ABSTRACT

Abstract Introduction Multiple myeloma is characterized by proliferation of clonal plasma cells. The identification of prognostics factors to identify patient's risk is important. Among the studied factors, it was identified of relevant importance the lactic dehydrogenase. Objectives To evaluate the impact of the value of DHL in combination with the score ISS in the medium patients overall survival (OS). Methods It is a retrospective cohort with 252 patients with MM recently-diagnosed that attendance in the institution of the study. Results To evaluate the association between DHL and ISS, we found 6 new groups to be analyzed: ISS I and normal DHL with medium overall survival not reached, and with DHL loud with medium OS of 69,8 months, ISS II and normal DHL with medium overall survival of 78,8 months and with DHL loud with medium OS of 73,9 months, ISS III and normal DHL with medium overall survival of 46,7 months and with DHL loud with medium OS of 45,5 months. Conclusion Through the association of ISS I and normal DHL, ISS III and high DHL and others combinations, we build a new score with superior impact prognostic in our population treated in real life.

ABSTRACT

Background: Laparoscopic surgery is associated with more favorable clinical outcome than conventional open surgery. This might be related to the magnitude of tissue trauma and tissue stress response. This study compares the intensity of tissue injury by assessing plasma C-reactive protein (CRP), lactic dehydrogenase (LDH), and cancer antigen 125 (CA 125) in patients undergoing laparoscopic hysterectomy compared to abdominal hysterectomy.Methods: This study was conducted at Ain Shams University Maternity Hospital from May 2018 to February 2020. 74 women candidate for hysterectomy were recruited and randomized into two groups: group A included 37 cases who underwent abdominal hysterectomy, and group B included 37 cases that underwent laparoscopic hysterectomy.Results: CRP, LDH and CA125 significantly increased post-operatively in both groups. Postoperative mean serum CRP, LDH and CA125 were significantly higher in the laparotomy group (10.84±2.47, 262.21±76.77, and 13.41±2.6 respectively) compared to laparoscopy group (7.92±2.25, 148.53±43.56, 11.79±2.77 respectively) (p<0.05).Conclusions: Surgery is a significant cause of tissue injury that can be assessed by specific markers. We suggest that laparoscopic surgery causes less tissue damage as assessed by lower postoperative CRP and LDH values, which might account for the earlier recovery and reduced hospital stay in laparoscopic surgery.

ABSTRACT

Molecular mechanisms whereby H2S influences its targets have been of intriguing interest. In this work, L-lactic dehydrogenase ( L-LDH) was used as the protein target, and three kinds of H2S-donor reagents ( NaHS, Na2S, and polysulfide) were chosen. The interactions of these H2S-donor reagents with L-LDH were disclosed by molecular fluorescent assays for real-time monitoring of L-LDH activity. The results of the SDS-PAGE showed that H2S might not interact with L-LDH to form disulfide/trisulfide bonding. Circular dichroism spectra assays revealed that H2S reagents could be likely to react with cysteine thiols to yield sulfurated thiol (-SSH) derivatives in L-LDH, and sulfur-containing PS ( polysulfide) was a stronger protein S-sulfurating agent than the other two sulfides. Matrix assisted laser desorptionionization time-of-flight tandem mass spectrometry ( MALDI-TOF-MS/MS) study showed partial S-sulfuration of the active cysteine sites existed in L-LDH. In conclusion, H2S exerts its biological effects as a gasotransmitter through its reactions with cysteine thiols in proteins by S-sulfuration.

ABSTRACT

Objective To study the accuracy of lactic dehydrogenase (LDH) in the diagnosis of pneumocystis pneumonia (PCP). Methods The data of this systemic review was retrieved from the PubMed, China Biology Medicine disc, Wanfang, Weipu and China National Knowledge Infrastructure (CNKI) databases from establishment till to October 31st, 2017. Case-control studies about the diagnosis of PCP were enrolled. Enrolled studies were required that patients in case group ware PCP and patients in control group were lung diseases other than PCP. The QUADAS tool was used to evaluate the quality of studies. The RevMan 5.3 software was used to draw a forest plot. The StataMP 14 software was used to make subgroup analyses by drawing receiver operator characteristic (SROC) curves for the whole group, the acquired immune deficiency syndrome (AIDS) group, and the not all-AIDS group, and calculating their diagnostic odds ratio (DOR) and 95% confidential interval (95%CI). Results Thirteen studies, all in English, were included. There were 825 patients in the case group, in which 650 patients were AIDS. There were 1 341 patients in control group, in which 888 patients were AIDS and most of them were Pulmonary Kaposi Sarcoma, bacterial pneumonia, pulmonary tuberculosis etc. Although there were different positive values of LDH in different studies, from 200 U/L to 598 U/L, sensitivities were good, especially in AIDS patients all values were above 80% (80%-100%). The specificities had big fluctuations, from 6% to 85%, which made them poor. The DOR (95%CI) of LDH in PCP diagnosis of all patients, AIDS patients and not-all AIDS patients were 6.73 (3.19-14.21), 9.17 (3.79-22.18) and 5.07 (1.30-19.80) respectively. Conclusions The sensitivity of LDH in the diagnosis of PCP is high, especially in AIDS group. In practice if LDH is negative, there should be more evidences to support the treatment of PCP.

Objective To study the effect of vibration on free hemoglobin(FHb), lactic dehydrogenase(LDH), and K+concentration of red blood cells(RBCs)stored for different lengths of time.Methods Thirty-five bags of RBCs stored for different lengths of time(3,7,11,26,and 35 d,7 bags per group)were chosen as the target of research.A wheeled vehicle that could vibrate in three directions(horizontal,longitudinal,and vertical)was used to simulate RBC transportation in addition to an electromagnetic vibration test system.The RBC samples were prepared and vibrated for 0 h,or 3 h for detection and analysis of their FHb, LDH, and K+concentration.Results The FHb concentration was significantly increased after vibration(P<0.05), and tended to increase with the extension of storage time.LDH(P<0.05)and K +concentrations(P<0.05)were obviously increased with the length of storage(3,7,11,26,and 35 d), and significantly increased after vibration of 3 h.Conclusion FHb concentration is increased after vibration, and the storage time could change FHb.The vibration and storage time have great effect on LDH and K +concentrations.

Objective: To study the effects of Sini Decoction before and after compatibilities on acute myocardial ischemia rats. Methods: The model of myocardial ischemia in rats was established by ig Pit. The effects of Sini Decoction, Aconiti Lateralis Radix Praeparata (ALRP)-Glycyrrhizae Radix et Rhizoma (GRR) decoction, ALRP-Zingiberis Rhizoma (ZR) decoction, ZR-GRR decoction, single ALRP decoction, single ZR decoction and single GRR decoction on the extension of myocardial infarction, lactic dehydrogenase (LDH), creatine kinase (CK), aspartate aminotransferase (AST), superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), and lactic acid in rats were evaluated. Results: Sini Decoction, ALRP-GRR decoction, ALRP-ZR decoction, and single ALRP decoction obviously decreased the extension of myocardial infarction (P ALRP-ZR decoction > ALRP-GRR decoction > single ALRP decoction. It reveals the scientific connotation of the compatibilities of Sini Decoction and accurate action about drug using of Sini Decoction from the perspective of efficacy.

ABSTRACT

Objective To explore the effect of rituximab on serum hepcidin (Hepc), soluble CD40 ligands (sCD40L), platelet derived growth factor-BB (PDGF-BB) and lactate dehydrogenase (LDH) in patients with malignant lymphadenoma.Methods 52 patients with malignant lymphoma from the hospital were randomly divided into control group and experimental group.The control group were treated by cyclophosphamide, pirarubicin, vincristine and prednisone and the experiment group were treated on the basis of control group with rituximab.The serum hepcidin, sCD40L, PDGF-BB and LDH levels were compared after treatment.Results Compared with pre-treatment, the serum hepcidin, PDGF-BB and LDH levels were lower( P<0.05), the sCD40L level was higher(P<0.05) post-treatment.The serum hepcidin, PDGF-BB and LDH levels in experimental group post-treatment were lower, sCD40L level was higher than those in control group(P<0.05).Conclusions The rituximab could reduce serum hepcidin, PDGF-BB and LDH levels and elevate serum sCD40L level in patients with malignant lymphadenoma.

ABSTRACT

OBJECTIVE: Peritonitis is one of major complication of peritoneal dialysis. It is the most important reason for removal of peritoneal catheter, death, or converting to hemodialysis. There is a debate that peritonitis decreases residual renal function. Thus, the authors carried out a study to grasp the prognosis of peritonitis which affects residual renal function (RRF) and assessed the risk factors of its clinical course. METHODS: Among 245 patients who had been on peritoneal dialysis in Soonchunhyang University Bucheon Hospital from April 2001 to March 2012, the author selected 91 patients who had kept on peritoneal dialysis for more than 3 months and experienced more than one episode of peritonitis. The medical records and laboratory findings were reviewed. RESULTS: During the average period of 36.3+/-22.8 months of peritoneal dialysis, total of 182 episodes of peritonitis developed in 91 study subjects. Among them, the 15 cases (8.2%) did not improve despite of intraperitoneal antibiotics and peritoneal catheter was subsequently removed. Eight patients were dead because of peritonitis related complications. The lactic dehydrogenase (LDH) level of peritoneal fluid at the third day of treatment had a significant correlation with peritonitis treatment failure (odds ratio [OR], 1.079; P=0.009) and death (OR, 1.071; P=0.049), respectively. The RRF after peritonitis became significantly low (P=0.011) compared to before peritonitis. But the slopes of declining rate of RRF were not different between before and after peritonitis (P=0.932). CONCLUSION: The LDH level of peritoneal fluid at the third day of treatment was correlated with treatment failure or death. The declining rate of RRF was not affected after peritonitis.

Objective To investigate the clinical significance of quantitatively detecting serum cancer antigen 125(CA125) ,lactate dehydrogenase(LDH) ,β2-microglobulin(β2-MG) and serum ferritin(SF) in primary gastric lymphoma(PGL) .Methods The clini-cal data in 41 cases of PGL confirmed by Pathological examination were retrospectively analyzed on the serum CA 125 ,LDH ,β2-MG and SF levels ,and their relationship with the pathological type ,clinical phase ,grouping ,international prognostic index (IPI) andHe-licobacter pylori infection were compared .Results The serum levels of CA125 ,LDH ,β2-MG and SF in the Ⅲ + Ⅳ stage were high-er than those in the Ⅰ + Ⅱ stage in PGL patients(P<0 .05);the IPI scores in the middle-higher risk + high risk group were obvi-ously higher than those in the lower risk + low-middle risk group (P<0 .01) .There were no significant differences in the patho-logical type ,grouping and Helicobacter pylori infection .Conclusion Detecting the levels of CA125 ,LDH ,β2-MG and SF has the im-portant clinical value to the clinical phase and the prognostic judgment in PGL .

ABSTRACT

ObjectiveTo investigate the changes of serum myocardial enzymes and high density lipoprotein cholesterol(HDL-C) in children with septic shock.MethodsFifty-two children with septic shock in our hospital from Jan 2006 to Mar 2011 were divided into severe septic shock group( n =27 ) and mild septic shock group ( n =25 ).The serum creatine kinase-MB ( CK-MB ),α-hydroxybutyrate dehydrogenase (α-HBDH),lactic dehydrogenase(LDH) and HDL-C were analysed in both two groups on 1,2,4,7,10,15 d,and compared with 32 normal children as control group.Meanwhile,the children's condition,death cases and cured time were recorded.ResultsThe levels of serum CK-MB [ ( 32.084 ± 4.595 ) U/L,( 61.481 ±5.639) U/L],α-HBDH/LDH(0.694 ±0.080,0.884 ±0.079) in mild and severe septic shock groups were higher than those of normal control group[ (21.675 ± 3.453) U/L ] and (0.443 ± 0.065 ) ( P < 0.01 ) ;the levels of serum HDL-C[ (0.646 ±0.067) mmol/L,(0.310 ± 0.124) mmol/L ] in the two septic shock groups were decreased significantly than that of control group[ ( 1.012 ±0.156) mmol/L] (P <0.01 ).Five cases died in the severe septic shock group while no cases died in mild septic shock group.The times of CK-MB recovered[ (9.82 ±1.76) d vs (4.68 ± 1.22) d],α-HBDH/LDH recovered[ (7.23 ± 1.38) d vs (3.76 ± 0.83 ) d] and HDL recovered[ ( 12.14 ± 2.21 ) d vs ( 6.48 ± 1.33 ) d ] were more slowly in severe septic shock group than those of mild septic shock group( P < 0.01 ).ConclusionThe higher of the serum CK-MB and α-HBDH/LDH,and the lower of HDL-C in septic shock children,the disease is more severe,the recovery time is longer and a worse prognosis may occur.

Se evaluaron 104 pacientes adultos con anemia drepanocítica clasificados en: grupo 1 (G1) pacientes sin hipertensión pulmonar (HTP) (n=74); y grupo 2 (G2) pacientes con HTP (n=30). Se observó disminución estadísticamente significativa para la hemoglobina, el hematócrito y la oximetría de pulso, y aumento significativo de los leucocitos, la deshidrogenasa láctica, la bilirrubina total e indirecta, la creatin-kinasa MB y la creatinina en los pacientes del G2. La prueba de X² mostró diferencia significativa entre los grupos para el síndrome torácico agudo (p=0,05) con mayor frecuencia en el G2. La frecuencia de HTP en el total de enfermos evaluados fue del 28,8 por ciento. Las extrasístoles ventriculares fueron las alteraciones más frecuentes en el electrocardiograma en el G1, y la hipertrofia del ventrículo izquierdo en el G2; mientras que en el ecocardiograma Doppler fue la dilatación del ventrículo izquierdo en ambos grupos. De los casos diagnosticados con HTP (n=30), 27 (73,3 por ciento) fueron clasificados como grado 1; 7 (23,3 por ciento) grado 2 y 1 (3,4 por ciento) grado 3


A total of 104 adult patients presenting with sickle cell disease were assessed and classified into: group 1 (G1) patients without pulmonary hypertension (PHT) (n = 74) and group 2 (G2) patients with PHT (n = 30). There was a statistically significant decrease for hemoglobin, hematocrit and pulse oximeter and a significant increase of leukocytes, the lactic dehydrogenase, the total and indirect bilirubin, la MB creatine kinase and the creatinine in the G2-patients. The X² test demonstrate a significant difference among groups for the acute thoracic syndrome (p = 0,05) more frequent in the G2. The PHT frequency in all patients assessed was of 28,8 percent. The ventricular extrasystoles were the more frequent alterations in the electrocardiogram in the G1 and the left ventricle hypertrophy in the G2; whereas in the Doppler echocardiogram the more frequent as the left ventricle dilatation in both groups. From the cases diagnosed with PHT (n = 30), 27 (73,3 percent) were classified as 1 degree; 7 (23,3 percent) 2 and 1 degrees (3,4 percent) 3 degree

ABSTRACT

As células-tronco mesenquimais (MSCs) são células com grande potencial de diferenciação e estão sendo recentemente introduzidas na clínica para tratamento de várias doenças. Possuem várias vantagens incluindo sua estabilidade fenotípica in vitro. OBJETIVO: isolamento das MSCs de líquido amniótico, sua expansão e a demonstração da sua capacidade de se diferenciar em células miogênicas e adipogênicas, sem alterar a estabilidade cromossomal em meio de cultura. MÉTODOS: a fim de avaliar a mudança funcional destas células, foram avaliados parâmetros bioquímicos nas células adipogênicas já diferenciadas e antes da diferenciação através da dosagem de triglicérides. A diferenciação em células musculares foi avaliada comparando os níveis de creatinofosfoquinase - CK, desidrogenase lática - LDH e aldolase produzidas por estas células antes e após diferenciação. RESULTADOS: os níveis de triglicérides foram significativamente maiores nas células diferenciadas, mostrando ainda a formação de grânulos intracitoplasmáticos. Todos os outros valores obtidos foram significativamente maiores nas células miogênicas diferenciadas quando comparadas às não diferenciadas. CONCLUSÃO: os resultados sugerem que estes protocolos podem ser usados para avaliar diferenciação de células-tronco em células adipogênicas e miogênicas, e que o líquido amniótico pode ser uma fonte para obtenção destas células.


The mesenchymals stem cells (MSCs) are cells with the great potential of differentiation are being introduced in the clinic for treatment of several diseases. Mesenchymal stem cells have several advantages including the stability of their phenotype in vitro. BACKGROUND: isolation of MSCs in amniotic fluid, its expansion and the demonstration of the capacity of these cells to differentiate in adipogenic and miogenic cells, without to change the chromosomal stability of the MSCs in culture. METHODS: in order to evaluate the functional change of these cells, were gotten values of the differentiated adipogenic cells and not differentiated through the dosage of triglycerides. The miogenic nature of the differentiated cells was analyzed comparing the creatine kinase - CK, lactic dehydrogenase - LDH and aldolase produced by the cells. RESULTS: the values of triglycerides were significantly higher in differentiated cells, showing intracitoplasmatic granule form after differentiation. All the biochemical characters were significantly higher in differentiated miogenic cells. CONCLUSIONS: this study suggests that the standardized protocol of differentiation can be used in the attainment of cells with characteristics of adipogenic and muscular cells, from amniotic fluid.

ABSTRACT

Objective To observe the effects of anti lipopolysaccharide (LPS) immunoglobulin Y (IgY) on LPS in vitro . Methods The direct in vitro neutralization of different concentrations of IgY and LPS was conducted for the detection of the remained amount of LPS to observe the dose effect relationship. The primary cultured human umbilical vein endothelial cells (HUVECs) were cultured for 48-72 h after addition of certain concentrations of fluorescein isothiocyanate LPS (FITC LPS) and IgY. Changes of the fluorescent intensity in HUVECs were observed at different time points. After addition of certain concentrations of LPS and IgY for incubation with HUVECs for 4, 8, and 12 h, the changes of lactate dehydrogenase (LDH) activity were measured. Results The dose effect relationship was satisfactory when the ratio was 16︰1. At 4, 8, and 12 h, fluorescent intensity was somewhat strong in FITC LPS group, but weak in IgY group, especially at 8 and 12 h. The antifatigued effect of Haimaichong on the rats has been studied.When the rats were given Haimaichong nutrient 15 days,the motion endurance,the activity of lactic dehydrogenase the removing speed of lactate and the contents of the glycogen in the liver and in the muscle have been enhanced markedly;The level of blood sugar during the long time of motion can be kept steadily;The product of urea nitrogen in the serum has been decreased notably in motion and removed swiftly after motion.The results suggest that Haimaichong nutrient have the antifatigued effect strikingly.

ABSTRACT

Objective To study the changes of lactic dehydrogenase (LDH) and ? hydroxybutyric acid (HBD) in atrophic gastritis type A(AAG) patients. Methods Auto biochemistry instrument was used to detect serum LDH and HBD. Hemoglobin, vitamin B 12 and gastrin were also examined routinely. Results All AAG patients had elevated levels of LDH and HBD in serum and 60 percent of patients had a rise of indirect bilirubin. In the first day after supplement with folic acid and vitamin B 12 , LDH and HBD decreased simultaneously by 30% along with a decrease of indirect bilirubin. The changes of LDH and HBD were earlier than changes of reticulocytes. Conclusions Serum LDH and HBD rise significantly in AAG patients. This might be associated with in situ hemolysis of bone marrow. And it may be a good index for differential diagnosis between AAG and atrophic gastritis type B.

@#ObjectiveTo study the relationship between the activity of serum lactic dehyrogenase (LDH) and retromorphosis osteoarthropathy.MethodsSerum lactic dehydrogenase of 148 cases with retromorphosis osteoarthropathy (retromorphosis group) were exzamed, comparing with LDH of 138 normal subjects(control group). ResultsThe mean value of LDH in retromorphosis group was 199.28±91.64 while that in the control group was 183.85±69.46.There was no significant difference between two groups(P>0.05).Conclusion Retromorphosis osteoarthropathy may not affect the activity of serum lactic dehydrogenase.

ABSTRACT

Sputum LDH determination can differentiate between uninfected and infected bronchial secretions and appears to be a useful objective index of response of bronchopulmonary infections to antibiotic therapy. Its behavior parallels that of the clinical, bacteriologic and radiologic picture in 20 patients in PGH treated for their infection. About one half of the patients had pneumonias secondary to diplococcus pneumonia for which doxycycline was effective. It is disturbing to note the emergence of 8 cases with gram negative infection, 5 of whom failed to respond to dexycycline, gentamycin and other antibiotics. (Author)

ABSTRACT

We evaluated the hypoxic effects of the gas permeable contact lens of varying oxygen transmissibilities on the normal rabbit and photorefractive excimer keratectomized cornea. Fifty rabbits were divided into two groups accordig to corneal condition. The first group was thirty normal rabbit group, the other was twenty photorefractive excimer keratectomized group. A month following PRK(photorefractive keratectomy), gas permeable contact lenses were put on both group with three different Dk values (less 60, 60-90,over 120) for 1, 3, 5and 7 days for extended wear. Corneal pathological findings such as abrasion, ulcer were found in proportion to the duration of contact lens wearing in the two groups. Over 4 days continous wearing, the RGP lens was no more tolerable in excimer treated group in consideration of corneal thickness. Gas permeable lens caused an increase in tear LDH activity from 1,190 U/L to 17,000 U/L from the first day in excimer treated group and low DK contact lens group. Light and electron microscopy showed in both group that the longer the wearing time, the more the desqumation of epithelia; these changes were more severe in case of the lens caused the corneal hypoxia and complications and were not tolerable in compromised cornea with excimer laser even in high Dk/L gas permeable contact lens.

Object It was reported that salvianolic acid A from Salvia miltiorrhiza Bge could improve memory dysfunction of mice with cerebral ischemia reperfusion injury The effects of total salvianolic acids (Sal), including salvianolic acid A and B, rosmaric acid and protocatechic aldehyde was now studied to observe its effect on cerebral ischemia reperfusion injury in hope of obtaining a preliminary view on its mechanism of action Methods The effects of Sal on lactic dehydrogenase (LDH), superoxide dismutase (SOD) and malondialdehyde (MDA) of cerebral ischemia reperfusion injured mouse and rat models were observed Results Sal 10, 20 mg/kg iv and 5, 10 mg/kg iv could markedly increase the activities of LDH and SOD and lower the production of MDA in the injured cerebral tissue On focal cerebral ischemia reperfusion injured rat, 24 h reperfusion after 2 h ischemic injury, 5, 10 mg/kg iv Sal could decrease infarct size, ameliorate neurological deficit with obvious inhibition of the decrease of LDH and SOD activities and the over production of MDA Conclusion Sal showed obvious protective effects on cerebral ischemia reperfusion injuries of mice and rats via attenuating oxygen free radical
