ABSTRACT

AIM:To explore the regulatory effect of chemokine CCL 3 on exosome secretion from human bone marrow mesenchymal stem cells(hBMSCs).METHODS: hBMSCs were stimulated with chemokine CCL 3 at different concentrations in vitro.The proliferation of hBMSCs was measured by CCK-8 assay and viable cell counting.Exosome se-cretion from hBMSCs was qualitatively analyzed by transmission electron microscope(TEM)and flow cytometry, and the quantitative analysis was carried out by flow cytometry and nanoparticle tracking analysis(NTA).RESULTS:Compared with control group,the viability of the hBMSCs detected by CCK-8 assay was increased when hBMSCs were treated with CCL3(P<0.05).The results of viable cell counting demonstrated that the number of hBMSCs was raised in CCL 3 group in a dose-dependent manner(P<0.05).The results of flow cytometry showed that hBMSCs expressed 3 CCL3-related spe-cific receptors,CCR1,CCR5 and CCR9.Compared with control group,the fluorescence intensity of CCR9 in CCL3 group was obviously enhanced.However,no significant difference of fluorescence intensity for CCR 5 and CCR1 was observed be-tween the 2 groups.The results of NTA demonstrated that the secretion capacity of CCL 3-induced hBMSCs was far less than that in control group(P<0.05).However, the microvesicles larger than 100 nm in CCL3 groups were increased(P<0.05).The above results indicated that the higher concentration of CCL 3 induced the lower secretion of exosomes.In addi-tion,the results of flow cytometry demonstrated that CCL 3-induced hBMSCs showed lower quantity of CD 9 +exosomes than those in control group(P<0.01).CONCLUSION:CCL3 promotes the proliferation of hBMSCs but depresses the secre-tion of exosomes in a dose-dependent manner.CCL3 affects the size distribution of exosomes and increases the number of nonfunctional microvesicles of larger than 100 nm in size.CCL3 induces the expression of CCR9 in hBMSCs.

ABSTRACT

The rapid recovery of postoperative gastrointestinal motility is the key factor to accelerate the recovery of patients. The recovery of gastrointestinal motility is often affected by various factors, such as anesthesia and operation. The present study shows that the inhibition of stress response caused by acute pain can improve the recovery of gastrointestinal motility. Howeve, the effects of perioperative intravenous analgesics on gastrointestinal motility are not consistent. So we reviewed the effects of different intravenous analgesics on gastrointestinal motility.

ABSTRACT

<p><b>OBJECTIVE</b>To understand the mechanism of invasion by Legionella dumoffii.</p><p><b>METHODS</b>The L. dumoffii strain Tex-KL was mutated using the Tn903 derivative, Tn903dIIlacZ. After screening 799 transposon insertion mutants, we isolated one defective mutant. We then constructed the gene-disrupted mutant, KL16, and studied its invasion of and intracellular growth in HeLa and A549 cells, and in A/J mice survival experiments. The structure of traC-traD operon was analyzed by RT-PCR.</p><p><b>RESULTS</b>The transposon insertion was in a gene homologous to Salmonella typhi traC, which is required for the assembly of F pilin into the mature F pilus structure and for conjugal DNA transmission. Results from RT-PCR suggested that the traC-traD region formed an operon. We found that when the traC gene was disrupted, invasion and intracellular growth of L. dumoffii Tex-KL were impaired in human epithelial cells. When mice were infected by intranasal inoculation with a traC deficient mutant, their survival significantly increased when compared to mice infected with the wild-type strain..</p><p><b>CONCLUSION</b>Our results indicated that the traC-traD operon is required for the invasion and intracellular growth abilities of L. dumoffii Tex-KL in epithelial cells.</p>

ABSTRACT

Objective To explore the clinical phenotype of airways disease in elderly patients using hierarchical cluster analysis.Methods A total of 67 elderly patients with respiratory symptoms were enrolled in a prospective study.Demographic and clinical data,such as respiratory symptoms,cumulative tobacco cigarette consumption,acute exacerbation,atopic symptoms and peak flow diary were collected.Pulmonary function tests,blood tests (total serum IgE level and blood eosinophil level) were performed in each patient during the stable stage.Then patients with different clinical phenotype were identified by hierarchical cluster analysis.Results Four clusters were identified with the following characteristics by hierarchical cluster analysis:cluster 1,atopic patients with no smoking,normal lung function,but increased total serum IgE levels and asthma symptom;cluster 2,patients with no smoking and normal pulmonary function with wheezing but without chronic cough;cluster 3,patients with chronic obstructive pulmonary disease and smoking,severe airflow limitation and poor quality of life;cluster 4,patients with asthma-chronic obstructive pulmonary disease overlap syndrome and smoking,airflow limitation and increased total serum IgE levels.The forced expiratory volume in 1 second (FEV1) / forced vital capacity (FVC) ratio,FEV1/predicted value,rate of FEV1 change,maximal mid-expiratory flow (MMEF)/ predicted value,the diffusion lung capacity for carbon monoxide (DLCO)/alveolar volume (VA)/predicted value,residual volume (RV)/ predicted value,total serum Ig E levels,cumulative tobacco cigarette consumption,the St.George's Respiratory Questionnaire (SGRQ) score had significant differences in patients before versus after treatment (all P<0.05 or P<0.01).Conclusions Based on hierarchical cluster analysis,distinct clinical phenotypes of airways disease in elderly patients can be identified.Conclusions With patients having asthma or COPD alone,patients with Asthma-COPD overlap syndrome (ACOS) always experience a more rapid decline in lung function and frequent exacerbations,having poor health-related quality-of-life (HRQOL) outcomes,which deserve our high attention.

Statement of Problem: Given that meticulous implant prosthodontic procedures are recommended to obtain the best possible intraoral fit, the die systems used for multi implant casts warrant further investigation. Die stone expansion and errors introduced by removable die casts may exceed the accuracy required for the passive fit of implant prosthesis. Purpose: The purpose of the study was to evaluate the linear dimensional accuracy between the implant master die and three conceptually different die systems such as Pindex system, Accu-trac precision die system, and Conventional brass dowel pin system. Materials and Methods: Thirty impressions of implant master die were made with polyether impression material. Ten experimental implant casts were fabricated for each of the three different die systems tested: Accu-trac precision die tray system, Pindex system, and conventional brass dowel pin system. The solid experimental casts were sectioned and then removed from the die system 30 times. Linear distances between all six possible distances were measured from one centre of the transfer coping to the other, using a co-ordinate measuring machine in millimeters up to accuracy of 0.5 microns. Data were tabulated and statistically analyzed by Binomial non parametric test using SPSS version 15. Results: Significant differences were found for distance A-B (P = 0.002), A-C ( P = 0.002), A-D (P value = 0.002), and B-D ( P = 0.021) in Conventional Dowel pin system however for Accu-trac precision die tray system, it was significant only for distance A-D (P = 0.002) but for Pindex system it was non-significant for all the distances measured. Conclusion: Within the limitations of this study, use of Pindex system is recommended when sectioned dies are needed for a multi implant retained prosthesis.

Por más de 30 años se han usado ampliamente las fluoroquinolonas a las cuales ha habidobuena tolerancia; son de amplio espectro contra gérmenes grampositivos y gramnegativos,aunque la actividad de la norfloxacina, la ciprofloxacina y la ofloxacina contra estreptococos yalgunos anaerobios es limitada; tienen, además, buena biodisponibilidad oral y adecuadapenetración en los tejidos. Se presentan reacciones características de hipersensibilidadaproximadamente en 1 por cada 50.000. tratamientos. Se describe un caso de desensibilizaciónoral exitosa con ciprofloxacina en una paciente con infección urinaria crónica parcialmentetratada. Esta es la primera desensibilización con quinolonas reportada en Colombia.


Desensitization with ciprofloxacin in a patient with partially treated urinary tract infectionFluoroquinolones have been widely used for over 30 years and tolerance to them has beengood. They are of broad spectrum against both gram positive and gram negative bacteria, althoughthe activity of norfloxacin, ciprofloxacin and ofloxacin against streptococci and some anaerobicbacteria is rather limited. We report a case of successful oraldesensitization with ciprofloxacin in a patient withchronic partially treated urinary tract infection. Only 1 case of common bile duct injury after laparoscopic cholecystectoy was death from the toxic shock, another case of bile duct injury after conventional cholecystectoy with the drainage of abdominal cavity was death from multiple system organ failure, all of the other patients were cured.ConclusionThe iatrogenic biliary-tract injury may be causes variously, and should be discovered as soon as possible and be operated in time.

ABSTRACT

This research aims to identify the prevalence of reproductive tract infections (RTIs) and to investigate the knowledge of married women with child bearing age about RITs as well as their practices in treatment and prevention of these diseases. The research was conducted in Thuy Dzung commune- Thai Thuy District - Thai Binh province in 1996-1997. Both quantitative and qualitative methods were applied in this research. In cross sectional study, clinical and para clinical examinations and questionnaires were applied in 609 married women aged 15-49. The prevalence of RTIs was 32.8% (95% CI: 32.8% +/- 3.7). Among these infections, 28.5% of the cases were due to candida, 44% due to bacteria and 27.5% due to both candida and bacteria. The qualitative study was implemented in 7 months in 70 married and some men, health workers.The result shows that both local people and health workers perceived STDs as separate diseases,meaning that RTIs do not include STDs and hardly related with them. Free drugs and free health services are not enough, it is more important that the health workers are better able to deal with comon RTIs, and to counsel and help local women to prevent these diseases.

TPN can not only improve the malnutrition of organism resulted from ma- lignant tumour,but also slimulate the growth of tumour.Therefore,how to make best advantages of TPN is of great concern. From July1 994to December1 995 ,we treat- ed73patietns with malignant tumour in di- gestive tract by applying TPN,and achieved very good curative effects. We sum up our experience as following:1 perioperation and chemotherapy is a good indication oTPN which can reduce post- operation com- plication and enhance effect of chemothera- py;2 Itis contraindication of TPN to termi- nal cancer patients without operation and chemotherapy patients,TPN support is helpless;3 Operation and chemotherapy may aggravate the patients stress and cause a high matabolic rate in organism,so application of TPN should stick to the prin- ciples of metabolic support;4Taking Glu- tamine(GLN) and Se will benefit the treat- ment of tumour;5 It amy be helpful to check the growth of tumour by increasing fat emulsion,decreasing or increasing cer- tain amino acid. The questions mentioned above need futher study.

ABSTRACT

This reseach was conducted in 4 mountainous communes inluding a Pako minority commune and a Van Kieu minority commune and 2 other communes of Kinh in Qu¶ng TrÞ province. 326 married women were randomly selected for attending and interview and examination. A cross-sectional survey was employed and both clinical and laboratory tests was utilized during data collection process. It was explored that 63.80% of the sample were suffering from RTIs. In addition, geographical, ethnic, educational level factors, age and time in marriage, abortion and some reproductive hygiene habits effected on the RTIs prevalence.

ABSTRACT

In this study, we determined the levels of 8 minerals (Na, K, Ca, Mg, Fe, Cu, Zn and Mn)in sera of 30 patients receiving total parenteral nutrition (TPN) for more than 5 d and tested the 8 mineral concentrations in the 16 drugs and solutions often usrd for TPN.The concentrations of Na, K, Ca, and Mg in the TPN drugs and solutions were much higher than those of the trace elements (Fe, Cu, Zn and Mn).Severe zinc deficiency was found in 2 patients receiving TPN for about 60 d and 171 d and their s:rum zinc concentrations lowered to 5.00 umol/L and 1.22 umol/L, respectively with clinical symptoms and signs induced by zinc deficiency.Our results show that mineral deficiencies do not occur at least within 2 weeks in short-term TPN patients, but in long-term TPN patients mineral deficiencies, especially zinc deficiency may appear.
