ABSTRACT

Objective To explore the effect and mechanism of Kuntai Capsule (KC) on premature ovarian failure in rats.Methods Totally 40 SD female rats were randomly divided into four groups:control group,model group,Kuntai Capsule (KC) group and conjugated estrogens tablets (CET) group.The premature ovarian failure model in rats was made by ig administration of 75 mg/kg tripterygium.After the model was established,rats were continually treated with 0.6 and 0.625 g/kg of KC and CET respectively by ig administration for 36 d.HE staining was used to observe the morphology of ovary and count the number of follicle.The enzyme-linked immunosorbent assay (Elisa) method was used for the detection of follicle stimulating hormone (FSH),luteinizing hormone (LH),serum estradiol (E2),and anti-Mueller tube hormone (AMH) level.The real-time fluorescence quantitative PCR (qRT-PCR) method was used to detect the mRNA expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF).Results Compared with control group,the number of primordial follicles,preantral follicles and antral follicles were significantly reduced,the number of atretic follicles was increased (P < 0.05),E2 and AMH significantly decreased (P < 0.05),FSH and LH levels increased significantly (P < 0.05),and the levels of VEGF and bFGF mRNA in model group were significantly decreased (P < 0.05).Compared with model group,the number of primordial follicles,preantral follicles,and antral follicles were significantly increased,the number of atretic follicles was significantly reduced (P < 0.05),E2 and AMH significantly increased (P < 0.05),FSH and LH levels significantly decreased (P < 0.05),and the levels of VEGF and bFGF mRNA in KC group were significantly increased (P < 0.05).Conclusion By regulating the level of hormone and up-regulating the expression of VEGF and bFGF,KC can repair the damaged ovarian tissue and promote the growth and development of the follicle,so as to inhibit the premature exhaustion of mRNA.

ABSTRACT

Objective To evaluate the value of serum E2 levels during COH in predicting IVF-ET outcome. Method Data from 311 IVF-ET cycles received long protocol were collected and analyzed according to E 2 levels 5 days after stimulation:Group A (E2≤500 pmol/L), Group B (500 2 000 pmol/L). Results In groups with E2>1 000 pmol/L, the conditions of oocyte and embryo are higher than E2 2 000 pmol/L, the pregnancy rate become lowest. Conclusion E2 levels among 1 000~1 500 pmol/L during early COH predicts a better pregnancy outcome.

ABSTRACT

It has been known that estradial(E2) level is concerned with the quality, number and growing of follicles. Determination of the serum levels of E2 has been used to predict the oocyte maturity. We experienced the rare case who has significantly low serum E2 level(551.26 pg/ml) in spite of multiple ovarian responses by sonographic finding, but from her we obtained 23 oocytes on MCD#16 after COH with ultrashort protocol(HP-FSH 150 IU, daily). The numbers of retrieved oocyte were 23 and fertilization rate was 56.6%, so the result of IVF-ET was successful twin pregnancy. Serum estradiol level and sonographic finding are essential to measure the number and growing of follicles, but E2 levels may be too unreliable to be used as sole parameter for the number and maturity of oocytes.

ABSTRACT

OBJECTIVE: To clarify whether hearing sensitivity in postmenopausal women is associated with serum estradiol level or bone mineral density (BMD) MATERIALS AND METHODS: From January 1999 to December 1999, serum concentration of estradiol, bone mineral densities of the lumbar vertebrae and femoral neck, and hearing sensitivity were measured in 1162 postmenopausal women. Serum concentration of estradiol was measured by radioimmunoassay. Dual energy X-ray absorptiometry (DEXA) was used to measure bone mineral densities of the lumbar vertebrae and femoral neck. Pure tone thresholds were evaluated by screening pure tone audiometry. All data were analyzed with Student's t-test, multiple logistic regression analysis, and multiple linear regression analysis. RESULTS: A total of 1155 patients were analyzed; 155 patients were in the hearing-loss group and 1000 patients were in the control group. On univariate analysis, significant differences were found in mean age, concentration of serum estradiol, and BMD of the femoral neck between the two groups (mean+/-SD, 58.1+/-5.2 vs 56.0+/-4.3, 11.6+/-6.0 vs 13.1+/-12.0, 0.67+/-0.11 vs 0.70+/-0.11; p-value: 0.0001, 0.01, 0.02). On multiple logistic regression analysis and multiple linear regression analysis, significant association was found between age and hearing loss and between age and pure tone threshold (p-value: 0.0001, 0.0001, respectively). However, neither seum concentration of estradiol nor BMD was associated with hearing loss or pure tone threshold. CONCLUSION: This study suggests that hearing sensitivity in postmenopausal women is not directly associated with serum estradiol level or BMD.
