ABSTRACT

A method was developed for the determination of furocoumarin compounds(psoralen, oxypeucedanin, imperatorin, isoimperatorin) in traditional Chinese medicine samples by dispersive liquid-liquid microextraction(DLLME) coupled with high performance liquid chromatography and the relation of sample solution volume, sediment phase volume with enrichment factors was deducted. Some important parameters that influenced the extraction efficiency were optimized: 50 μL carbon tetrachloride and 300 μL acetonitrile were rapidly injected into 1.5 mL sample solution containing 2.5%(w/V) NaCl. After centrifugation at 3500 r/min for 3 min, the sedimented CCl_4 phase was pumped and recorded the volume, then dissolved with methanol and injected into the HPLC for analysis. Under the optimum conditions, a good linear relationship was obtained in the range of 0.006 6.00 mg/L of both psoralen and oxypeucedanin and 0.006-12.0 mg/L of both imperatorin and isoimperatorin. The limits of detection were 1.0-3.0 μg/L(S/N=3). The relative standard deviations were 2.3%-5.4%(n=5). The enrichment factors ranged from 12.6 to 38.5 folds. The average recoveries ranged from 97.5% to 114.8%. The four kinds of furocoumarin compounds in Radix Angelicae dahuricae and Yuanhu Zhitong tablet were determined by the proposed method with satisfactory results.

ABSTRACT

Objective To establish a HPLC method for the determnation of serum concentration of isoimperatorin in rats and study pharmacokinetic characteristic of isoimperatorin in rat after drug admiaistraction. Method The concentrations of isoimperatorin in rats were determined after drug administration and the pharmacokinetic parameters of isoimperatorin were estimated by DAS2.0 computer program. Result The concentration-time profiles were fitted with open two compartment model system. The main pharmacokinetic parameters were as follows:Tmax =(1.6?0.000)h, Cmax =(15.842?0.35)mg/L, AUC0-t=(80.761?10.03)mg/(L?h), AUC0-∞=(89.45?9.34)mg/(L?h), t1/2=(5.101?0.24)h. Conclusion The method is appropriate for the determination and pharmacokinetic study of isoimperatorin in rats after drug administration.

ABSTRACT

Objective To optimize the alcohol extraction technique of the medical herbs such as Rhizoma Corydalis and Radix Angelicae Dahuricae. Methods To optimize the alcohol extraction technique with L9(34) orthogonal test. The content of the tetrahydropalmatine in Rhizoma Corydalis,which was used as evaluating criteria,was determined by HPLC. Results The optimized extraction technique was as follows: the medicinal material was extracted for 3 times with 3 times amount of 75% alcohol,1.5 hours per time. Conclusions The optimized technique is reasonable and the extraction rate was high.

ABSTRACT

Objective: To build an extraction method of coumarins from Radix Angelicae Dahuricae. Methods: The solvent varieties, polarities, and extraction methods were systematically studied by TLC with imperatorin as marker.Results: The more effective way to extract coumarins from Radix Angelicae Dahurica was refluent extraction with 4 times amount of 95% alcohol twice (one hour each time).Conclusion: The Solvent varieties and extraction method showed greater effects than others on the extraction efficiency of imperatorin.

ABSTRACT

AIM:To compare the quality of Radix Angelicae dahuricae Dispensing Granules produced by different pharmaceutical factories.METHODS:The main coumarin compounds were identified by TLC and HPLC respectively;imperatorin was determined by HPLC.RESULTS:Imperatorin and other main coumarin compounds of Radix Angelicae dahuricae Dispensing Granules in the market could not be determined because of low content.CONCLUSION:Radix Angelicae dahuricae Dispensing Granules produced by different pharmaceutical factories in the market have distinct differences in quality,and were of poor quality in the mass.
