ABSTRACT

Objective To determine the effects of Tempol, one of the nitroxides, in the presence of ultraviolet-AI (UVA1, 340 nm -400 nm) on superoxide enzyme (SOD) activity, lipid peroxidation, and expression of collagen Ⅰ, collagen Ⅲ and matrix metalloproteinases (MMP)-1, MMP-3 in human dermal fi broblasts in vitro. Methods Fibroblasts were irradiated by a single exposure to UVA1 and at the same time incubated with or without Tempol, and detected 24 h later. SOD activity and lipid peroxidation, as shown by accumulation of malondialdehyde (MDA), were detected by biochemical assay. Expression of collagen Ⅰ, collagen Ⅲ (protein levels) and MMP-1, MMP-3 (mRNA level) was examined by ELISA and semi-quantitative RT-PCR, respectively. ABSTRACT

Aim To investigate the protective effects of ginsenosides ( GS) on reactive oxygen species-induced oxidative damage in mouse spermatogonial cells. Meth-ods Mouse spermatogonial cell oxidative stress model was established and the attenuating effects of ginsen-osides on germ cell oxidative damage were evaluated by determination of cell viability,malondialdehyde( MDA) formation,superoxide dismutase ( SOD) activity and glutathione ( GSH) level. Results The exposure to hypoxanthine/xanthine oxidase ( HX/XO) induced an elevation in MDA,while a decrease in germ cell viability,SOD activity and GSH level. However,supplementation with GS ( 10 mg?L -1) restored HX/XO-induced decrease in cell viability,SOD activity and GSH level and HX/XO-induced increase in MDA formation. Conclusion GS may exert antioxidant activity to attenuate reactive oxygen species-induced oxidative damage in mouse spermatogonial cells.

ABSTRACT

A number of Chinese herbs have been indicated to have protective effects beforecerebral ischemic attacks in man. Polydatin, a glycoside isolated from the roots of Polygonum cuspidatum, is one of the major active ingredients of the Chinese herb. To investigate the effects of polydatin on free radicals in cerbral ischemia, a model of ischemia/reperfusion in rat brain was used. After polydatin administration, the contents of brain water and malondialdehyde (MDA) decreased, and the activities of superox-ide dismutase(SOD), catalase (CAT) and glu-tathione peroxidase (GSH-Px) were elevated to certain extent in cerebral cortex and hippocampus of the rats. Protective action of polydatin in ischemia/reperfusion induced brain injury has been suggested. The effect of polydatin was re-latd to the concentrations of the polydatin administered. The results showed that 12 mg ?kg-1 polydatin (iv) had the most significant effect on free radicals in ischemic brain injury. The results of this study may be useful in designing new drugs in the treatment of strokes and cerebral vascular diseases.

ABSTRACT

Objective To investigate the influence of military exercise on some biochemical indexes in soldiers.Methods The level of serum soluble tumor necrosis factor receptor-1(sTNFR-1)and soluble intercellular adhesion molecule-1(sICAM-1)were determined with ELISA in peripheral blood of 133 servicemen who participated in the excercise before and after the military exercise.At the same time,the levels of serum gastrin and tetosterone were determined by radioimmunassay(RIA),the level of superoxide dismutase(SOD)and malonyl dialdehyde(MDA)were determined by biochemical methods.The correlation between the changes in these contents and psychological stress was studied.Results The levels of serum sTNFR-1,sICAM-1,testosterone and MDA of 133 servicemen increased after the military exercise compared with that of before the exercise(P0.05).Conclusion As a strong stress,military exercise can inhibit the immunological function and produce changes in emotional state,gastroenterological hormones,sex hormones etc.It is necessary to carry out mental and physiological training,so that the ability of servicemen to withstand the military stress can be improved.
