ABSTRACT

BackgroundBreast cancer has been considered a public health problem and homeopathic treatments are becoming increasingly recommended due to its ways of action and absence of adverse effects. MCF-7 is an adenocarcinoma of human breast cell line useful as preclinicalmodel to screen therapeutic agents such as ultra-diluted Viscum album, an European plant which extract is commonly used in cancer therapy. AIMS MCF-7 and mesenchymal stem cells (MSC) were used to evaluate the in vitrocytotoxicity of homoeopathic Viscum album 1x10-3(VA3X). Methodscells were cultured for 24 hours in controlled environment (37.5oC and 5% CO2) in 96-well plates. After this time, VA3X was added to the culture medium in concentrations varying from 10 to 100 L/mL.A control group was maintained with culture medium only. Cells were cultivated for 48 hours in these conditions for evaluation of cell viability by MTT assay. ResultsHigher cytotoxicity was observed in MCF-7 when compared to MSC, as the lower concentration of VA3X was capable of inducing tumor cell death and not healthy cell death. The MTT assay results were that 42 L/mL of VA3X reduced MCF-7 cells viability to 50% and 62 L/mL reduced MSC cells to the same percentage, what means that tumor cells are more sensible to VA3X than heathy cells. ConclusionViscum albumpresented higher cytotoxic action on human breast cancer cell line culture than on mesenchymal stem cells. This medicine is extensively used against cancer, and the use of the homoeopathic form of it brings new possibilities as no or fewer adverse effects would be present.(AU)

ABSTRACT

Only 1% of all pituitary surgeries are performed to treat metastatic tumors in this region. The most common primary sites are breast and lung respectively. In this article we describe a patient with previous breast adenocarcinoma presenting with oculomotor palsy sudden right. The objective is to describe a case of breast metastasis to the sellar region, showing clinical and radiological findings that make this plausible diagnosis.

ABSTRACT

Aim Toinvestigatewhetherexposureto Sanguinarine (SAN ) can inhibit cell proliferation in human mammary adenocarcinoma cells (MCF-7 ) and thepossiblemechanism. Methods WeexposedMCF-7 to anticancer compound SAN,cell viability was as-sessed by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT ) reduction assay. ROS was measured using confocal microscopy,expres-sion of caspase-3 ,caspase-8 and caspase-9 were calcu-latedusingchemiluminescencemethod.Results SAN remarkably inhibited growth of human mammary adeno-carcinoma MCF-7 cells by decreasing cell proliferation. ROS release and caspase-3,caspase-8,caspase-9 ex-pression were stimulated by SAN in MCF-7 ,and these changes were abolished by the antioxidant,N-acetyl-cysteine(NAC).Conclusion Regulationofcaspases expression and release from MCF-7 cells are possibly e-voked by SAN through reactive oxygen species.

ABSTRACT

An 8-year-old female, German Shepherd dog was presented with history of dyspnea and seizure. One Year Previously a lumpectomy had been performed for surgical resection of mammary mass. In serum chemistry, severe hypoglycemia and elevation of aspartate aminitransferase and creatine kinase were shown. In thoracic radiography, there were variable sized nodules in the overall lung field. On computed tomography examination, the mass was shown in left longissimus lumborum muscle. Histopathological examination revealed adenocarcinoma derived from the mammary gland. Muscular metastasis of mammary gland tumor is uncommon. This is a rare observation and could easily be overlooked or misinterpreted.

ABSTRACT

Objective To observe inhibition rate of mouse mammary adenocarcinoma(EMT6) and inhibition effect on tumor angiogenesis of Panax notoginseng F.H. Chen and Cordyecps sinensis Sacc. powder. Methods BALB/C mice as bearing cancer animals with subcultured mouse-transplantable EMT6 cell train had accepted intervention treatment by Panax notoginseng F.H. Chen and Cordyceps sinensis Sacc. powder. In this way, inhibition rate of the mouse-transplanted EMT6 by the powder was recorded and the expression of microvessel density(MVD),vascular endothelial growth factor(VEGF) and tissue inhibitor of metalloproteinase-2(TIMP-2) in the tumor tissue after paraffin imbedding and section were detected through immunohistochemistry technique. Results The tumor inhibition rates of two designed dose groups were separately 48.95% and 37.64%. MVD and VEGF expressions of tumor tissue in the two group were significantly suppressed while the TIMP-2 expression increased. Conclusions Panax notoginseng F.H.Chen and Cordyceps sinensis Sacc. powder can restrain the growth of EMT6,and the pathway of this function is likely related to inhibiting tumor angiogenesis.

ABSTRACT

OBJECTIVESTo determine the effect of cis-9, trans-11-conjugated linoleic acid on the cell cycle of mammary cancer cells (MCF-7) and its possible mechanism of inhibition cancer growth.

METHODSUsing cell culture and immunocytochemical techniques, we examined the cell growth, DNA synthesis, expression of PCNA, cyclin A, B(1), D(1), p16(ink4a) and p21(cip/wafl) of MCF-7 cells which were treated with various c9, t11-CLA concentrations (25 mM, 50 mM, 100 mM and 200 mM) of c9, t11-CLA for 24 and 48 h, with negative controls (0.1% ethanol).

RESULTSThe cell growth and DNA synthesis of MCF-7 cells were inhibited by c9, t11-CLA. MCF-7 cells, after treatment with various c9, t11-CLA doses mentioned above for 8 days, the inhibition frequency was 27.18%, 35.43%, 91.05%, and 92.86%, respectively and the inhibitory effect of c9, t11-CLA on DNA synthesis (except for 25 mM, 24 h) incorporated significantly less(3)H-TdR than did the negative control (P<0.05 andP<0.01). To further investigate the influence on the cell cycle progression, we found that c9, t11-CLA may arrest the cell cycle of MCF-7 cells. Immunocytochemical staining demonstrated that MCF-7 cells preincubated in media supplemented with different c9, t11-CLA concentrations at various times significantly decreased the expressions of PCNA, and Cyclin, A, B(1), D(1) compared with the negative controls (P<0.01), whereas the expressions of p16(ink4a) and p21(cip/wafl), cyclin-dependent kinases inhibitors (CDKI), were increased.

CONCLUSIONSThe cell growth and proliferation of MCF-7 cells is inhibited by c9, t11-CLA by blocking the cell cycle, which reduces expressions of cyclin A, B(1), D(1) and enhances expressions of CDKI (p16(ink4a) and p21(cip/wafl)).

ABSTRACT

Aim To establish a new paclitaxel resistant human mammary adenocarcinoma cell subline(MCF-7/Taxol) and investigate its characteristics. Methods A paclitaxel resistant human mammary adenocarcinoma cell subline(MCF-7/Taxol) was developed by gradually increasing the concentration of Taxol from the parent cell line MCF-7 in vitro. The multidrug resistance of MCF-7/Taxol to anticancer agents was evaluated by SRB assay;the distribution of their cell cycles was detected by flow cytometry;the positive expression rate of P-gP、LRP、ToPoII、GST?、ER and PR was measured by S-P immunohistochemistry;the intracellular accumulation of Taxol was assessed by HPLC;the morphological features and the celluar ultrastructure characteristics were observed respectively by light microscopy and scanning electron microscopy. Results MCF-7/Taxol was resistant to several chemotherapy agents, such as Hydroxycamptothecine,Epirubicin,Doxorubicin,Mitoxantone and so on.The IC50 of MCF-7/Taxol to Taxol was 525 times higher than that of MCF-7,and the IC50 of MCF-7/Taxol stopped administrating Taxol for three months was 150 times higher than that of MCF-7.The multiplication time of MCF-7/Taxol was longer than that of the natural cell and the proportion of cells in S-phase increased while that in G1-phase decreased. The expression levels of P-gp、LRP and GST? increased, and ER and PR were not observed. The morphology of MCF-7/Taxol became larger and irregular, and the surface of natural cell was in the shape of floss, but MCF-7/Taxol was in the shape of bead. The intracellular of Taxol was observed in both MCF-7/Taxol and stoppage administrating. Conclusions MCF-7/Taxol cell subline was a typical multidrug resistant cell line which had basic characteristics of drug resistance cells. It was supposed that there was a cell subline which was tumor stem cell of MCF-7 included in this multidrug resistant cell line.

ABSTRACT

The ultrastructural characteristics of the mouse mammary adenocarcinoma cell line (MA782/5S-8102) have been studied by transmission electron microscope. The results show that the cells were irregular in the shape, with abundant microvilli on cell surface. The nucleus was abnormal in shape with the nuclear membrane sunk or swollen, the nucleus/cytoplasmic ratio (N/C) was high. There were many free ribosomes, abundant mitochondrias, dilated rough endoplasmic reticulum in the cytoplasm. Golgi apparatus, lysosomes and lipid droplets were present. Moreover, many cytoplasmic type-A particles and a few immatural type-B particles at the surface of the cell were observed.
