ABSTRACT

Herbicide(s) commonly known as weedkillers, are chemical substances used to control unwanted plants. Paraquat(PQ) (1, 1-dimethyl 4, 4- bipyridillium dichloride) is the active component of the herbicide (Parae Force) used inthis study.This study was performed on 25 male Wistar rats that were divided into 5 groups (A, B, C, D and E), Group A servedas the control group that received only distilled water, Group B, C, D & E served as the treated groups that received25%, 50%, 75%, & 100% leachate of Paraquat dichloride concentration respectively for 8 weeks.This study investigated the effect of paraquat dichloride on the testes, sperm characteristics and body weight inadult male Wistar rats. Histological analysis of the testes as well as semen analysis was carried out.Histomorphometry study of the testes showed deformed seminiferous tubules with irregular shape andspermatogenesis was arrested. Interstitial space was comparatively loose and disrupted Leydig cells are alsovisible. Seminiferous tubules also exhibiting degenerated basement membrane along with degenerated interstitialcells, degenerated spermatogonia and damaged Sertoli cells. Consequently, the sperm count from the Paraquatdichloride exposed semen revealed significant decreased results, the sperm morphology and sperm progressivitywere also affected negatively.In conclusion, the present study indicates that Paraquat dichloride demonstrated toxic effects on malereproductive parameters.

ABSTRACT

Objective Establish a method for qualitative and quantitative analysis of paraquat dichloride in organs in rat by UPLC-MS/MS and study the rat animal model poisoned by intragastric administration of paraquat dichloride to investigate the postmortem distribution of paraquat dichloride in poisoning death rat. Methods The rats were given an intragastric administration of 1/2LD50 Paraquat dichloride. The rats were dissected at 0.5h, 2h, 4h, 8h, 12h, 24h, 48h, 72h respectively after the intragastric administration. The specimens of -the heart, liver, spleen, lung, kidney, brain, muscle, bladder and stomach-were collected and analyzed immediately. Qualitative and quantitative analysis were performed by UPLC-MS/MS. Results Within 4h, stomach is the main distribution organ. The content of paraquat dichloride is the highest in stomache and relatively low in other organs. The concentration of organization except stomach changed little within 4h. The concentration of stomach has a sharp decline after 4h. The concentration in organs except stomach has a sharp rise after 4h. There is a significant difference(P<0.05) between each organs and brain. Conclusion There was a postmortem maldistribution of paraquat dichloride in poisoning death rats and the concentration in organs changes with time. The analysis method of UPLC-MS/MS and postmortem distribution of paraquat dichloride can be applied to the forensic identification of paraquat dichloride poisoning death and provide direction for delete this part toxicology analysis.

ABSTRACT

São apresentados dois casos de pacientes com lesões viscerais do fígado e rins consequentes ao manuseio inadequado de um agrotóxico, o dicloreto de paraquat 1,1'-dimetil-4,4'-dipiridilio dicloreto (Gramoxone) e chamam a atenção para um tipo especial de hepatite tóxica; descrevem o tipo de lesão hepática Q sua topografia intralobular, assim como lesões graves nos túbulos renais, com formação de sais de cálcio. São feitas considerações sobre as vias de absorção do tóxico e das áreas de predileção de atuação dentro do lóbulo hepático, assim como sobre o diagnóstico diferencial com outros tipos de hepatite tóxica (AU).
