ABSTRACT

AIM:To investigate the effect of flavonoids from stem and leaf of Scutellaria baicalonsis Georgi(SSF)on paired helical filament(PHF)abnormality and the regulatory mechanism of protein phosphatase(PP)in rats' brain induced by okadaic acid(OA).METHODS:Male Sprague-Dawley(SD)rats were microinjected with OA(200 ng/kg)by the lateral ventricle to establish a memory impairment model.Morris water maze was used to screen the memory impairment model.The successful model rats were continuous intragastric infusion(ig)SSF for 36 days.The relative pro-tein expression of PHF,PP1,PP2A-Cα,PP2A-Cβ,PP2CA and PP2CB in the rat cerebral cortex and hippocampus were detected by Western blot.GinKgo biloba leaf flavonoids(GLF)were used as positive control drug.RESULTS:Compared with the sham-operated rats ,the relative protein expression of PHF in the cerebral cortex and hippocampus and PP 1 in cor-tex of model rats were significantly increased(P<0.01),and the protein expression of PP2A-Cα,PP2A-Cβin the cere-bral cortex and hippocampus and PP2CB in the hippocampus were decreased(P<0.05),while the relative protein expres-sion of PP2CA and PP2CB in the cortex were significantly increased(P<0.01).SSF reversed the abnormality in the pro-tein expression of PHF,PP2A-Cαand PP2A-Cβin rat cortex and hippocampus and PP1 in rat cortex induced by OA(P<0.01),which had no significant effect on the relative protein expression of PP 2CA and PP2CB.GLF also showed similar results to SSF.CONCLUSION:SSF significantly reduces the abnormal formation of PHF in rats ' brain induced by OA ,which may be related to the regulation of PP 1,PP2A-Cαand PP2A-Cβexpression,but not with PP2CA and PP2CB ex-pression.

ABSTRACT

By measuring the growth data of Scutellaria baicalensis in different cutting-seedling and determined active ingredient contents by HPLC and ultraviolet spectrophotometric determination. such as flavonoids. baicalin. wogonoside. baicalein. wogonin. oroxylin A. scutellarin. luteolin. and apigenin in the whole plant. Under circumstances of guaranteeing the quality and yield of medicinal materials. the yield of medicinal materials. and stems and leaves reached 193.60,63.21 kg/mu after twice cutting seedling. Not only yield but also active ingredient contents have been improved to some extent. the contents of flavonoids. baicalin. wogonoside. baicalein. wogonin. oroxylin A reached 18.52%. 15.13%. 4.03%. 1.04%. 1.04%. 0.12%. respectively in roots. Luteolin was not detected in young stems and leaves of S. baicalensis,the contents of other active ingredients such as scutellarin. luteolin and apigenin reached 7.00%. 0.96%. 0.04% respectively under twice cutting seedling. Therefore. regular cutting seedling could be regard as a new cultivation technique for wider range of promotion. And gaining high quality and yield of medicinal materials and tea with the purpose of rational utilization of natural resources and promoting the development of integration of herbal combination.

ABSTRACT

Objective To explore liver protective effects of the stems and leaves and roots of Scutellariae Radix from different areas of Gansu Province;To discuss differences in liver protection of the stems and leaves and roots of Scutellariae Radix. Methods All Kunming mice were randomly divided into 11 groups:normal group, model group, positive control group, Scutellariae Radix root groups and Scutellariae Radix stem-leaf groups of Longxi, Minxian, Weiyuan, Zhangxian. The mice received gavage with relevant medicine once a day for 7 days. 2 hours after the last medication, the mice were given peritoneal injection with 0.1%CCl4 and olive oil to establish the models with liver injury. Hepatic index, the activity of serum ALT and AST, the levels of T-AOC in the liver tissue were calculated in 18 hours after establishing the models. Results Compared with the model group, hepatic index, the activity of serum ALT and AST in the mice of Scutellariae Radix root groups from difference producing areas decreased significantly (P<0.05);the levels of T-AOC in the liver tissue was raised notably (P<0.05). Compared with stem-leaf groups from the same producing area, hepatic index, the activity of ALT and AST in the mice of Scutellariae Radix root groups were remarkably lower than these of Scutellariae Radix stem-leaf groups (P<0.05);the levels of T-AOC in the liver tissue was evidently higher than that of Scutellariae Radix stem-leaf group (P<0.05). Conclusion Scutellariae Radix root water decoction can prevent and protect CCl4-induced acute liver injury to a certain degree, while Scutellariae Radix stem-leaf water decoction shows no significant protective effect on acute liver injury.

ABSTRACT

AIM: To study the exciting effects of ethanol extract of capitate phrynium (Phrynium Capitatum Willd) stem and leaf on the uterine smooth muscle of rat,mice,guinea pig and rabbit pretreated by diethylstilbestrol. METHODS: Conventional method was adopted to prepare animal uterine specimens both in vivo and in vitro. Different doses of the extract were given to the specimens and recorded their tensility, frequency and activity of contraction with the help of BL-420 biological experimental system. IC_(50) of the extract was calculated, with oxytocin as standard comparison. RESULTS: 95% ethanol extract and 50% ethanol extract of capitate phrynium and leaf had obviously stimulating action on the uterine smooth muscle of rat, mice, guinea pig and rabbit in a dose-dependent manner in vitro, whichinclude accelerating contraction frequency, raising tension (P<0.05, P<0.01). In addition, 50% ethanol extract also had a significant stimulating action on uterine smooth muscle of rabbit in vivo(P< 0.01). CONCLUSION: Ethanol extracts of capitate phrynium have an effect on rodent uterine. Smooth muscle indicates that clinical application of Chinese practitioner used to treat the dysfunctional uterine bleeding proves to be effective.

ABSTRACT

Objective To optimize the extracting technology of total flavones in powder of Fagopyrum tataricum stem and leaf by enzymatic treatment. Methods The powder was treated by cellulase before extracted using water, the effects of enzyme dosage, treatment temperature, treatment time, and pH value on the extract rate of total flavones were studied. Results The optimum extracting technology was as follows: enzymatic treatment temperature: 55℃, enzyme dosage: 3.0 ?L, pH value: 6.5, treatment time: 90 min, extracting 3 times at: 90℃ for 30 min once. The extract rate of total flavones was 1.47% by this technology. Conclusion Cellulase could be applied in the assisstant extraction of total flavones in powder of F. tataricum stem and leaf.

ABSTRACT

Thirty two rabbits were divided into 4 groups: (1) Normal diet control group, ( 2 ) Hyperlipid diet control group, ( 3 ) Hyperlipid diet group injected with 30 mg/kg GSL, ( 4 ) Hyperlipid diet group injected with 60 mg/kg GSL, The whole expeiment took about 42 days. On 24d all hyperlipid diet were removed, and only normal diet given. At the end of this experiment all rabbits were killed by bleeding and the results showed that GSL could decrease the serum lipids GSL could increase the HDL - C in the rabbits injected with 60 mg/kg GSL. GSL could protect the walls of aortas from the damage of hyperlipid serum. The contents of lipid and MDA in the Walls of aortas were decreased. GSL had the effects of protection on the heart and liver by decreasing of Total Cholesterol and MDA. So we conclude that GSL has the effects of lipid regulation and lipid antipeoxidation on the chronic hy-perjipidemic rabits.

ABSTRACT

Aim To investigate the the effect of ginsenosides of stem and leaf(GSL) on mouse fatty liver and its mechanism.Methods Mice were randomly divided into normal control group(NC,fed with normal diet) and model group(fed with high-fat diet).Twelve weeks later,when the establishment of experimental model of fatty liver was confirmed,the model mice were subdivided into 4 subgroups: HF group(fed continuously with high-fat deit),NT group(fed with normal deit),GSL1 group(fed with normal deit and treated intragastrically with GSL 50 mg?kg-1?d-1),GSL2 group(fed with normal deit and treated intragastrically with GSL 100 mg?kg-1?d-1).Mice in NC,HF and HT groups were given distilled water by gastric perfusion.Two weeks later,all mice were killed,and blood was collected for measuring serum TC,TG,HDL-C,LDL-C,ALT contents,liver tissue for determining hepatic TC,TG,MDA SOD levels.In addition,liver index and pathology were also observed.Expression of PPAR? and CYP2E1 mRNA in liver was examined by RT-PCR.Results In GSL2 group,GSL significantly reduced liver index,serum lipid,hepatic lipid and MDA contents,and elevated SOD activity.Moreover,GSL obviously improved pathological change,increased PPAR? mRNA and suppressed CYP2E1 mRNA levels in the liver.In NT group,hepatic lipid and MDA contents remained high and SOD activity is low,although liver histology somewhat improved.In GSL1 group the results were similar to those in NT group.Conclusions These results suggest that GSL might be effective in the treatment of hepatic adipose infiltration of mice.It may be associated with increasing PPAR? mRNA,then decreasing serum lipid and hepatic lipid;and inhibiting CYP2E1 mRNA expression in the liver,thus suppressing lipid peroxidation.

ABSTRACT

AIM To observe the synergical effects of ginsenoside of stem and leaf(GSL)in combination with choline on learning and memory of Alzheimers disease(AD). MEHODS AD animal models were made by damaging nucleus basalis of Meynert with quinolinic acid. One time training passive aviodance step-down and water-maze spatial localization task were used to observe the ability of learning and memory.RESULTS Treatment with combination of GSL(400 mg?kg -1 ?d -1 ,ig) and choline(200 mg?kg -1 ?d -1 ,ig) on AD rats decreased significantly the number of errors on step down (ig for 13 days) and the training times to reach the criterion on water maze (ig for 16 days). The effect of GSL in combination with choline was more remarkable than that of GSL or choline and proved no obvious difference compaired with that of 1,2,3,4-tetrahydroacridine (10 mg?kg -1 ?d -1 ,ig). Value Q was more than one after administrition of both GSL and choline. CONCLUSION GSL in combination with choline may synergically improve the impairement of learning and memory of AD.
