ABSTRACT

Objective To develop a method for the measurement of the content of paeoniflorin and albiflorin in red peony root (the roots of Paeonia lactiflora, RPR) by near-infrared spectroscopy (NIR) for quality rapid assessment. Methods A total of 65 RPR samples were collected from Sichuan, Zhejiang and Anhui of China, and then quantified the content of paeoniflorin and albiflorin by NIR and ultra-high performance liquid chromatography (UPLC). Fifty-three samples were chosen as the calibration set, while the remaining 12 samples were assigned to external validation set. The calibration model of paeoniflorin and albiflorin was further developed by modified partial least squares (MPLS) using UPLC data as reference based on optimized spectral regions, pre-treament of spetrum and the number of principal divisors. The content of paeoniflorin and albiflorin in unknown samples (validation model) was predicted according to the validation set of NIR. Results The standard errors of prediction (SEP) for the contents between the predicted value of NIR method and the estimated value of UPLC method were 1.437 2% for paeoniflorin and 0.784 3% for albiflorin, and their correlation coefficient (R) were 0.990 2 for paeoniflorin) and 0.994 4 for albiflorin in 53 calibration set samples. Moreover, the SEP and R of that in 12 validation set samples were 0.714 5%, 0.988 0% for paeoniflorin, and 0.632 4, 0.994 6 for albiflorin, respectively. The sum of paeoniflorin and albiflorin in RPR cultivated in Sichuan, Anhui, and Zhejiang province in China were 5.49%, 5.33%, and 4.81%, respectively. Conclusion NIR can quantify the amounts of paeoniflorin and albiflorin quickly and simultaneously in RPR for quality rapid assessment. The quality of RPR cultivated in Sichuan, Anhui, and Zhejiang is similar based on the sum of these two bioactive compounds without significant differences.

ABSTRACT

Objective To observe the therapeutic effect of red peony root decoction (RPRD) on model rats with acute pancreatitis (AP),and the effect on the expression of NF-κB,TNF-α and IL-6.Methods Forty SD rats were randomly divided into five groups:normal control group (group A),AP model groups (group B,24 h; group C,72 h) and RPRD treatment groups (group D,24 h; group E,72 h).There were 8 rats in each group.The AP models were induced by intraperitoneally injecting with L-arginine.After the model was established,rats of group B and group C were treated with intra-gastric instillation of NS,and rats in group D and group E with RPRD.At 24 and 72 hours after induction of AP,rats in each group were sacrificed and blood samples were taken for the measurement of serum amylase,TNF-α and IL-6.The expression of NF-κB p65 protein in the pancreas were detected by Western Blot,and pathological status of the pancreas were analyzed.Results The serum amylase level in group B and group D was significantly higher than that in group A (P<0.05),and the level in group D was obviously lower than that in group B (P<0.05).The serum amylase level in group C and group E was decreased,and there was no difference between group A (P>0.05).The pathological severity and pathological scores of the pancreas in group D and group E were more lighter than those in group B and group C (P<0.05).The level of serum TNF-α and IL-6 was significantly decreased in RPRD treatment groups (P<0.05),group D compared with group B,[ (12.54±2.12)pg/ml,(552.88±81.27)pg/ml] vs.[ (20.53±4.16)pg/ml,(1255.75± 118.82)pg/ml]; group E compared with group C,[ (11.01 ±2.02)pg/ml,(582.87± 88.86) pg/ml] vs [ (23.85 ± 5.59) pg/ml,(1366.33 ± 163.16) pg/ml].The expression levels of NF-κBp65 protein in the pancreas of group D and group E were significantly lighter than those in group C at both time points (group D compared with group B,(0.07±0.055) vs (0.71 ±0.029),P<0.05; group E compared with group C,(0.15±0.038) vs (0.65±0.026).Conclusion RPRD may alleviate the pathologic changes in AP

ABSTRACT

Objective To determine and compare the contents of oxypaeoniflorin and paeoniflorin of Paeoniae Radix,and to improve the quality standard of Radix Paeonia.Methods The contents of oxypaeoniflorin and paeoniflorin in different kind of White and Red Peony Root were successfully analyzed by HPLC.Results The content of oxypaeoniflorin in Red Peony Root is more than White Peony Root at 0.8361% ;The content of paeoniflorin in White Peony Root is more than Red Peony Root at 0.2157%.The proportion of paeoniflorin and oxypaeoniflorin in White Peony Root is more than Red Peony Root.Conclusion With HPLC technology,quantitative analysis of active constituents in Paeoniae Radix was performed accurately.

ABSTRACT

In China, the roots of several species of Paeonia plants in the Paeoniaceae family have been used as crude drugs under the names of Ji-shao-yao, or the red peony, and Bai-shao-yao, or the white peony, since olden times. While in Japan, the simply dried root of Paeonia lactiflora Pallas has been used as the Chinese crude drug Shao-yao, or Shakuyaku in Japanese. As for the origins of the modern “red” and “white” peony names, there have been a variety of theories, e.g. the names were perhaps derived from differences in their root or flower colors, or whether they were wild or cultivated. Based on our herbological study, we have concluded that the dried root with a cork surface was named the red peony, and those peeled cork layers, the white peony. During the Ming Dynasty, in China, the root of wild peonies such as Paeonia veiitchii and P. obovata, whose flowers are reddish, were processed into the red peony, while cultivated peony root of the white flowered variety, P. lactiflora, was processed into the white peony drug. Because of this coincidence in flower color and name of the processed product, red flowered varieties or wild plants came to be called the plant origin of the red peony, while the white flowered varieties or cultivated plants came to be called the white peony.

ABSTRACT

AIM:To establish an HPLC fingerprint spectrum of Red Peony Root ( Radix paeonia rubra). METHODS:Ten batches of samples were extracted by 50% ethanol and extract liquor was evaporated to dryness,resolved with methanol. The HPLC analysis was performed on a Diamonsil C18 column with the eluting system of gradient acetonitrile and water. MS was applied to the determination of possible chemical structure. RESULTS:Nine main characteristic peaks were selected from the fingerprint spectrum of 10 batches of samples. Five constituents of which were identified as gallic acid,peoniflorin albiflorin,benzoylpaeoniflorin and paeonol based on the MS spectra. CONCLUSION:The method is valuable for quality conthol of Red Peony Root.
