ABSTRACT

This study investigated the chemoprotective effects of Punica granatum L. (Punicaceae) fruits alcoholic extract (PGE) on mice exposed to hexavalent chromium [Cr(VI)]. Animals were pretreated with PGE (25, 50 or 75 mg/kg/day) for 10 days and subsequently exposed to a sub-lethal dose of Cr(VI) (30 mg/kg). The frequency of micronucleated polychromatic erythrocytes in the bone marrow was investigated and the Cr(VI) levels were measured in the kidneys, liver and plasm. For the survival analysis, mice were previously treated with PGE for 10 days and exposed to a single lethal dose of Cr(VI) (50 mg/kg). Exposure to a sub-lethal dose of Cr(VI) induced a significant increase in the frequency of micronucleated cells. However, the prophylactic treatment with PGE led to a reduction of 44.5% (25 mg/kg), 86.3% (50 mg/kg) and 64.2% (75 mg/kg) in the incidence of micronuclei. In addition, the 50 mg/kg dose of PGE produced a higher chemoprotective effect, since the survival rate was 90%, when compared to that of the non-treated group. In these animals, reduced amounts of chromium were detected in the biological materials, in comparison with the other groups. Taken together, the results demonstrated that PGE exerts a protective effect against Cr(VI)-induced genotoxicity.


Este estudo investigou os efeitos quimioprotetores do extrato alcoólico dos frutos da Punica granatum L. (Punicaceae) (EPG) em camundongos expostos ao cromo hexavalente [Cr(VI)]. Os animais foram pré-tratados com o EPG (25, 50 ou 75 mg/kg/dia) durante 10 dias e subsequentemente expostos a uma dose subletal de Cr(VI) (30 mg/kg). A frequência de eritrócitos policromáticos micronucleados na medula óssea foi investigada e os níveis de Cr(VI) foram quantificados nos rins, fígado e plasma. Para a análise de sobrevida, os camundongos foram previamente tratados com EPG durante 10 dias e expostos a única dose letal de Cr(VI) (50 mg/kg). A exposição à dose subletal de Cr(VI) induziu aumento significativo na frequência de células micronucleadas. Entretanto, o tratamento profilático com EPG levou à redução de 44,5% (25 mg/kg), 86,3% (50 mg/kg) e 64,2% (75 mg/kg) na incidência de micronúcleo. Além disso, a dose de 50 mg/kg de EPG produziu maior efeito quimioprotetor, uma vez que a taxa de sobrevivência foi de 90%, quando comparada àquela do grupo não tratado. Nesses animais, quantidades reduzidas de cromo foram detectadas nos materiais biológicos, em comparação com os outros grupos. Em conjunto, os resultados demonstram que o EPG exerce efeito protetor contra a genotoxicidade induzida pelo Cr(VI).

ABSTRACT

Punica granatum has been used for centuries to confer health benefits in a number of inflammatory diseases. Based on its usage in Ayurvedic and Unani medicine, dietary supplements containing pomegranate extract are becoming popular for the treatment and prevention of arthritis and other inflammatory diseases. Pet-ether, dichloromethane and methanol fractions of flower part were chosen for pharmacological screening and analgesic and anti-inflammatory activities in animal model. The anti-inflammatory activity was assessed using the carrageenan-induced rat paw edema model. The analgesic effect was measured in mice using the acetic acid-induced writhing test. In the acetic acid-induced writhing test in mice, pet-ether, dichloromethane and methanol fractions at 200 mg/kg doses level showed 75.77% (p<0.001), 68.56% (p<0.001), 54.64% (p<0.001) inhibition of writhing, respectively. In rat paw edema model induced by carrageenan, pet-ether, dichloromethane and methanol fractions were found to reduce significantly (p<0.001) the formation of edema at the 100 mg/kg dose level and showed 26.92% (p<0.001), 27.97%(p<0.001), 21.85%(p<0.001) inhibition respectively of edema volume at the end of 4 h. Punica granatum possesses evident analgesic and anti-inflammatory activities. The results signify the traditional uses of Punica granatum for inflammation and pain.

ABSTRACT

Punica granatum Linn. (Punicaceae) commonly known as Pomegranate is a dark greenish large deciduous shrub or small tree, about 5-10 m height. Flower buds are traditionally used in the treatment of asthma and allergy. The aim of this study was to validate the traditional antiallergic property using milk-induced leucocytosis and milk-induced eosinophilia in rats. Flower buds of the plant were extracted successively using various solvents to obtain the respective extracts. These extracts were administered to mice at the dose of 50 and 100 mg/kg, orally. Only the ethanol extract showed significant reduction in leukocyte and eosinophil count, these results are a validation of the use of the extract of polar compounds of P. granatum flower buds as an antiallergic agent.


Punica granatum Linn. (Punicaceae) comúnmente conocida como granada es un pequeño árbol o arbusto grande de hoja caduca, de unos 50-10 m de altura y de color verde oscuro. Los botones florales se utilizan tradicionalmente en el tratamiento del asma y la alergia. El objetivo de este estudio fue validar la propiedad tradicional antialérgica utilizando ratas con leucocitosis y eosinofilia inducida por leche. Los botones florales de la planta se extrajeron sucesivamente con varios solventes para obtener los extractos respectivos. Estos extractos se administraron a los ratones a dosis de 50 y 100 mg / kg, por vía oral. Sólo el extracto obtenido con etanol mostró una reducción significativa en el recuento de leucocitos y eosinófilos, estos resultados son una validación del uso del extracto de compuestos polares de los botones florales de P. granatum como un agente antialérgico.

ABSTRACT

Punica granatum L., Punicaceae, amplamente usada no Brasil, foi avaliada quanto ao seu potencial antitumoral in vitroe in vivo. Investigou-se in vitro a citotoxicidade do extrato etanólico do fruto e folha da P. granatumutilizando células K-562 e células do Tumor Ascítico de Ehrlich (TAE), pelos métodos de redução do MTT e exclusão do azul de tripano. Nos estudos in vivoavaliou-se o aumento da sobrevida de animais portadores do TAE e tratados, por via oral, com diferentes doses dos extratos etanólicos da P. granatum(12,5; 25; 50 e 100 mg/kg) por dez dias consecutivos. Além disso, nestes animais analisou-se o potencial de inibição tumoral e a atividade antiangiogênica da P. granatum. Os resultados dos estudos in vitrodemonstraram uma redução na viabilidade das células K-562 e do TAE, concentração-dependente, nos métodos investigados. Os resultados in vivo demonstraram aumento da sobrevida dos animais portadores do TAE tratados, de forma dose-dependente. Em paralelo, observou-se diminuição do número de células tumorais na cavidade peritoneal dos animais portadores e tratados. Além disto, os tratamentos empregados reduziram o padrão de vascularização da parede abdominal. Dessa forma, os dados apresentados revelaram que o extrato de P. granatumpossui atividade antitumoral in vitroe in vivo em paralelo a redução da angiogênese peritoneal.


Punica granatum L., a plant widely used in Brazil, was tested for its antitumor and antiangiogenic activities in vitroand in vivo. In this work, the in vitrocytotoxicity was evaluated using the K-562 cell line and Ehrlich ascites tumour cells, by MTT tetrazolium reduction test and the trypan blue exclusion test. In vivostudies investigated the increase in the survival time of Ehrlich tumour-bearing mice after treatment with different doses of Punica granatumL. ethanol extract (12.5; 25; 50 e and 100 mg/kg), by gavages, for ten consecutive days. In addition, we also investigated the tumour inhibition potential and antiangiogenic activity of this plant. In vitroresults demonstrated a decrease of K-562 and Ehrlich ascites tumour cells viability, with both methods used, in a dependent-manner concentration. In vivoresults showed a significant antitumor activity against Ehrlich ascites tumour growth, increasing survival time. In parallel, we detected a significant inhibition of the tumour growth, along with a decrease in the vascular pattern of the peritoneal wall. Thus, the data presented herein clearly showed that Punica granatum L. has antitumor and antiangiogenic activities.
