ABSTRACT

Objective To investigate the effects of polysaccharide sulfate(PSS)on lipopolysaccharide(LPS)-induced mice pulmonary microvascular endothelial cell(PMVEC) injury in vitro .Methods PMVEC were divided into the blank control group (C) ,LPS stimulation group (L) and PSS+ LPS group (LP) .The effect of PSS on the viability of LPS-induced PMVEC was ob-served by MTT assay .The influence of polymorphonuclear(PMN) on the PMVEC adhesion number was measured by the rose Ben-gal staining .The concentrations of TNF-αand ICAM-1 in culture supernatant of PMVEC were detected by ELISA assay .Results PSS could inhibit the decrease of PMVEC viability caused by LPS(P=0 .001) ,compared with the group C ,the adhesion number of PMVEC and PMN in the group L was significantly increased(P=0 .000) ,the expression of TNF-αand ICAM-1 was significantly increased(P=0 .000);compared with the group L ,PSS pretreatment for 1 h could significantly decrease the LPS caused adhesion of PMVEC and PMN(P=0 .000) ,the expression of TNF-αand ICAM in the group LP was significantly decreased (P<0 .05) .Con-clusion PSS can inhibit LPS-induced PMVEC injury and adhesion of PMVEC and PMN ,its mechanism may be related with the de-crease of ICAM-1 and TNF-αexpression .

ABSTRACT

Polysaccharide sulfate (PSS) is a new type of antiatherosclerotic medicine for its effects of anticoagulation, anti-thrombosis and modulation of dyslipidemia. However, it is still uncertain whether PSS could modulate the diabetic dyslipidemia or not. Here, the rat model of diabetic dyslipidemia was developed and the effects of PSS on glucose and lipid levels were investigated in this animal model. Wistar rats were iv injected with streptozotocin 20 mg·kg-1 after feeding with high fat diet for one and a half month. Then, rats received orally PSS (30, 90, and 180 mg·kg-1) for 1 month. After oral treatment with PSS (90 and 180 mg·kg-1) for 1 month, the levels of triglyceride (TG), total cholesterol (TC), low density lipoprotein-cholesterol (LDL-C) were significantly reduced and the level of high density lipoprotein-cholesterol (HDL-C) increased, compared with diabetic control rats. Moreover, PSS (30, 90, and 180 mg·kg-1) had a tendency to reduce glucose and insulin levels, and significantly increased insulin sensitivity index. Our results suggest that PSS could improve insulin sensitivity and relieve dyslipidemia in diabetic dyslipidemic rats.

ABSTRACT

OBJECTIVE:To study the general pattern and characteristics of adverse drug reactions(ADR)induced by polysaccharide sulfate(PSS).METHODS:A retrospective study was carried out to collect and statistically analyze62cases of ADR induced by PSS reported in domestic academic journals between January1994and December2005.RESULTS:The ADR to PSS were independent of patient’s sex and dependent of patient’s age,administration routes and dosage.The ADR occurred at different intervals after drug application.Clinical manifestations of ADR were multiple involving several organs and systems,mainly damage of immune system and musculoskeletal system.CONCLUSION:Clinical doctors and pharmacists should under-stand the pattern and characteristics of ADR induced by PSS,and strengthen monitoring of its use in order to reduce occurrence of ADRs.

ABSTRACT

Objective To investigate the effects of polysaccharide sulfate(PSS)on the growth of vascular endothelial cell cultured in vitro and the adhesion of vascular endothelial cell to polymorphonuclear neutrophils(PMN).Methods ECV-304,a kind of human umbilical vein endothelial cells,was cultured in vitro with different concentrations of PSS(0,50,100,150,200,250 g?mL-1),the growth of ECV-304 was observed by means of MTT.Hypoxia/ reoxygenation(H/R) model of ECV-304 was established,the adhesion rate of PMN from patients with cerebral infarction to cultured ECV-304 was detected with or without treatment of PSS and H/R.Results The growth of ECV-304 in PSS-treated subgroups(final concentrations: 50,100,150,200 g?mL-1) were much better than in non-PSS subgroup,especially in subgroup with PSS final concentration of 100 g?mL-1.The adhesion rate of cultured ECV-304 to PMN from patients with cerebral infarction was higher significantly than that from the healthy subject,which was increased when ECV-304 being treated with H/R.The adhesion rate of cultured ECV-304(whether treated with H/R or not) to PMN was decreased significantly when ECV-304 being treated with PSS(100 g?mL-1).Conclusion The experiments in vitro showed that PSS could promote the growth of cultured ECV-304 and decrease the adhesion rate of H/R ECV-304 to PMN.

Thirty -nine cases of hypertension with blood hyperviscosity and deformed wave C dy/dt max,diagnosed by admittance cardiogram,were divided into two randomized groups,eighteen patients received PSS therepy while the other groups treated with Dan -Shen Co and Xin Nao Shu Tong. ABSTRACT

Objective To investigate the effects and mechanism of action of PSS on the treatment of patients with early DN. Methods 76 patients with early DN were given PSS 100mg(iv gtt, qd) for 14 days. ET-1, TXB_2, 6-Keto-PGF_(1a), blood viscosity TG, TC, HDL-C, LDL-C and urine protein quantity(24h UPQ) were determined and compared between before and after treatment. Results All indexes above were improved significantly after therapy. Conclusion PSS may modulate ET-1 secretion and the ratio of TXB_2/6-Keto-PGF_(1a), reduce blood lipid content and viscosity, improve hemorrheology in patients with early DN.

ABSTRACT

Objective:To investigate if a plant-derived polysaccharide sulfate(M33A) binding to HIV1 gp120 may induce the exposure of neutralization epitopes of gp120,and if M33A-bound HIV-1ⅢB antigens may be used as a AIDS vaccine for inducing neutralizing antibodies against HIV-1.Methods:Whole-inactivated M33A-bound HIV-1ⅢB antigens were prepared and used to immunize mice after mixing with FCA or FIA.The titers of anti-HIV-1 IgG antibodies in immunized mouse plasma were detected by ELISA,and the HIV-1 neutralization by those plasma was detected by the improved microtiter neutralization assay.Results:M33A-bound HIV-1 antigens induced higher titers of anti-HIV-1 IgG antibodies(group C:1.5?10~6;group D:1.5?10~6) than HIV-1 antigens alone(4.9?10~5),and female mice produced 3 times higher titers of anti-HIV-1 IgG antibodies than male mice after immunized with various HIV-1 antigens.All three immunization schemes did not induce the production of anti-HIV-1 neutralizing antibodies.Conclusion:M33A binding does not induce gp120 to expose neutralization epitopes.However,M33A may improve the level of mice immune responses to HIV-1 antigens,suggesting M33A may enhance immune response to HIV-1 antigens.
