ABSTRACT

Objective To study the mechanism of IL-6 stimulated the secretion of IL-10 in nasopharyngeal carcinoma CNE-2 cells.Methods Nasopharyngeal carcinoma CNE-2 cells were cultured with recombination cytokine IL-6 in vitro,or anti-IL-6 receptor antibody and signal pathway inhibitor were pre-incubated for 1 hour,and then IL-6 were added,the mRNA and protein level of IL-10 were separately detected by RT-PCR and enzyme-linked immunosorbent assay.Results Compared with the control group,the expression and secretion of IL-10 in IL-6 stimulated group were significantly increased,which was in a dose and time dependent way,the difference was significant(P < 0.01).Additionally,IL-6 stimulated the expression and secretion of IL-10 by CNE-2 cells were significantly decreased in following pre-incubated with anti-IL-6 receptor antibody or NF-κB inhibitor,the difference was significant(P < 0.01),but such effect was not detected when CNE-2 cells were pre-incubated with the PI3K inhibitor,p38/MAPK inhibitor,JNK inhibitor,MEK1/2 inhibitor and STAT3 inhibitor.Conclusion IL-6 can induce the expression and secretion of IL-10 in nasopharyngeal carcinoma CNE-2 cells via IL-6R/NF-κB signal pathway,and blocking IL-6 signal may be useful for the immunotherapy of nasopharyngeal carcinoma.

ABSTRACT

Objective To study the effects of containing bismuth sequential therapy on interleukin-10 (IL-10) and interleukin-17 (IL-17) in patients with peptic ulcer.Methods 162 patients with peptic ulcer were randomly divided into two groups,the observation group(n =81 cases) and the control group (n =81 cases).The patients in the observation group were treated through containing bismuth sequential therapy,while the patients in the control group were treated through traditional triple therapy.IL-10 and IL-17 were detected before and after treatment.Results The total effective rates were 97.5% in the observation group and 87.7% in the control group,which of the two groups had significantly difference (x2 =3.96,P < 0.05).Hp eradication rate of the observation group was 96.3 %,and which of the control group was 80.2%,and there was significant difference between the two groups (x2 =4.17,P < 0.05).IL-10 and IL-17 in both groups were significantly decreased after treatment.There was a significant difference between two groups after treatment (t =3.7244,9.3422,all P < 0.05).Conclusion Bismuth-containing sequential therapy can significantly reduce IL-10 and IL-17 in the serum of patients with peptic ulcer.

ABSTRACT

Objective To observe the effect of rosiglitazone on the concentration of interlukin (IL)-6 and IL-10 in lung tissues of diabetic rats.Methods The experimental diabetic rats were yielded by injecting streptozotocin(STZ) and feeding with high fat and high glucose food.We observed lung morphology in control group,diabetes mellitus(DM) group,and rosiglitazone group at 10 week and 20 week respectively under light microscope.Alteration of IL-6 and IL-10 in lung was measured by immunohistochemistry.Results The optical density values of IL-6 in the control group,the DM group and the roggerosiglitazone treatment group were 0.15 ±0.01,0.16 ±0.01;0.22 ±0.02,0.31 ±0.04;0.22 ±0.03,and 0.20 ±0.02 at 10 week and 20 week respectively (Fwithin =216.89,P < 0.01 ; Fbetween =342.62,P < 0.01 ; Finteraction =341.51,P < 0.01).Any two groups had significant difference(P < 0.05) except the comparison of the IL-6 values at 10 week and 20 week in the control group (P > 0.05).The absorbance values of IL-10 in the three groups were 0.13 ± 0.01,0.15 ±0.02;0.20 ±0.01,0.21 ±0.01;0.20 ±0.02,and 0.17 ±0.01 at 10 week and 20 week respectively (Fwithin =14.612,P <0.01 ;Fbetween =909.19,P <0.01 ;Finteraction =210.55,P <0.01).Any two groups had significant difference(P <0.05) except the comparison of the IL-6 values at 10 week and 20 week in the control group.Conclusion The elevated levels of IL-6 and IL-10 in lung tissue of dibtetic rats might be related to the inflammation of lung tissues.Rosiglitazone may alleviate lung inflammation by regulating the levels of IL-6 and IL-10.

ABSTRACT

Objective To investigate the effects of interlukin-10 (IL-10) on lipopolysaccharide (LPS)-induced liver injury in rats.Methods One hundred male Wistar rats,aged 10-14 weeks,weighing 250-300 g,were randomly divided into 5 groups (n =20 each):control group (C group),LPS group,IL-10 group,HO-1 inducer cobalt protoporphyrin-Ⅸ group (Co group) and HO-1 inhibitor zinc protoporphyrin-Ⅸ group (Zn group).The animals in LPS,IL-10,Co and Zn groups received intraperitoneal LPS 20 mg/kg.IL-10,Co and Zn groups received recombinant human IL-10 1 μg at 3 h before LPS injection.Co and Zn groups received cobalt protoporphyrin-Ⅸ and zinc protoporphyrin-Ⅸ 25 mg/kg at 2 h before administration of recombinant human IL-10.Ten rats in each group were chosen at 24 h after LPS injection and blood samples were collected from the heart for determination of the levels of serum alanine aminotransferase (ALT),aspartate transaminase (AST),tumor necrosis factor-α (TNF-α) and interlukin-1β (IL-1 β).The animals were then sacrificed and lungs removed for determination of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activities,malonodialdehyde (MDA) content and expression of HO-1 mRNA in lung tissues.The left 10 rats in each group were chosen and the survival rates within 72 h after LPS administration were recorded.Results Compared with C group,the levels of serum ALT,AST,TNF-αand IL-1 β and MDA content in lung tissues were significantly increased,and GSH-Px and SOD activities in lung tissues and survival rates were decreased in LPS,IL-10,Co and Zn groups (P < 0.05).Compared with LPS group,the levels of serum ALT,AST,TNF-α,IL-1β and MDA content in lung tissues were significantly decreased,GSH-Px and SOD activities in lung tissues and survival rates were increased,and the expression of HO-1 mRNA was up-regulated (P < 0.05).Compared with IL-10 group,the levels of serum ALT,AST,TNF-α and IL-1β and MDA content in lung tissues were significantly increased,and GSH-Px and SOD activities in lung tissues and survival rates were decreased in group Zn (P < 0.05),and no significant change in the parameters mentioned above was found in Co group (P > 0.05).Conclusion IL-10 can attenuate LPS-induced liver injury in rats by inducing the expression of HO-1.

ABSTRACT

Distribution and characterization of interlukin-10 (IL-10)-secreting cells in lymphoid tissues of pigs naturally infected with porcine circovirus type 2 (PCV2) were evaluated in accordance with PCV2 antigen detection. After screening a total of 56 pigs showing the symptoms of postweaning multisystemic wasting syndrome (PMWS), 15 pigs were PCV2 positive and 5 pigs, which showed stronger positive signals over multiples tissues were further investigated. This study showed that in PCV2-infected lymphoid tissues, particularly mandibular lymph node, spleen and tonsil, IL-10 expression was mainly localized in T-cell rich areas but rarely in B cell rich areas. IL-10 was highly expressed in bystander cells but rarely in PCV2-infected cells. Elevated IL-10 expression was predominantly associated with T cells, but rarely with B cells or with macrophages. The results of this study provide evidence for the role of IL-10 in chronic PCV2 infection and its relation to PCV2 antigen in affected tissues. Constantly elevated levels of IL-10 lead to immunosuppression in persistent and chronic viral infections. The increased IL-10 expression observed in PCV2 infection in this study suggests that IL-10-mediated immunosuppression may play an important role in the pathogenesis and maintenance of naturally occurring PCV2 infection.

ABSTRACT

Objective To explore the influence of three SNPs within interlukinc (IL) 10 promoter region on the expression of interlukin 10 mRNA in childhood systemic lupus erythematosus (SLE),then to determine if the SNPs do associate with the pathogenesis of childhood SLE.Methods The levels of IL 10 mRNA were detected in children with SLE and their parents by Taqman PCR,then the correlations between levels of IL 10 mRNA and haplotypes were assessed.Results The results showed that haplotype GCC was associated with higher level of IL 10 mRNA within children with SLE.Conclusion Haplotype GCC is associated with high secretion of IL 10 within childhood SLE,but not operate independently,and maybe it regulates the high secretion of IL 10 with other susceptible genes of SLE.

ABSTRACT

Objective To investigate interleukin 10 (IL 10,Th2 type cytokines) and interferon gamma (IFN ?,Th1 type cytokines) secreting capacity by peripheral blood mononuclear cells (PBMCs) in 30 patients with systemic lupus erythematousus (SLE) and 10 healthy individuals,and try to understand the role of Th1/Th2 balance in the pathogenesis of SLE.Method Sandwich ELISA was used to measure IL 10 and IFN ? level in the supernatant of culture from PBMCs.Results The level of IL 10 produced spontaneously by PBMCs in active stage was significantly higher than in static phase and healthy person ( P 0 05).Conclusion These data suggest that the Th2 type cytokine (IL 10) was increased in patients with SLE,and an imbalance towards Th2 predominance may play a significant role in the pathogenesis of SLE.Regulating IL 10 level may provide a new approach for the treatment of SLE.

ABSTRACT

Objective To investigate the protective effects of different concentrations of propofol on thelungs apainst acute injury induced by lipopolysaccharide (us) in rats.Methods Twenty-four male Wistar ratsweighing 150-250g were randomly divided into 4 groups with 6 animals in each group : (1) control group receivedonly normal saline; (2) LPS group received LPS 5 mg?kg~(-1)i. v.; (3) propofol group 1 received a bolus 5 mg?kg~(-1) after LPS followed by propofol infusion at 5 mg?kg~(-1) ; (4) propofol group Ⅱ received a bolus of propofol 10mg.kg~(-1) after LPS followed by propofol infusion 10 mg?kg~(-1). Blood samples were obtained from femoralartery for determintiion of serum concentrations of TNF-?, IL-? and IL-10 at 1, 2, 3, 4 h after LPS injection. Theanimals were then killed by exsanguination. The lungs were removed. Left lung was lavaged and the broncho-alveolar lavage fluid (BALF) was collected for determination of its neutrophil count, and protein, TNF-?, IL-I?and IL-10 levels. Right lung was used for measurement of wet / dry lung weight ratio. Results In LPS group thewet/dry lung weight ratio, BALF neutrophil counts and protein contents and BALF and serum TNF-?, Ib-I? andIL-10 levels were significantly increased compared with control group (P
