ABSTRACT

OBJECTIVE To study the differentiation of PC12 cells induced by total salvianolic acid (Tsa) and the mechanism. METHODS MTT assay was used to detect the effect of Tsa 0.01, 0.1 and 1.0 μg·L-1on proliferation of PC12 cells and on the cells damaged by oxygen and glucose deprivation (OGD).The number of projections of PC12 cells was statistically analyzed.Western blotting was applied to detect the levels of microtubule-associated protein2 (MAP-2), extracellular signal-regulated kinase1/2 (ERK1/2), phosphorylated ERK1/2(p-ERK1/2), mitogen-activated protein kinase kinase1/2(MEK1/2) and p-MEK1/2 proteins.MEK inhibitor U0126 was examined for its effect on expressions of p-ERK1/2 and ERK1/2 protein in PC12 cells induced by Tsa 1.0 μg·L-1.RESULTS Compared with normal control group, Tsa 1.0 μg·L-1could promote PC12 cell proliferation, and the survival rate was increased by 90%, but the survival rate of PC12 cells was not affected by Tsa 0.01 or 0.1 μg·L-1. Compared with OGD injured group,PC12 cells injured by OGD could be repaired by Tsa 0.1 or 1.0 μg·L-1,and the survival rate was increased to (47.7±1.8)% and (63.2±13.0)%, respectively (P<0.05, P<0.01). Compared with normal control group,Tsa 0.01,0.1 and 1.0 μg·L-1could promote the growth of PC12 cell projections (P<0.01). Western blotting results showed that Tsa could promote the expressions of MAP-2, p-ERK1/2 and p-MEK1/2 proteins, and this effect could be blocked by U0126 inhibitor (P<0.01). CONCLUSION Tsa can induce the proliferation and differentiation of PC12 cells, the mechanism of which is possibly the activation of p-MEK and p-ERK1/2.

ABSTRACT

Objective: To study the influence of total salvianolic acid(TSA) on the renal interstitial fibrosis caused by unilateral ureteral obstruction (UUO) in rats. Methods: Forty healthy Sprague-Dawley rats were randomly divided into 4 groups: the UUO group, sham operation group, angiotensin-converting enzyme inhibitor ( ACEI)-treated group (positive control), and TSA-treated group. The rats in UUO group and sham-operation group received gastric gavage with normal saline for 8 days before operation; rats in ACEI-treated group and TSA-treated group received ACEI (6 mg/kg by daily gastric gavage for 8 days) and TSA (20 mg/kg by daily gastric gavage for 8 days), respectively. All rats were killed 7 days after operation and the expression of transforming growth factor-β 1 (TGF-β1) was detected by immunohistochemical method. The dynamic histological changes of renal interstitial tissues were observed by H-E and Masson staining. Results: TGF-β1 expression in TSA-treated (1.38 ± 0.26) and ACEI-treated (1.38 ± 0.26) groups was significantly lower than that in UUO group (P < 0.05). TSA obviously reduced TGF-β1 expression and collagen deposition in the renal interstitial tissues and improved the renal pathological changes in UUO rats. Conclusion: TSA can evidently inhibit UUO-induced renal interstitial fibros is in rats, which might be related with the down regulation of TGF-β1 expression.

AIM: To study the protective effect of total salvianolic acid against cerebral ischemia-reperfusion injury. METHODS: The cerebral ischemia-reperfusion model in mice was made by means of ligating bilateral common carotid arteries in mice. After reperfusion, latency, error number of step down test and the gasping time after cutting head in ischemia-reperfusion mice were recorded. Spectrophotometric assay were used to measure the activity of superoxide dismutase (SOD), the contents of malondialdehyde (MDA) and glutathione peroxidase (GSH) in brain of experimental mice brain homogenate. RESULTS: In step-down test, the ischemia-reperfusion impaired the function of learning and memory in mice. The total salvianolic acid markedly improved the function of learning and memory, reduced the error number and extended the latency in ischemia-reperfusion mice. The total salvianolic acid also significantly inhibited the changes of SOD, MDA and GSH in the cerebrum induced by ischemia-reperfusion. CONCLUSION: The total salvianolic acid has protective effects against cerebral ischemia-reperfusion injury via its antioxidant activity.

ABSTRACT

AIM: To study the technological parameters of the purification process of total salvianolic acid(TSA) of Radix Salvia Miltiorrhizae(RSM) with D101 macroreticular resin. METHODS: Based on orthogonal experiment,the reservation rate of TSA in macroreticular resin columniation was took as index to investigate the chief influential factors in the purification of TSA. RESULTS: The volume of distilled water and the content of TSA in RSU solution had significant effect on the reservation rate of TSA in macroreticular resin columniation. With the help of macroreticular resin,the reservation rate of TSA could reach 87.52%.Its purity was 77.74%,comparing with crude extract,increased by 3.6 times. CONCLUSION: The method of purifing TSA of RSM with D101 macroreticular resin is feasible,it could be applied to industrialized production of TSA of RSM.

Results: The releasing rate of the tablets was conformed to zero kinetics. Conclusion: The study was found to be effective in the process of total salvanolic acid gastric residential tablets.
