ABSTRACT

Objective To investigate the expression and distribution of RTCB in various tissues of mice. Methods We detected the expression of RTCB by Western blotting and the location of the expression of RTCB in each tissue by immunohistochemistry. Results RTCB protein was all expressed in 17 tissues of mice. The relative expressions of oviduct, skeletal muscle and kidney were high, while the relative expressions of bladder and small intestine were low. In addition, the immunostaining of RTCB was different in different tissues or in different cells of the same tissue. The immunostaining was stronger in some specific cells of testis, epididymis, ovary, uterus, fallopian lube and small intestine. Conclusion RTCB may participate in the maintenance of reproductive function in mice.

ABSTRACT

Objective@#To explore the feasibility of application of multi-point puncture immunohistochemical to determine the invasion extent of the epidermis in Kaposiform hemangioendothelioma before surgery and choose the surgical method.@*Methods@#From July 2013 to April 2016, 17 patients with Kaposiform hemangioendothelioma were admitted to the People′s Hospital of Zhengzhou University, including 7 males and 10 females. The first consultation age was 75.60±31.55 days. Preoperative multi-point pathological puncture D2-40, CD31, CD34 immunohistochemical staining was to test the invasion extent of the epidermis in Kaposiform hemangioendothelioma and to determine whether undergo autologous skin grafting. Skin graft survival areas were recorded postoperatively. Platelet values were examined after 1 week, 4 weeks, 12 weeks and 24 weeks.@*Results@#In these 17 cases, 15 cases with epidermis and dermis unaffected was performed in situ grafting. 2 cases with the tumor invasion, superficial skin grafting were repaired. In these 15 cases of in situ grafting, platelet values returned to normal at 1 week after the operations, and platelet values were higher than 100×109/L at 4 weeks, 12 weeks and 24 weeks. Among them, 12 cases of epidermal survival area were greater than 90%, 2 cases were 70%-90%, and 1 cases was about 50%.@*Conclusions@#During the complete resection of Kaposiform hemangioendothelioma, the multi-puncture D2-40, CD31 and CD34 staining are used to determine invasion extent of the tumor to the skin, which can maximize the retention of normal tissue surface, reduce postoperative complications. This is a new multi-disciplinary surgical approach.

ABSTRACT

Objective To explore the radiological features of intracerebroventricular central neurocytoma and so as to improve the diagnosis of this disease. Methods Thirty patients with pathologically confirmed intra-cerebroventricular central neurocytoma in Nanfang Hospital in past 7 years were analyzed retrospectively for their clinical,radiological and pathological features combined with literatures review. Results Lesions of central neuro-cytoma located at the Monro's foramen areas,the anterior horns and bodies of lateral ventricles.Twenty eight of the patients underwent MRI(with 4 patients received MRS)and 11 patients underwent CT.All cases revealed unilateral or bilateral hydrocephalus of the lateral ventricles and ventriculomegaly.Plain CT revealed heterogeneous isodensity or slighly high density lesions with multiple cystic degeneration,necrosis and calcification while a few lesions(4 le-sions)showed hemorrhage.Heterogeneous isointense or mild hypointense were demonstrated on T1-weight MRI and heterogeneous isointense or slightly hyperintense on T2-weight MRI.Multiple cystic degeneration(always showed as distinctive "soap bubble sign"), necrosis and calcification could also be found within the tumor. The solid compo-nent of tumor revealed slightly high to high signal intensity on DWI while the cystic and necrosis components showed low signal intensity.Most of the tumors showed mild to moderate enhancement with vessels passing through on contrast scan. All cases with 1H MRS showed with obviously increased in Cho peak and decreased in Cr and NAA peaks.In immunohistochemical assay,most of the tumors revealed positive expressions of Neu-N,Synaptophy-sin, GFAP and hypo-level Ki-67. Conclusions The intracerebroventricular central neurocytoma has some typical imaging manifestations.Comprehensive analysis of CT and MRI can be conducive to preoperative diagnosis and dif-ferential diagnosis with other tumors in the lateral ventricle.

ABSTRACT

Purpose To investigate the expression of cy-toskeletal protein 4. 1N in Ewing's sarcoma and the correlation with cell proliferation as well as clinicopathological characteris-tics. Methods Immunohistochemical EnVision two-step stai- ning was used to detect the expression of 4. 1N in 52 Ewing's sarcoma cases, which 47 cases of them were performed to detect the expression of Ki-67. The relationship between the expression of 4. 1N and Ki-67, and the correlation with the pathological fea- tures including prognosis were analyzed. Results The positive expression of 4. 1N was found in 35% (18/52) of Ewing's sar-coma. Lower frequency of 4. 1N strong expression (1. 9% , 1/52) was detected in the cohort. Ki-67 labeling index in Ewing's sarcoma varied from 3% to 85% . 4. 1N expression displayed a negative correlation with Ki-67 index ( P <0. 05). Among the follow-up of 27 cases, Kaplan-Meier survival analysis suggested that patients with 4. 1N-negative had shorter survival period than patients with 4. 1N positive expression (P <0. 05). However, the expression of 4. 1N was not statistically correlated with sex, age, tumor's localization and metastasis. Conclusion 4. 1N mainly showed negative or lower expression in Ewing's sarcoma and was related to cell proliferation index Ki-67, which implied a potential function in regulating proliferation in Ewing's sarcoma. 4. 1N expression has potentially predictive value in outcome of E-wing's sarcoma.

ABSTRACT

Purpose To investigate the expression of pro-grammed death1 (PD-1), programmed death-ligand1 ( PD-L1) in gastric carcinoma and its correlation with clinicopathological characteristics and prognosis. Method Immunohistochemistry was used to dectect the expression of PD-1/PD-L1 in 75 cases of gastric cancer and adjacent tissues of cancer, without chemother-apy and radiotherapy from 2011 to 2016 in The Affiliated Jiangsu Shengze Hospital of Nanjing Medical University. To analyzed the relationship between expression of PD-1/PD-L1 in gastric carci-noma and clinicopathologic characteristics, and analyzed rela-tionship of overall survival time in all patients. Result The tumor cells expressed 37. 3% (28/75) PD-L1 positive rate in 75 cases, which associated with age, tumor diameter and differ-entiation. PD-1 positive rate in the tumor infiltrating lympho-cytes was 46. 7% (35/75), which associated with pTNM stag-ing. The positively rate of PD-L1 expression in tumor cells and PD-1 expression in the tumor infiltrating lymphocytes were sig-nificantly higher than that in the adjacent tissues of carcinoma (P<0. 001). Kaplan-Meier and Long-rank results showed the overall survival time was significantly related with tumor diame-ter, differentiation, invasion depth, metastasis and pTNM stag-ing ( P <0. 05), but unrelated with PD-1 and PD-L1 expres-sion. But if the follow-up time was 14 months, the prognosis of PD-L1 positive patients were significantly lower than that of the negative ones. Conclusion Expression of PD-1/PD-L1 in cancer cells are significantly higher than that in para-carcinoma tissue. Positive expression of PD-L1 in tumor cells may contrib-ute to evaluation of short-term prognosis, and has nothing to do with the overall survival rate.

Objective To explore the correlation between chemokine receptor 4(CXCR4) and vascular en-dothelial growth factor (VEGF) and ovarian cancer.Methods 59 cases of oophorectomy were selected from February 2014 to April 2015.According to the pathological results,the patients were divided into ovarian canc-er group(group A) with 32 cases and benign tumor group (group B) with 27 cases.Another 35 patients un-dergoing non ovarian surgery were selected as control group (group C).The expressions of CXCR4 and VEGF in ovarian tissues of each group were detected,and the correlation with ovarian cancer was analyzed.Results The expression levels of CXCR4 and VEGF in the three groups were in the turn of A,B and C,and the differ-ence was statistically significant (P<0.05).The positive rate of VEGF was higher in low differentiation canc-er tissues,and the difference was statistically significant (P<0.05);the positive rate of VEGF was relatively high in the cancer tissues of clinical stage Ⅲ and IV,and the difference was statistically significant (P< 0. 05);the positive rates of CXCR4 and VEGF were higher in ovarian cancer with peritoneal metastasis or lymph node metastasis,and the difference was statistically significant (P<0.05).The expression of CXCR4 in ovarian canc-er tissues was positively correlated with the expression of VEGF (P<0.05).Conclusion CXCR4 and VEGF were highly expressed in ovarian cancer tissues,and the two involved in the development,invasion and metastasis of tumors.

ABSTRACT Objective: To evaluate the diagnostic utility of the p16ink4a protein expression as a marker for adenocarcinoma of the cervix. Methods: In a cross-sectional study, p16ink4a expression was evaluated in 30 cervical biopsies from patients diagnosed with invasive adenocarcinoma from 2 reference clinics in Brazil, and compared with 18 biopsies of endocervical polyps (control cases). The performance of the tests for p16ink4a was evaluated using a conventional contingency table, and the Kappa (k) index was used to evaluate the agreement of the marker with the tissue diagnosis. Results: In total, 66% of the invasive adenocarcinoma cases were positive for p16ink4a. All of the adenomatous polyps cases used as negative controls were shown to be negative for p16ink4a. The marker showed a high sensitivity and a high negative predictive value. The Kappa index was good for p16ink4a (k 1/4 0.6). Conclusion: Considering the strong association between the p16ink4a marker and the cervical adenocarcinoma, its use represents an important tool for reducing incorrect diagnoses of adenocarcinoma and thereby avoiding overtreatment.


RESUMO Objetivo: Avaliar a utilidade diagnóstica da expressão da proteína p16ink4a como marcador de adenocarcinoma do colo. Métodos: Em estudo transversal, a expressão de p16ink4a foi avaliada em 30 biópsias cervicais de pacientes diagnosticadas com adenocarcinoma invasivo de colo uterino provenientes de dois serviços de referência no Brasil, comparando com achados em 18 biópsias de pólipos endocervicais (grupo de controle). Para avaliar a performance do teste, foi utilizada tabela de contingência convencional, e para avaliar a concordância com o diagnóstico, foi aplicado o índice de Kappa (k). Resultados: No total, 66% dos casos de adenocarcinoma invasivo foram positivos para p16ink4a. Todos os pólipos adenomatosos foram negativos para p16ink4a. O marcador mostrou uma alta sensibilidade e alto valor preditivo negativo. O índice de Kappa foi bom para p16ink4a (k 1/4 0.6). Conclusion: Considerando a forte associação entre o marcador p16ink4a e o adenocarcinoma cervical, seu uso representa uma ferramenta importante para reduzir o risco de diagnóstico incorreto de adenocarcinoma e, por conseguinte, evitar o excesso de tratamentos.

In this study, the expression of extra cellular matrix proteins was immunohisto chemically studied and compared with the histological grading of squamous cell carcinomas of the lower lip and tongue. Material and Methods: The lower lip carcinomas (n =12) and the tongue carcinomas (n = 12) were histopathologically graded according to Bryne’smethod. The immunohisto chemical technique used specific antibodies for collagen IV and laminin. Histopathological and immunohisto chemical analysis were carried-out at the invasive tumor front. Results: Most of the lower lip carcinomas(91.7%) were classified with lower scores and all tongue carcinomas (100%) with high-grade malignant scores (p < 0.01). Collagen type IV expression was absent in the peri tumoral basement membrane in 50% of lower lip carcinomas and in66. 7% of tongue carcinomas (p = 0.09). Laminin expression was absent in the peritumoral basement membrane in 66.7% of lower lip carcinomas and in 58.3% of tongue carcinomas (p = 0.48). When these two glycoproteins were expressed, they showed alinear, thin and discontinuous pattern and a weak intensity of expression. Conclusion: The high gradem alignancy score of the tongue carcinomas was associated with the pattern of expression of the matrix proteins studied. This suggested that tonguesquamous cell carcinomas have more invasive potential and more aggressive biological behaviorthan the lower lip carcinomas...


Neste estudo, a expressão das proteínas da matriz extracelular foi estuda da imunoisto quimicamente e comparada com a classificação histológica dos carcinomas de células escamosas do lábio inferior e língua. Material e Métodos: Os carcinomas de lábio inferior (n = 12)e os carcinomas de língua (n = 12) foram graduados histopatologicamente de acordo com o método de Bryne. A técnica de imunoistoquímica utilizou anticorpos específicos para colágeno IV e laminina. Análises histopatológica e imunoistoquímica foram conduzidas na frente invasiva tumoral. Resultados: A maioria dos carcinomas de lábio inferior (91,7%) foi classificada em baixo grau e todos os carcinomas de língua (100%) em alto grau de malignidade (p < 0,01). A expressão de colágeno tipo IV estava ausente na membrana basal peri tumoral em 50% dos carcinomas de lábio inferior e em 66,7% dos carcinomas de língua (p = 0,09). A expressão de laminina estava ausente na membrana basal peri tumoral em 66,7%dos carcinomas do lábio inferior e em 58,3% dos carcinomas de língua (p = 0,48). Quando estas duas glicoproteínas foram expressas, mostraram-se comum padrão linear, fino e descontínuo e uma fraca intensidade de expressão. Conclusão: O alto grau de malignidade dos carcinomas de língua associou-se com o padrão de expressão das proteínas de matriz estudadas. Isso sugere que carcinomas de células escamosas de língua têm comportamento biológico mais agressivo e potencial mais invasivo do que os carcinomas de lábio inferior...

ABSTRACT

Objective:To investigate the expression of Survivin and programmed cell death 5(PDCD5)protein in mucoepidermoid carcinoma(MEC).Methods:Survivin and PDCD5 were detected by immunohistochemical staining in 20 cases of normal parotid tis-sue(group A),40 cases of pleomorphic adenoma(group B)and 45 cases of MEC tissues(group C).Results:The positive expres-sion ratio of Survivin in group A,B and C was 10.0%,27.5% and 55.6% respectively(χ2 =14.556,P <0.01),while that of PD-CD5 was 85%,65% and 33.3% respectively (χ2 =17.439,P <0.01).The expression of survivin or PDCD5 was related with dif-ferentiation,lymph node metastasis and TNM staging of MEC.Survivin and PDCD5 showed a negative correlation in MEC(χ2 =4.500,P =0.034,γs =-0.316).Conclusion:Survivin over-expression and PDCD5 down-expression may play a role in the de-velopment and progress of mucoepidermoid carcinoma.

AIM: To investigate the significance of NAD (P)H-quinone oxidoreductase 1 (NQO1) protein overexpression for prognostic evaluation of ovarian mucinous cystadenocarcinoma .METHODS:NQO1 protein was detected in 162 cases of ovarian mucinous cystadenocarcinoma , 35 cases of ovarian mucinous cystadenoma and 29 samples of normal ovarian epithelial tissues by the method of EnVision immunohistochemical staining .The correlation between high expression of NQO1 protein and clinicopathological features of ovarian mucinous cystadenocarcinoma was also evaluated .Overall sur-vival and disease-free survival rates of ovarian mucinous cystadenocarcinoma patients were calculated by Kaplan -Meier method.RESULTS:The positive rate and strongly positive rate of NQO 1 protein were 85.8%and 64.2%in ovarian mu-cinous cystadenocarcinoma , respectively , which are significantly higher than those in ovarian mucinous cystadenoma , and normal ovarian epithelial tissues ( P <0.01 ) .NQO1 expression was significantly correlated with the histological grade (P<0.05) and clinical stage (P<0.01) of ovarian mucinous cystadenocarcinoma .Kaplan-Meier survival analysis showed that the overall survival rate and disease-free survival rate were significantly higher in ovarian mucinous cystadenocarcinoma patients with high NQO1 expression than those with low NQO1 expression (P<0.01).CONCLUSION:NQO1 expression is closely correlated with the progression and prognosis of the patients with ovarian mucinous cystadenocarcinoma .High ex-pression of NQO1 protein may be used as an important indicator for the patients with poor prognosis of ovarian mucinous cystadenocarcinoma .

ABSTRACT

Objective To investigate the expression levels and clinical significance of programmed cell death 1 ligand (PD-L1) and programmed cell death factor-1 (PD-1) in peripheral T-cell lymphoma (PTCL). Methods Immunohistochem?istry was used to detect expression levels of PD-L1 and PD-1 in PTCL (test group, n=51) and benign proliferative lesions of lymph node tissues (control group, n=20). The correlations of PD-L1 and PD-1 expressions with clinical pathological param?eters and prognosis were analyzed between two groups. Results The expression level of PD-L1 was significantly higher in PTCL group than that in control group (74.51%vs 35.00%,χ2=9.662, P<0.05). The positive expression of PD-1 was signifi?cantly higher in PTCL group than that in control group (66.67%vs 25.00%,χ2=10.074, P<0.05). There were significant dif?ferences in PD-L1 and PD-1 expressions between different peripheral lactate dehydrogenase (LDH) levels of PTCL group (P<0.05). After two cycles of CHOP or ECHOP treatments, the response rate (RR) was higher in PD-L1 negative group than that in positive group (84.6%vs 47.4%,χ2=5.478, P<0.05), and RR was higher in PD-1 negative group than that in positive group(82.4%vs 44.1%,χ2=6.755, P<0.05). The median overall survival (OS) time was higher in PD-L1 negative group than that in positive group (29.8 months vs 17.6 months,χ2=4.413, P<0.05) and the median OS time was higher in PD-1 negative group than that in positive group (29.8 months vs 17.6 months,χ2=8.293, P<0.05). Conclusion There are high expression levels of PD-L1 and PD-1 in peripheral T-cell lymphoma, which is closely related with the elevated LDH in peripheral blood, poor response rate and shorter OS. Therefore, the expression levels of PD-L1 and PD-1 can be used as factors of worse effect of chemotherapy and poor prognosis.

ABSTRACT

Antecedentes: la incidencia y mortalidad por cáncer de mama han presentado un aumento neto. Uno de los tipos de esta heterogénea enfermedad se caracteriza por la amplificación y alta expresión del gen ERBB2, que codifica el receptor tipo 2 del factor de crecimiento epidérmico humano (HER2). Este receptor estimula procesos oncogénicos y dichos tumores se asocian a un peor pronóstico. El objetivo del estudio fue analizar las características de tumores HER-2+ en un grupo de pacientes con carcinoma de mama. Métodos: se estudió los casos de mujeres con biopsia diagnóstica de cáncer de mama registrados durante 2006 en los archivos del Servicio de Anatomía Patológica del Hospital San Juan de Dios; entre los que contaban con estudios inmunohistoquímicos, se estudió la expresión de HER-2 y se realizó análisis estadísticos. Resultados: se halló 34 tumores HER-2+ (15,7%), 24 con expresión fuerte (11,1%) y 10 con débil (4,6%). La edad media al diagnóstico de las pacientes con expresión fuerte fue de 46,9 años (42,5-51,1 IC 95%), y con expresión débil fue de 54,4 años (46,7-62,1 IC 95%); la edad promedio para los tumores HER-2- fue de 58,1 años (56,2-60,0 IC 95%). Las pacientes <50 años tuvieron un OR=3,477 de tener HER-2+. De 21 tumores, el 90,5% presentó un grado histológico alto, y de 16 casos, únicamente 3 tenían un tamaño <2 cm. Conclusión: se encontró una asociación del tipo HER-2+ con pacientes jóvenes, tumores de mayor tamaño y alto grado histológico. Los datos encontrados en este primer reporte son similares a las últimas estimaciones mundiales.


Background: The incidence and mortality due to breast cancer has suffered a net increase. One type of this heterogeneous disease is characterized by an amplification and higher expression of the ERBB2 gene that codifies for human epidermal growth factor receptor type 2 (HER-2). This receptor mediates oncogenic processes and such tumors have been associated with a worse prognosis. The aim of this study was to analyze the characteristics of HER-2+ tumors in a group of patients with breast cancer. Methods: This study included cases of breast cancer diagnosed through biopsy, registered during 2006 in the archives of the Pathology Department of the San Juan de Dios Hospital. The HER-2 expression was studied and statistical analyses were performed in cases that had immunohistochemical studies. Results: Thirty-four HER-2+ tumors were found (15.7 %), 24 with a strong expression (11.1%) and 10 with a weak expression (4.6%). The average age at diagnosis of patients with a strong expression was 46.9 years (42.5-51.1 IC 95%) and in those with a weak expression, 54.4 years (46.7-62.1 IC 95%). In the case ofHER-2- tumors, de mean age at diagnosis was 58.1 years (56.2-60.0 IC 95%). Patients <50 years had an OR=3.477 of having HER-2+. Out of 21 tumors, 90.5% showed a high histologic grade and out of 16 cases, only 3 measured <2 cm. Conclusion: The data found is similar to the latest global statistics. An association was found between HER-2+ type tumor and young patients, larger size tumors and higher histologic grade.

ABSTRACT

Objective To investigate the expressions of GDNF and GFRα1 in childhood nephroblastoma, and therefore its clinical significance. Methods The expression levels of GDNF and GFRα1 were examined by immunohistochemistry of 30 parrafin samples from nephroblastoma section as well as 16 parrafin samples of peritumor renal tissue. Results The pos?itive expression rate of GDNF and GFRα1 in nephroblastoma were higher than those in normal renal tissue(66.7%vs 6.3%;63.3%vs 6.3%). GDNF and GFRα1 were strongly expressed in epithelium and embryo bud, but weakly expressed in lobus intermedius of nephroblastoma. There was no correlation between the expressions of GDNF and GFRα1 with histological types, clinical stage and metastasis of nephroblastoma of patients. Conclusion High expression of GDNF and GFRα1 in childhood nephroblastoma might indicate its role in tumor development.

ABSTRACT

Purpose To explore the accuracy of ALK fused gene expression by immunohistochemistry ( IHC) in non-small cell lung cancer ( NSCLC) patients, and to investigate the clinical and pathological features of ALK-positive NSCLC patients. Methods By u-sing rabbit monoclonal D5F3 antibody, ALK IHC was performed on 234 NSCLC patients. ALK positive cases were confirmed by reverse transcription-polymerase chain reaction ( RT-PCR) . Results The positive incidence of ALK by IHC in 234 NSCLC specimens was 8. 97% (21/234), the positive rate of ALK fused gene verificated by RT-PCR was 5. 98% (14/234). There was significant difference with histological type, age, stage (P120, the consistency rate was 100%. Conclusion Although immunohistochemical expres-sion of ALK fused gene may have a certain false positive, IHC or immunohistochemical score> 120 show very high value for ALK fused gene RT-PCR followed by ALK immunohistochemistry in lung cancer is a economical and feasible method for the valuation of ALK fused gene.

ABSTRACT

ObjectiveTo investigate the effects of dopamine D1 receptor (D1R) tool drugs and combined acupuncture and medicine on striatal expressions of D1R and dopamine transporters (DAT) in middle cerebral artery occlusion (MCAO)-reperfusion rats.MethodForty-seven male SD rats were randomly grouped, used to make a model and given corresponding interventions. The materials were taken and fixed six hrs later. Striatal D1R and DAT expressions were detectedby an immunohistochemical method in different groups.ResultThe neurological deficit score was significantly higher in the model group than in the blank group. Electroacupuncture treatment decreased the score significantly (P0.05). DAT expression was significantly down-regulated in the other groups compared with the model group (P0.05).ConclusionCerebral ischemia-reperfusion can result in high D1R and DAT expressions in rat striatum on the ischemic side. Electroacupuncture, D1R antagonists and a combination of the two can significantly down-regulate D1R expression and have a protective effect on the brain. The effects of electroacupuncture and D1R antagonists can not be added to each other. D1R signaling pathway may be one of ways by which electroacupuncture produces a protective effecton the brain.

Objective To explore the expression of matrix metalloproteinase-14(MMP-14)protein in the human stomach carcinoma tissues and its correlation with carcinoma invasiveness and metastasis.Methods The MMP-14 protein expression was detected by immunohistochemistry in 59 cases of stomach carcinoma tissues (observation group)and 20 cases of normal stomach tissues (control group,the adjacent normal tissues from the tumor margin of 5 cm confirmed by pathology),and its correlation with the clinically pathological parameters was analyzed.The expression characteristics of MMP-14 among various TNM stages of stom-ach carcinoma were also analyzed.Results The positive rate of MMP-14 expression was 50.85%(30/59)in the observation group and 5.00% (1/20)in the control group,the positive rate of the observation group was significantly higher than that of the control group (P <0.01);the expression level of MMP-14 was correlated with the differentiation degree,regional lymph node metastasis degree,invasion depth,lymphatic invasion and TNM stage,which showing the statistical difference(P < 0.01);the expression of MMP-14 protein was up-regulated and showed the transferring trend from cytoplasm to cellular membrane along with the progres-sion of TNM stage.Conclusion The overexpression of MMP-14 protein exists in stomach carcinoma tissues,which contributing to the invasion and metastasis of stomach carcinoma cells.

ABSTRACT

Objective To investigate the effects of exercise on expressions of brain-derived neurotrophic factor (BDNF) and tyrosine kinase receptor B (TrkB) in spinal cord of spinal cord injury (SCI) rats. Methods Spinal cord injury models were produced by universal spinal cord impact system. Twenty-four Sprague-Dawley rats were randomly divided into 3 groups, exercise group (SCI-induction and exercises, n=8), control group (SCI-induction without exercises,n=8) and sham-operation group (no operation, without SCI nor exercises, n=8). Exercise training began from the 7th day after injury for 4 weeks. The locomotor function was assessed by Basso-Beattic-Bresnahan (BBB) scale before injury and at the 1st, 2nd, 3rd, 4th and 5th week post injury. The expressions level of BDNF and TrkB in spinal cords were detected by immunohistochemis-try and Western blot. Results Since the 3rd week after injury (after 2 weeks of exercise training), BBB scores in exercise group were higher than that in control group (P < 0.05). In immunohistochemistry test, positive immunologic reaction to BDNF protein was mainly located in anterior horn of spinal cord, and the expression level in exercise group was significantly higher than that in sham-operation group and control group (P<0.05). In immunohistochemistry test, positive immunologic reaction to TrkB protein was located in anterior horn and dorsal horn and central canal of spinal cord, and the expression lev-el in exercise group was significantly higher than that in sham-operation group and control group (P<0.05). Expression of BDNF and TrkB in exercise group rats after exercise for 4 weeks were significantly increased, compared with that in control group. Conclusion Exercise training may effectively induce BDNF and TrkB expression in spinal cords and promote the re-covery of locomotor function of SCI rats.

ABSTRACT

Objective To investigate the expression of survivin/Human leukocyte antigen class I ( HLA-Ⅰ) proteins and its physiological significance in clear cell renal cell carcinoma ( CCRCC ) . Methods Immunohistochemistry was used to analyze survivin/HLA-Ⅰ protein expression in 90 cases of CCRCC and 10 normal tissues to study relationships with clinical symptoms and disease prognosis . Resutl s The level of survivin protein expression was found to be significantly higher in CCRCC tissues 82.2%( 74/90) than in normal tissues( 0/10).However, the relative amount of HLA-Ⅰprotein in colorectal cancer tis-sue was also found to be significantly lower 67.8%(61/90) than in normal tissues 90%(9/10).Survivin expression was associated with tumor grade , stage,and lymph node metastasis ( P=0.000, P=0.016, and P=0.001, respectively ) .Conversely , lost HLA-Ⅰexpression did not have any associations with clinicopath-ological data (P>0.05).Survivin negative patients (25.0%, 4/16) had a higher tumor-free survival rate than patients (52.7%, 39/74)with survivin expression (P=0.037).Patients (27.6%, 8/29) with normal HLA-Ⅰlevels had a higher tumor-free survival rate than those ( 60.7%, 37/61) with reduced HLA-Ⅰlev-els (P=0.020).The univariate and multivariate analyses indicated that expression of survivin and HLA indi -vidually and in combination were independent predictors for CCRCC patient survival . Conclusions Over-expression of survivin but reduced HLA-Ⅰ expression is associated with CCRCC development and progres-sion.

ABSTRACT

Objective To detect the expression of apelin and angiotensin I receptor related protein (APJ) in rats with chronic obstructive pulmonary disease (COPD), and to explore the role of apelin-APJ system in the occurrence and development of COPD. Methods A rat model of COPD was established using the method of smoke exposure. SD rats were randomly divided into a control group and a COPD group. The rats in COPD group were exposed to secondhand cigarette smoke in a fume box twice a day for 4 months. Expressions of apelin mRNA and APJ mRNA in rat lung tissues were detected by real-time quantitative polymerase chain reaction (qRT-PCR). Expressions of apelin protein and APJ protein were determined by immunohistochemistry. Results The expressions of apelin mRNA and APJ mRNA in COPD group were decreased by 44% and 13% as compared with those in the control group. The mRNA expressions of apelin and APJ in rat lung tissues were negatively correlated with RV/(LV + S) (r = -0.454 and -0.448, P < 0.05), and positively related with FEV0.3/FVC (r = 0.529 and 0.475;P < 0.05). Apelin and APJ were mainly expressed in the epithelial cells of the bronch and lung, alveolar macrophages, vascular endothelial cells, and the membrane and cytoplasm of some alveolar wall cells. The expression of apelin and APJ protein was lower in COPD group than in the control group (P < 0.05). Conclusions The mRNA and protein expressions of apelin and APJ are decreased in the lung tissues in rats with COPD , which may be an important factor leading to the development of COPD. Apelin-APJ system might be a new target for prevention and treatment of pulmonary hypertension and right ventricular hypertrophy induced by COPD.

ABSTRACT

Purpose To investigate the pathological characteristics of loco-regional recurrent nasopharyngeal carcinoma ( rNPC ) . Methods Nasopharyngeal biopsy specimens of 46 rNPCs and 63 primary NPCs were collected. HE staining, immunohistochemistry and EBV small RNAs ( EBERs) in-situ hybridization were performed. Results The over-expression rates of both p63 and CK5/6 in rNPC were significantly higher than those of primary NPCs (P=0. 005, P=0. 026), while no statistical significance of Ki-67 over-ex-pression existed between the two groups ( P=0. 387 ) . More necrotic tissues, inflammatory exudates, giant bizarre carcinoma cells, desmoplastic stroma, giant bizarre tumor cells and higher degree of squamous differentiation were found in rNPCs. The carcinoma cells of 5 rNPCs were negative for both EBERs in-situ hybridization and LMP-1 immunohistochemical staining. Conclusion The loco-re-gional rNPC has two peaks of latency interval:2~5 and 9~11 years. The loco-regional rNPC cells have higher degree of squamous differentiation with higher expression of p63 and CK5/6, as well as more invasive ability. In addition, both EBERs in-situ hybridization and LMP-1 immunostaining are negative in 10. 87% (5/46) of loco-regional rNPC.
