ABSTRACT

Introduction: One of the main bottlenecks in restoration projects based on sexual reproduction is post-settlement survival, mainly due to competition for substrate with fleshy algae and predation. Therefore, substrates of different shapes and materials have been created and tested, seeking to optimize these processes with attractive surfaces for the larvae and structures where the recruits are protected from predation, and competition is reduced. Objective: To improve settlement and post-settlement survival of two important Caribbean reef-building corals, using different coatings on substrates. Methods: To determine whether substrate coatings properties are favourable to larval settlement in Orbicella annularis, and O. faveolata, collected in Puerto Morelos, Mexican Caribbean, we evaluated their settlement for three weeks on six coatings with a combination of properties. Each coating was designed to provide a combination of two out of three properties: 1) water repellence (hydrophobicity), 2) phosphorescence-based colour, and 3) mineral-enriched surface chemistry. In a separate experiment larvae settlement was tested using coatings with a single property. Finally, we determined the post-settlement survival of O. annularis and O. faveolata on the different coatings for seven weeks. Results: The combination of high hydrophobicity and light blue phosphorescent microparticles and high hydrophobicity and red-orange phosphorescent microparticles resulted in a higher settlement of O. annularis and O. faveolata when compared with other coatings (30.8 - 66.7 % higher). No significant differences were found in the number of larval settled when the water-repellence and the phosphorescence-based were evaluated independently. Post-settlement survival time on substrates was low, with a maximum of 34 days after settlement for O. annularis and 42 days for O. faveolata. Conclusions: In terms of the larval settlement, the combination of the coatings properties appears to play an essential role in the choice of microhabitat for both O. annularis and O. faveolata. But individually these properties did not generate an advantage in the larval settlement. Moreover, some chemical components associated with the coatings may be counterproductive to the survival of the polyps over time.


Introducción: Uno de los principales cuellos de botella en proyectos de restauración basada en reproducción sexual es la supervivencia de las larvas posterior al asentamiento, principalmente por la competencia por el sustrato con algas filamentosas y la depredación. Por ello, se han creado y analizado sustratos de diferentes formas y materiales, buscando optimizar estos procesos con superficies atrayentes para las larvas, y estructuras donde los reclutas se encuentran protegidos de la depredación y se disminuya la competencia. Objetivo: Mejorar el asentamiento y la supervivencia de dos importantes corales formadores de arrecifes del Caribe, utilizando diferentes recubrimientos en sustratos. Métodos: Para determinar si las propiedades de la superficie del sustrato son favorables para el asentamiento de larvas de Orbicella annularis y O. faveolata, recolectadas en Puerto Morelos, Caribe mexicano, evaluamos su asentamiento durante tres semanas en seis recubrimientos con una combinación de propiedades. Cada recubrimiento fue diseñado para proporcionar una combinación de dos de tres propiedades: 1) repelencia al agua (hidrofobicidad), 2) fosforescencia y 3) química superficial enriquecida con minerales. En un experimento separado se evaluó el asentamiento de larvas en sustratos con recubrimientos de una sola propiedad. Finalmente, se determinó la supervivencia posterior al asentamiento de O. annularis y O. faveolata sobre los diferentes recubrimientos durante siete semanas. Resultados: La combinación de alta hidrofobicidad y micropartículas fosforescentes azules y alta hidrofobicidad y micropartículas fosforescentes rojo-naranja dio como resultado un mayor asentamiento de O. annularis y O. faveolata en comparación con otros recubrimientos (30.8 - 66.7 % mayor). No se encontraron diferencias significativas en el número de larvas asentadas cuando se evaluaron de forma independiente la repelencia al agua y la fosforescencia. El tiempo de supervivencia posterior al asentamiento en los sustratos fue bajo, con un máximo de 34 días después del asentamiento para O. annularis y 42 días para O. faveolata. Conclusiones: En el asentamiento de larvas, la combinación de las propiedades del recubrimiento parece desempeñar un papel importante en la elección del microhábitat tanto para O. annularis como para O. faveolata. Pero de forma individual estas propiedades no generaron una ventaja en el asentamiento larvario. Además, algunos componentes químicos asociados con los recubrimientos pueden ser contraproducentes para la supervivencia de los pólipos a lo largo del tiempo.

ABSTRACT

Pneumocystis, an important opportunistic fungal pathogen that parasitizes in multiple mammalian lungs, may cause life-threatening Pneumocystis pneumonia (PCP) and even death among immunocompromised individuals. With the rapid development of high-throughput sequencing and multi-omics technologies, systematic comparative analyses of genome, transcriptome, and whole-genome sequencing results demonstrate that Pneumocystis is a type of obligate biotrophic fungi, and requires obtaining nutrition from hosts. In addition, sexual reproduction is an essential process for Pneumocystis survival, production and transmission, and asexual reproduction facilitates Pneumocystis survival, which provides new insights into understanding of the whole developmental process of Pneumocystis in the host lung and inter-host transmission of Pneumocystis. This review summarizes the advances in the reproduction mode of Pneumocystis and underlying mechanisms, which provides insights into prevention and treatment of PCP, notably for the prophylaxis against nosocomial transmission of PCP.

ABSTRACT

Apomixis or asexual reproduction through seeds occurs in about 400 species of flowering plants producing geneticallyuniform progeny. During apomixis, meiosis is bypassed and embryos develop by parthenogenesis. However, the endospermcould form either autonomously without fertilization or sexually, depending on the plant species. Most probably, aheterochronic expression of sexually expressed genes is one of the reason that causes apomixis. A better understanding ofthe genetic components regulating apomixis is important for developmental and evolutionary studies and also for engineering apomixis traits into crop plants that may realize a possibility to propagate hybrid vigor in a range of subsequentgenerations.

ABSTRACT

A reprodução sexuada já foi considerada universal, e posteriormente, a forma mais perfeita de reprodução. Todavia, a partir de meados do século XIX, pesquisas no nível celular colocaram em xeque a ideia de que tipos de reprodução assexuadas fossem primitivos ou inferiores. Ao longo do século XX, e adentrando no XXI, hipóteses foram levantadas para explicar as vantagens da reprodução sexuada sobre a assexuada assim como o que permitiria a reprodução sexuada se manter quando seria mais vantajoso se reproduzir de forma assexuada. A mais importante e conhecida é a hipótese da Rainha Vermelha. Paralelamente, vários trabalhos procuraram entrever as pressões ecológicas que permitiram e favoreceram o aparecimento da reprodução sexuada em um cenário situado há cerca de dois bilhões de anos. O objetivo desse trabalho é revisar respostas históricas que marcaram o estudo da origem, da evolução e da manutenção da reprodução sexuada, identificando algumas das principais questões que a comunidade científica elaborou nos últimos duzentos anos.


Sexual reproduction has already been considered universal, and subsequently, the most perfect form of reproduction. However, since the mid-nineteenth century, research at the cellular level has questioned the idea that asexual reproduction types are primitive or inferior. During the twentieth century, and entering the XXI, hypotheses were raised to explain the advantages of sexual reproduction over the asexual as well as what would allow sexual reproduction to be maintained when it would be more advantageous to reproduce asexually. The most important and known is the Red Queen hypothesis. At the same time, several studies have sought to understand the ecological pressures that allowed and favored the appearance of sexual reproduction in a scenario that was around two billion years ago. The aim of this work is to review historical responses that marked the study of the origin, evolution and maintenance of sexual reproduction, identifying some of the main questions that the scientific community has elaborated over the last two hundred years.

ABSTRACT

Fungal pathogens represent an important group of human pathogenic microbes that lead to an unacceptably severe global burden especially due to exceptionally high mortality. For many fungal pathogens, they are widespread saprophytes and human host is not the exclusive niche for their proliferation. Their exceptional capability to survive and thrive within infected host likely stems from their sophisticated strategies in adaptation to diverse biotic and abiotic stressors from natural niches or predators. Among these 'environmental pathogens', Cryptococcus neoformans as a model organism claims the lives of more than half a million annually. Some recent studies indicate that cryptococcal survival both inside and outside of hosts can be coordinated by a combination of social behaviors. In this review, we describe and discuss the social behaviors employed by C. neoformans and address their significant impact on biofilm formation, sexual reproduction and pathogenicity.

ABSTRACT

Body size is one of the most important factors regarding herbaceous perennial plants life-histories, and several fitness components of these organisms are related to size. Clonal plants show distinct kinds of reproduction and can develop offspring by sexual or asexual ways. We aimed to understand how body size affects Comanthera nivea (Eriocaulaceae) sexual reproduction and to verify how clonal growth is related to flower head production in this species. We sampled 600 rosettes in rupestrian grasslands and performed linear regression analysis between body size and number of produced flower heads. We also compared the flower head production between isolated rosettes and rosettes within clones. Our results showed that body size was significantly related, but explained only a small part of flower head production. The flower head production was higher in rosettes within clones than in isolated ones. The clones presented a rosette or a small group of rosettes that concentrated the sexual reproduction. Clonality was positively associated with sexual reproduction. Clonality can represent an important way of allowing the persistence of plants by sexual reproduction in markedly seasonal stressful environments. The cases of clonality enhancing the sexual reproduction must be considered and put in focus on reproductive biology research.


O tamanho de corpo é um dos atributos de história de vida mais importantes para plantas herbáceas perenes e muitos componentes da aptidão desses organismos são relacionados ao tamanho. As plantas clonais apresentam diferentes tipos de reprodução e podem gerar prole por vias sexuadas ou assexuadas. Nosso objetivo foi compreender como o tamanho de corpo afeta a reprodução sexual em Comanthera nivea e verificar como o crescimento clonal relaciona-se à produção de capítulos dessa espécie. Nós amostramos 600 rosetas em campos rupestres e utilizamos regressões lineares para verificar a relação entre o tamanho de corpo e o número de capítulos produzidos pela planta. Nós também comparamos a produção de capítulos entre rosetas isoladas e rosetas pertencentes a clones. Nossos resultados mostram que o tamanho de corpo tem influência positiva significativa na produção de capítulos, mas explica apenas parte da variação nos dados. A produção de capítulos é maior em rosetas pertencentes a clones que em rosetas isoladas. Os clones geralmente apresentam uma roseta, ou um grupo de rosetas, que concentra a reprodução sexual. A clonalidade apresentou ligações significativas com a reprodução sexuada. Em ambientes sazonais a clonalidade pode representar uma importante forma de persistência das plantas já que pode permitir um melhor desempenho da reprodução sexuada em vista dos estresses ambientais. Os casos nos quais a clonalidade representa uma via de melhor desempenho para a reprodução sexuada devem ser considerados e colocados em foco na pesquisa em biologia reprodutiva.

ABSTRACT

Fluctuations in world harvests of Artemia sp cysts have increased prices sharply in the past years. Several organisms have been tested as alternative sources for a total or partial replacement of Artemia sp, among which the microcrustacean (Anostraca) Dendrocephalus brasiliensis stands out. The objective of this study was to investigate the reproductive strategy of D. brasiliensis by means of AFLP markers. The distinction between sexual and parthenogenetic reproduction was conducted using 7 EcoRI and MseI primer combinations in a full-sib family composed of 13 progenies. Forty polymorphic markers were obtained, of these, 35% were from paternal origin and 65% were maternally inherited. Results showed mendelian segregation in all markers through the chi-square at P≤0.05. Sex changes were observed after 72 hours of grouping individuals of the same sex in a container, suggesting a possible sex density-dependent behavior. Paternal inheritance revealed that this species adopts sexual reproduction under experimental conditions.


Há vários anos que a produção de cistos de Artemia sp. apresenta flutuações, o que aumenta consideravelmente o seu valor. Vários organismos têm sido testados para a substituição total ou parcial da Artemia, dentre os quais se destaca o microcrustáceo (Anostraca) Dendrocephalus brasiliensis. O objetivo do presente trabalho foi investigar a estratégia reprodutiva de D. brasiliensis por meio de marcadores de AFLP. A distinção entre uma reprodução sexuada e partenogenética foi feita a partir da genotipagem de sete combinações de primers de AFLP em uma família composta de 13 progênies. Quarenta marcadores polimórficos foram obtidos, dos quais 35% foram de origem paterna e 65%, materna. Os resultados mostraram a segregação mendeliana de todos os marcadores através do teste de qui-quadrado a P≤0.05. Mudanças de sexo foram observadas após 72 horas do agrupamento de indivíduos de mesmo sexo em um recipiente, sugerindo uma possível razão sexual populacional densidade-dependente. A herança paterna mostrou que a reprodução sexual é adotada por essa espécie sob condições experimentais.

ABSTRACT

Objetivo. Evaluar el efecto de dos inductores hormonales en la reproducción inducida de nicuro Pimelodus blochii. Materiales y métodos. Para los procesos experimentales fueron utilizados adultos sexualmente maduros, sometidos a tres tratamientos aplicados vía intramuscular, en dosis única de 0.25 mL/kg Ovaprim® (OVAP) (T1), 0.5 mL/kg de OVAP (T2) y 6.25 mg/kg de Extracto de Hipófisis de Carpa (EHC) (T3), para este último tratamiento la inyección fue dividida en 20 y 80%, con un intervalo de 12 h entre aplicaciones. Previo a la extracción de los gametos, los animales fueron tranquilizados por inmersión en una solución de Metanosulfonato de Tricaina (90 mg/L). El desempeño reproductivo fue evaluado mediante el índice de ovulación (hembras ovuladas/hembras tratadas), fecundidad absoluta (Fa) (ovocitos/hembra), fecundidad relativa (Fr) en función del número de ovocitos desovados por gramo de peso. La fecundación se realizó en seco y seis horas post-fecundación (HPF) se determinó la tasa de fertilidad. Resultados. La ovulación (ºh) para el T1 fue a las 297.1±30.0, T2 294.6±32.9 y T3 247.3±13.1 ºh. En todos los tratamientos se obtuvieron hembras ovuladas, donde los mayores índices de ovulación fueron obtenidos con Ovaprim® (T1 y T2) con 36.4 y 50%, respectivamente. Las tasas de fecundación obtenidas fueron mayores a un 50%, para el tratamiento 1 y 2, con valores de 74.5 y 32.7%, respectivamente. Conclusiones. El uso de inductores hormonales puede ser efectivo para garantizar la reproducción inducida del nicuro, en dosis única de 0.25 y 0.5 mL/kg de Ovaprim®.


Objective. Evaluate the effect of two hormonal inducers in induced breeding of nicuro Pimelodus blochii. Materials and methods. The experimental process used sexually mature adults exposed to three intramuscular treatments, a single dose of 0.25 mL/g Ovaprim® (OVAP) (T1), 0.5 mL/kg of OVAP (T2) and 6.25 mg/kg of pituitary carp extract (EPC) (T3), for the latter treatment the injection was divided into 20 and 80%, with an interval of 12 hours between applications. Prior to the removal of gametes, the animals were tranquilized by immersion in a Tricaine Methanesulfonate (90 mg/L) solution. Reproductive performance was evaluated using ovulation rates (females ovulated/treated females), fertility (Fa) (eggs/female), relative fertility (Fr) depending on the number of eggs spawned per gram of weight. The dry method of fertilization was used and the fertility rate was determined six hours post-fertilization (HPF). Results. The Ovulation (ºh) was: for T1 297.1±30.0, T2 294.6±32.9 and T3 247.3±13.1ºh. Ovulated females were obtained in all treatments, where the highest rates of ovulation were obtained with Ovaprim ® (T1 and T2) with 36.4 and 50% respectively. Fertilization rates were higher than 50% for treatments 1 and 2, with values of 74.5 and 32.7% respectively. Conclusions. The use of hormonal inducers may be effective to ensure nicuro induced reproduction with a single dose of 0.25 and 0.5 mL/kg Ovaprim®.

ABSTRACT

Since cerrado fires may impede the growth of seedlings into trees, they may shape the population of woody species. In this study, we assessed the effects of a severe fire on the population structure and spatial distribution of Zanthoxylum rhoifolium, a widespread cerrado tree. We were interested to know the importance of the resprouting and sexual reproduction in regenerating the population. The study area had been for about six years protected from fire, before a severe fire at the end of the dry season in 2006. We sampled and measured all individuals of Z. rhoifolium found in 80 plots of 25 m². We found 149 individuals before the fire and 112 after the fire, of which 77 were resprouts from burned seedlings and saplings. We did not find significant differences between the population structure before and after the fire. The spatial distribution of the population remained clumped after the fire. Thus, the Z. rhoifolium population was very resilient to a severe fire. We did not find any new seedlings. As a consequence, resprouting seems to be more important than sexual reproduction in promptly regenerating the Z. rhoifolium populations.


Uma vez que as queimadas no cerrado podem impedir o crescimento de plântulas, elas podem moldar a população de espécies arbóreas. Neste estudo, avaliamos os efeitos de uma queimada severa na estrutura e na distribuição espacial da população de Zanthoxylum rhoifolium, uma espécie de árvore comum no cerrado. O objetivo era saber a importância relativa do rebrotamento e da reprodução sexuada na regeneração da população. A área de estudo, há aproximadamente seis anos sem queimar, sofreu uma queimada severa no final da estação seca de 2006. Amostramos todos os indivíduos de Z. rhoifolium encontrados em 80 parcelas de 25 m². Encontramos 149 indivíduos antes do fogo e 112 depois dele, dos quais 77 eram rebrotas de plântulas e jovens queimados. Não encontramos diferenças significativas entre a estrutura da população antes e após o fogo. A distribuição espacial da população permaneceu agregada após a queimada. Portanto, a população de Z. rhoifolium foi resiliente ao fogo. Não foi encontrada nenhuma nova plântula. Consequentemente, o rebrotamento parece ser mais importante que a reprodução sexuada na regeneração imediata das populações de Z. rhoifolium.

Dypsis decaryi (Jum.) Beentje & J. Dransf. é uma importante palmeira comercializada para fins ornamentais. Embora seja propagada sexuadamente, não há informações na literatura sobre os fatores que afetam a germinação das sementes dessa espécie. Objetivou-se, neste trabalho estudar o efeito da temperatura e do substrato na germinação de sementes de Dypsis decaryi. O delineamento experimental foi o inteiramente casualizado em esquema fatorial 6 x 2 (seis temperaturas: 25ºC, 30ºC, 35ºC, 20-30ºC e 25-35ºC e ambiente combinadas com dois substratos: vermiculita e areia), com 5 repetições de 20 sementes. Anotou-se, diariamente, o número de sementes germinadas adotando como critério de germinação a protrusão do botão germinativo. Após 35 dias houve estabilização dos dados e foi calculado o Índice de Velocidade de Germinação (IVG) e a porcentagem de germinação (dados transformados em arc sen √x/100) cujas médias foram comparadas pelo teste de Scott-Knott, a 5 por cento de probabilidade. Observou-se que maiores porcentagens de germinação de sementes de Dypsis decaryi foram obtidas quando colocadas para germinar em areia nas temperaturas ambiente de laboratório (93 por cento), 25-35ºC (92 por cento) e 25ºC (86 por cento), e em vermiculita, nas temperaturas de ambiente (91 por cento), 20-30ºC (87 por cento), 30ºC (83 por cento), 25-35ºC (80 por cento) e 25ºC (76 por cento). As sementes germinaram mais rápido em condições de ambiente de laboratório independentemente do substrato. Na temperatura de 35ºC a porcentagem de germinação foi bastante reduzida tanto em areia (21 por cento) como em vermiculita (38 por cento); as sementes germinaram mais lentamente e a germinação foi desuniforme.


Dypsis decaryi (Jum.) Beentje & J. Dransf. is an important palm with ornamental commercial value. Although it reproduces sexually, there are no data available as far as what are the factors that affect this species seed germination. The purpose of this work is to study the potential effects of temperature and substrate in Dypsis decaryi seed germination. The research framework is based on randomization, using a 6 X 2 factorial schema (six different temperatures, 25ºC, 30ºC, 35ºC, 20-30ºC, 25-35ºC and room temperature, combined with two types of substratum, sand and vermiculite) with 5 repetitions of 20 seeds. The researcher made daily records on the number of germinated seeds, and the criteria used was protusion of the germination plug. After 35 days data become stable. The Index of Germination Speed (IVG) and germination percentage were then calculated (data were converted to sen). The averages for these two indexes were compared using the Scott-Knott method with a probability of 5 percent. The data show that the higher percentages of germination were obtained with a sand substratum at temperatures of 25ºC (86 percent), 25-35ºC (92 percent) and lab room temperature (93 percent), and with a vermiculite substratum, at temperatures of 25ºC (76 percent), 30ºC (83 percent), 20-30ºC (87 percent), 25-35ºC (80 percent) and room temperature (91 percent). The seeds germinated sooner in the lab environment, for both substrates. The percentage of germination at 35ºC was markedly lower, both on sand (21 percent) and vermiculite (38 percent). In this case, the seeds took longer to germinate and germination was not uniform.
