -1 ),medium -dosage group (injected with SLV320, 5.0μg·kg -1 ·d -1 ),low -dosage group (injected with SLV320,2.5μg·kg -1 ·d -1 )and furosemide group (fed with furosemide,2.0mg·kg -1 ·d -1 ).Each group had 10 rabbits and continuous treated for one week.The indexes of plasma renin activity (PRA),angiotensinⅡ (AngⅡ),aldosterone (ALD)and beta ntriuretic peptide (BNP)were detected at pre -and post -treatment,and compared among 4 groups.The indexes of left ventricular end -systolic dimension (LVESD),left ventricular end -diastolic dimension (LVEDD),left ventricular posterior wall (LVPW), left ventricle ejection fraction (LVEF),left ventricular fractional shortening (LVFS)and E /A at pre -and post -treatment were detected by echocardiography and compared among 4 groups.The wet weigh of the left and right ventri-cle were weigh accurately.And the indexes of left ventricle weight index (LVWI)and the body weight index (BWI) were calculated and compared among 4 groups.Results The plasma levels of PRA,AngⅡ,ALD and BNP were no different among 4 groups before study (all P >0.05).The sequence of 4 groups on the plasma levels of RAAS indexes was high -dosage group 0.05).The sequence of 4 groups on the levels of LVEF and LVFS was high -dosage group >medium -dosage group >low -dosage group >furosemide group,and the sequences of LVESD,LVEDD,LVPW and E /A were high -dosage group

ABSTRACT

Objective To explore the relationship among genetic polymorphism of angiotension Ⅱ type 1 re-ceptor(AT1 R) A1166-C, angiotensin converting enzyme (ACE) insertion/deletion (I/D), aldosterone synthase (CYP11B2)-344T/C and hypertensive disorder complicating pregnancy.Methods Polymerase chain reaction-re-striction fragment length polymorphism (PCR-RFLP) assay was used to detect the genotypes of AT1 R A1166-C ,ACE (I/O) ,CYP11B2 -344T/C in 86 cases of hypertensive disorder complicating pregnancy and 175 cases of normal control.Results There was 18 combined types in hypertensive disorder complicating pregnancy cases and normal control cases.Compared to AT1R-AA + ACE-Ⅱ + CYP11B2-TT, Odds ratios (OR) of AT1R-AA + ACE-DO +CYP11B2-TC,AT1 R-AC + ACE-ID+CYP11B2-TC and AT1R-AC+ACE-DD+CYP11B2-TC are 7.289,5.315 and 5.694 respectively.There was no statistical significance among the others.Conclusion In all 18 kinds of combined types, AT1 R-AA + ACE-DO + CYP11B2-TC,AT1R-AC+ACE-ID+CYP11B2-TC and AT1 R-AC + ACE-DD +CYP11B2-TC might increase the susceptibility of hypertensive disorder complicating pregnancy.It is possible that multigenes are interacted in the etiology of hypertensive disorder complicating pregnancy.

ABSTRACT

Background: Prenatal exposure to Dutch famine was associated with glucose intolerance in adults. There is a line of evidence implicating the involvement of renin-angiotensin system in fetal development, but it is not known whether a genetic variant of reninangiotensin system may modify the long-term outcome in offspring born to mothers exposed to famine. Method: To examine whether an Insertion/Deletion (I/D) polymorphism in the angiotensin-converting enzyme (ACE) gene is associated with glucose intolerance in subjects born during the famine in the Pacific War, 8,657 healthy men were studied. ACE genotypes were determined using polymerase chain reaction. Subjects with fasting glucose more than 6.1 mmol/l or those diagnosed as diabetes mellitus or current user of oral hypoglycemic agents or insulin were considered as glucose intolerance. Result: Among overall subjects, logistic regression analysis did not show a significant risk of ACE DD genotype for glucose intolerance (men OR 1.03; 95% CI, 0.88-1.21), but showed 42% increased risk in men (adjusted OR: 1.42; 95% CI, 1.04-1.93, p=0.019), born during the war period, between 1941 and 1944. Conclusion: A genetic variant of renin-angiotensin system may modify the glucose intolerance as the long-term outcome in offspring born during the war-induced famine. Finding provides an evidence for the interaction of fetal ACE genotype and intrauterine environmental factor in predisposition to type 2 diabetes mellitus.

ABSTRACT

The renin-angiotensin system plays an important role in the progression of chronic renal diesases. At present, the main approach for treating the chronic progressive renal disease is to block the role of renin-angiotensin system. Recently, angiotensin receptor blockers have being realized in width with its few side effect and good safety. A number of experimental and clinical resultsin recent 10 years have indicated that ARB can prevent or even reverse the progression of chronic renal diseases in many ways effectively and provided great evidences for current therapy of CPRD.
