ABSTRACT

Herbicide(s) commonly known as weedkillers, are chemical substances used to control unwanted plants. Paraquat(PQ) (1, 1-dimethyl 4, 4- bipyridillium dichloride) is the active component of the herbicide (Parae Force) used inthis study.This study was performed on 25 male Wistar rats that were divided into 5 groups (A, B, C, D and E), Group A servedas the control group that received only distilled water, Group B, C, D & E served as the treated groups that received25%, 50%, 75%, & 100% leachate of Paraquat dichloride concentration respectively for 8 weeks.This study investigated the effect of paraquat dichloride on the testes, sperm characteristics and body weight inadult male Wistar rats. Histological analysis of the testes as well as semen analysis was carried out.Histomorphometry study of the testes showed deformed seminiferous tubules with irregular shape andspermatogenesis was arrested. Interstitial space was comparatively loose and disrupted Leydig cells are alsovisible. Seminiferous tubules also exhibiting degenerated basement membrane along with degenerated interstitialcells, degenerated spermatogonia and damaged Sertoli cells. Consequently, the sperm count from the Paraquatdichloride exposed semen revealed significant decreased results, the sperm morphology and sperm progressivitywere also affected negatively.In conclusion, the present study indicates that Paraquat dichloride demonstrated toxic effects on malereproductive parameters.

ABSTRACT

Objective To investigate the correlation between three sperm indexes and the outcome of in vitro fertilization and embryo transfer(IVT-ET) before and after semen treatment.Methods The semen derived from 298 male patients treated with IVT-ET from the Affiliated Hospital of Southwest Medical University was retrospectively analyzed.Routine analysis of semen before treatment was performed according to the WHO standard.Vitrolife density gradient centrifugation combined with upstream method was used to analyze sperm parameters after sperm analysis,and the observation and statistics were performed after fertilization and sequential culture to day 3.Results The total viability of spermatozoa before and after semen treatment,the percentage of normal spermatozoa,sperm density,and other differences were statistically significant (P<0.05);The normal sperm morphology rate was positively correlated with fertilization rate(r=0.487,P<0.01) and cleavage rate(r=0.250,P<0.05);The sperm vitality of post-processed semen was positively related to the fertilization rate (r =0.249,P<0.05,);and significantly cleavage rate (r =0.272,P<0.05);After treatment,sperm density was positively correlated with 2 PN(r=0.609,P<0.01) and multiple PN fertilization rates(r=0.243,P<0.05).Conclusion Our experimental results indicate that the parameters of post-processed sperm treated at the day when the oocytes were collected may be more helpful in predicting the fertilization rate and cleavage rate of IVF-ET.Therefore,the IVF-ET treatment protocols should be chose based on the normal sperm morphology rate and sperm vitality after semen treatment.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the health effects of parental dietary exposure to GM rice TT51 on the male reproductive system of rat off spring.</p><p><b>METHODS</b>Rice-based diets, containing 60% ordinary grocery rice, MingHui63, or TT51 by weight, were given to parental rats (15 males/30 females each group) for 70 days prior mating and throughout pregnancy and lactation. After weaning, eight male offspring rats were randomly selected at each group and fed with diets correspondent to their parents' for 70 days. The effects of exposure to TT51 on male reproductive system of offspring rats were assessed through sperm parameters, testicular function enzyme activities, serum hormones (FSH, LH, and testosterone levels), testis histopathological examination, and the relative expression levels of selected genes along the hypothalamic-pituitary- testicular (HPT) axis.</p><p><b>RESULTS</b>No significant differences were observed in body weight, food intake, organ/body weights, serum hormone, sperm parameters, testis function enzyme ACP, LDH, and SDH activities, testis histopathological changes, and relative mRNA expression levels of GnRH-R, FSH-R, LH-R, and AR along the HPT axis.</p><p><b>CONCLUSION</b>The results of this study demonstrate that parental dietary exposure to TT51 reveals no significant differences on the reproductive system of male offspring rats compared with MingHui63 and control.</p>

ABSTRACT

Objective To study the influences of the length of sexual abstinence on sperm parameters and seminal plasma biochemical indicator.Methods Sperm concentration and motility were detected by computer assisted sperm analysis(CASA).The sperm morphology was analyzed using semiautomatic sperm morphology analyzer.Seminal plasma fructose concentration,neutral a-glycosidase,zinc,and acid phosphates concentration were measured by spectrophotometer.Improved DTNB was conducted for detection of the content of seminal plasma novain.Prostatespecific antigen(PSA)was measured with commercial kit.Three groups were defined according to the days of sexual abstinence:G1(1-3 days),G2(4-5 days)and G3(>6 days).Results Sperm concentration of G2 was significantly higher than that of G1(P<0.01),and of G3 than that of G1 and G2(P<0.01).Sperm motility and per centage of morphologically normal sperm of G3 were significantly lower than that of G1(P<0.01).Seminal plasma fructose concentration of G3 was significantly lower than that of G1 and G2(P<0.01,P<0.05).Seminal plasma neutral a-glycosidase,carnutine,zinc concentration of G3 was significantly higher than that of G1 and G2(P<0.01,P<0.05).Conclusion The length of sexual abstinence can influence sperm parameters and seminal plasma biochemical indicator.

ABSTRACT

OBJECTIVE To study the relationship between Gardnerella vaginalis and male infertility. METHODS To test G.vaginalis in semen by nested PCR and to carry out correlation analysis with sperm parameter,volumetric count of leukocytes and AsAb.Totally 383 male cases with infertility,set up into positive group,other 297 cases set up into negative group. RESULTS G.vaginalis in semen was positive in 86 cases in positive group,their semen concentration,motility,a+b grade motile,and the swelling rate of hypoosmotic swelling test were observed to be evidently lower compared to negative group(P0.05). CONCLUSIONS G.vaginalis has an effect on sperm quality.
