ABSTRACT

@#The lymphatic system is the main way of tumor metastasis and diffusion. Esophageal cancer is one of the typical cancers that are prone to metastasis through the lymphatic system. At present, an increasing number of studies show that the interaction between tumor cells and lymphatic endothelial cells is the first step in tumor lymphatic metastasis, but the underlying molecular mechanism is unclear. This article reviews the role and changes of tumor-related lymphatic vessels and lymphatic endothelial cells in the process of tumor lymphatic metastasis, which lays a foundation for further study of the specific molecular mechanism of esophageal cancer lymphatic metastasis and provides a new treatment direction for esophageal cancer patients.

ABSTRACT

Priapism is a rare pathological penile erection, and there are some inadequacies in its definition, classification, diagnosis, and therapeutic strategies. In this article, we sum up our years of experience with priapism and put forward some new views and ideas about its definition, classification, pathophysiologic process, pathological change, diagnostic essentials, therapeutic measures, indications of successful treatment, and post-therapeutic rehabilitation of erectile function. We also describe the clinical features, diagnosis and treatment of some special types of priapism, such as intermittent seizure, sleep-related painful erection, and tumor-related priapism, hoping to help urologists and andrologists in the further understanding and management of priapism.

ABSTRACT

BACKGROUND: In this manuscript, we assessed tumor recurrence and tumor‑related mortality in a clinical series of endometrial cancer patients. MATERIALS AND METHODS: A retrospective evaluation of 276 patients (mean age 64 years) with histologically confirmed endometrial cancer treated at a single hospital in Madrid (Spain) was conducted. The median follow‑up was estimated using the inverse Kaplan–Meier method. RESULTS: Salient findings were endometrioid carcinoma (84.8% of cases), grade G1 (48.9%) and stages IB (35.1%) and IC (23.2%). Myometrial infiltration >50% was documented in 31.2% of cases and lymphovascular space invasion in 11.9%. After surgery, 52.5% of patients were classified into the low risk group, 21.4% into the intermediate risk group and 26.1% into the high risk group. Tumor recurrence occurred in 14.5% of patients, with an estimated median follow‑up of 45 months (95% confidence interval (CI): 41.2–48.8), locoregional recurrence in 42.5% and distant recurrences in 57.5%. Furthermore, 40% of tumor recurrences developed during the first year after primary treatment and 90% over the first 3 years of follow‑up. The tumor‑related mortality rate was 15.9%. The estimated median follow‑up was 46 months (95% CI: 43.0–49.0). Furthermore, 5.07% of death because of tumor developed during the first year after primary treatment and 13.77% over the first 3 years of follow‑up. CONCLUSION: The rates of tumor‑related death and tumor recurrence in endometrial cancer patients are low, with the highest percentages occurring within 3 years of primary treatment. Most of the recurrences occur outside the pelvis.

ABSTRACT

The Nrf-2/ARE signaling pathway is not only affected by itself but also influenced by multiple tumor-related signals. To expand our understanding about the Nrf-2/ARE signaling pathway and lay a theoretical foundation for further research, we reviewed the effects of multiple tumor-related signals on the Nrf-2/ARE signaling pathway and studied the possible underlying mechanisms.

ABSTRACT

Jumonji Domain Containing 2A (JMJD2A) may be a cancer-associated gene involved in human breast cancer. With a view to investigating expression of JMJD2A in human breast cancer and benign lesion tissues as well as relationship between JMJD2A and tumor related proteins, histological and immunohistochemical analysis, Western blot and quantitative real-time PCR in infiltrating duct carcinoma and fibroadenoma for JMJD2A and immunohistochemical analysis and quantitative real-time PCR in infiltrating duct carcinoma for tumor related proteins (ARHI, p53, ER, PR and CerbB-2) were performed. Histological examination validated the clinical diagnosis. The JMJD2A positive rate of infiltrating duct carcinoma was significantly higher than fibroadenoma by immunohistochemical analysis. The mean optical density of JMJD2A in infiltrating duct carcinoma was higher than fibroadenoma by western blot. JMJD2A mRNA level in infiltrating duct carcinoma was higher than fibroadenoma by quantitative real-time PCR. Spearman correlation analysis revealed that the expression of JMJD2A was associated with ARHI, p53 and ER from immunohistochemical results respectively. Pearson correlation analysis revealed that the expression of JMJD2A was associated with ARHI, p53 and ER from quantitative real-time PCR results respectively. Expression of JMJD2A in infiltrating duct carcinoma was higher, and associated with ARHI, p53 and ER. The results may take JMJD2A as a potential diagnostic and therapeutic target in human breast cancer.

ABSTRACT

Objective To investigate the effects of H_2O_2 on growth and proliferation in CNE-2Z cells. Methods To identify the effects of H_2O_2 on the growth and proliferation in CNE-2Z cells,Cell growth curves,MTT assay and plate colony formation rate were used;Meanwhile,Cell cycle distribution, proliferation index and apoptosis analysis were performed by flow cytometry(FCM) in human NPC cells CNE-2Z in vitro, before and after the action of H_2O_2;the protein expressions of Egr-1,p53,p16,Cyclin D1 were detected by Western-Blotting analysis. Results There were no EGR-1 and P16 proteins expressions in CNE-2Z cells by Western-Blotting analysis before the action of H2O2;the growth and proliferation of CNE-2Z cells were inhibited significantly by H_2O_2 compared with the control cells, and the apoptosis cells were increased. The increased expressions of EGR-1,P53,and Cyclin D1 protein were also shown in CNE-2Z cells by means of Western-Blotting analysis after the action of H_2O_2;but no P16 protein expression; The expression of EGR-1 protein was significant correlation with P53 protein expression, but no significant correlation with Cyclin D1 protein expression. Conclusion There was no EGR-1 protein expression in CNE-2Z cells, and EGR-1 protein can be induced expression rapidly and transiently by H_2O_2,which probably have an important!role in the growth inhibition and apoptosis. The promotor of Egr-1 gene also can be induced activation by radiation(ROS,reactive oxygen species),there is an important application in therapy of NPC and other tumors.

ABSTRACT

Objective The development and progression of intestinal metaplasia ( IM ) and expression of tumor-related proteins in dog’ s stomach were observed. Methods IM animal model was established in Beagle’ s stomach by feeding N-methyl-N’-nitro-N-trosoguanidine( MNNG ),ranitidine, and local X-irradiation. Expression of APC, p53, K-ras and bcl-2 genes in canine gastric lesion were determined by immunohistochemistry. Resullts IM canine model was successfully induced, with which sequential pathology studied. It was proved that progression of normal epithelial cells to IM cells might undergo several stages including superficial gastritis, chronic atrophic gastritis, mild focal IM, moderate, and severe IM.Aberrant bcl-2 protein was detected in atrophic gastric epithelium and abnormal expression of APC, K-ras, and bcl-2 found in IM mucosa. Conclusion Canine gastric IM was induced by the method mentioned above. The development of normal gastric mucosa to IM in dog seemed to resemble that of human being. The presence of tumor-related proteins might indicate the transformation of IM to malignancy.
