ABSTRACT

Objective To study the effect of batroxobin combined with Yinxingdamo injection on serum endothelin(ET),nitric oxide(NO),superoxide dismutase(SOD),vascular cell adhesion molecule-1(VEGF-1)in patients with sudden deafness(SVCAM-1),to explore the best treatment for patients with sudden deafness.Methods Ninety patients with sudden deafness from June 2014 to June 2015 were recruited as the subjects of this study.The patients in control group were treated with batroxobin and batroxobin and ginkgo dipyridol.The levels of serum ET,NO,SOD,sVCAM-1,time of symptom recovery and the curative effect were observed.Results After treatment,the levels of ET,NO and sVCAM-1 in the observation group were significantly lower than those in the control group [(64.28±5.72)pg/mL vs(67.36±6.31)pg/mL,(43.08±9.53)μmol/(93.24±11.25)NU/mL](P<0.05).The results showed that there was no significant difference between the two groups.The recovery time of tinnitus,auria and vertigo in the observation group were less than those in the control group [(3.86±1.02)d vs(5.97±1.34),(5.03±1.24)d vs(7.37±2.01)d,(8.09±2.10)d vs 9.07±2.37)d](P<0.05).The total effective rate in the observation group was better than that in the control group(93.33%vs 75.55%,P<0.05).Conclusion Batroxobin combined with ginkgo dipyridolum injection can decrease the level of ET,NO and sVCAM-1,improve the level of SOD,and improve the microcirculation of the inner ear.Compared with the single effect of Batroxobin More desirable,worthy of promotion.

ABSTRACT

OBJECTIVE:To investigate the effects of doxorubicin perfusion therapy on therapeutic efficacy and related indexes in patients with superficial bladder cancer underwent transurethral resection of bladder tumor (TURBT).METHODS:Medical records of 96 patients with superficial bladder cancer were analyzed retrospectively and divided into observation group and control group according to drug use,with 48 cases in each group.Observation group was given perfusion therapy of 0.9% Sodium choride solution 40 mL containing 20 mg doxorubicin 7 d after surgery.Control group was given TURBT combined with equal amount of 0.9% Sodium chloride solution.Both groups received treatment for consecutive 10 months,once a week in first 8 weeks,later once a month.The recurrence rate and disease progression of 2 groups were observed and compared,and the levels of sICAM-1 and sVCAM-1,the levels of tumor marker DKK1 and VEGF were observed before and after treatment.The occurrence of ADR was recorded.RESULTS:After 24 months of treatment,remission rate and control rate of observation group were significantly higher than control group,with statistical significance (P<0.05).There was no statistical significance in the progressive survival rate between 2 groups (P>0.05).Before treatment,there was no statistical significance in the levels of sICAM-1、sVCAM-1、DKK1 and VEGF between 2 groups (P>0.05);after treatment,the levels of sICAM-1、sVCAM-1 、DKK1 and VEGF in 2 groups were significantly lower than before treatment,the observation group was significantly lower than the control group,with statistical significance (P< 0.05).The recurrence rate,the incidence of frequent/urgent urination,hematuria and dysuria in observation group after treatment were significantly lower than control group,with statistical significance (P<0.05).CONCLUSIONS:For patients with superficial bladder cancer underwent TURBT,doxorubicin perfusion therapy can significantly reduce recurrence rate,relieve tumor deterioration and reduce tumor activity with good safety.

ABSTRACT

Objective To analyse the effect of ursodeoxycholic acid on serum levels of cholyglycine ( CG ) , conjugated bile acid ( CBA ) and soluble vascular cell adhesion molecules (sVCAM-1) in patients with intrahepatic cholestasis of pregnancy.Methods 56 patients who were diagnosed with intrahepatic cholestasis of pregnancy in our hospital were collected.All patients were randomly divided into experimental group and control group, 28 cases in each group. The control group were treated with dexamethasone, and the experimental group were treated with the treatment of ursodeoxycholic acid, after 7d of treatment, the serum levels of glucocholic acid , CBA, ALT, AST and sVCAM-1 were detected in all patients. Results After treatment, compared with control group, the serum CG,TBA,ALT,AST and sVCAM-1 levels were significantly lower in the experimental group,and the difference was statistically significant (P<0.05).Conclusion The ursodeoxycholic acid can significantly reduce the serum CG,TBA, ALT,AST and sVCAM-1 levels in patients with intrahepatic cholestasis of pregnancy,improve pregnancy outcome,with guidance significance for clinic.

ABSTRACT

Objective To analysis the effect of Longdanxiegan capsule on serum sVCAM-1, immune globulin and SOD level in patients with sudden deafness and tinnitus.Methods 48 patients who were diagnosed with sudden deafness and tinnitus were collected.All patients were randomly divided into experimental group and control group,24 cases in each group.Patients in the control group received conventional western medicine treatment, patients in the experimental group were given Erlongzuoci pill on the basis of control group treatment , after the treatment, the serum levels of sVCAM-1, IgG, IgA, IgM and SOD were detected in all patients.Results After treatment, compared with control group, the serum levels of sVCAM-1 was lower in the experimental group,and the difference was statistically significant(P<0.05);the serum IgG, IgA and IgM levels were lower in the experimental group (P<0.05); the serum SOD level was higher in the experimental group (P<0.05).Conclusion The Longdanxiegan capsule can reduce significantly the serum sVCAM-1, IgG, IgA and IgM levels, improve serum SOD level in patients with sudden deafness and tinnitus, improve immune function and the ability to remove free radicals , and have a guiding significance for clinica .

ABSTRACT

Objective To investigate the changes of VCAM-1 and ICAM-1 levels in tumor and peripheral blood in pancreatic cancer patients. Methods 28 cases with pancreatic cancer were selected from August 2013 to Octorber 2014 as experiment group,28 cases with acute pancreatitis who needed removal of pancreas as control group,28 healthy people as normal group.ICAM-1 and VCAM-1 in tissue were compared and sVCAM-1 and sICAM-1 in serum were compared.Results ICAM-1 and VCAM-1 in tissue of experiment group were significantly higher than normal group(P0.05;r=0.093,P>0.05),there were positive correlation between CA19-9 and sICAM-1,sVCAM-1 in serum of experiment group(r=0.573,P<0.01;r=0.435,P<0.05).Compared with pre-operation, the serum concentrations of sVCAM-1 and sICAM-1 were lower(P<0.05).Conclusion ICAM-1 and VCAM-1in pancreatic cancer patients are significantly higher than that in acute pancreatitis patients and healthy people, the correlation of ICAM-1 and VCAM-1 expression in pancreatic cancer, early diagnosis and treatment of pancreatic cancer has important significance.

BACKGROUND: Soluble adhesion molecules including soluble vascular cell adhesion molecule-1 (sVCAM-1) are released during an infalmmatory process such as artherosclerosis. Elevated sVCAM-1 also has been reported in diabetic nephropathy. But, the clinical significance of elevated of sVCAM-1 is not certain. We measured serum sVCAM-1 for the purpose to validate the clinical usefulness in diabetic nephropathy. METHODS: In this study, we measured serum sVCAM-1 in 12 normal subjects and 64 type 2 diabetic patients with proteinuria over 300 mg/day [median 24-h urine protein (range): 2.2 (0.3-18.7) g/day]. We evaluated the relationship of serum sVCAM-1 with lipoproteins including total cholesterol, LDL, oxidized LDL (oxLDL), HDL, and lipoprotein(a) (Lp(a)), with markers of inflammation including high-sensitivity CRP (hs-CRP), serum albumin and fibrinogen, and with renal parameters including 24-h urine protein, serum creatinine and homocysteine. RESULTS: In patients with diabetic nephropathy, median sVCAM-1 was 561 ng/mL (range 183-1304), which was significantly higher than that of normal subjects (324 ng/mL; 213-760, p< 0.05). In the diabetic nephropathy patients, sVCAM-1 was positively correlated with serum creatinine (r=0.34, p< 0.01), serum Lp(a) (r=0.27, p< 0.05) and 24-h urine protein (r=0.26, p< 0.05). In a multiple linear regression analysis, 24-h urine protein and serum Lp(a) were associated with an increased level of sVCAM-1 (r2=0.22, p=0.003). CONCLUSION: In summary, a positive correlation of sVCAM-1 with 24-h urine protein suggests that high sVCAM-1 may reflect increased production of sVCAM-1 due to more advanced renal injury. A positive association of sVCAM-1 and serum Lp(a) also suggests increased release of sVCAM-1 from associated atherosclerotic lesions in these patients. These results suggest that sVCAM-1 may be closely related with the renal function in patients with overt diabetic nephropathy.

ABSTRACT

BACKGROUND: Cell adhesion molecules (CAMs) have been shown to be highly expressed in atherosclerotic lesions. Membrane-bound CAMs allow the tethering and rolling of monocytes and lymphocytes as well as the firm attachment and transendothelial migration of leukocytes. Soluble forms of CAMs may serve as monitors for increased expression of membrane-bound CAMs and thus may reflect progressive formation of atherosclerotic lesions. We assessed the role of the solu-ble CAMs in patients with type 2 Diabetes. METHODS: Serum levels of soluble E-selectin (sE-selectin), soluble intercellular adhesion molecule-1 (sICAM-1), and soluble vascular cell adhesion molecule-1 (sVCAM-1) were measured by enzyme immunoassay (R and D Systems, Minneapolis, USA) in patients with type 2 Diabetes (n=69) and normal control subjects (n=38). RESULTS: Fasting blood sugar, serum cholesterol, and blood pressure were significantly (P < 0.001) higher in diabetic patients than in control subjects. Serum sE-selectin, sICAM-1, and sVCAM-1 concentrations in diabetic patients were significantly (P < 0.001) higher than in the control subjects (69.7 +/- 32.0, 257.1 +/- 73.0 and 813.8 +/- 322.6 ng/mL versus 43.3 +/- 19.5, 173.1 +/- 66.8, and 400.4 +/- 77.4 ng/mL, respectively). The serum sICAM-1 concentrations in diabetic patients with microalbu-minuria were significantly (P=0.004) higher than in those patients without microalbuminuria (311.3 +/- 79.0 ng/mL versus 245.2 +/- 60.2 ng/mL). However, the sE-selectin and sVCAM-1 concentrations in diabetic patients with microalbuminuria were only slightly (P < 0.10) higher than in the patients without microalbuminuria. CONCLUSIONS: These results suggest that three kinds of circulating CAMs measured in this study increased significantly in patients with type 2 Diabetes. It is considered that circulating CAMs may be markers for atherosclerotic lesions in patients with type 2 Diabetes with symptomatic and asymptomatic atherosclerosis.

ABSTRACT

Soluble forms of ICAM-1 (sICAM-1) and VCAM-1 (sVCAM-1) have been reported from the supernatant of cytokine-activated endothelial cells, cancer cells and from sera of cancer patients. We measured sICAM-1 and sVCAM-1 from the serum of 20 healthy volunteers and 142 gastric cancer patients by ELISA assay. Ninety-five patients were operable and 47 patients were in-operable at the time of this study. Particularly in the 28 operable patients, we sampled both portal and peripheral blood simultaneously and measured the levels of the soluble forms of cell adhesion molecules (sCAMs). The sCAMs level and sero-positivity rate increased with cancer progression in order of the healthy controls, operable patients, and inoperable patients. In in-operable cancer, the sICAM-1 level increased more with liver metastasis. sICAM-1 and sVCAM-1 did not correlate with each other in either portal or peripheral blood. A total of 58.3% of patients with liver metastasis and 22.9% of patients without liver metastasis showed synchronous expression of both sCAMs (p = 0.03). Synchronous sero-positivity of sCAMs and alpha FP was higher with liver metastasis (p = 0.01). The median overall survival duration which co-expressed both sCAMs was 9 months. This showed a significant difference compared with the sICAMs non-expressing group, where the median survival was not reached until 24 months follow-up (p = 0.002). The synchronous expression of sCAMs was an independent risk factor in gastric cancer patients. We raise the possibility that synchronous sICAM-1 and sVCAM-1 elevation may be a useful monitor to determine tumor burden in gastric cancer.

ABSTRACT

BACKGROUND: Leukocyte-endothelial adhesion molecules have been implicated in the pathogenesis of inflammatory disease. ICAM-1, VCAM-1 and E-selectin are cell surface adhesion molecule on vascular endothelial cells. They are up-regulated by inflammatory cytokines and regulate the adhesion and migration of leukocytes across the endothelium. Tuberculosis, a granulomatous disorder is an infection caused by Mycobacterium tuberculosis. The clinical manifestations of tuberculosis are dependent on the cellular immune response to tubercule bacilli. Circulating adhesion molecules are probably formed by cleavage and release into the circulation of the extracellular domain of the membrane bound form. The elevated levels of circulating adhesion molecules have been reported in numerous disease state. To evaluate their role as markers of disease activity in tuberculosis, we measured a sE-selectin, sVCAM-1 and sICAM-1 levels in the serum with seventies of mild, moderate and far advanced pulmonary tuberculosis. METHODS: The control and test groups were divided as follows. Group I: control(n=5), Group II: atients with mild pulmonary tuberculosis(n=12), Group III: patients with moderate pulmonary tuberculosis(n=20), Group IV patients with far advanced pulmonary tuberculosis(n=19). Serum sICAM-1, sVCAM-1 and sE-selectin were measured by ELISA kit. RESULTS: Serum soluble adhesion molecules are elevated in patients with pulmonary tuberculosis. Circulating ICAM-1 levels were significantly elevated in patients with moderate and far advanced pulmonary tuberculosis when compared with control group. When compared with control group, serum sVCAM-1 levels showed significant elevation in patients with mild, moderate and far advanced pulmonary tuberculosis. Serum sE-selectin levels were significantly elevated in patients with far advanced pulmonary tuberculosis when compared with control group. CONCLUISON: These results suggest that sICAM-1, sVCAM-1, and sE-selectin may be invloved lix the pathogenesis of tuberculosis. And, particularly, sICAM-1 and sVCAM-1 may be useful markers of the disease activity.

ABSTRACT

Objective To explore the acute effects of nickel in air PM2.5 on the cardiovascular system of rats.Methods The air PM2.5 of nickel-contaminated area and control area were collected and determined for some major metal elements.42 male Wistar rats were randomly divided into 10 groups,6 rats in each,the rats were instilled through trachea with saline,PM2.5 suspension of nickel-contaminated area and the control area respectively,at different doses(low,moderate and high dose).24 hours later,all the rats were killed for the determination of sVCAM-1 in the blood and MCP-1 in the myocardial tissue.Results The concentration of nickel in the air PM2.5 of nickel-contaminated area was found to be about 50 times higher than that in the control area.A good dose-response relationship was found in MCP-1 determination of the heart tissue in nickel-contaminated groups,and there were significant differences between nickel-contaminated area group and other groups(P﹤0.05).As for the sVCAM-1 in the serum,it was significant higher in the nickel-contaminated group and control group than the saline control group(P﹤0.05).Among the groups of nickel-contaminated area and control area,significant differences were found between the moderate and high dose groups.Conclusion The air PM2.5 from nickel-contaminated area has an obvious effect on the MCP-1 and sVCAM-1 of rats,the nickel in the air PM2.5 is likely the main contributing factor.

ABSTRACT

Objective To study the effects of rAAV2/1-Acrp30 on sICAM-1 and sVCAM-1 level as well as NF-?B expression in GK rats with diabetic arteriosclerosis so as to explore the effect of adiponectin on diabetic macroangiopathy. Methods A total number of 30 atherosclerotic GK rat models were randomly divided into three groups:① Model group one (M1):hind limb intramuscular injection of normal saline; ② Model group two (M2):hind limb intramuscular injection of vacuity virus rAAV2/1; ③ Treatment group (T):hind limb intramuscular injection of rAAV2/1-Acrp30 at a dose of 1?1012mg/L. After 8 weeks' treatment,the rats were killed,and serum sVCAM-1 and sICAM-1 level as well as aortic NF-?Bp65mRNA expression were measured in each group. Aortic NF-?Bp65mRNA expression was measured by RT-PCR.Results Compared with those in control model groups (M1 and M2),sVCAM-1 and sICAM-1 levels were decreased significantly in the treatment (T) group (P
