ABSTRACT

Objective To observe the therapeutic effect of sinomenine(SN) in adjuvant induce arthritis(AIA) model rats and its influence on inflammation related factors expressions.Methods The AIA rat model was duplicated by adopting complete Freund's adjuvant(FCA) induction method.After successfully constructing the model,the rats were randomly divided into the model group,methotrexate group,SN low,middle and high dose groups.The rats without constructing model were taken as the normal control group.Taking materials was performed after 21 d continuous medication gavage.The rat joint injury was observed by HE staining and the pathological semiquantitative scoring was performed.The serum levels of rheumatoid factor(RF),C-reactive protein (CRP),interleukin-1(IL-1),interleukin-6(IL-6),and interleukin-10(IL-10) were tested by using the automatic blood biochemical analyzer.Results Comparing with the model group,the joint injury degree in the methotrexate group,SN low and middle dose groups were obviously mild and the pathological semiquantitative score was significantly decreased(P<0.05),serum CRP level was significantly deceased(P<0.05),the serum inflammation related factors IL-1 and IL-6 levels were significantly decreased,while the IL-10 level was significantly increased (P<0.01).Conclusion Low and middle dose SN can regulate the imbalance of proinflammatory factors and inflammation-suppressing factors expression levels,thus significantly improve the joint pathological injury degree in rheumatoid arthritis.

ABSTRACT

Objective To investigate the effect of rosmarinic acid on the behavioral changes in enhanced single prolonged stress (ESPS) model rats and the levels of interlukin-1β (IL-1β) and interlukin-6 (IL-6) in the hippocampus.Methods 48 adult male Sprague Dawley rats were randomly divided into six groups (n =8):Control group,Control + RA (L) group,Control + RA (H) group,ESPS group,ESPS + RA (L) group and ESPS + RA (H) group.Behavioral changes of these rats were analyzed by open field test and elevated plus-maze.The levels of IL-1 β and IL-6 were measured by enzyme linked immunosorbent assay (ELISA).Results (1) Open field test showed that the number of central region entering and the fraction of time exploring in center of ESPS group were significantly reduced than that of Control group ((18.13 ± 10.15) times,(26.68 ± 10.06) %) and ESPS + RA (H) group ((16.88 ± 8.81) times,(25.08 ± 8.52) %) (P < 0.05).And it showed no significant difference among Control + RA(L) group,Control + RA(H) group and Control group.Meanwhile,there was also no statistic difference between ESPS group and ESPS + RA(L) group.(2) Elevated plus-maze test showed that percentages of open arm entries and fraction of time exploring in open arm in reference to total number of entries into all arms and total time spent on all arms in ESPS group were significantly reduced than that of Control group((37.38 ± 8.24)%,(17.63 ±4.74)%) and ESPS + RA(H) group((33.72 ±9.49)%,(16.99 ±4.28)%) (P < 0.05),but there was no significant difference between that of ESPS group and ESPS + RA(L) group(P>0.05).It also showed no significant difference among Control + RA (L) group,Control + RA (H) group and Control group.(3) Compared with ESPS group,RA(10mg/kg) reduced the levels of IL-1β and IL-6 in the hippocampus,but RA(5mg/kg) did not have this effect.(4) Correlation analysis results showed the level of IL-1β in the hippocampus was negatively related with the ameliorated PTSD-like behaviors of ESPS exposure rats.Conclusion RA can ameliorate PTSDlike behaviors of ESPS exposure rats,and this effect may be carried out by down-regulating the levels of IL-1β and IL-6 in the hippocampus,especially the IL-1β.

ABSTRACT

Objective To investigate the possible mechanisms of the attribution of psychological stress to the temporomandibular joint disorder(TMD) ,through the evaluation of the animal model and detection of the proinflammatory cytokines in the TMJ.Methods The animal models of communication box were built to mimic the psychological stress.The concentration of the serum Cor and ACTH was detected in the control group, Psychological Stress group ( PS group), and diazepam ( anti-anxiety drug) group ( PS + DI group).The expression of the proinflammatory cytokines IL-1 and IL-6 in the rat TMJ in the different phases of psychologicalstress was detected by RT-PCR.Results The results of the serum concen- tration of Cor and ACTH showed that there was significant difference between the control group and the PS group(P<0.01 ) ,while no significant difference between the control group and the PS + DI group(P>0.05).The expressions of IL-1 and IL-6 were comapared in all group.The expressions of IL-1 in CON group were (0.453± 0.021 ) mg/L, (0.439 ± 0.028 ) mg/L and (0.454 ± 0.023 )mg/L.These values were markedly increased compared with those of the PS group(0.981 ±0.024)mg/L, (0.746±0.017)mg/L and (0.510 ±0.016)mg/L respectively, P<0.01 ) ,but no significant differences compared with PS + DI group(0.549 ± 0.014 ) mg/L, ( 0.498 ± 0.014 ) mg/L and ( 0.444 ± 0.022 ) mg/L respectively, P > 0.05).Similar changes were observed in expressions of IL-6.The expressions of IL-6 in the CON rats were (0.525 ±0.028)mg/L,(0.515 ±0.028)mg/L and (0.518 ±0.022)mg/L,respectively,while those of PS group were(0.820 ± 0.023 ) mg/L, (0.694 ± 0.019 ) mg/L and (0.579 ± 0.015 ) mg/L, respectively, which were significan- tly higher in the PS groups(P< 0.05 ).But there were no significant differences between CON group and PS + DI group( (0.599 ±0.015)mg/L, (0.541 ±0.015)mg/L, (0.487 ±0.008)mg/L respectively, P>0.05).Conclusion The psychological stress can play important role in the formation of TMD.

ABSTRACT

Objective To investigate the role of psychological intervention in the prevention of the temporomandibular joint disease (TMD) , through the observation of the relative changes in the rat TMJ under psychological stress after psychological intervention. Methods The rat model of communication box was built to exert the psychological stress. The antianxiety agent was applied before stress, and the stressor was removed after stress. The expression of the proinflammatory cytokines IL-1 and TNF-α in the mandibular condylar chondrocytes in rat TMJ was detected by ELISA and RT-PCR. Results The RT-PCR results showed that the expression of IL-1 mR-NA increased into the peak in the 1st week, weakened in the 3rd week, and returned to normal in the 5th week, while the TNF-αmRNA peaked in the 1st week, returned to normal in the 3rd week. The ELISA results showed that there was no significant difference of the OD value of the serum IL-1 and TNF-α(0. 095 ±0. 006,0. 077 ± 0.007,0.069 ±0.009 ;0.079 ±0.010,0.075 ±0. 009,0.079 ± 0.012) in the antianxiety agent group (0. 107 ± 0.024,0. 101 ±0.005,0.088 ±0.010)and the stressor removal group(0. 090 ±0.016,0. 088 ±0.005,0.089 ± 0.011) , compared with the control group(0.087 ±0.004,0.090 ±0.009,0.089 ±0.010;0.074 ±0.008,0.069 ±0.015,0.068 ±0.011) (P>0.05), while significant differences were observed when compared with the psychological stress group(0.282 ±0.045,0.226 ±0.021,0.092 ±0.002;0. 164 ±0.009,0.123 ±0.013,0.091 ± 0.006) (P<0.05 ). Conclusion Application of the antianxiety agent and stressor removal could effectively counter the influence of psychological stress to TMJ, which provides good experience for the clinical prevention of TMD.

ABSTRACT

BACKGOUND: Recently it has been reported that several cytokine gene polymorphisms regulate cytokine production and play an important role in immune and inflammatory response. We evaluated IL-1beta IL-1Ra, and TNF-alpha gene polymorphism in patients with primary glomerulonephritis to determine the association between cytokine polymorphism and disease susceptibility. METHODS: In this study, we enrolled 118 patients with primary glomerulonephritis and healthy 300 persons who had visited the health screening center. We analyzed -511C/T polymorphism of IL-1beta tandem repeats polymorphism in intron 2 of IL-1Ra and -308G/A polymorphism of TNF-alpha We classified primary glomerulonephritis according to pathologic finding and clinical diagnosis. RESULTS: There were no differences with IL-1betaand TNF-alpha gene polymorphism between patient and control group. The carriage of IL1RN*2 was significantly associated with an increased risk of primary glomerulonephritis (patients:control=12.75:5.4%, p<0.01). IL1RN*2 was significantly frequent in patients with membranous GN or minimal change disease (p<0.05). When we classified glomerulonephritis according to clinical diagnosis, IL1RN*2 carriage rate was higher in patients with nephrotic syndrome and RPGN or acute nephritic syndrome than patients with asymptomatic urinary abnormalities (p<0.05). IL-1beta(TT) genotype was more prevalent in acute glomerulonephritis (68.4%) than asymptomatic urinary abnormalities or other glomerulonephritis. TNF2 carriage rate showed a lower tendency in patients with asymptomatic urinary abnormalities. CONCLUSION: IL1RN*2 is significantly associated with an increased risk of development of primary glomerulonephritis. We suggest cytokine gene polymorphism is also related to clinical manifestations of glumerulonephritis.

ABSTRACT

Objective To investigate the protective effects of different concentrations of propofol on thelungs apainst acute injury induced by lipopolysaccharide (us) in rats.Methods Twenty-four male Wistar ratsweighing 150-250g were randomly divided into 4 groups with 6 animals in each group : (1) control group receivedonly normal saline; (2) LPS group received LPS 5 mg?kg~(-1)i. v.; (3) propofol group 1 received a bolus 5 mg?kg~(-1) after LPS followed by propofol infusion at 5 mg?kg~(-1) ; (4) propofol group Ⅱ received a bolus of propofol 10mg.kg~(-1) after LPS followed by propofol infusion 10 mg?kg~(-1). Blood samples were obtained from femoralartery for determintiion of serum concentrations of TNF-?, IL-? and IL-10 at 1, 2, 3, 4 h after LPS injection. Theanimals were then killed by exsanguination. The lungs were removed. Left lung was lavaged and the broncho-alveolar lavage fluid (BALF) was collected for determination of its neutrophil count, and protein, TNF-?, IL-I?and IL-10 levels. Right lung was used for measurement of wet / dry lung weight ratio. ABSTRACT

Objective: To study the effect of Baihuzhuifeng Pills (BHZFP) on interleukin 1?(IL 1?) secreted by synoviocytes in adjuvant arthritis (AA) rats. Methods: Synovium of knee joints from AA rats were digested by collagenase and tripsin to prepare DSSC(digested synovial single cells). The content of the supernatant fluid was assayed by the radioimmunoassay. Results: BHZFP down regulated IL 1? productions of DSSC.Conclusion: It is suggested that BHZFP be probably inhibit the over proliferation of synoviocyte in AA rats by diminishing IL 1? secretion.
