ABSTRACT

Objective To analyze the epidemiological characteristics and related traditional Chinese medicine (TCM) constitution and emotional characteristics.Methods The convenient sampling method was adopted,and sub-healthy subjects receiving outpatient physical examinations served as the respondents.The Individual Physical and Moral Health Investigation Scale was used to conduct the questionnaire investigation.The Epidata3.0 software was used to build the database and SPSS18.0 was used to do statistical analysis.Results The psychological sub-health was mainly concentrated in young people below 35 years old,which accounted for 74.10%;385 cases (63.00%) were female;562 cases (92.40%) were the college degree or above.And the number of people with general economic status was 475 (78.60%).In the TCM constitution classification of the mental sub-health population,384 cases (62.85 %) of biased constitution were significantly higher than those of somatic sub-health (P<0.05).The biased constitution was dominated by qi-deficiency,yangdeficiency and qi-stagnation.In the aspect of emotional characteristics,the psychological sub-healthy population mainly manifested by anxiety,depression,compulsion and fear,the difference between these four emotional problems and somatic sub-health was statistically significant (P<0.05),but in the emotional problems of hostility,interpersonal sensitivity and paranoid,which had no statistically significant differences compared with somatic sub-healthy people (P>0.05).Conclusion The psychological healthy intervention needs to play emphasis on the emotional problems occurred in young females with higher educational background and general economic status and pay attention to regulate qi-deficiency and yang-deficiency by TCM.

ABSTRACT

This study was designed to screen the targets of bioactive ingredients of Baihe Dihuang Tang, and investigate the "multi-components, multi-targets and multi-pathways" intervention mechanism of Baihe Dihuang Tang on psychological sub-health. The ADME/T calculation method was used to screen the active ingredient of Baihe Dihuang Tang, and then using ADME/T calculation method to filtrate the active components of Baihe Dihuang Tang, then Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), PharmMapper database and Medical Subject Headings (MeSH) were combined to forecast and filtrate the targets of the main active ingredients. In addition, the predicted targets were verified by the Surflex-dock in Sybyl. The Cytoscape software was used to construct the Baihe Dihuang Tang ingredients-targets-disease network, while ClueGO software was used to analyze the molecular function and biological process of the targets. There are total 11 active ingredients and 21 targets in Baihe Dihuang Tang. A good interaction between them was supported by the score. The 21 targets were mainly involved in gamma-aminobutyric acid signaling pathway, cAMP metabolic process and monoamine transport relevant biological processes. Thus, Baihe Dihuang Tang may play a role in the intervention of psychological sub-health by regulating the activity of G-protein coupled amine receptor and the expression of monoamine neurotransmitter, which reflects the features of traditional Chinese medicine multi-components, multi-targets and multi-pathways. This research provides evidences on the pharmacological mechanism of Baihe Dihuang Tang effect on psychological sub-health.

ABSTRACT

Objective: To establish the animal model of rat with psychological sub-health and carry out the comprehensive evaluation by the methods of behavior, metabonomics, and factor analysis. Methods: Simulating human to cause the psychological sub-health state in the fierce competitive social environment due to improper diet, irregular work and rest, overload mental stress, and social failure. Modeling procedures by using composite and non-somatic stimuli factors including stimulation of empty bottle stress, on the sidelines of shock stress, improper diet stress, round-the-clock stress, and social failure stress. The following behavioral experiments were conducted such as sucrose preference rate, high plus maze, light/dark box, and open field test. Body mass and organ index were observed in rats. The factor analysis was used based on behavioral and metabolomic evaluate model. Results: The body weight and organ index of rats did not change significantly; But sucrose preference rate decreased significantly (P<0.05), the number of open arms entries and the proportion of time in the open arms decreased significantly (P<0.01); Error count and the time into black box increased significantly (P<0.01). Factor analysis dug up three common factors from the macroscopic behaviors, namely depression and anxiety, memory, and other factors. Metabolomic analysis showed that the control group and model group clearly separated in the PLS-DA plot, and identified seven biomarkers: high density lipoprotein, acetic acid, glutamic acid, creatine, choline, betaine, and threonine. Conclusion: The establishment of the rat model with psychological sub-health can reflect the state of psychological sub-health of human being. And the evaluation method of model possesses scientific rationality, feasibility, and reliablity. It can provide the reference for further study on its mechanism and the related drugs action.
