ABSTRACT

Objective To investigate the expression and significance of the adaptor protein epsin 3 (EPN3) in colorectal cancer in order to provide reference for further stud)' of EPN3. Methods GEPIA and GEDS were used to analyze the expression of EPN3 in colorectal cancer tissues and cells. SMART and cBioPortal databases were used to analyze the relationship between EPN3 gene metfrylation and cop)' number variation and its expression level. Metascape was used to complete analysis of gene ontology functional annotation and related pathways of EPN3 related genes and BioPlex was applied to construct a protein network in HCT116 cell. Thirteen pairs of colorectal cancer adjacent tissue and cancer tissue specimens were collected, and EPN3 mRNA expression were detected by Real-time PCR. The effect of abilities of cell proliferation, clone formation and migration via silencing EPN3 in HCT116 and HT29 were observed. Results GEPIA, GEDS, SMART and cBioPortal analyses showed that EPN3 was highly expressed in colorectal tumor tissues (P<0. 01), and was related to methylation and copy number variation. The enrichment result of EPN3 related genes showed that it was mainly related to cell adhesion. And a protein interaction network constructed by CCDC130, TNFAIP1, PHGDH, EPN2, etc. was related to protein ubiquitination. Real-time PCR result showed that EPN3 was highly expressed in tumor tissues (P<0. 05). Silencing EPN3 inhibited the proliferation, clony formation and migration abilities of HCT116 and HT29 cells. Conclusion EPN3 is highly expressed in colorectal cancer tissues and is related to cell adhesion and protein ubiquitination. Down-regulated EPN3 can inhibit abilities of proliferation, clony formation and migration of HCT116 and HT29 cells, and this could provide a reference for further research on EPN3.

ABSTRACT

PURPOSE: The purpose of this study was to evaluate the effect of the zirconia surface architecturing technique (ZSAT) on the bond strength between veneering porcelain and zirconia ceramic. MATERIALS AND METHODS: 20 sintered zirconia ceramic specimens were used to determine the optimal surface treatment time, and were randomly divided into 4 groups based on treatment times of 0, 1, 2, and 3 hours. After etching with a special solution, the surface was observed under scanning electron microscope, and then the porcelain was veneered for scratch testing. Sixty 3 mol% yttria-stabilized tetragonal zirconia polycrystal ceramic blocks were used for tensile strength testing; 30 of these blocks were surface treated and the rest were not. Statistical analysis was performed using ANOVA, the Tukey post-hoc test, and independent t-test, and the level of significance was set at α=.05. RESULTS: The surface treatment of the zirconia using ZSAT increased the surface roughness, and tensile strength test results showed that the ZSAT group significantly increased the bond strength between zirconia and veneering porcelain compared to the untreated group (36 MPa vs. 30 MPa). Optimal etching time was determined to be 2 hours based on the scratch test results. CONCLUSION: ZSAT increases the surface roughness of zirconia, and this might contribute to the increased interfacial bond strength between zirconia and veneering porcelain.

ABSTRACT

O objetivo do presente estudo foi avaliar os parâmetros de adesão entre glaze e full-contour zirconia (FCZ) e FCZ glazeada e cimento resinoso. Zircônia e glaze tiveram as suas propriedades caracterizadas. Amostras de FCZ foram divididas em 5 grupos: (GL) aplicação de glaze; (AL+GL) jateamento com óxido de alumínio + glaze; (CJ+GL) partículas de óxido de alumínio modificada por sílica(Cojet®) + glaze; (SP+GL) tratamento químico com solução piranha + glaze; (CJ) Cojet. Microscopia eletrônica de varredura (MEV) e de força atômica (MFA) foram realizadas para avaliar a topografia das amostras. Scratch test foi realizado para avaliar o tipo de união entre zircônia e glaze após os diferentes tratamentos [(GL), (AL), (CJ) e (SP). Durante a cimentação, os grupos glazeados [(GL), (AL+GL), (CJ+GL) e (SP+GL)] foram tratados com ácido fluorídrico (HF) e silano; e o grupo não glazeado (CJ) apenas com silano. O cimento resinoso foi unido à zircônia utilizando um dispositivo específico para o teste de cisalhamento (SBS). Para a realização do teste SBS, metade dos espécimes de cada tratamento foi ensaiada 24 h após a cimentação e a outra metade foi envelhecida (armazenagem e termociclagem). Para a avaliação da retenção, foram confeccionados coroas FCZ e preparos para coroa total em substrato deresina epóxica (G10) (simulando um dente). Os grupos que receberam tratamento na superfície interna da coroa [(GL), (CJ+GL), (SP+GL) e (CJ)] foram cimentados com cimento resinoso sem MDP (Multilink Automix, 3M). O grupo que não recebeu tratamento na superfície interna (PF) foi cimentado utilizando um cimento resinoso contendo MDP (Panavia F, Kuraray). Os dados de adesão foram analisados estatisticamente (ANOVA e Tukey test). A full-contour zirconia apresentou alta densidade, microestrutura homogênea e propriedades mecânicas comparáveis à zircônia tradicional. The aim of the present study was to evaluate the adhesive parameters between glaze and full-contour zirconia (FCZ) and overglazed FCZ and resin cement. Zirconia and glaze were tested to determined theirs properties. FCZ samples were divided into 5 groups: (GL) glaze application; (AL+GL)sandblasting with aluminum oxide + glaze; (CJ+GL) tribochemical silica coating (Cojet®) + glaze; (SP+GL) chemical surface treatment with piranha solution+glaze; (CJ) Cojet. Electron scanning microscopy (SEM) and atomic force microscopy (MFA) were performed to evaluate the FCZ topography.Scratch test was executed to evaluate the bonding between FCZ and glazeafter the different surface pretreatment. For the bonding procedures the glazed groups [(GL), (AL+GL), (CJ+GL) and (SP+GL)] were etched with hydrofluoric acid (HF) and treated with a silane coupling agent and the group without glaze (CJ) was treated only with a silane. The resin cement was bonded to the zirconia surface using a dedicated device for the shear test(SBS). For the SBS test, half of the samples were tested after 24 h and theother half after storage and thermocycling. To evaluate the tensile retention of FCZ crowns cemented on epoxy resin (G10) specimens simulating fullcontour crowns were made. Pretreated groups [(GL), (CJ+GL), (SP+GL) and (CJ)] were bonded with a resin cement without MDP (Multilink Automix, 3M). The group without surface pretreatment (PF) was bonded with a MDP-based resin cement (Panavia F, Kuraray). The adhesive data were statistically analysed (ANOVA e Tukey test). FCZ showed high density, homogeneous microstructure and mechanical properties similar to the traditional zirconia. FCZ glaze was characterized as a leucite with irregular shape and size morphology. The scratch test pattern was similar for all the evaluated groups and classified as a cohesive failure of the glaze. ABSTRACT

STATEMENT OF PROBLEM: Regardless of any restoration, most of case, we used in screw connection between abutment and implant. For this reason, implant screw loosening has been remained problem in restorative practices. PURPOSE: The purpose of this study was to compare surface of coated/plated screw with titanium and gold alloy screw and to evaluate physical property of coated/plated material after scratch test in FESEM investigation. MATERIAL AND METHODS: GoldTite, titanium screw provided by 3i (Implant Innovation, USA) and TorqTite, titanium screw by Steri-Oss (Nobel Biocare, USA) and gold screw, titanium screw by AVANA (Osstem Implant, korea) - were selected for this study. Each abutment screw surface was observed at 100 times, and then screw crest, root, and slope were done more detailed numerical value, at 1000 times with FESEM. A micro-diamond needle was also prepared for the scratch test. Each abutment screw was fixed, micro-diamond scratch the surface of head region was made at constant load and then was observed central region and periphery of fine trace through 1000 times with FESEM. RESULTS: The surface of GoldTite was smoother than that of other kinds of screw and had abundant ductility and malleability compared with titanium and gold screw. The scratch test also showed that teflon particles were exfoliated easily in screw coated with teflon. Titanium screw had a rough surface and low ductility. CONCLUSION: It was recommended that the clinical use of gold-plated screw would prevent a screw from loosening.

ABSTRACT

A 24-year-old woman complained of chronic eczematous lichenified skin lesions with itching on both hands. The lesions were distributed mainly on the lateral and dorsal sides of the fingers. She had a history of atopic dermatitis and noticed itching sensation followed by erythema within minutes of contact with salmon at the respective contact sites, and subsequent formation of papules. Additionally, the ingestion of salmon resulted in swelling on her lips and throat. She had been working as a cook for 2 years and her sister had suffered from allergy to metal. Patch test with Korean standards and bakery series yielded positive to nickel and cobalt without clinical relevance. In a standardized skin prick test, strongly positive reactions to raw salmon and red snapper were observed after 20 min, but a patch test with the same substances was negative. We report a case of protein contact dermatitis due to salmon and red snapper.

ABSTRACT

Objective:To investigate the incidence of cedar pollinosis in district of Wuhan. Method..A surveyrelative to cedar pollinosis was carried out in 309 students in Hubei Medical University in 1999,including ques-tionnaire investigation, nasal inspection and scratch test. From February to April of 2000,a clinical investigationof cedar pollinosis was adopted anong 205 patients of allergic rhinitis including scratch test and nasal mucosatest. Result:①The positive rate of cedar pollen scratch test of 309 students was 7.8%. ②59 cases (28.8%00) washypersensitive to cedar pollen clinically in allergen test. Conclusion : Cedar pollinosis is one kind of importantpollinosis in district of Wuhan and should be paid high attention to.
