ABSTRACT

Purpose: Dry eye syndrome (DES) is a global issue occurring due to tear deficiency or excessive tear evaporation. It is associated with a variety of symptoms causing ocular discomfort. The purpose of the study was to evaluate causative factors, treatment modalities, quality of life, and preservatives used in eye drops. Methods: This prospective, follow?up study was conducted in the ophthalmology outpatient department of a tertiary care teaching hospital. Patients older than 18 years of age of either sex diagnosed with DES and willing to give written informed consent were included. The patients were subjected to the Ocular surface disease index Questionnaire (OSDI Questionnaire) twice; at the time of the first visit and at 15 days follow?up. Results: A male preponderance was observed with a 1.86:1 male?to?female ratio. The mean age of the study population was 29.15 ± 10.07 years. The most common presenting complaints were symptoms related to dryness of the eyes followed by refractive error. Exposure to TV/computer screen for more than 6 hours is the most common causative factor. There was a statistically significant improvement in overall quality of life (QoL) in patients of DES on treatment. However, no significant difference was seen in the improvement of quality of life in comparison with different preservatives used in prescribed eye drops for the treatment of DES. Conclusion: DES can adversely affect the quality of life of patients. Prompt treatment of this condition can significantly improve the patient’s QoL. Physicians should be encouraged to perform quality of life evaluations for patients with DES to guide them in treatment with more individual?specific treatment options.

ABSTRACT

This study explored the preservation effect of strigolactone analogs on Gastrodia elata tubers and screened out the suitable preservation measures of G. elata to provide a safer and more effective method for its storage and preservation. Fresh G. elata tubers were treated with 7FGR24, 2,4-D isooctyl ester, and maleic hydrazide, respectively. The growth of flower buds, the activities of CAT, and MDA, and the content of gastrodin and p-hydroxybenzyl alcohol were measured to compare the effects of different compounds on the storage and preservation of G. elata. The effects of different storage temperatures on the preservation of 7FGR24 were compared and analyzed. The gibberellin signal transduction receptor gene GeGID1 was cloned, and the effect of 7FGR24 on the expression level of GeGID1 was analyzed by quantitative polymerase chain reaction(qPCR). The toxicity of the G. elata preservative 7FGR24 was analyzed by intragastric administration in mice to evaluate its safety. The results showed that compared with 2,4-D isooctyl ester and maleic hydrazide, 7FGR24 treatment had a significant inhibitory effect on the growth of G. elata flower buds, and the CAT enzyme activity of G. elata was the highest, indicating that its preservation effect was stronger. Different storage temperatures had different effects on the preservation of G. elata, and the preservation effect was the strongest at 5 ℃. The open reading frame(ORF) of GeGID1 gene was 936 bp in length, and its expression level was significantly down-regulated after 7FGR24 treatment, indicating that 7FGR24 may inhibit the growth of flower buds by inhibiting the gibberellin signal of G. elata, thereby exerting a fresh-keeping effect. Feeding preservative 7FGR24 had no significant effect on the behavior and physiology of mice, indicating that it had no obvious toxicity. This study explored the application of the strigolactone analog 7FGR24 in the storage and preservation of G. elata and preliminarily established a method for the storage and preservation of G. elata, laying a foundation for the molecular mechanism of 7FGR24 in the storage and preservation of G. elata.

ABSTRACT

Purpose: To evaluate the role of McCarey-Kaufman (MK) medium in maintaining the integrity of donor corneal epithelium. Methods: Nineteen corneal buttons were harvested and stored in MK media at 2°C–8°C for four days. Serial photographs were done every day till the 3rd day, and images were then analyzed with ImageJ software (LOCI, University of Wisconsin, USA). The area of exposure and epithelial defect (ED) was calculated every day for each corneal button. Results: The average age of the donors was 56.5 ± 22.7 years and mean time from death to preservation of the corneal buttons was 7.7 ± 3.1 hours. The average corneal area was 145.6 ± 18.8 mm2. The total mean area of exposure was 3.6 ± 4.8, 7.2 ± 9.2, and 9.0 ± 11.9 mm2, and ED was 1.7 ± 4.6, 2.8 ± 5.3, and 3.3 ± 5.9 mm2 on days 1, 2, and 3, respectively. The percentage of increase in the area of exposure and ED in MK media was 3.71% and 1.1% from day 1 to day 3, respectively. Six out of 19 corneal buttons (31.57%) were utilized for keratoplasties, of which two were utilized in house and four were distributed outside. Of the two utilized corneas, none had epithelial defect on postoperative day 1. Rest 13 corneas were either used for training and research purposes, stored in glycerol media, or discarded. Conclusion: Since the percentage change in area of exposure/ED is not much at the end of day 3, corneas stored in MK media can be safely used even after three days of storage. Hence, MK medium serves as an excellent medium in maintaining the integrity of donor corneal epithelium

ABSTRACT

Purpose: To evaluate the efficacy of voriconazole and amphotericin B in McCarey–Kaufman (MK) media. Methods: MK media vials were supplemented with either voriconazole at 1, 2, 20, 50, 100 ?g/mL or amphotericin B at 0.5, 1, 2, 10, 20 ?g/mL. The standard inoculum of the American Type Culture Collection (ATCC) strain of Candida albicans, Aspergillus flavus, and Fusarium keratinoplasticum was added to the set of vials. The efficacy outcomes were calculated as ‘viable fungal colony counts’ determined from the samples taken on Days 0 and 4. MK media containing fungal inoculum but without antifungal supplements were used as control. Results: In the voriconazole arm, on Day 4, a reduction in the colony count was observed for Candida albicans (1 ?g/mL, 36%; 100 ?g/mL, 100%), Aspergillus flavus (1 ?g/mL, 53.8%; 100 ?g/mL, 80.4%), and Fusarium keratinoplasticum (1 ?g/mL, 39.0%; 100 ?g/mL, 72.2%). Similarly, in the amphotericin B arm, on Day 4, a reduction in the colony count was observed for Candida albicans (0.5 ?g/mL; 99.9%; 20 ?g/mL, 100%), Aspergillus flavus (0.5 ?g/mL, 65.2%; 20 ?g/mL, 84.8%), and Fusarium keratinoplasticum (0.5 ?g/mL, 90.1%; 20 ?g/mL, 100%). Conclusion: Compared to voriconazole, the addition of amphotericin B significantly reduces fungal contamination in MK media.

SUMMARY Introduction: Nitrates and nitrites can be found in meat and dairy products, vegetables, and fruits. The consumption of these preservatives has been associated with the emergence of gastric, colorectal cancer and non-Hodgkin's lymphoma, although studies are still inconclusive. Methodology: It is a review of literature of the narrative type, in which there was a recovery of articles published in English and Portuguese, in the databases SciELO, Lilacs, Science Direct and Capes journals. Articles related to nitrates and nitrites in foods were included, as well as articles that correlated the consumption of these preservatives with the appearance of cancers, regardless of the year of publication, although articles published between the years 2009 to 2019 were prioritized. Results: Nitrates and nitrites are generally associated with industrialized food products; therefore, it is possible to observe the presence of these salts in a wide variety of foods. The excessive consumption of nitrates and nitrites, whether through water or food, has been associated with a great diversity of diseases, stimulating the development of several studies investigating, including, the correlation between the consumption of these preservatives and the appearance of cancers. Conclusion: It is observed that nitrates and nitrites form compounds with a carcinogenic potential, making them interesting to avoid excessive consumption of these preservatives.


RESUMEN Introducción: Los nitratos y nitritos pueden ser encontrados en carnes y productos lácteos, verduras y frutas. El consumo de estos conservantes ha sido asociado con la aparición de cáncer gástrico, colorrectal y linfoma no Hodgkin, aunque los estudios aún no son concluyentes. Metodología: Se hizo una revisión de tipo narrativo de la literatura, en la que se recuperaron artículos publicados en inglés y portugués, en las bases de datos SciELO, Lilacs, Science Direct y revistas Capes. Se incluyeron artículos relacionados con nitratos y nitritos en alimentos, así como artículos que correlacionaban el consumo de estos conservantes con la aparición de cánceres, independientemente del año de publicación, aunque se priorizaron artículos publicados entre los años 2009 a 2019. Resultados: Los nitratos y nitritos se asocian generalmente con productos alimenticios industrializados; por tanto, es posible observar la presencia de estas sales en una amplia variedad de alimentos. El consumo excesivo de nitratos y nitritos, ya sea a través del agua o de los alimentos, se ha asociado a una gran diversidad de enfermedades, estimulando el desarrollo de varios estudios que investigan, entre ellos, la correlación entre el consumo de estos conservantes y la aparición de cánceres. Conclusión: Así, se observa que los nitratos y nitritos, forman compuestos con potencial carcinogénico, por lo que resulta interesante evitar el consumo excesivo de estos conservantes.

Metodologia: Trata-se de uma revisão de literatura do tipo narrativa, na qual houve a recuperação de artigos publicados em inglês e português, nas bases de dados SciELO, Lilacs, Science Direct e Periódicos Capes. Foram incluídos artigos relacionados a nitratos e nitritos em alimentos, bem como artigos que correlacionassem o consumo desses conservantes com o aparecimento de cânceres, independentemente do ano de publicação, embora tenham sido priorizados os artigos publicados entre os anos de 2009 a 2019. Resultados: Nitratos e nitritos são geralmente associados a produtos alimentícios industrializados; portanto, é possível observar a presença desses sais em uma grande variedade de alimentos. O consumo excessivo de nitratos e nitritos, seja pela água ou pelos alimentos, tem sido associado a uma grande diversidade de doenças, estimulando o desenvolvimento de diversos estudos que investiguem, inclusive, a correlação entre o consumo desses conservantes e o aparecimento de cânceres. Conclusão: Assim, observa-se que nitratos e nitritos formam compostos com potencial carcinogê-nico, por isso é interessante evitar o consumo excessivo desses conservantes.

ABSTRACT

Natamycin is a polyene macrolide antibiotics with strong and broad spectrum antifungal activity. It not only effectively inhibits the growth and reproduction of fungi, but also prevents the formation of some mycotoxins. Consequently, it has been approved for use as an antifungal food preservative in most countries, and is also widely used in agriculture and healthcare. Streptomyces natalensis and Streptomyces chatanoogensis are the main producers of natamycin. This review summarizes the biosynthesis and regulatory mechanism of natamycin, as well as the strategies for improving natamycin production. Moreover, the future perspectives on natamycin research are discussed.

ABSTRACT

Vessel allograft preservation is essential for vascular reconstruction surgery. The preservation solution is crucial inextending the preservation period while maintaining vascular endothelial function. Currently, a 0.9% normal salinesolution (NSS) is widely used as a storage solution although its protective effect on endothelial preservation islimited. In this study, we determine the beneficial effect of recombinant human secretory leukocyte protease inhibitor(rhSLPI), supplemented to 0.9% NSS, for isolated aortic preservation. The thoracic and abdominal aorta were isolatedfrom Wistar rats (n = 6), and the aortic rings were subsequently cut and preserved in 0.9% NSS in the presenceand absence of 1 µg/ml rhSLPI for 0, 6, 24, and 48 hours. The preservative solution was collected to determine thereleased lactate dehydrogenase (LDH) activity and pro-inflammatory cytokine levels, including tumor necrosis factorα (TNF-α) and interleukin-6 (IL-6). Furthermore, the appearance and severity of vessel degeneration were subjected toblind histopathological assessment by pathologists. The results indicated that 0.9% NSS, supplemented with rhSLPI,significantly reduced the released LDH activity, TNF-α, and IL-6 levels and could attenuate endothelial detachment,endothelial degeneration, and endothelial necrosis. This study demonstrated for the first time that adding rhSLPI to asaline preservative solution could prevent vascular injury and possibly extend the graft storage duration before clinicalintervention.

ABSTRACT

Recentemente, produtos farmacêuticos e cosméticos com concentrações mínimas de parabenos e outros conservantes ganharam e apelo comercial e de segurança, devido à controvérsia sobre a segurança dos conservantes. No entanto, o uso de conservantes é essencial para garantir a conservação microbiana de produtos cosméticos e farmacêuticos durante o seu uso. Neste trabalho, desenvolveu-se um método quimiométrico de espectroscopia no infravermelho com Fourier transform near-infrared (FTIR) para prever a eficácia de sistemas conservantes em produtos farmacêuticos e cosméticos tópicos usando os conceitos de Quality by design (QbD) e Process Analytical Technology (PAT). A abordagem de QbD foi usada para determinar a eficácia antimicrobiana frente aos microrganismos: Candida albicans (ATCC 10231), Escherichia coli (ATCC 8739) e Staphylococcus aureus (ATCC 6538), em funções das concentrações de parabenos, e determinar a região de Design Space, empregando o delineamento de compóstio central (CCD) Todas as 15 formulações preparadas foram analisadas utilizando um espectrofotômetro (FTIR) equipado com aparato de Attenuated Total Reflectance (ATR). Os modelos de regressão por Partial Least Squares (PLS) para predição dos "slopes" das curvas de morte microbiana em função dos espectros ATR/FTIR foram bem ajustados, com R2 e R2-predição de 0,9937 e 0,8921, 0,9947 e 0,8783, e 0,9957 e 0,9222 para Candida albicans (ATCC 10231), Escherichia coli (ATCC 8739) e Staphylococcus aureus (ATCC 6538), respectivamente. O método FTIR proposto aplicado em uma abordagem de PAT foi capaz de prever a eficácia do sistema conservante em tempo reduzido. Este método de predição de silício permitirá um controle lote-a-lote da eficácia do sistema conservante de produtos farmacêuticos e cosméticos


Recently, pharmaceuticals and cosmetics with minimal concentrations of parabens and other preservatives have gained and commercial and safety appeal due to controversy over the safety of preservatives. However, the use of preservatives is essential to ensure the microbial conservation of cosmetic and pharmaceutical products during use. In this work, a chemometric method of infrared spectroscopy with Fourier transform (FTIR) was developed to predict the effectiveness of preservative systems in pharmaceutical products and topical cosmetics using the concepts of Quality by design (QbD) and Process Analytical Technology (PAT). The QbD approach was used to determine antimicrobial efficacy against candida albicans (ATCC 10231), Escherichia coli (ATCC 8739) and Staphylococcus aureus (ATCC) microorganisms 6538), in functions of paraben concentrations, and determine the Design Space region, employing the design of central composite (CCD). All 15 prepared formulations were analyzed using a spectrophotometer (FTIR) equipped with Attenuated Total Reflectance (ATR). The Partial Least Squares (PLS) regression models for the prediction of the slopes of microbial death curves as a function of ATR /FTIR spectra were well adjusted, with R2 and R2-prediction 0.9937 and 0.8921, 0.9947 and 0.8783, and 0.9957 and 0.9222 for Candida albicans (ATCC 10231), Escherichia coli (ATCC 8739) and Staphylococcus aureus (ATCC 6538)respectively. The proposed FTIR method applied in a PAT approach was able to predict the effectiveness of the preservative system in reduced time. This method in silico prediction will allow a batch-to-lot control of the effectiveness of the preservative system of pharmaceuticals and cosmetics

ABSTRACT

PURPOSE: To compare the intraocular pressure (IOP) in diabetic macular edema (DME) patients and macular edema associated with branch retinal vein occlusion (BRVO) patients after intravitreal preservative-free Triamcinolone injection.METHODS: This study included 36 patients diagnosed with DME and 44 patients diagnosed with BRVO with macular edema. Both groups were treated with intravitreal preservative-free Triamcinolone (Maqaid®, Wakamoto Pharmaceutical Co., Ltd., Tokyo, Japan) injection, and we compared the IOPs of the two groups determined before injection and at 1, 2, 3, and 6 months after injection. We also compared the IOP elevation ratios and durations, and central foveal thickness (CFT) changes using optical coherence tomography.RESULTS: In both groups, there was no statistical significance in the IOP before injection and at 1, 2, 3, and 6 months after injection. However, the IOP elevation ratio in the DME patients (38.9%) was significantly higher than that in the BRVO patients (15.9%) (p = 0.02). The duration of IOP elevation in the DME patients (1.14 ± 1.85 months) was significantly longer than that in the BRVO patients (0.30 ± 1.20 months) (p = 0.03). When the IOP was not controlled, we used IOP-lowering agents, and two patients in the DME were treated with glaucoma surgery. There was no statistical significance in the CFT before injection and at 1, 2, or 3 months after injection between the two groups (p = 0.72, p = 0.26, p = 0.66, p = 0.34, respectively). However, the CFT after 6 months was 328 ± 103 µm in the DME group and 434 ± 189 µm in the BRVO; this difference was significant (p < 0.01).CONCLUSIONS: Intravitreal injection of preservative-free Triamcinolone was effective in the treatment of both DME patients and macular edema patients associated with BRVO. Furthermore, Triamcinolone was more safely injected in macular edema associated with BRVO patients than in DME patients.

ABSTRACT

Este trabalho propôs o uso do fármaco quelante mesilato de desferroxamina (DFO) como agente adjuvante para estabilização química e microbiológica de formulações. Soluções de ácido ascórbico (AA) 5,0% (p/v) foram preparadas com sistemas antioxidantes constituídos por diferentes combinações de DFO, ácido etilenodiamino tetra-acético (EDTA) e metabissulfito de sódio, cada adjuvante na concentração máxima de 0,1% (p/v). Os sistemas foram testados previamente quanto à atividade antioxidante, mediante adição de um complexo de ferro (III) redox-ativo e ensaio baseado em fluorescência. Os sistemas também foram associados ao metilparabeno e avaliados quanto à atividade antimicrobiana pelo método turbidimétrico, utilizando-se a técnica de microdiluição em meios líquidos e cepas padrão de bactérias e fungos, incluindo S. aureus (ATCC 6538), E. coli (ATCC 8739), P. aeruginosa (ATCC 9027), C. albicans (ATCC 10231) e A. brasiliensis (ATCC 16404). As soluções de AA foram expostas a condições de teste de estabilidade acelerada e avaliadas quanto à estabilidade química, empregando-se método volumétrico validado para quantificar AA. Verificou-se que o EDTA foi o agente quelante que melhor contribuiu na estabilidade química da solução de AA, entretanto, o DFO apresentou desempenho muito superior ao EDTA para bloquear a atividade pró-oxidante do ferro. Além disso, o DFO foi fator relevante na inibição do crescimento microbiano e demonstrou sinergia com o metilparabeno. A otimização estatística dos resultados indicou que o uso do DFO nos sistemas antioxidante e conservante pode reduzir consideravelmente a concentração dos adjuvantes convencionais, EDTA, metabissulfito e metilparabeno, os quais são muitas vezes associados a reações de hipersensibilidade ou a danos ao meio ambiente


In this work it was proposed the use of the chelating drug desferroxamine mesylate (DFO) as adjuvant for chemical and microbiological stabilization of formulations. Ascorbic acid (AA) solutions 5.0% (w/v) were prepared with antioxidant systems containing different combinations of DFO, ethylenediaminetetraacetic acid (EDTA) and sodium metabisulphite, using a maximum concentration of 0.1% (w/v) for each adjuvant. Previously, the systems were spiked with a redox-active iron (III) complex and tested for antioxidant activity by fluorescence-based assay. The systems also were associated with methylparaben and evaluated for antimicrobial activity by turbidimetric method, using the microdilution technique and standard strains of bacteria and fungi, including S. aureus (ATCC 6538), E. coli (ATCC 8739), P. aeruginosa (ATCC 9027), C. albicans (ATCC 10231) and A. brasiliensis (ATCC 16404). The AA solutions were exposed to accelerated stability test conditions and evaluated for chemical stability, using a volumetric method that was validated to quantify AA. It was found that EDTA was the chelating agent that most contributed to the chemical stability of AA solution, however, DFO demonstrated a much higher performance to EDTA to block the pro-oxidant activity of iron. In addition, DFO was a relevant factor in the inhibition of microbial growth and showed synergy with methylparaben. The statistical optimization of the results indicated that the use of DFO in the antioxidant and preservative systems might considerably reduce the concentration of the conventional adjuvants, EDTA, metabisulphite and methylparaben, which are often associated with hypersensitivity reactions or environmental damage

RESUMEN En la industria avícola, las infecciones por Salmonella son un problema, debido a los efectos sobre la producción y los riesgos a la salud pública. En las aves infectadas puede ocurrir una diseminación sistémica de las bacterias, que afecta los órganos y favorece, tanto la transmisión entre las aves como la contaminación de los productos avícolas, tales como las carnes, durante el proceso de sacrificio de los animales. Se decidió evaluar el efecto que diferentes combinaciones de glucosa oxidasa (GOX) y glucosa tienen sobre el crecimiento de diversos serotipos de Salmonella aislados de aves en crecimiento y de engorde, y determinar su potencial como antibacteriano. Se tomaron muestras de corazón y de ciego de aves de 7 y 42 días de edad y se aislaron e identificaron S. typhimurium y S. enteritidis. Estas bacterias fueron cultivadas en medio Luria-Bertani (LB) en presencia de GOX (0,5; 1 y 2U/mL) y glucosa (0,5, 1 y 2%), en combinaciones bajo un diseño factorial 32. El crecimiento fue monitoreado por el cambio de absorbancia a 600 nm, durante 6 horas de incubación. Se observó una reducción significativa del crecimiento de ambos serotipos al utilizar 2U/mL de GOX y diferentes concentraciones de glucosa, lo cual, demostró la capacidad antibacteriana que posee el sistema GOX/glucosa sobre este género, por lo que se podría emplear como un aditivo para la conservación de carnes de aves y productos derivados, a fin de alargar su vida de anaquel y minimizar riesgos a la salud.


SUMMARY In the poultry industry, Salmonella infections are a problem due to the effects on production and risks to public health. In infected birds, a systemic spread of bacteria can occur, which affects the organs and favors both the transmission between birds and the contamination of poultry products, such as meat, during the animal´s slaughter process. It was evaluated the effect that different combinations of glucose oxidase (GOX) and glucose have on the growth of different Salmonella serotypes isolated from broilers, and thus determine their potential as antibacterial. Heart and caecal samples were taken from birds 7 and 42 days old. S. typhimurium and S. enteritidis were isolated and identified. These bacteria were cultured in Luria-Bertani medium (LB) in the presence of GOX (0.5, 1 and 2U/mL) and glucose (0.5, 1 and 2%) in combinations under a factorial design 32. Growth was monitored by absorbance change at 600nm for 6 hours of incubation. A significant reduction of the growth of both serotypes was observed when using 2 U/mL of GOX and different concentrations of glucose. This demonstrated the antibacterial capacity that the GOX/glucose system has on this genus, so it could be used as an additive for the preservation of poultry meats and derived products, in order to lengthen its storage time and minimize health risks.

ABSTRACT

ABSTRACT: The waste in the fruit production chain, including the juice and pulp industries, produces large quantities of leftover husks, seeds and bagasse. This volume of waste generates huge environmental and economic impact. The objective of this research was to determine the potential of using residues from passion fruit (Passiflora edulis) and apple (Malus domestica) varieties in the production of functional flours. Passion fruit flour showed greater reduction of DPPH (EC50%: 50.4μg/mL) radicals, showing antioxidant potential, as well as a more efficient inhibition of Staphylococcus aureus (71.3±1.2μg/mL), with a modest; however efficient, inhibition of acetylcholinesterase (10%). All Apple flours were good antioxidants and the fuji apple flour stood out inhibiting Pseudomonas aeruginosa (78.6±3.1μg/mL). All the residues showed potential for use as a functional product either as a source of antioxidants, a natural (antimicrobial) preservative for dry foods or supplementary use by patients with Alzheimer's disease.


RESUMO: O desperdício na cadeia produtiva de frutos, incluindo as indústrias de suco e polpa, produz grandes quantidades de resíduos, como cascas, sementes e bagaços. Esse volume de resíduos gera impacto ambiental e econômico. O objetivo deste trabalho foi determinar o potencial de aproveitamento de resíduos de maracujá (Passiflora edulis) e de variedades de maçã (Malus domestica) na produção de farinhas funcionais. A farinha de maracujá apresentou maior redução de radicais DPPH (EC50%: 50,4μg/mL), demonstrando potencial antioxidante, maior inibição de Staphylococcus aureus (71,3±1,2μg/mL) e da acetilcolinesterase (10%). Todas as farinhas de maçã foram boas antioxidantes e a fuji destacou-se inibindo Pseudomonas aeruginosa (78,6±3,1μg/mL). Todos os resíduos mostraram potencial para aproveitamento como produto funcional, seja como fonte de antioxidantes, conservante natural (antimicrobiano) para alimentos secos ou uso suplementar no tratamento de Alzheimer.

Abstract Pyroligneous extract is applied in diverse areas as an antioxidant, an antimicrobial, and an anti-inflammatory agent. The discovery of new cost-effective antimicrobial agents of natural origin remains a challenge for the scientific community. This study aimed to conduct a systematic review and a technological forecasting of the existent evidence regarding the use of pyroligneous extract as a potential antimicrobial agent. Studies were identified through an investigation of various electronic databases: PubMed, SciFinder, Web of Science, Scopus, Scielo, Google scholar, and ProQuest. Patents were searched through INPI, Google patents, Espacenet, Patents online, USPTO, and WIPO. The literature on antimicrobial activity of pyroligneous extract are limited given the short duration of studies and variability in study design, use of pyroligneous preparations, and reports on results. However, evidence suggests the potential of pyroligneous extract as a natural antimicrobial agent. The most studied activity was the role of PE as a food preservative. However, pyroligneous extracts are also effective against pathogenic bacteria in the oral microflora and treatment of candidal infections. Further research is needed using standardized preparations of pyroligneous extracts to determine their long-term effectiveness and ability as antimicrobial agents.

ABSTRACT

Recently, there has been a growing demand for natural preservatives because of increased consumer interest in health. In this study, we produced Lactobacillus rhamnosus cell-free supernatant (LCFS) and evaluated and compared its antimicrobial activity with existing natural preservatives against pathogenic microorganisms and in chicken breast meat contaminated with Escherichia coli and Staphylococcus aureus. Lactobacillus rhamnosus cell-free supernatant possessed 30 units of lysozyme activity and contained 18,835 mg/L of lactic acid, 2,051 mg/L of citric acid and 5,060 mg/L of acetic acid. Additionally, LCFS inhibited the growth of fourteen pathogenic bacteria, S. aureus, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Listeria innocua, S. epidermidis, L. ivanovii, E. coli, Pseudomonas aeruginosa, Shigella sonnei, Shi. flexneri, Proteus vulgaris, Pseudomonas fluorescens, and Klebsiella pneumoniae. The antibacterial activity of LCFS was stronger than that of egg white lysozyme (EWL), Durafresh (DF) and grapefruit seed extract (GSE). Additionally, LCFS maintained its antimicrobial activity after heat treatment at 50℃~95℃ and at pH values of 3~9. Moreover, LCFS inhibited the growth of E. coli and S. aureus in chicken breast meat. In conclusion, it is expected that LCFS, which contains both lysozyme and three organic acids, will be useful as a good natural preservative in the food industry.

OBJECTIVE: The study was conducted to determine the preservative activity of ethanolic extract of mangosteen (Garcinia mangostana L.) pericarp and its compatibility in an antacid suspension.METHODS: The extract was subjected to phytochemical screening and was used as preservative in a formulated antacid suspension. Compatibility with the active pharmaceutical ingredient (API) and excipients were analyzed using fourier transform-infrared spectroscopy. Preservative activity of the formulation against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa was assessed using the United States Pharmacopoeia (USP) antimicrobial effectiveness test, with methylparaben as positive control and suspension without preservative as negative control.RESULTS: The extract exhibited pharmaceutical compatibility with API and excipients. The formulation revealed comparable reduction in microbial count of E. coli, S. aureus, and P. aeruginosa with positive control at Day 14 (p=0.916, 0.624, 0.335). At Day 28, comparable activity with positive control was only observed against E. coli and S. aureus (p=0.999, 0.854). However, it displayed significant increase in activity against P. aeruginosa (p=0.010) at Day 28. These activities may be attributed to glycosides and reducing substances present in the extract.CONCLUSION: The ethanolic extract from Garcinia mangostana L. pericarp acted as a preservative in the formulation of an antacid suspension. It conformed to the USP criteria for antimicrobial effectiveness test on bacteria.

ABSTRACT

In2O3nanoparticles-modified multiwalled carbon nanotubes (In2O3-MWCNTs) were successfully prepared as solid phase extraciton (SPE) adsorbent for the determination of three food preservatives(benzoic acid,sorbic acid,and methylparaben) in beverage samples coupled with high performance liquid chromatography (HPLC).Because of the large surface area and amount of hydroxyl groups on the surface of In2O3nanoparticles,In2O3nanoparticles could interact with specific functional groups.In2O3-MWCNTs adsorbent was prepared based on the large surface area of In2O3and superior thermal and chemical stability of MWCNTs.The obtained adsorbent was characterized via transmission electron microscope (TEM),thermogravimetric analysis (TGA),and Fourier transform-infrared (FT-IR) spectroscopy.Several experimental parameters affecting the extraction efficiency were investigated by single-factor experiments,such as adsorbents amount,sample volume,eluent volume,sample pH,and eluent type.The first four factors were then further evaluated by means of a Taguchi's L16(44) orthogonal array experimental design.According to the results of orthogonal array experiment,the optimal conditions were chosen as follows: 0.15 g of adsorbent amount,5.0 mL of sample volume,0.6 mL of ACN-H2O (60:40,0.1% formic acid,V/V) as eluent,and sample pH 4.0.Under the optimum conditions,the LODs (S/N=3) and LOQs (S/N=10) for three preservatives were in the range of 0.004-0.012 μg/mL and 0.012-0.038 μg/mL,respectively.The recoveries ranged from 70.2% to 109.4%.The method is rapid,sensitive,and suitable for the determination of preservatives in food samples.

ABSTRACT

PURPOSE: To compare the degrees of conjunctival injection and corneal toxicity between preservative and preservative-free topical 0.1% fluorometholone after strabismus surgery. METHODS: A randomized, prospective clinical study was performed to compare the degrees of conjunctival injection and corneal toxicity between preservative and preservative-free topical 0.1% fluorometholone after strabismus surgery. Sixty-one patients with intermittent exotropia were included in this study. They were told to apply antibiotic eye drops (ED) and either preservative or preservative-free topical 0.1% fluorometholone (F1) three times a day. Measurements of the degrees of conjunctival injection and corneal toxicity were performed at postoperative 1 week and 3 weeks each. RESULTS: Seventeen patients (34 eyes) were included in group 1 (preservative F1) and twenty patients (40 eyes) were included in group 2 (preservative-free F1). The average pixel value (measured via the Image J software) representing the degree of conjunctival injection was 31,732 ± 9,946 in group 1 and 38,347 ± 12,189 in group 2 at postoperative 1 week, while the average pixel value was 10,150 ± 4,493 in group 1 and 11,836 ± 4,290 in group 2 at postoperative 3 weeks. There was a significant difference between the decrease in pixel value for the two groups (p = 0.040). There was no significant difference in the mean value of the Oxford stain score between the two groups at postoperative 3 weeks, however the mean questionnaire scores in group 2 were significantly lower than in group 1 (p = 0.001). CONCLUSIONS: Preservative-free 0.1% fluorometholone ED demonstrated a larger decrease in the degree of conjunctival injection than for preservative ED after strabismus surgery. Therefore, the use of preservative-free steroid ED may be beneficial for decreasing both conjunctival injection and postoperative discomfort following strabismus surgery.

ABSTRACT

Objective:To analyze the causes of microbial contamination in methadone hydrochloride oral solution used for the ma-intenance treatment of heroin addict in Guangxi and propose the relative countermeasures. Methods:The samples of methadone hydro-chloride oral solution were obtained from 10 maintenance treatment clinics, and then the microbial limit test and identification were per-formed,the content of preservative was detected and the relationship between the preservative content and microbial contamination was studied. Results:The total number of aerobe in the samples of methadone hydrochloride oral solution from the 10 clinics was all over the standard limit value of 100 cfu·ml-1 ,and the main contaminating germ was Burkholderia cepacia. The contents of sorbic acid were 78. 2%-98. 3%,the four batches with the lowest concentration of preservative were detected with higher total number of aerobe. Con-clusion:The microbial contamination in methadone hydrochloride oral solution from the 10 maintenance treatment clinics is severe,and the environment of medicine packaging, content of preservatives, packaging machine and storage conditions are the primary causes for the microbial contamination. To ensure drug quality safety, it is suggested that the environment cleanliness of medicine packaging be improved, suitable content of preservatives be added, packaging machine be clean, the products be stored in shade and the quality be tested regularly.
