ABSTRACT

Objective To explore the expression and meaning of heat shock protein 90 (Hsp90) in pancreatic cancer and its correlation with the occurrence,development and metastasis of the tumor tissues and its influence on prognosis. Methods Via immunohistochemical staining, this study used the commercial tissue chip to determine the expression of Hsp90 in 63 pancreatic cancer resection specimens and their para-cancerous counterparts, which were pathologically confirmed from January 2009 to August 2013. We also analyzed the correlation with patient's clinical parameters by using one-way ANOVA or t test. Results Hsp90 expression in pancreatic cancer tissues was significantly higher than tissue adjacent to carcinoma, and it was associated with histological grade, lymph node metastasis,clinical and TNM phase of pancreatic cancer (P<0.05). Conclusions Hsp90 is highly expressed in pancreatic cancer tissues,and its expression is associated with invasion and metastasis of pancreatic cancer. There-fore,Hsp90 may serves as a prognostic indicator of pancreatic cancer while its high expression may represent poor prognosis of pancreatic cancer patients.

ABSTRACT

Objective:To investigate effects of depside salt from salvia miltiorrhiza(DSSM)on the dental pulp of rats under repeated exposure to +Gz acceleration.Methods:27 male SD rats were randomly divided into 9 groups(n =3).The rats in negative control group was only fixed with rats kept-keeling device,in positive control group were exposed to +5 Gz and +10 Gz respectively for 30 s each time,5 times per day,4 days per week,for 3 consecutive weeks.The rats in other groups were respectively given DSSMat 2,6 and 18 mg/kg respectively 30 min before each +Gz exposure.The rats in negative and positive control groups were injected with 0.9%sodium chloride solution 30 min prior to +Gz exposure.After the experiment,The pulp changes were observed by HE staining.Real time qPCR was applied to determine the relative expression of heat sheck protein(HSP70)mRNA.Results:+Gz exposure decreased the activity degree of the rats,and resulted in damage of rat dental pulp.With the increase of +Gz value,the relative expression of HSP70 mRNA was increased(P <0.05).HSP70 mRNA expression in drug groups was less than that in their own positive control groups,but showed statistical significance only in the 18 mg/kg and +10 Gz group(P <0.05).Conclusion:+Gz exposure may dam-age dental pulp.DSSMhas certain protective effect under +Gz exposure.

ABSTRACT

OBJECTIVE: To compare the distribution and expression of hsp60 and hsp27 in placental trophoblast between preterm and term placenta and to observe hsp immune response in relation to the pathogenesis of preterm birth. METHODS: 22 cases of preterm trophoblast, between 24 weeks and 36 weeks gestation, which were developed spontaneous onset or less than 24 hours after rupture of membrane were obtained. And aged-matched, 22 cases of normal term trophoblast, as control were also obtained after informed consent from each patient. The protein extraction form trophoblast was stained by immunohistochemical methods and was measured by the assay of Western blots. And the density of band using Image-writer were taken and statistical assay were performed as significance <0.05. RESULTS: The expressions of hsp60 and hsp27 in trophoblast of preterm and term placenta were identified by immunohistochemical staining method. The hsp60 had significantly higher expression in trophoblast of preterm birth than in that of term birth (P<0.001) and the hsp27 also had significantly higher expression in trophoblast of preterm birth than in that of term birth (P<0.02) CONCLUSION: The higher expression of hsp60 and hsp27 in trophoblast of preterm birth might be suggested the development of immune response to occur preterm labor Further study are necessary to determine the exact actions of hsp60 and27 in trophoblast and to understand the immune mechanism of preterm birth.

ABSTRACT

HSP70 is a kind of stress protein that takes care of protein through its life, and it has attracted attention as a factor to promote health. This protein is known for its induction route through hyperthermia stimulation. We compared differences between the effects of carbonic warm water bathing and tap warm water bathing, and the report obtained interesting results. The subjects are six healthy adults (average age: 23.8±5.5 years, each three from males and females), applied full immersion bathing at 41°C for 10min in both ca. 1, 000ppm of high concentration CO<sub>2</sub> warm water and tap warm water, and compared HSP70 before the bathing and one day after the bathing. During the observation of 24h, external thermal stimulation such as warm bathing was banned. 3 persons took warm bathing in CO<sub>2</sub> water first and the other 3 persons took tap water first. There was 10 days interval between the bathing in both types of bathing.<br>The results showed that an increase in precordial temperature measured with a deep-body thermometer was 1.0°C in tap warm water bathing and 2.3°C in CO<sub>2</sub> warm water bathing. The change in HSP70 was 3.31→4.35 (AU/mg protein: p=0.08) in tap warm water bathing and 3.42→5.04 (p<0.05) in CO<sub>2</sub> warm water bathing. Although a slight increase was recognized in tap warm water bathing, a significant increase in HSP70 was recognized in CO<sub>2</sub> warm water bathing.

ABSTRACT

PURPOSE: The purposes of study were to assess the expression patterns of heat shock protein (HSP) after glutamine and glutamine with non- lethal heat shock treatment, to evaluate the protective effects of heat shock protein from apoptosis in cultured human corneal epithelial cell. METHODS: The cultured human corneal epithelial cells were divided into two group. One group was treated with 0, 10, 20, 30, 40, 50 mM of glutamine and the other group was exposed to 43 degrees C (heat shock) for 30 minutes with same concentration of glutamine. After glutamine and heat treatment, the expression patterns of Hsp 27, 70 were examined by western blot and immunohistochemistry. Apoptosis was induced with 80uM of etoposide. The viability (cell protection rate of heat shock protein) against apoptosis after etoposide treatment was measured by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay. RESULTS: Expression of Heat shock protein 70 was not significantly effected in only glutamine treatment, but was remarkably increased in heat shock with glutamine treatment group. The increased cell number (viability= antiapoptotic effect of heat shock protein)of glutamine with heat shock group after etoposide treatment suggested that Hsp 70 appeared to be a major role in protection of Human corneal epithelial cell from apoptosis. The expression of Heat shock protein 27 was not effected in only glutamine and heat with glutamine treatment group. CONCLUSIONS: These data suggest that induced heat shock protein protect etoposide-generated apoptosis in human corneal epithelial cell.

ABSTRACT

This study was designed to evaluate the expression of heat shock protein in tooth and surrounding tissue during the experimental movement of rat incisors, by LSAB(labelled streptavidine biotin) immunohistochemical staining for heat shock protein. Twenty seven Sprague-Dawley rats were divided into a control group(3 rats), and 6 experimental groups(24 rats), to which 75g of force was applied from helical springs across the maxillary incisors. Rats of experimental groups were sacrificed at 0.5, 1, 4, 7, 14 and 28 days after force application, respectively. And the periodontal tissues of a control group and experimental groups were studied immunohistochemically. The results were as follows : 1. In control group, the expression of HSP47 was rare in gingiva, dentin and cementum, and mild in periodontal ligament and alveolar bone. But it was more evident than that of HSP70. 2. The expression of HSP47 or HSP70 was rare or mild in dentin, cementum and odontoblast of experimental group, regardless of the duration of force application, which was not different from that of control group. 3. In experimental group, the expression of HSP47 got to the highest degree in periodontal ligament and alveolar bone at 4 days after force application, and then decreased. And the expression was more evident in the pressure side than in the tension side of periodontal ligament 4. The expression of HSP70 began to increase at 12 hours after force application and got to the highest degree at 4 days, in the capillary of pulp and periodontal ligament. And the expression was more evident in the pressure side than in the tension side of periodontal ligament 5. The expression of HSP70 in alveolar bone of experimental group was rare, which was similar to that of control group.

ABSTRACT

PURPOSE: To evaluate the relationships between the expression of HSP 70 and prognostic factors, including T-stage, lymph node metastasis, distant metastasis, stage, differentiation and survival rate in colorectal cancer. METHODS: We studied 62 patients with colorectal cancer in order to evaluate the relationships between the expression of HSP 70 and multiple prognostic factors, including T-stage, lymph node metastasis, distant metastasis, stage and differentiation. RESULTS: There was no statistically significant correlation among HSP 70, T-stage, lymph node metastasis, distant metastasis, stage and differentiation. However, in our study, the expression of HSP 70 was slightly elevated in case with no lymph node metastasis, a lower stage and well differentiated colorectal cancer. In regards to survival, the overall survival rate was lower in the group with positive HSP 70 vice the negative group, and this was statistically significant. CONCLUSION: HSP 70 expression and the prognostic factors of colorectal cancer appear not to be correlated to each other, although HSP 70 may play a certain role in colorectal cancer. Further studies are required for determining the prognostic factors in colorectal cancer.

ABSTRACT

BACKGROUND: An accurate and rapid method for specise identification of coagulase negative staphylococci(CNS) has been increasingly necessary for the clinical significance and planning the management of patients with staphylococcal infections. Recently, it has been reported that there is a highly conserved area on their 60KDa heat shock protein(HSP60) gene sequences between the interspecies of CNS and it can be amplified by a set of universal degenerate primer. This led us our attention to focus on whether the PCR-based RFLP method using Mse / restriction enzyme could be a useful tool for the species identification of CNS. METHODS: In the present study, we performed PCR-based RFLP analysis using a set of degenerate primers covering HSP60 and Mse / restriction enzyme on the reference strains and 25 clinical isolates(10 of S. epidermidis, 10 of S. haemolyticus, 4 of S. lugdunensis and 1 of S. warneri) which were previously identified by the API-STAPH, Vitek GPI card and/or with conventional biochemical test. RESULT: All the seven reference strains revealed that each strain has a distinct electrophoresed band patterns with combination of different number (up to 8) and size of fragments. And these distinct band patterns showed remarkable concordance with the seven reference strains and 25 clinical isolates. CONCLUSION: These result strongly suggest that the PCR-RFLP method using degenerate primers covering the HSP60 gene and Mse / digestion enzyme offer a convenient and accurate tool for species-specific identification of CNS.

ABSTRACT

To determine whether fiber type-specific expression of heat shock protein (HSP, or stress protein) occurs in unstressed rat skeletal muscle, the medial gastrocnemius of adult female Sprague-Dawley rats was subjected to immunohistochemical analysis. Antibodies against 5 types of anti-myosin heavy chain (MHC) were used to classify the type of fibers, and 2 types of anti-HSP antibodies were employed to analyze the fiber type-specific expression.<BR>Serial cross-sections of 10 μm thick cut by a cryostat were incubated with primary anti-MHC or anti-HSP 60 and 72 antibodies, followed by biotinylated secondary anti-mouse antibodies, and avidin-biotin complex solution. A peroxidase DAB substrate kit (Vector SK-4100) or BCIP/NBT solution was used to visualize the immunoreaction of each fiber type.<BR>By using the 5 types of anti-MHC antibodies, fibers were classified into 4 types : slow-type I, fasttypes IIA, IIX, and IIB. Anti-HSP 72 antibody reacted with many, but not all, type I and IIA fibers, whereas anti-HSP 60 antibody reacted specifically with type I fibers. Neither type IIX nor IIB fibers showed immunoreactivity with anti-HSP 60 or 72 antibodies. These results suggest that the expression of HSP 60 protein is related to that of type I MHC, and that the expression of HSP 72 protein may be related to that of types I and ha MHC, in unstressed rat skeletal muscle.

ABSTRACT

Behcet's disease is recognized as a systemic inflammatory disease of unknown etiology. The disease has a chronic course with periodic exacerbations and progressive deterioration. Previous reports have shown at least three major pathophysiologic changes in Behcet's disease; excessive functions of neutrophils, vasculitis with endothelial injuries, and autoimmune responses. Many reports suggested that immunological abnormalities and neutrophil hyperfunction may be involved in the etiology and the pathophysiology of this disease. HLA-B51 molecules by themselves may be responsible, in part, for neutrophil hyperfunction in Behcet's disease. T cells in this disease proliferated vigorously in response to a specific peptide of human heat shock protein (hsp) 60 in an antigen-specific fashion. T cells reactive with self-peptides produced Th1-like proinflammatory and/or inflammatory cytokines. This leads to tissue injury, possibly via delayed-type hypersensitivity reaction, macrophage activation, and activation and/or recruitment of neutrophils. These data shed new light on the autoimmune nature of Behcet's disease; molecular mimicry mechanisms may induce and/or exacerbate Behcet's disease by bacterial antigens that have activated T cells which are reactive with self-peptide(s) of hsp. This would lead to positive selection of autoreactive T cells in this disease.
