ABSTRACT

Peroxisome proliferators-activated receptor-gamma(PPAR?),one of members of the nuclear hormone receptor superfamily of ligand-activated transcription factors, may affect the ocurrence and development of atherosclerosis(AS) through effective modulate lipocyte differentiation, improve glucose and lipoprotein decompensation and control inflammation reaction and so on.This review will focus on the recently development that relation on PPAR? and AS.

ABSTRACT

Aim To explore the role of a peroxisome proliferators-activated receptor gamma ligand,Rosiglitazone,in angiogenesis in human lung adenocarcinoma cells with reference to the regulation of vascular endothelial growth factor(VEGF).Methods Human lung adenocarcinoma A549 cell line was cultured in vitro.Expression of PPAR? and VEGF in A549 lung adenocarcinoma cells treated with different concentrations of Rosiglitazone was examined by semi-quantitative RT-PCR and immunohistochemical staining. Results PPAR? protein levels were higher in cultured A549 cells.RT-PCR and immunohistochemical staining showed that PPAR? mRNA and protein were enhanced in cells treated with Rosiglitazone in a dose-dependent manner, compared with those of untreated cells.Rosiglitazone had a potent inhibitory effect on the expression of VEGF in A549 cells dose-dependently in a range of concentrations. The effect was maximal with 10 ?mol?L~(-1) RSG and weaken over 20 ?mol?L~(-1) RSG,whereas 100 ?mol?L~(-1) RSG didnot have this down-regulation.GW9662,a PPAR? antagonist, partially blocked this effect of Rosiglitazone. Conclusions Activation of PPAR? suppresses angiogenesis in human lung adenocarcinoma. This action is probably associated with the down-regulation of angiogenic factor,VEGF,which may be modulated by PPAR?.These results suggest that PPAR? might be a novel molecular target for angiogenesis lung adenocarcinoma.

ABSTRACT

Objective To investigate the relationships of peroxisome proliferators activated receptor ?(PPAR?) and matrix metallo protease-7 ( MMP-7) with gastric cancer ( including low and high grade atypical hyperplasia). Methods One hundred and twenty gastric tissue specimens (four groups) were collected by gastroscopy or surgical operation, including 31 patients with chronic gastritis, 25 patients with low-grade atypical hyperplasia, 27 patients with high-grade atypical hyperplasia, and 28 patients with gastric cancer. The expression of PPAR? and MMP-7 were observed by immunohistochemical technique and analyzed by statistic methods. Results Both of them have high expression in the groups of gastric cancer respectively, and the high combined expressions of them were found in gastric cancer and high-grade atypical hyperplasia. Conclusion PPAR? and MMP-7 were the important differential signs of the gastric cancer and precancer by inference, the combined expression of PPAR? and MMP-7 was the evidences in differential diagnosis among gastric cancer, high and low grade atypical hyperplasia and gastritis.

ABSTRACT

PPAR? is a kind of nuclear receptors responsible for the regulation of glucose and lipid metabolisim and adipocyte difference, which is related with the pathogenesis of obesity, insulin resistance and type 2 diabetes mellitus.PPAR? agonists as insulin sensitizer have been used in the clinical treatment for type 2 diabetes for more than decades.Recently, with the further research plunging into the PPAR signaling pathway and insulin sensitizer, the recognition of this crucial nuclear receptor has been renewed and developed. The aim of this review is to report the update research outcomes.
