ABSTRACT

Introducción. Los tumores neoplásicos se caracterizan por su invasividad y metástasis. Las células neoplásicas tienen heterogeneidad genética, por lo cual pueden desarrollar resistencia a los quimioterápicos. Por esta razón, las plantas continúan siendo una fuente importante de nuevos productos anticancerígenos. Objetivo. Evaluar la actividad citotóxica y antiproliferativa de un extracto rico en fucoidan de Lessonia trabeculata nativa (EFLt) sobre la línea celular de adenocarcinoma mamario murino, triple negativo 4T1. Métodos. La citotoxicidad y la IC50 se determinaron en monocapas de 4T1 empleando el reactivo MTT. Para demostrar la actividad antiproliferativa se aplicaron los métodos de cierre de herida y anticlonogénico utilizando las IC50 del EFLt y Dox (doxorubicina). El cierre de herida fue evaluado mediante barrido de tiempos discretos; el efecto anticlonogénico fue evaluado 7 días postratamiento mediante el conteo de colonias y se determinó la fracción de sobrevivencia. Adicionalmente, se evaluaron la citotoxicidad y actividad antiproliferativa combinando las IC50 de EFLt y Dox. El porcentaje de migración y conteo de colonias se realizó con el programa ImageJ. Resultados. La IC50 del EFLt (950 μg/mL) produjo 56% de citotoxicidad, 80,3% de inhibición de la migración celular, 68% de inhibición en la formación de colonias. La IC50 de Dox fue 0,5 μg/mL. Conclusiones. El EFLt ejerce citotoxicidad dependiente de la concentración y tiene efecto antiproliferativo sobre 4T1. Se requiere continuar los ensayos en modelos de mayor complejidad que permitan esclarecer el potencial antitumoral del EFLt.


Introduction. Neoplastic tumors are characterized by invasiveness and metastasis. Neoplastic cells are genetically heterogeneous and can develop resistance to chemotherapeutic agents. For this reason, plants continue to be an important source of new anticancer products. Objective. To determine the cytotoxic and antiproliferative activity of a fucoidan-rich extract of native Lessonia trabeculata (EFLt) on the tripe negative murine mammary adenocarcinoma cell line 4T1. Methods. Cytotoxicity and IC50 were determined in 4T1 monolayers using the MTT reagent. To demonstrate antiproliferative activity, "wound" closure and anticlonogenic methods were applied using the IC50 of EFLt and Doc (doxorubicin). "Wound" closure was evaluated by discrete times sweep to determine percentage inhibition of cell migration; the anticlonogenic effect was evaluated by colony counting 7 days after treatment and the survival fraction was determined. In addition, cytotoxicity and antiproliferative activity were evaluated by combining the IC50 of EFLt and Dox. Percent migration and colony counts were performed using ImageJ software. Results. The IC50 (950 μg/mL) of EFLt was 56% cytotoxicity, 80,3% inhibition of cell migration, 68% inhibition of colony formation.The ICof Dox was 0,5 μg/mL. Conclusions. EFLt exerts concentration-dependent cytotoxicity and antiproliferative effect on 4T1. Further studies in more complex models are needed to elucidate the anti-tumor potential of EFLt.

Tubeimoside-1 (TBMS1) is a triterpene saponins active components with large content in Cucurbitaceae plant Fritillaria, which is water-soluble and stable. It has a broad inhibitory effect on lung cancer, colorectal cancer, breast cancer, gastric cancer and other tumors. The mechanism is mainly related to the inhibition of tumor cell growth, induction of tumor cell apoptosis, inhibition of tumor cell invasion and metastasis, induction of cell autophagy, and inhibition of tumor angiogenesis.

ABSTRACT

This study aims to evaluate effects of inclusion of moringa in Japanese quail diets on laying performance, egg quality, blood parameters, serum biochemical profile, and behavior. One hundred and forty-four Japanese quails, approximately 35-d-old, were distributed in a completely randomized design with four treatments and six replications. Treatments were diet inclusion of 0, 2, 4 and 6% of dried and ground moringa leaves. The inclusion of moringa in diets reduces the quadratic feed intake up to the level of 1.20%, increases weight of eggs with a quadratic behavior up to 3.80%, and linearly increases yolk weight. Yolk color changes with higher levels of inclusion of moringa and resulted in more intense colors. The biochemical profile of quails changed slightly but remains within the normal range. The inclusion of 4% of moringa in diets increases alkaline phosphatase. Regarding cholesterol and triglycerides, diet with 6% moringa inclusion was lower when compared to the others. The behavior of laying quails does not change due to inclusion of moringa in diets. Up to 3.83% of Moringa oleifera can be included in Japanese quail diet to improve egg quality without compromising performance, biochemical profile, blood parameters and behavior.(AU)


O objetivo deste estudo foi avaliar a inclusão de Moringa oleifera na dieta de codornas japonesas sobre o desempenho, a qualidade de ovos, os parâmetros sanguíneos, o perfil bioquímico e o comportamento. Foram utilizadas 144 codornas japonesas, com aproximadamente 35 dias de idade, as quais foram distribuídas em delineamento inteiramente ao acaso, com quatro tratamentos e seis repetições cada. Foram fornecidas dietas com 0, 2, 4 e 6% de folhas desidratadas e moídas de moringa. A inclusão de moringa nas dietas reduziu quadraticamente o consumo até o nível de 1,2%, aumentou o peso dos ovos com comportamento quadrático até o nível de 3,8% e aumentou linearmente o peso da gema. A coloração da gema se intensificou com maiores inclusões de moringa. O perfil bioquímico sofreu alterações leves, mas não saiu dos padrões normais para codornas. A inclusão de 4% de moringa nas dietas aumentou os níveis de fosfatase alcalina. Já a inclusão de 6% reduziu níveis de colesterol e triglicérides. O comportamento das codornas não se alterou com a inclusão de moringa nas dietas. A inclusão de até 3,83% de Moringa oleifera na dieta de codornas japonesas aumenta a qualidade de ovos sem comprometer o desempenho, o perfil bioquímico, os parâmetros sanguíneos e o comportamento delas.(AU)

ABSTRACT: This study aimed to analyze the effect of phytogenic additives based on cardol, cardanol, and ricinoleic acid on the physico-chemical qualities and biochemical parameters of Holstein cow milk. Nineteen animals were divided into the control (GI) and treatment (GII) groups. Prior to the beginning of the experiment, sodium monensin was removed from the feed provided to the animals. This study consisted of two phases. In the first, animals from GI received the standard diet, without additives, while animals from GII received the standard diet supplemented with 10 g of phytogenic additives for 60 days. At the end of the first stage, the animals remained 30 days without receiving additives and in the second phase the groups were inverted, and the GII again received the phytogenic additive during the subsequent 60 days. Physical-chemical analyzes of milk were performed at moments 0, 15, 30, 45 and 60 days of the experiment and the blood parameters at times 0 and 60 days of the experiment were evaluated. There was no significant difference for the treatment x time interaction in any of the variables, but there was a trend between groups for the CCS parameter and there was a statistical difference for the protein, as well as for the AST. In this study it is observed that the addition of phytogenic additive modulating rumen fermentation based on cardol, cardanol and ricinoleic acid does not compromise, in general, the quality of milk, with positive results for some specific parameters such as protein content and aminotransferase.


RESUMO: Esse estudo teve por objetivo avaliar o efeito de um aditivo fitogênico a base de cardol, cardanol e ácido ricinoléico na qualidade físico-química do leite e perfil bioquímico de vacas da raça Holandês. Dezenove animais foram divididos em dois grupos: Grupo Controle (GI) e Grupo Tratado (GII). Previamente ao início do experimento, retirou-se a monensina sódica da ração fornecida aos animais. Este estudo consistiu de duas fases, sendo que na primeira fase, os animais do GI receberam a dieta padrão, sem aditivos, enquanto os animais do grupo GII receberam a ração padrão suplementada com 10 g dos aditivos fitogênicos durante 60 dias. Ao final da primeira etapa, os animais permaneceram 30 dias sem receber aditivos e na segunda fase os grupos foram invertidos, sendo que o GII novamente recebeu o aditivo fitogênico durante os 60 dias subsequentes. Foram realizadas análises físico-químicas do leite nos momentos 0, 15, 30, 45 e 60 dias do experimento e avaliados os parâmetros sanguíneos nos momentos 0 e 60 dias do experimento. Não se observou diferença significativa para a interação tratamento x tempo em nenhuma das variáveis, mas tendência entre grupos para o parâmetro de CCS, e houve diferença estatística para a proteína, assim como para a AST. Neste estudo observa-se que a adição de aditivo fitogênico modulador de fermentação ruminal a base de cardol, cardanol e ácido ricinoléico não compromete, de forma geral, a qualidade do leite, tendo resultados positivos para alguns parâmetros específicos como teor de proteína e aspartato aminotransferase.

ABSTRACT: The food industry is in a continuous search for strategies of reducing of antibiotic residues in livestock production, given the development of antibiotic-resistant bacteria. In that context, there is a consistent increasing in the usage of alternative feed phytogenic additives. Due to the pharmacological, the plants Marrubium vulgare and Leonurus sibiricus, are both potential replacements for growth-promoting antibiotics. This study evaluated the productive performance and analyze the sensory qualities in the meat of broiler that were administered M. vulgare and L. sibiricus infusions. A total of 320 chicks from an poultry farm were distributed in 8 groups groups according treatments by 28 days: Control Group (Saline); Positive control (tylosin 1%, 50 ppm); Groups of M. vulgare (MV) infusion: 2% (MV2), 4% (MV4) and 6% (MV6); and groups treated with L. sibiricus (LS) infusion: 2% (LS2), 4% (LS4) and 6% (LS6), all of them received relative doses corresponding to the live weight percentage in milliliter. There was weght gain in MV6 and LS6 group and a better food conversion in MV4, MV6, LS4, and LS6. In addition, weight gain and enhancement of the pectoral muscles were observed in these groups. The sensory analysis revealed that the broilers' meat of MV had a more intense flavor. For marketing purposes the meat from birds that received LS had a better demand. Improvement of productive performance and sensory qualities of the broilers meat might be associated with the phytobiotic activity in it. Therefore, MV and LS can be used as natural growth promoter in broilers.


RESUMO: A indústria de alimentos busca a redução de resíduos de antibióticos na criação intensiva de animais de produção devido ao desenvolvimento de bactérias resistentes. Nesse contexto, tem havido um aumento no uso de aditivos fitogênicos alternativos nas rações. Devido às suas propriedades farmacológicas demonstradas anteriormente as plantas Marrubium vulgare e Leonurus sibiricus tem potencial como aditivos em substituição aos antibióticos promotores de crescimento. O objetivo desse estudo foi avaliar o desempenho produtivo e analisar as qualidades sensoriais da carne de frangos que receberam a administração da infusão da M. vulgare (MV) e da L. sibiricus (LS). 320 pintainhas de um criatório industrial foram distribuídas em oito grupos experimentais: Grupo Controle (Salina); Controle Positivo (tilosina 1%, 50 ppm); Grupos tratados com MV: 2% (MV2), 4% (MV4) e 6% (MV6); e grupos tratados com a LS: 2% (LS2), 4% (LS4) e 6% (LS6), e todos eles receberam doses relativas correspondendo à percentagem de peso vivo, em mililitros. Houve maior ganho de peso no 28o dia nos grupos MV6 e LS6 e melhor conversão alimentar em MV4, MV6, LS4 e LS6. Além disso, foi observado aumento no peso e circunferência dos músculos peitorais nesses grupos. A análise sensorial revelou que amostras de carne do grupo MV tiveram sabor mais intenso do que no grupo controle. Na intenção de compra, a carne dos animais que receberam LS teve maior preferência. A melhoria do desempenho produtivo e das características sensoriais da carne de frangos pode estar associada à atividade fitobiótica. Assim, MV e LS podem ser usados como promotores de crescimento naturais para frangos de corte.

Abstract Background: Coccidiosis is the infectious disease with the greatest economic impact in poultry production. Additionally, chemotherapeutic growth promoters are being substituted for safer alternative strategies. Objective: To evaluate the effect of feeding oleoresins from chili pepper (Capsicum annuum) and turmeric (Curcuma longa L.) on the performance, survival rate, and the Productive Efficiency Index of broilers from 1 to 21d challenged by Eimeria sp. Methods: A total of 700 male 1-day-old chicks (Cobb 500) challenged with Eimeria sp, were assigned to a completely randomized design with four treatments: (1) negative control diet, without anticoccidial agent or growth promoters; (2) control diet+0.05% salinomycin; (3) negative control diet+100g/ton; and (4) negative control diet +140g/ton of test product (Curcuma longa L. plus Capsicum annuum oleoresins). Results: Body weight, weight gain, feed conversion and Productive Efficiency Index did not differ between broilers fed anticoccidial chemotherapeutics and oleoresins. In addition, there was no difference in feed intake and survival rate between dietary treatments. Conclusion: Broilers supplemented with chili pepper and turmeric oleoresins from 1 to 21d present similar body weight, weight gain, feed conversion, and Productive Efficiency Index compared to broilers supplemented with chemotherapeutic anticoccidials without affecting feed intake or survival rate.


Resumen Antecedentes: El impacto económico generado por la coccidiosis en el sector avícola es el mayor en relación con otras enfermedades. Adicionalmente, los promotores de crecimiento quimioterapéuticos se han venido reemplazando por estrategias alternativas. Objetivo: Evaluar el efecto de oleorresinas de pimiento (Capsicum annuum) y cúrcuma (Curcuma longa L.) sobre el desempeño zootécnico, tasa de supervivencia y el Índice de Eficiencia Productiva en pollos de engorde de 1 a 21 dias desafiados con Eimeria sp. Métodos: Se utilizaron 700 pollos machos (Cobb500) de 1 día de edad, distribuidos en un diseño completamente al azar con cuatro tratamientos: (1) dieta control negativo sin agentes anticoccidianos ni promotores de crecimiento; (2) dieta control+0,05% de salinomicina; (3) dieta control negativa+100g/ton; y (4) dieta control negativa +140g/ton del producto a evaluar (oleorresina de Curcuma longa L. y Capsicum annuum). Resultados: Se obtuvieron resultados similares repecto a peso corporal, ganancia de peso, conversión alimenticia e Índice de Eficiencia Productiva en los tratamientos que contenían anticoccidiano quimioterapéutico y los que contenian oleorresinas. Además, no se observaron diferencias en el consumo de alimento ni en la tasa de supervivencia entre los tratamientos dietarios. Conclusión: Los pollos de engorde suplementados entre 1 y 21 d con oleorresinas de pimiento y cúrcuma presentan similar peso corporal, ganancia de peso, conversión alimenticia e Índice de Eficiencia Productiva en comparación con los suplementados con anticoccidianos quimioterapéuticos, sin afectar el consumo de alimento ni la viabilidad.

Objetivo: Avaliar o efeito das oleoresinas da pimenta Chilli (Capsicum annuum) e açafrão-da-terra (Curcuma longa L.) no desempenho zootécnico, taxa de sobrevivência e Índice de Eficiência Produtiva de frangos de corte de 1 a 21d desafiados por Eimeria sp Métodos: Foram utilizados 700 pintos de 1d, machos (Cobb500) distribuídos em delineamento inteiramente casualizado, em quatro tratamentos: (1) dieta controle negativo, sem agente anticoccidiano ou promotores de crescimento; (2) dieta controle+0,05% de salinomicina; (3) dieta controle negativa+100g/ton; e (4)+140g/ton de produto teste (oleoresina de Curcuma longa L. mais Capsicum annuum) e desafiados por Eimeria sp. Resultados: Houve resultados similares para peso, ganho de peso, conversão alimentar e Índice de Eficiência Produtiva entre frangos alimentados com dietas contendo anticocidiano quimioterápico e dietas contendo oleoresinas. Além disso, não houve diferença para consumo de ração e a taxa de sobrevivência entre os tratamentos dietéticos. Conclusão: Frangos de corte alimentados com dietas suplementadas com oleorresinas de pimenta chili e açafrão-da-terra apresentaram resultados similares para peso, ganho de peso, conversão alimentar e Índice de Eficiência Produtiva que frangos de corte alimentados com dietas suplementadas com anticoccidianos quimioterápicos de 1 a 21d, sem afetar o consumo de ração e a taxa de sobrevivência dos animais.

ABSTRACT

In present experiment 288 day-old commercial broiler chicks were subjected to eight dietary treatments consisting of three replicates of twelve chicks in each replicate were reared for a period of 6 weeks in winter season. The dietary treatments includes: T0- Negative control, T1-control, T2-0.5 % Amla + 0.5 % Ashwagandha + 0.25 % Turmeric powder, T3-0.25 % Amla + 0.5% Ashwagandha + 0.25% Turmeric powder, T4-0.125% Amla + 0.5 % Ashwagandha + 0.25 % Turmeric powder, T5- 0.5 % Amla + 0.25 % Ashwagandha + 0.25 % Turmeric powder, T6-0.5 % Amla + 0.125 % Ashwagandha + 0.25 % Turmeric powder and T7-0.5 % Amla + 0.5 % Ashwagandha + 0.125 % Turmeric powder. Standard managemental practices were followed during the experimental period. Supplementation of phytogenic mixture consisting of 0.5 % amla and 0.5 % Ashwagandha with 0.25% or 0.125 % turmeric powder (T2 and T7) gave best results and improved the giblet percentage, eviscerated weight percentage and drawn percentage in winter season. The cost of production of drawn weight (`/kg) of broilers at six weeks of age was reduced as much as ` 10.23 (T2) and ` 9.48 (T7) in winter season. The study concluded that addition of 0.5 % amla and 0.5 % Ashwagandha with 0.25% or 0.125 % turmeric powder can be effectively supplemented as an alternative to antibiotics growth promoter in poultry ration in winter for improving the carcass characteristics and such ration was found more economical in comparison to that having antibiotic growth promoter

@#Objective To investigate the inhibitory effect and molecular mechanism of delphinidin (Dp) on HER-2 positive breast cancer. Methods The HER-2 positive breast cancer cells (MDA-MB-453 and BT-474) were treated with different concentrations of delphinidin (10,20,40,80 and 160 μmol/L) for 48 hours. The same concentration of DMSO was used as the solvent control. CCK-8 method was used to measure the effect of Dp on cell activity, and half maximal inhibitory concentration (IC50) was calculated to determine the concentration of subsequent experiments. The cells were treated with different concentrations of Dp (20, 40 and 80 μmol/L) for 48 hours. HE staining was used to observe the cell morphological changes. Flow cytometry was used to analyze the cell cycle. Cell apoptosis rate was detected by TUNEL assay. The phosphorylation levels of NF - κB signaling pathway related proteins were determined by Western blot assay. Results Delphinidin inhibited the proliferation of MDA-MB-453 and BT-474 in the concentration ranges of 10,20,40,80 and 160 μmol/L (IC50: 41.02 and 60.97 μmol/L). In the concentrations of 20,40 and 80 μmol/L, compared with the control group, it was found that some cells were detached, floated and lysed, and the cell volume was decreased, the proportion of cells in G2/ M phase and the apoptosis rate were increased in DP treatment groups. Compared with the control group, the expression levels of p-NF-κB/p65, p-IκBα, p-IKKα/β and p-PKCα were significantly decreased in the 40 and 80 μmol/L Dp treatment groups, while the expression levels of IκBα, IKKα, IKKβ and PKCα were increased in the Dp treatment groups (P<0.05). Conclusion Delphinine can inhibit the proliferation of breast cancer cells by blocking the NF-κB signaling pathway and inducing the G2/M cycle arrest and apoptosis of MDA-MB-453 and BT-474 cells. ABSTRACT

Objective: To investigate the expression level of chromosomal region maintenance 1 (CRM1) in mantle cell lymphoma, and to explore the possible mechanism of naturally derived sulforaphene LFS-01 inhibiting the proliferation of mantle cell lymphoma cells. Methods: The expression of CRM1 in mantle cell lymphoma was analyzed by Oncomine data mining. After treatment with LFS-01, the viability of mantle cell lymphoma JeKo-1 cells was detected by CCK-8 assay, the nuclear transport function of CRM1 protein was measured by laser confocal microscopy, the expression of CRM1 protein was detected by Western blotting, the cell cycle arrest and apoptosis were analyzed by FCM and transmission electron microscopy, the expression change of apoptosis pathway-related proteins was detected by Western blotting, and the impact of caspase inhibitor z-VAD-FAM on the effects of LFS-01 was detected by CCK-8 assay and FCM. Lentiviral infection was used to establish a stable JeKo-1 cells expressing mutant CRM1 (C528S), then the effects of LFS-01 on the nuclear transport function of CRM1 and the proliferation of JeKo-1 cells were detected by laser confocal microscopy and CCK-8, respectively. The transcriptional level in JeKo-1 cells after LFS-01 treatment was detected by RNAseq, and the viability of JeKo-1 cells treated with Toll-like receptor (TLR) inhibitor TAK-242 and LFS-01 was measured by CCK-8 assay. Results: CRM1 was overexpressed in mantle cell lymphoma (P < 0.05). LFS-01 inhibited the proliferation of JeKo-1 cells, and the 50% inhibitory concentration (IC50) at 24 h and 48 h were 5.81 μmol/L and 9.09 μmol/L, respectively. 20.0 μmol/L LFS-01 inhibited the nuclear transport function of CRM1 (P < 0.05), 6.0 μmol/L LFS-01 down-regulated the expression level of CRM1 (P < 0.05), 10.0 μmol/L LFS-01 induced cell cycle arrest at G2/M phase and induced apoptosis (both P < 0.05), and 4 μmol/L LFS-01 up-regulated the expression levels of poly ADP-ribose polymerase (PARP), cleaved caspase-3 and cleaved caspase-9 (all P < 0.01). The caspase inhibitor z-VAD-FAM reversed the apoptosis-induction effect of LFS-01 (P < 0.01). CRM 1 mutation eliminated the effects of LFS-01 on CRM1 function and cell proliferation (both P < 0.05). LFS-01 inhibited the cell proliferation-related pathways (P < 0.01), and TLR inhibitor TAK-242 combined with LFS-01 had synergetic inhibitory effect on JeKo-1 cells (P < 0.01). Conclusion: LFS-01 can inhibit the growth of mantle cell lymphoma cells through inducing cell cycle arrest and apoptosis, and CRM1 is essential in this process. In addition, LFS-01 and TLR inhibitor TAK-242 have synergetic inhibitory effect on mantle cell lymphoma cells.

The inhibiting mechanisms of brucine to cancer cells included inducing the apoptosis of cancer cells, inhibiting the proliferation of cancer cells, preventing cell adhesion and transfer. This paper reviews the function of brucine and its anti-tumor preparations, and compares the efficacy and adverse reactions between brucine related preparations and traditional drugs to evaluate its anti-tumor effects. Furthermore, more exploration on the inhibiting mechanisms is necessary for the patients to obtain safe and effective drugs.

ABSTRACT The aim of this study was to evaluate the effect of Syzygium cumini leaves (SCL) in laying hens diets on productive performance, egg quality and lipid oxidation of yolk. A total of 108 Hysex White laying hens were distributed in a completely randomized design with three treatments and six replicates of six birds each. The treatments consisted of SCL inclusion at dietary levels of 0, 5 and 10 g/kg. There was no significant effect of SCL inclusion on feed intake, laying percentage, weight and egg mass, feed conversion ratio, Haugh units, specific gravity, percentage of yolk, albumen and egg shells and shell thickness. However, the inclusion of SCL significantly influenced the yolk color and yolk lipid oxidation measured by TBARS values. Yolk color increased and TBARS values ​​decreased with the inclusion of SCL. The inclusion of SCL in laying hens diets improves pigmentation and lipid stability of yolk.

ABSTRACT

Objective To isolate and identify anti-tumor constituents from Lysimachia christinae Hance.Methods The compounds were isolated and purified by chromategraphy on AB-8 macroporous resin, silica gel, Sephadex LH-20, ODS and preparative HPLC.Their structures were elucidated using chemical and spectroscopic methods.All thecompounds obtained were evaluated on their growth inhibitory effects in vitro using the MTT method.Results Eleven compounds were isolated from the whole plant of L.christinae Hance and were identified as 3,3'-dimethoxy-6,6'-di[(Z)-pentadec-10-en-1-yl]-[1,1'-bi(cyclohexane)]-3,3',6,6'-tetraene-2,2',5,5'-tetraone(1),physcion(2),munjistin(3), 9,16-dioxooctadec-10,12,14-trienoic acid(4),cyclamiretinA-3-O-{β-D-xylopyranosyl-(1→2)-β-D-glucopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→4)]-α-L-arabinopyranoside(5),lysikoianoside(6),quercetin-3-O-glucopyranoside(7),isorhamnetin3-O-β-D-galactopyranoside(8),3',4',5',5,7-pentahydroxyflavone(9), rutin(10), and quercetin(11).The screening results of antitumor activity in vitro showed that IC50 of compound 5 for human cervical carcinoma cells HeLa, human osteosarcoma cells U20S, human lung adenocarcinoma cells PC-9,and human colorectal carcinoma cells CT-26 was 2.29, 1.22, 1.43, and 1.29 μmol/L respectively.Conclusion Compound 1 is a new compound and compounds 2-4 are isolated from the plants of Lysimachia for the first time.Compound 5 shows obvious inhibitory effects on tumor cell growth in vitro.

Dois experimentos foram realizados com o objetivo de avaliar o uso das sementes secas de Piper cubeba nas dietas de frangos de corte de 1 a 21 dias de idades e seus efeitos sobre o perfil bioquímicos do sangue e na biometria dos órgãos das aves. Em cada experimento 240 pintos de corte machos, com um dia de idade da linhagem Cobb foram distribuídos em um delineamento inteiramente casualizado, com cinco tratamentos e quatro repetições de 12 aves por parcela experimental. No primeiro experimento foi avaliado o uso da Piper cubeba em dietas consideradas de alta digestibilidade, a base de milho e farelo de soja, e no segundo avaliou-se o uso da pimenta em dietas de baixa digestibilidade, as quais foram obtidas com a inclusão de farinha de carne e ossos. Com relação aos dados de perfil bioquímico sanguíneo do experimento I, com exceção da gama glutamil transferase, todos os demais níveis séricos se apresentaram dentro dos limites recomendados para aves, e apenas os níveis de triglicerídeos diferiram entre os tratamentos (P<0,05). No experimento II houve diferença significativa para os níveis de albumina, colesterol e ácido úrico, porém isso não foi resultado de desordens metabólicas, visto que com exceção da gama glutamil transferase, todos os níveis encontraram-se dentro dos limites recomendados. Em ambos os experimentos não foram observadas diferenças (P>0,05) para a biometria dos órgãos. Nas condições em que os experimentos foram realizados a inclusão das sementes de Piper cubeba na ração para frangos de corte não proporcionou alterações bioquímicas e biométricas que possam limitar seu uso como material vegetal para os estudos fitogênicos.(AU)


Two experiments were conducted to evaluate the use of dried seeds of Piper cubeba in the diets of 1 to 21-day-old broilers and its effect on biochemical blood profile and biometry of the organs. In each experiment, 240 one-day-old male broiler chicks Cobb were distributed in a completely randomized design with five treatments and four replicates of 12 birds per experimental plot. The first experiment evaluated the use of Piper cubeba in considered highly digestible diets based on corn and soybean meal, and the second evaluated the use of pepper in low digestibility diets, which were obtained with the inclusion of meat and bone meal. Regarding the blood chemistry profile data of the experiment I, with the exception of gamma glutamyl transferase, all other serum levels were within recommended limits for poultry, and only triglyceride levels differed between treatments (P <0.05). In the second experiment there was significant difference in albumin levels, cholesterol and uric acid, but this was not the result of metabolic disorders, because except for the gama glutamyl transferase levels, all variables were within recommended levels. In both experiments, there were no differences (P> 0.05) for the biometry of organs. The conditions under which the experiments were performed at inclusion of Piper cubeba seeds in feed for broilers has not provided any toxicity to poultry.(AU)

Based on our research and other references,this article discussed the antitumor effect of Ganoderma mediated by immunological mechanism, including promoting the function of mononuclear-macro?phages,and natural killers,promoting maturation and differentiation of dendritic cells,increasing its antigen presentation,activating lymphocytes and increasing cytotoxicity of cytotoxin T lymphocyte, promoting production of cytokines,inhibiting tumor escape from immune surveillance. Also, clinical studies with immunological indexes were reviewed.

ABSTRACT

Objective: To investigate the antitumor effect of sanguinarine (SAN) in vivo and its possible mechanism. Methods: The tumor-bearing BALB/c male nude mouse models were established by subcutaneous inoculation of gastric cancer GTL-1 6 cells. The mouse models were divided into three groups [14 mice in each group were intragastrically administered with 0.9% NaCl solution (as the control group), 2.5 mg/kg SAN and 5.0 mg/kg SAN every other day for 7 times, respectively]. The general status of the mice was observed, and the tumor volume was measured. After 28 days, the mice were sacrificed, then the tumor volume and weight were recorded. The histopathologic examination of tumor tissues was performed using hematoxylin-eosin (HE) staining. Then the protein expression of epidermal growth factor receptor (EGFR) in tumor tissues was detected by Western blotting. Results: As compared with the control group, the tumors grew slowly in SAN treatment groups. At autopsy, the volume and weight of tumors in SAN treatment groups were all smaller than those in the control group (P < 0.01), and the tumor volume and weight were smallest in 5.0 mg/kg SAN group (P < 0.01). The result of histopathologic examination revealed that SAN treatment could reduce the accumulation of tumor cells and epidermal invasion of tumor cells. The expression levels of EGFR were significantly lower in SAN treatment groups than that in the control group (P < 0.05). Conclusion: SAN inhibits the growth of tumor in nude mice inoculated with gastric cancer GTL-16 cells, and which may be related to the downregulation of EGFR expression.

Background: The rainforest is an important source of natural compounds with therapeutic properties. Although there are many anti-inflammatory and antineoplastic drugs available to the clinician, there is an ongoing need for new therapeutic drugs with fewer serious adverse effects. Aim: To evaluate the in vitro cytotoxic effects of lupeol and casearin G on tumor cells, on phagocytic activity and nitric oxide (NO) production by blood mononuclear cells. Material and Methods: The cytotoxic effect of these compounds on cell lines MCF-7 (human breast adenocarcinoma) and PC-3 (human prostate cancer) was measured by a colorimetric assay (MTS/PMS) and the sulphorhodamine B assay. Peripheral blood mononuclear cells were obtained from eight healthy volunteers. The effect of these compounds on nitric oxide (NO) production was measured using the Griess reaction. Their effect on phagocytic activity of PBMC was also evaluated. Results: Lupeol (≥ 2 mM) resulted in a reduction of both the phagocytic index and the percentage of phagocytic monocytes and macrophages. Treatment of monocytes/macrophages with lupeol (72 µM) and casearin G (4 µM) reduced the production of NO. Neither lupeol (< 969 µM) nor casearin G (< 55 µM) had cytotoxic effects on PBMC. Casearin G showed both cytotoxic (IC50, LC50) and cytostatic (GI50) effects against tumor cells, PC-3 (IC50 = 12.5 µM; GI50 = 13.3 µM; LC50 = 51.9 µM) and MCF-7 (IC50 = 112.8 µM; GI50 = 11.8 µM; LC50 = 49.4 µM), as well as a hemolytic effect (≥ 182 µM). Conclusions: These observations indicate that lupeol and casearin G might be useful compounds in the preparation of anti-inflammatory drugs, whereas casearin G might be useful in the elaboration of antitumor drugs.

Plant extracts are kinds of important anticancer drugs.Anticancer drugs research has focused on several important plant extracts such as resveratrol,ricin,epigallocatechin-3-gallate,oridonin,juglone,quercetin,matrine,nordihydroguaiaretic acid.In vitro and in vivo studies have shown that their antitumor mechanisms including induction of apoptosis,cytotoxicity,induced redifferentiation,antiangiogesis,antiinvasion and anti-metastasis.Plant extracts have multi-link and multi-target.Part of them have been used in preclinical studies.In-depth study of the anti-tumor mechanisms of plant extracts on cancer treatment is important,and the full mechanism needs further study.

Hedgehog signaling pathway is not only related with the development of embryo, formation of organ, and the regeneration and repair of tissues in adults, but also plays an important role in the growth and metastasis of tumor cells. Aberrant activation of this pathway has been found to be involved in the development of cancer. Understanding the mechanism of Hedgehog signaling pathway in various tumors is critical to the development and the use of specific agents targeting this pathway. GDC-0449 is the first small-molecule Smo-inhibitor entering clinical trials, and it shows clinical benefit. This article summarizes the relationship between the Hedgehog signaling pathway and the progression of tumor, and addresses the practical application of this pathway in tumor therapy.

ABSTRACT

Objective: To investigate the pro-apoptotic effect of gambogenic acid (GNA) on human colonic carcinoma HCT116 cells, and to explore the possible molecular mechanism. Methods: MTT assay was performed to detect proliferation inhibition effect of GNA on HCT116 cells. After staining with DAPI, the pro-apoptotic effect of GNA on HCT116 cells was observed under a fluorescence microscope. The changes in cell cycle distribution of HCT116 cells induced by GNA were examined by FCM. The expressions of cyclin D1, cyclin E, P21, P27 and poly (ADP-ribose) polymerase (PARP) proteins in HCT116 cells were detected by Western blotting. Results: GNA exerted an significant inhibitory effect on HCT116 cell proliferation in a time-and dose-dependent manner, and it could induce the typical nuclear apoptotic morphology. Under a fluorescence microscope, DAPI staining results showed that GNA could obviously induce the apoptosis of HCT116 cells. FCM showed that GNA could significantly increase the percentage of cells at G0/G1 phase, whereas obviously decrease the percentage of cells at S phase, which indicated that the cell cycle was arrested at G 0/G1 phase. Western blotting analysis showed that GNA could efficiently down-regulate the expression levels of cyclin D1 and cyclin E proteins, whereas up-regulate the expression levels of P21 and P27 proteins, and also promote the cleavage of PARP. Conclusion: GNA can induce the cell cycle arrest at G0/G1 phase by down-regulating the expression levels of cyclin D1 and cyclin E, and up-regulating the expression levels of P21 and P27. Therefore, GNA can efficiently promote the cell proliferation inhibition and the apoptosis of HCT116 cells. Copyright© 2011 by TUMOR.
