ABSTRACT

Objective To observe the clinical effect of continuous vacuum sealing drainage (VSD) in trea-ting diabetic foot and its influence on the expression of serum insulin-like growth factor(IGF1) and nitric ox-ide(NO) .Methods Sixty-eight cases of diabetic foot in this hospital from June 2014 to December 2015 served as the research subjects and randomly divided into the control group and observation group .The control group took the common wound dressing treatment ,while the observation group adopted the VSD treatment ;the clin-ical efficacies after three courses of treatment were compared between the two groups .The treatment time , number of dressing change and wound healing time in the two groups were performed the statistics .The data after completely statistics were performed the contrastive analysis ;glycosylated hemoglobin (HbA1c) ,IGF1 and NO expression levels before and after treatment were compared between the two groups .The correlation between clinical effect with the levels of IGF1 ,NO and HbA1c was analyzed .Results The clinical cure rate of the observation group was 52 .94% ,the total effective rate was 94 .12% ,the clinical cure rate of the control group was 35 .29% ,the total effective rate was 67 .65% ,and the results of curative effect evaluation in the ob-servation group was better than that of the control group ,the difference was statistically significant ( P<0 .05) .The observation group was significantly better than the control group in the treatment time ,dressing change frequency and wound healing time ,the difference was statistically significant (P<0 .05) .After three courses of treatment ,the average expression level of IGF1 and NO in the observation group were higher than those in the control group ,the difference was statistically significant (P<0 .05) .There was a negative correla-tion between the curative effect and the level of serum IGF1 and NO (r= -0 .997 ,-0 .923 ,P< 0 .05) ,and there was a positive correlation between the curative effect with the HbA 1c level(r=0 .816 ,P<0 .05) .Conclu-sion VSD has an ideal effect in the treatment of diabetic feet ,can effectively shorten the treatment time ,and has the recovery of serum IGF1 and NO expression level .

ABSTRACT

Objective To evaluate the expression and the relativity of PMS2,Akt1 and P-AktS473 protein in A2780,Caov3,C13* and ES2 ovarian cancer cell lines .Methods The expression of PMS2 ,Akt1 and P-Akt S473 protein in A2780 ,Caov3 ,C13*and ES2 ovarian cancer cells was detected by Western Blot .After treated with IGF-1 (Akt1 activator) and API-2 (specific Akt1 in-hibitor) ,Caov3 ,ES2 and A2780 cells were collected and the level of PMS2 was detected by Western Blot .Results PMS2 ,Akt1 and P-Akt S473 proteins were detected in all of the four ovarian cancer cell lines with varied expression levels ,and the activity of Akt1 was inversely related to PMS2 expression in ovarian cancer cells .Exposed to Akt kinase stimulator IGF-1 ,ES2 and Caov3 cells were detected with a dramatically PMS2 decreasing .Meanwhile ,the decreasing of PMS2 protein was time-dependent on IGF-1 .Treated with API-2 ,Akt kinase specific inhibitor ,A2780 was detected with PMS2 dramatically increasing ,and the increasing of PMS2 pro-tein was time-dependent on API-2 .Conclusion In ovarian cancer cells ,PMS2 expression could be directly regulated by activated Akt1 .

ABSTRACT

Objective To study the insulin‐like growth factor binding protein (IGFBP)‐2 and IGFBP‐6 expression and clini‐cal significance in colorectal adenomas .Methods A collection of Chengde Medical College Hospital from July 2012 to March 2013 after surgical treatment of colorectal cancer confirmed by pathology (colorectal cancer ,CRC) tissue samples of 50 patients ,colorec‐tal adenomas (colorectal adenoma ,CRA) 50 cases ,20 cases of colorectal normal mucosa .Immunohistochemistry and RT‐PCR were used to detect the expression of IGFBP‐2 and IGFBP‐6 protein and mRNA ,combined with clinical and pathological data were statis‐tically analyzed .Results IGFBP‐2 protein positive expression and the amount of mRNA expression in the CRA group compared with normal colorectal mucosa had a rising trend;While compared with CRC group had a tendency to reduce ,and the differences are obvious statistical significance (P0 .05) ,but with the degree of hyperplasia had sig‐nificant statistical difference(P<0 .05);In the CRA group ,IGFBP‐2 and IGFBP‐6 expression were negatively related to each other , and the difference was statistically significant (P<0 .01) .Conclusion Colorectal adenomas in normal colorectal mucosa of colorec‐tal cancer to the middle part of the transformation process ,and in its occurrence and development process ,insulin‐like growth factor family (IGFs) and IGFS‐R axis plays an important and irreplaceable role ,so IGFBP‐2 ,IGFBP‐6 may be used as diagnostic colorectal adenomas and early predictors of prognosis ,clinical studies on colorectal adenomas is important .
