ABSTRACT

@#Objective To screen a kind of solid culture medium without animal-derived materials for resuscitating Streptococcus pneumoniae(Spn).Methods Spn 19F was selected for the preliminary screening of 9 kinds of formula culture media without animal-derived materials,and then other 23 serotypes of Spn were cultured for verification.The effect of the selected medium on the hereditary stability of various serotypes was identified by strain identification and capsular polysaccharide antigen gene sequence analysis of the 15-generation strains.The strains resuscitated by this medium were fermented according to the production procedure,and the culture situation and the yield and quality of capsular polysaccharide antigen were analyzed to determine the production applicability of the screened medium.Results The number of viable bacteria of various types of Spn was desired after 11~15 h of incubation on the selected solid medium and there was no difference in strain identification and antigen gene sequence between the final 15 generation and the control(strains cultured in sheep blood solid medium).After fermentation and culture of the strain resuscitated by this medium,the cell growth and the capsular polysaccharide production increased slightly,and all the related verification indicators of capsular polysaccharide met the requirements in Chinese Pharmacopoeia(Volume Ⅲ,2020 edition).Conclusion The selected solid culture medium without animal-derived materials has good applicability and can replace blood-derived culture medium for the resuscitation of Spn strain for vaccine production.

ABSTRACT

Objective To determine the effects of different strength of Murashige and Skoog (MS) media (full, ½ and ¼) in solid and liquid media on in vitro growth of Typhonium flagelliforme (T. flagelliforme), whereby an optimum media composition can be provided for mass propagation of T. flagelliforme. Methods Rhizome bud of T. flagelliforme was obtained from the axenic in vitro established T. flagelliforme plantlets in Plant Tissue Culture Laboratory, Universiti Teknologi MARA, Shah Alam. Rhizome bud was used as explant and cultured onto shoot proliferation medium under different strength of MS media (full, ½, ¼) in solid and liquid culture media. Results After 6 weeks of culture, the number of shoot, number of leaf, number of root, height of shoot, fresh weight, dry weight and chlorophyll content of T. flagelliforme were analyzed. A comparison was made between liquid and solid culture media. The results revealed that the liquid culture media were more effective for all the growth parameters (shoot height, shoot number, leaf number, root number, fresh weight, dry weight, chlorophyll a and chlorophyll b content) compared to solid culture media. Apart from that, this study revealed the positive relationship between strength of MS media and type of culture media (solid and liquid media) to the growth of T. flagelliforme. Growth of T. flagelliforme was improved when MS strength was increased in liquid media. In contrast, growth of T. flagelliforme was improved when MS strength was decreased in solid media. Conclusions Through this study, an optimum media composition for mass propagation of T. flagelliforme had been established by observing effects of MS media strength and type of culture media (solid and liquid media) on the growth of T. flagelliforme.

ABSTRACT

Objective To study the characteristic 2-(2-phenylethyl)chromone components of endophytic fungal strain of Aquilaria sinensis by solid culture. Methods The compounds were isolated by various chromatographic methods such as silica gel, reverse-phase silica gel, Sephadex-LH20 column chromatography as well as crystallization. Results Seven 2-(2-phenylethyl)chromone analogues were isolated from the solid culture of Botryosphaeria rhodina A13. Their structures were established by spectral data as well as physicochemical properties, and identified as 6-hydroxy-7-methoxy-2-(2-phenylethyl)chromone (1), 6,7-dimethoxy-2-(2-phenylethyl) chromone (2), (5S,6R,7S,8R)-2-(2-phenylethyl)-5,6,7,8-tetrahydrchromone (3), 6-hydroxy-2-(2-phenylethyl)chromone (4), 4′-hydroxy-2-(2-phenylethyl)chromone (5), 6-methoxy-2-phenethyl-4H-chromen-4-one (6), and 6-methoxy-2-(4′-methoxy-phenethyl)-4H-chromen-4-one (7). Conclusion All of the compounds are isolated for the first time from the genus Botryosphaeria. This research opens up a new vista to produce the characteristic components of agarwood by endophytic fungi.

ABSTRACT

Objective To evaluate the BacT/ALERT 3D liquid rapid culture system for the rapid detection of mycobacterium tuberculosis(MTB)and early diagnosis of pulmonary tuberculosis.Methods BacT/ALERT 3D liquid culture mediums and lowenstein-jensen(L-J)solid culture mediums were applied to detect Mycobacterium tuberculosis sputum specimens respectively.Analysis of detection results and detection time were also performed.Results Average positive days of BacT/ALERT 3D liquid culture medium was(13 ±2.05)days,which was shorter than that of L-J solid culture mediums(34.7 ±4.76 )d (P<0.05 ).Compared to the L -J solid culture mediums,BacT/ALERT 3D liquid culture medium had higher positive rate for 59 patients whose sputum smear test was positive,and the positive rate were 71.18%(42/59)and 67.80%(40/59)respectively(P<0.05).BacT/ALERT 3D liquid culture medium had higher positive rate than L-J solid culture mediums for 106 patients whose sputum smear test was negative,and the positive rate were 39.62%(42/106)and 26.42%(28/106)respectively(P<0.05).Conclusion Compared to the traditional L-J solid culture system,BacT/ALERT 3D liquid culture system can shorten detection time and improve the positive detection rate of MTB in specimens with low concentration.

ABSTRACT

OBJECTIVE: To optimize the solid culture condition of three fungi which can induce Aquilaria sinensis to produce agilawood, thus to provide data for the production and application of the fungi. METHODS: Single factor experiment method was applied in the study. RESULTS: The optimal solid culture medium formula, temperature, pH value, moisture, the density of materials and illumination conditions for three fungi were obtained. CONCLUSION: The optimized method can provide high quality fungi for prompting Aquilaria sinensis to produce agilawood at larger scales. Copyright 2012 by the Chinese Pharmaceutical Association.

ABSTRACT

Avaliou-se o processo de enriquecimento protéico da palma forrageira (Opuntia ficus-indica Mill) com levedura Sacharomyces cerevisiae em cultivo semi-sólido, visando melhorar o valor nutritivo da palma para ser utilizada na alimentação de ruminantes. A levedura foi utilizada nas concentrações de 1, 2 e 3 por cento em base úmida no substrato formado pela palma forrageira, incubada em biorreatores durante 6, 12, 24 e 36 horas de fermentação. O delineamento experimental foi inteiramente ao acaso, em arranjo de parcelas subdivididas com quatro repetições. O conteúdo de proteína bruta quando se utilizou concentração de 3 por cento de inóculo, no período de seis horas, aumentou de 4,4 por cento na forma in natura para 10,4 por cento após o processamento. Os teores protéicos na concentração de 1 por cento do inóculo foram de 6,1, 8,1, 8,1 e 9,2 por cento; na concentração de 2 por cento, 9,6, 9,7, 9,8 e 9,8 por cento e na concentração de 3 por cento, 10,4, 10,4 7,9 e 7,9 por cento, nos períodos de 6, 12, 24 e 36 horas de fermentação, respectivamente. Uma fonte alternativa para arraçoamento de ruminantes, pode ser obtida pela bioconversão da palma forrageira.


The process of protein enrichment of the forage palm (Opuntia ficus-indica Mill) using the Saccharomyces cerevisiae yeast in a semi-solid culture to improve the nutritional value of forage palm for ruminants feeding was evaluated. The yeast concentrations of 1, 2 and 3 percent (wet basis) in the forage palm substrate were used. The periods of incubation were of 6, 12, 24, and 36 hours. A complete randomized experimental design in a split plot arrangement with four replicates was used. The crude protein content increased from 4.4 percent (in natura) to 10.4 percent when 3 percent of inoculums were used and the processing period was of 6 hours. The observed protein contents for 1 percent of the inoculum, used for the fermentation periods of 6, 12, 24, and 36 hours were 6.1, 8.1, 8.1, and 9.2 percent, respectively. These values were 9.6, 9.7, 9.8, and 9.8 percent for 2 percent of the inoculum, and 10.4, 10.4, 7.9, and 7.9 percent for 3 percent of the inoculum, respectively. An alternative for ruminant feeding can be obtained by bioconversion of forage palm.
