ABSTRACT

Objective To establish and evaluate a rat model presenting symptoms of arthritis-hypertension disease (AHD).Methods A total of forty healthy 5-6 week-old male SD rats were used in this study.Hypertension was induced by constriction of the left renal artery by two kidney one clip (2K1C) with a 0.25 mm silver clamp, and AHD model was established by injecting 0.1 mL complete Freund adjuvant to the left hind paw.Tail artery pressure was measured with a non-invasive blood pressure measurement system.The degree of swelling in the non-inflammatory joint of rats was measured with a paw volume measuring instrument, the arthritis index and incidence of inflammation were evaluated.The rats were sacrificed on the 35th day.The thoracic aorta, ankle joint and spleen tissues were examined by pathology using HE staining.Results The joint of AHD model rat was significantly swollen, extensive synovial cell hyperplasia, inflammatory cell infiltration, vascular pannus formation, and bone and cartilage destruction.The number of germinal centers in spleen was increased, and a large number of lymphocyte infiltration, diffuse proliferation of white pulp, and red pulp infiltration were present.The arthritis index, incidence of inflammation and histopathological scores of the joint and spleen were significantly higher than adjuvant arthritis (AA) rats;meanwhile, the blood pressure of AHD model rat was significantly increased, the thickness and cross-sectional area of thoracic aorta were significantly increased, while the lumen diameter was significantly reduced.The blood pressure and vascular injury were significantly increased or aggravated compared with the HT rats.Conclusions A rat model of arthritis-hypertension disease is successfully established by using complete Freund adjuvant intradermal injection to the footpad and surgery to narrow the left renal artery.

ABSTRACT

Objective To observe the clinical effect and safety of labetalol combined with magnesium sulfate in treating moderate and severe pregnancy-induced hypertension disease.Methods 78 cases with moderate and severe pregnancy-induced hypertension disease were divided into the control group and observation group according to random number table method,39 cases were in each group.The patients in the control group were given the magnesi-um sulfate only,and the patients in the observation group were given the treatment of labetalol combined with magnesi-um sulfate.After treatment,the average blood pressure,24h urine protein,maternal heart rate,fetus heart rate and pregnancy outcomes were detected.Results After treatment,the average blood pressure[(134.5 ±9.7) vs (85.7 ± 5.9)]mmHg of observation group was significantly lower than the control group(t=4.60,7.21,all P0.05);the termination of pregnancy(7.7%),placental abruption(0.0%),fetal distress(5.1%),incidence of neo-natal asphyxia(7.7%),postpartum hemorrhage(12.8%),low birth weight(12.8%) were significantly lower than the control group(33.3%,12.8%,20.5%,25.6%,33.3%,35.9%) (χ2 =7.86,5.34,4.13,4.52,4.62,5.64,all P<0.05);the total effective rate of observation group with 37 cases (94.9%) was significantly higher than the con-trol group with 31 cases (79.5%) (χ2 =4.13,P<0.05);the incidence of adverse reactions of observation group with 15.4%was significantly lower than the control group with 35.9%(χ2 =4.30,P<0.05).Conclusion Labe-talol combined with magnesium sulfate in treating moderate and severe pregnancy-induced hypertension disease could effectively reduce the average blood pressure,the 24h urine protein and improve the pregnancy outcomes,which was worthy of clinical popularization and application.

ABSTRACT

Objective To explore the corelations between trace protein in urine and early kidney damage in gestational hypertension disease.Methods 126 patients with gestational hypertension disease were selected as the observation group,and 100 cases of healthy pregnant women over the same period were selected as the control group.The urinary albumin content of pregnant women in the two groups was detected and compared.Results In the observation group,the content of urinary albumin was (34.86 ± 18.41) mg/L,and positive patients in 56 cases,negative in 70 cases.In the control group,the content of urinary albumin was (18.64 ± 16.45)mg/L,and positive patients in 12cases,negative in 88 cases.The differences of the urinary albumin content and positive rates between the two groups were statistically significant(t =6.89,x2 =27.90,all P < 0.05) ;The systolic blood pressure of urinary albumin-positive patients and negative patients in the observation groups were (155.6 ± 18.4) mm Hg,(146.3 ± 18.7)mm Hg,respectively,and the difference was statistically significant (t =2.79,P < 0.05).Conclusion Urinary albumin testing during pregnancy can effectively determine the existence of early renal damage in patients with gestational hypertension disease,and it can help to judge the severity of gestational hypertension disease.

ABSTRACT

@#ObjectiveTo observe the effect of Indapamide on the rehabilitation of cardiac function in patients with primary hypertension disease.Methods56 patients with primary hypertension disease were randomly divided into the Indapamide group and Hydrochlorothiazide group with 28 cases in each group, and treated with Indapamide and Hydrochlorothiazide respectively assisted with conventional non-drug treatment such as dietetic and weight control. The cardiac functions of patients in two groups were examined with echocardiography before and two months after treatment.ResultsAfter treatment, the left ventricular ejection fraction (LVEF) raised and relative wall thickness (RWT) declined in patients of two groups, but the effect of the Indapamide group was better than that of the Hydrochlorothiazide group ( P<0.05).ConclusionThe effect of Indapamide on cardiac function and structure is better than Hydrochlorothiazide.

ABSTRACT

AIM: To research the effect of serum from patients with blood stasis syndrome(BSS) associated with hypertension disease on vWF、TM、EPCR excreted by ECV-304 and Tanshinone Ⅱ_A's intervention. METHODS: Cultivated human umbilical vein endothelial cells(CRL-1730) were divided into four groups,according to different serum of patients with BSS associated with hypertension disease,BSS(model) group,non-blood stasis group,health group and control group.The incubation time was 24 h.Tanshinone Ⅱ_A was of different concentrations(40、20、10、5 ?g/mL),all Tanshinone Ⅱ_A groups' cells were incubated by patients' serum and Tanshinone Ⅱ_A for 24 h.vWF,TM and EPCR were assayed by enzyme-linked immunosorbent assay(ELISA). RESULTS: vWF,TM and EPCR secreted were higher in the model group,the non-blood stasis group and the health group than those in the control group;vWF,TM and EPCR secreted were higher in the model group,the non-blood stasis group than those in the health group;Thereinto,the difference was distinct between the model group and the health group(P
