ABSTRACT

Due to the special geographical location and the complex ecosystem types, plateau wetlands play important ecological roles in water supply, greenhouse gas regulation and biodiversity preservation. Napahai plateau wetland is a special wetland type with low latitude and high altitude, and its microbial diversity was rarely studied. The diversity of microbial communities in the Napahai plateau wetland was analyzed using metagenomics method. Among the microbes detected, 184 phyla, 3 262 genera and 24 260 species belong to the bacterial domain, 13 phyla and 32 genera belong to the archaeal domain, and 13 phyla and 47 genera belong to the fungal domain. Significant differences in species diversity between soil and water were observed. Acidobacteria, Proteobacteria and Actinobacteria were dominant phyla in soil, while Proteobacteria and Bacteroides were dominant phyla in water. Since the carbon and nitrogen metabolism genes were abundant, the pathways of carbon fixation and nitrogen metabolism were analyzed. Calvin cycle, reductive tricarboxylic acid cycle and 3-hydroxypropionic acid cycle were the main carbon fixation pathways, while Proteobacteria, Chloroflexi, and Crenarchaeota were the main carbon-fixing bacteria group. As for the nitrogen cycle, nitrogen fixation and dissimilatory nitrate reduction were dominant in water, while nitrification and denitrification were dominant in soil. Proteobacteria, Nitrospirae, Verrucomicrobia, Actinobacteria, Thaumarchaeota and Euryarchaeota contributed to the nitrogen cycle. The study on microbial diversity of Napahai plateau wetlands provides new knowledge for the comprehensive management and protection of wetland environment in China.

ABSTRACT

Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease worldwide, and its incidence is increasing year by year. The mechanism of NAFLD is not fully understood, and it lacks effective prevention and treatment. Recent studies have found that butyrate, as a short-chain fatty acids (SCFAs), plays an important role in gene regulation, immunoregulation, inhibition of tumor, regulation of intestinal mucosal barrier, and reduction of oxidative stress. Several studies have shown that butyrate could alleviate NAFLD. This article reviewed the mechanism of butyrate in the pathogenesis of NAFLD and its application in the treatment of NAFLD, so as to provide a new idea for the prevention and treatment of NAFLD.

ABSTRACT

Objective To explore the feasibility of bioelectrical impedance analysis in assessing resting energy metabolism(REM)and the association of REM with anthropometry in hemodialysis patients.Methods Adult hemodialysis patients from 11 hemodialysis facilities in Guizhou Province were enrolled in this study.Bioelectrical impedance analysis was used to measure the rest metabolic rate(RMR)and body composition of 765 patients undergoing regular dialysis.A standard method was used to measure their height,weight,upper arm circumference,triceps skinfold thickness,left calf circumference and grip strength.The upper arm muscle circumference was also calculated.The level of RMR and body composition in hemodialysis patients were compared by gender grouping.The patients were then divided into four groups according to the RMR quartile.Spearman correlation analysis and multiple linear regression analysis were derived to analyze the relationship between RMR and anthropometry.Results The level of RMR in male patients was significantly higher than that in female patients[1591(1444,1764)Rcal/d and 1226(1104,1354)Rcal/d,P < 0.001].Compared with the first quartile of RMR group,upper arm circumference,upper arm muscle circumference,left calf circumference and grip strength in the fourth quartile group were increased significantly(all P < 0.05).Multiple linear regression analysis showed that height(β=0.572),left calf circumference(β=0.273),upper ann muscle circumference(β=0.092)and suffering from protein energy wasting(PEW,β=-0.184)were independent influential factors of RMR(all P < 0.05).Conclusions RMR levels in patients with maintenance hemodialysis are associated with left calf circumference and upper ann muscle circumference,which may become a new index to evaluate energy consumption and malnutrition in MHD patients.

AIM To study the interactions of component (group) compatibility of Sanwu Huangqin Decoction in incubation model of rat liver microsomes.METHODS With maackiain and ephedrine as internal standards,HPLC was adopted in the simultaneous content determination of total tlavonoids (baicalin,wogonoside,baicalein,wogonin and oroxylin A) from Scutellariae Radix and total alkaloids (oxymatrine,oxysophocarpine and matrine)from Sophorae flavescentis Radix in whole prescription component (group) compatibility and single medicinal material part (group) at seven time points (0,15,30,45,60,90 and 120 min),followed by the calculation of their in vitro metabolic rates.RESULTS In whole prescription component (group) compatibility,the contents of baicalin and wogonoside were first increased (0-0.5 h) and then decreased (0.5-2.0 h),those of baicalein,wogonin and oroxylin A showed increasing trends (more obvious within 0.5 h).The stable and gende metabolisms of various alkaloids reached balance within 20 min.CONCLUSION The bioavailability improvement and efficacy enhancement of total flavonoids from Scutellariae Radix may attribute to the compatibility with total alkaloids from Sophorae flavescentis Radix and total polysaccharides from Rehmanniae Radix in Sanwu Huangqin Decoction

ABSTRACT

Background Thyroid associated ophthalmopathy (TAO) is an autoimmune disease.Current research on the pathogenesis focuses on common autoantigen.Insulin-like growth factor-1 receptor (IGF-1R) is necessary for the function of IGF-1,also IGF-1 plays an important role in signaling pathway of thyroid stimulating hormone receptor (TSHR).Objective This study was to investigate the effects of IGF-1 on the proliferation,expression of IGF-1R and TSHR on cultured orbital fibroblasts (OFs) derived from TAO.Methods Human orbital tissue was obtained from 17 TAO patients who received orbital adipectomy and 4 normal controls who received cosmetic surgery in West China Hospital from March 2016 to June 2016.OFs were cultured by explant culture with DMEM/F12 containing 5% fetal bovine serum and identified by immunochemistry.The OFs were treated with different concentrations of IGF-1.IGF-1 at different concentrations (0,50,100,125 μg/L) was added into the medium,respectively,and the proliferation of the cells (absorbancy) was detected by MTS.The percentages of IGF-1R and TSHR expressions in the cells were assayed by flow cytometry.Results Cultured cells appeared to be spindle-like in shape and grew well with abundant cytoplasm.The characteristics of the cells derived from TAO patients were consistant with normal ones.The cells showed the positive response for vimentin and absent respose for desmin,S-100,myoglobin and cytokeratin.The proliferative values of OFs were gradually elevated with the increase of IGF-I dose in both TAO group and normal group (Fgroup =219.639,P<0.001;F ion =17.752,P<0.001) with the optimal effects in 100 μg/L IGF-1.The expression levels of IGF-1R in the OFs were (0.009 1 ±0.008 7)%,(0.095 3±0.023 3) %,(0.083 7±0.022 7) % and (0.070 9 ± 0.024 1) % in the TAO group,and those in the normal group were (0.0023± 0.0006)%,(0.0093±0.0012)%,(0.0073±0.0015)% and (0.0083±0.0012)% after treatment of 50,100,125 μg/L IGF-1.The expression levels of IGF-1 R were significantly higher after treatment of 50,100 and 125 μtg/L IGF-1 than those treatment of 0 μg/L IGF-1 in both TAO group and normal group,and the expression levels of IGF-1R in the OFs were significantly increased in the TAO group compared with the normal group (all at P<0.05).No statistical difference was seen in the TSHR expression between the TAO group and normal group after treatment of 0,50,100 and 125 μg/L IGF-1 (Fgroup =0.133,P > 0.05;F ion =0.004,P > 0.05).Conclusions IGF-1 can promote the proliferation of OFs and up-regulate the expression of IGF-1R in OFs.However,IGF-1 dose not play a regulating effect on the expression of TSHR in OFs.

ABSTRACT

A colite por exclusão é descrita como processo inflamatório que ocorre nos segmentos colorretais desprovidos do trânsito fecal. A deficiência dos ácidos graxos de cadeia curta vem sendo considerada como principal fator causal. OBJETIVO: O objetivo do presente estudo foi avaliar, em modelo experimental de colite de exclusão, a importância da irrigação do segmento desprovido de trânsito com soluções nutricionais na prevenção e tratamento do processo inflamatório. MÉTODO: Foram utilizados trinta ratos Wistar, machos, com peso inicial variando entre 350 e 500 gramas, submetidos à derivação do trânsito intestinal através da realização de colostomia proximal e fístula mucosa distal. Os animais foram divididos em três grupos de 10 animais segundo a irrigação do segmento excluso de trânsito ter sido realizada, empregando-se: Grupo SF: solução fisiológica a 0,9 por cento; Grupo GH: solução de glicose a 50 por cento; e Grupo AG: solução de ácidos graxos de cadeia curta. Em todos os animais, a irrigação do colo excluso foi realizada em intervalos de quatro dias sendo sacrificados sempre no 21º pós-operatório. Os fragmentos removidos dos segmentos intestinais foram corados pelas técnicas da hematoxilina-eosina e tricrômio de Masson. As variáveis histológicas estudadas foram: espessura da túnica mucosa, congestão vascular; infiltrado inflamatório e a deposição de colágeno. Os resultados encontrados foram submetidos a estudo estatístico considerando nível de significância de 5 por cento (p< 0,05). RESULTADOS: Verificou-se que no grupo onde se irrigou o cólon excluso com solução de ácidos graxos de cadeia curta houve menor congestão vascular, menor infiltrado inflamatório e menor deposição de colágeno quando comparado aos demais grupos experimentais. CONCLUSÃO: Os resultados do presente trabalho mostram que a irrigação de segmentos desprovidos de trânsito fecal com ácidos graxos de cadeia curta, encontra-se relacionada à melhora no processo inflamatório...


Diversion colitis is described as an inflammatory process that occurs in the colorectal segments excluded of the fecal stream. The deficiency of the short-chain fatty acids inside of intestinal lumen was considered the main etiologic factor. AIM: The aim of the present study was valued, in experimental model of diversion colitis, the importance of the irrigation of the segment without fecal stream with nutritional solutions in prevention and treatment of the colic mucosa inflammation. METHOD: There were used thirty Wistar males rats, with initial weight varying between 350 and 500 grammas, submitted to the derivation of the fecal stream through proximal colostomy and distal mucous fistula. The animals were divided in three groups with 10 animals according to the irrigation of the excluded colic segment had been carried out with: SF Group: 0.9 percent physiologic solution; GH Group: hypertonic glucose solution of 50 percent and AG Group: solution of short-chain fatty acids. In all the animals, the irrigation of the excluded colon was carried each four days and the animals were sacrificed ever in 21st post-operative day. The removed fragments of the intestinal segments were colored by the techniques of hematoxylin-eosin and Masson trichromium. The histological variables studied were: thickness of the mucous layer, vascular congestion; inflammatory infiltrate and the level of tissue collagen. The considered results were subjected to statistical study considering signification level of 5 percent (p <0.05). RESULTS: The obtained results showed that in the group where the excluded colon was irrigated with solution of short-chain fatty acids there was less vascular congestion, less inflammatory infiltrated and less collagen deposition when compared to another experimental groups.
