ABSTRACT

Abstract This study aimed to determine the antiulcerogenic and antioxidant activities of Psyllium (Plantago ovata Forssk) seed ethanolic extract in rats. We assessed the antioxidant potential using free radical scavenging on DPPH, -carotene bleaching activity, ferric reducing power, and hydroxyl radical scavenging activity. In the antiulcerogenic study, pre-treatment with Plantago ovata seeds ethanolic extract (POE) (400 mg/kg b.wt) significantly protected against ethanol-induced gastric ulcer in rats by decreasing the ulcer index value and preserving the integrity of the gastric mucosa. The oxidative stress status in the stomach tissues showed a significant increase in the antioxidant enzyme levels of superoxide dismutase, catalase, and glutathione peroxidase with a significant decrease in lipid peroxidation during pre-treatment with POE. In conclusion, the POE protects against gastric ulcer due to its antioxidant potential and presence of bioactive molecules.


Resumo O presente estudo teve como objetivo determinar as atividades antiulcerogênica e antioxidante das sementes de Psyllium (Plantago ovata Forssk) em ratos. O potencial antioxidante foi avaliado utilizando o método do sequestro do radical livre DPPH, autooxidação do -caroteno, poder redutor de ferro e atividade de sequestro do radical hidroxila. No estudo antiulcerogênico, o pré-tratamento com o extrato etanólico das sementes de Plantago ovata (POE) (400 mg/Kg b.wt) reduziu a úlcera gástrica induzida pelo etanol em ratos, diminuindo o valor do índice de úlcera e preservando a integridade da mucosa gástrica. O estudo do estresse oxidativo nos tecidos estomacais mostrou um aumento significativo dos níveis das enzimas antioxidantes superóxido dismutase, catalase e glutationa peroxidase, com uma diminuição significativa da peroxidação lipídica enquanto pré-tratamento com POE. Em conclusão, o POE protege contra úlcera gástrica devido aos seus potenciais antioxidantes e à presença de moléculas bioativas.

O presente estudo teve como objetivo determinar as atividades antiulcerogênica e antioxidante das sementes de Psyllium (Plantago ovata Forssk) em ratos. ABSTRACT

Aim To explore the effect of cuminaldehyde in cumin fruit on gastric ulcer and the protective mechanism via establishing the gastric ulcer model of rats was by ethanol injury. Methods Thirty-six male R. norregicus were divided into six groups: control group, model group, omeprazole positive control group and cuminaldehyde low, medium and high dosage groups. After seven days of continuous intragastric administration, the acute gastric ulcer of R. norregicus was tested by absolute alcohol. Gastric ulcer area, inhibition rate, gastric tissue antioxidant activity, serum inflammatory factors and gastric mucosal protective factors were detected in different groups. Results The results showed that cuminaldehyde significantly reduced the area of gastric ulcer and increased the inhibition rate of gastric ulcer. The inhibition rate of cuminaldehyde at high dose group was up to 74.65%, the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH) in gastric tissue significantly increased, and the contents of serum prolandin E

Background: Gastric mucosal ulceration is the most common adverse effect with NSAIDS. Antacids, H2 blockers and PPIs are considered novel in treating ulcers but are not devoid of side effects. Hence, there a need for a drug which is effective against NSAID induced ulcers with no side effects. Coccinia grandis plant is traditionally used for the treatment of gastric/peptic ulcers. Hence, this study has been undertaken to scientifically validate the antiulcer activity of Coccinia grandis leaves against indomethacin induced gastric ulcer model.Methods: Following preparation of the extract, 24 Wistar rats were divided into 4 groups with 6 rats in each group (n=6). Group 1 received 1% CMC, group 2 received 1% CMC +indomethacin 40 mg/kg, group 3 received ethanolic leaf extract of Coccinia grandis 200 mg/kg +indomethacin 40 mg/kg and group 4 received omeprazole (2 mg/kg) +indomethzacin 40 mg/kg for 7 days. Calculation of ulcer score was done using ulcer index and percentage protection.Results: The ulcer index score (2.12�21) and percentage protection (69.71%) was comparable with the standard drug (1.76�11, 74.85%) respectively.Conclusions: The ethanolic leaf extract of Coccinia grandis showed significant antiulcer activity against indomethacin-induced gastric ulcer. Further studies are warranted to elucidate the exact mechanism of antiulcer activity.

ABSTRACT

Background: Most important adverse effect of NSAID is peptic ulceration. Even though H2 blockers and proton pump inhibitors are effective in preventing NSAID associated peptic ulceration, they are not without side effects. Hence there is a need for drugs which are effective in preventing NSAID induced peptic ulcer without producing side effects. Two plant products Aloe vera leaf extract and Aegle marmalos leaves are commonly used in Indian traditional medicine for treatment of peptic ulcers. Hence this study is undertaken to assess the antiulcerogenic potential of combination of these two drugs in comparison with ranitidine in preventing NSAID induced peptic ulcers.Methods: 18 albino rats were divided into 3 groups of 6 rats each. Group A: received ulcerogen only. Group B: pretreated with ranitidine before exposing to ulcerogen. Group C: pretreated with combination of Aloe vera and Aegle marmelos before exposing to ulcerogen. Two doses of indomethacin were administered at an interval of 15 hrs. Animals were sacrificed 6 hrs after the second dose of Indomethacin. Number of ulcers was noted, and ulcer index was calculated.Results: There was significant reduction in total score, mean score and ulcer index in ranitidine pretreated group and test compound group as compared to control group. Even though the total score and ulcer index in test group were lesser as compared to standard control group, it was not statistically significant.Conclusions: Combination of Aloe vera leaf extract and Aegle marmelos leaf extract produced very significant protection against indomethacin induced gastric ulcer.

Objective: To investigate the pharmacodynamics of modified Xiaochaihutang granules (MXG) and its active ingredients group (AIG) on gastric ulcer in order to provide scientific basis for the clinical effect and lay foundation for the development of related hospital preparations.Methods: Gastric ulcer models induced by pyloric ligation,acetic acid and ethanol were applied to observe the curative effects of MXG and its AIG, and the effects of MXG and its AIG on gastric secretion, gastric acid content, pepsin activity, superoxide dismutase (SOD) and malondialdehyde(MDA) contents were also studied.Results: Compared to the model control group,MXG and AIG could significantly reduce the ulcer index(P<0.01), total acidity(P<0.01),and pepsin activity (P<0.05 or 0.01), increase the SOD activity (P<0.05)and inhibit the increase of lipid peroxide MDA.Conclusion: MXG and AIG show better control effects on acute and chronic gastric ulcer, and the effects are related to reducing the activity of pepsin, increasing the SOD activity, inhibiting the increase of lipid peroxide MDA and so on.

Objective To determine the effects of non-thermal biological effect of low frequency pulsed electromagnetic field (LFPEF) with 15 Hz and 30 mT on ulcer wound during the plateau stress ulcer cure.Methods Thirty male Sprague-Dawley rats were assigned to three equal groups:control,normal cure group (NC) and exposure treatment groups (ET).The plateau rats stress ulcer models were constructed with the combination method of a hypoxic and low-pressure acclimation and the stimulation of absolute ethyl alcohol irrigating stomach.The LFPEF equipment generated the LFPEF of 15 Hz,40% duty cycle and 30 mT to expose and treat the rats' stomach of ET group with the frequency of 3 hours each day.After 6 days of exposure treatment,the stress ulcer cure situation of each group was evaluated by the methods of macroscopic observation,the gastric juice pH values detection and ulcer index (UI) measurement.Results After 6 days of model,the macroscopic observation showed that the ulcer of NC group was worse than the that of ET group (P<0.05).The pH values and UI of ET group were significantly different with those of NC group,and closed to those of the control group.Conclusion The nonthermal biological effect of LFPEF could promote the plateau stress ulcer cure.

Methods: A total of 138 SD rats were selected to make rat models with gastric ulcer induced by acetic acid (24 rats with sham operation served as sham operation group), and were randomly divided into model group (n = 30), western medicine group (n = 30), traditional Chinese medicine (TCM) group (n = 24) and combination group (combined western medicine and TCM group, n = 30). Western medicine group was gavaged with omeprazole in the morning and with iso-volumetric distilled water in the afternoon; TCM group and TCM sham operation group were gavaged with iso-volumetric distilled water in the morning and with Qi-Replenishing and Blood-Activating Formula in the afternoon; combination group was gavaged with omeprazole in the morning and with Qi-Replenishing and Blood-Activating Formula in the afternoon; sham operation group and model group were gavaged with iso-volumetric distilled water both in the morning and afternoon. Ulcer indexes and degree of mucosal degree in rats at different time points after gavage were observed. Twenty-eight days after gavage, interleukin (IL)-1β was given to induce ulcer recurrence so as to observe the recurrent severity and rate of ulcer in each group. Results: Compared with model group and western medicine group, treatment in combination group could prominently reduce the ulcer index of rats with peptic ulcer, and increase the healing rate and inhibition rate of peptic ulcer. After IL-1β-induced ulcer recurrence, combination group was significantly superior to model group and western medicine group in ulcer recurrent rate [50% (3/6) vs. 100% (6/6)] and severity. Conclusions: Basic acid-suppression therapy combined with Qi-Replenishing and Blood-Activating Formula can effectually improve the ulcer healing quality and reduce ulcer recurrence.

ABSTRACT

Objective To observe the effects of acupuncture on the sleeping quality of male Wistar rats with stress induced gastric mucosal injury from the viewpoint of the brain-intestine axis. Methods Male Wistar rats were randomized into normal control, model, ST36-CV12, BL62-KI6, and combined groups, with 8 rats in each group. The model was established by using intragastric perfusion of ethanol in the rats. Acupoints of Zusanli (ST36) and Zhongwan (CV12) on both sides were used for the ST36-CV12 group. Acupoints of Shenmai (BL62) and Zhaohai (KI6) on both sides were used for the BL62-KI6 group. All four acupoints were used for the combined group. 0.22 mm × 13 mm stainless steel filiform needles were inserted 5-10 mm deep, and were left untouched for 20 minutes before withdrawal. The acupoints were stimulated each day for five days and the paw withdraw thermal latency (PWTL) was detected after the last stimulation. 2%pentobarbital sodium was then injected (40 mg/kg), and sleeping time of rats was detected. Gastric mucosa was observed through naked eyes. Gastric mucosal ulcer Index was determined by using the Guth method. 5-HT and DA contents were detected by using HPLC-ECD method. Results Compared with the normal control group, PWTL and sleeping time decreased (P<0.01), and UI, 5-HT, and DA content increased in model group (P<0.05, P<0.01). Compared with the model group, PTWL and sleeping time increased (P<0.05, P<0.01), and UI decreased in all treatment groups (P<0.01). 5-HT content in ST36-CV12 and combined groups decreased significantly, and DA content in ST36-CV12 group dropped (P<0.01). Compared with other treatment groups, PWTL and sleeping time increased significantly (P<0.01), and UI decreased obviously in combined group (P<0.01). Conclusion Stimulating acupoints of Zusanli, Zhongwan, Shenmai, and Zhaohai (KI6) may promote the recovery of gastric mucosal injury while at the same time increase sleeping time, with four acupoints used in combination having the best efficacy. The mechanism may be related to adjusting contents of 5-HT and DA in corpus striatum.

Peptic ulcer disease is a common condition of present day life, and its incidence is markedly increasing. The available drugs for the treatment are associated with adverse effects. Hence, there is a need for newer and better treatment for the same. Objectives: To evaluate the anti-ulcer activity of ethanolic extract of Vitis Vinifera (seed) by Water Immersion stress induced gastric ulcer model in Wistar albino rats using two doses (100, 200mg/kg body weight.) Methods: The antiulcer activity was assessed by estimating gastric volume, pH, free and total acidity; ulcer number and its inhibition, gastric lesion, ulcer severity and ulcer index; and comparing it with the control groups. Results: The present study revealed that grape seed extract of 100mg/kg and 200mg/kg showed a significant (p<0.05) reduction in the gastric volume, ulcer number, gastric lesion and severity. The ulcer index was 45.33 and 36.66 for 100 and 200mg/kg respectively. Ulcer inhibition for 100 and 200 mg/kg group was 32.79% and 44.86% when compared with the control group. Ulcer protection with 100 and 200mg/kg was 29.21% and 39.82% when compared with the control group. Conclusion: This present study indicates that Vitis vinifera seed extract have potential anti-ulcer activity in the model tested.

Aim of the study was to investigate Gastroprotective and anti-secretory effect of Pep-Up Tablet on pyloric ligation-induced gastric ulcer model in rats. Pep-Up Tablet is an Ayurvedic proprietary formulation which is manufactured and marketed by Vasu Healthcare Pvt. Ltd., Vadodara. The selected animals were divided into two groups and each group consisted of six animals. Group-I was considered as disease control and Group-II was as test drug (Pep-Up Tablet) treated group. Pep-Up Tablet (200 mg/kg/day, p.o.) was administered for 7 days by oral route in Group-II. Pep-Up Tablet was studied for its effect on ulcer index, gastric wall secretory parameters and mucin activity. Pretreatment of Pep-Up Tablet showed significant reduction in ulcer index, gastric acid secretion and pepsin activity. Pep-Up Tablet significantly increased mucin activity (TC:TP ratio) as well which was due to significant increase in the total carbohydrate content. From the available data of present study, it can be concluded that Pep-Up Tablet plays important role mainly on inhibition of acid secretion and in increase of mucin secretion which in turn enhances the stability of gastric mucosal barrier. Pep-Up Tablet provided significant gastric cytoprotection against pyloric ligation-induced gastric ulceration in rats.

Aims: Barleria prionitis L. (Family Acanthaceae) is a medicinal plant found road side in India and whole plant or its various parts like leaves, root, bark, stem and flowers are used traditionally for various treatments like toothache, inflammation, boils, glandular swellings and ulcer. Leaf juice is useful in gastric ulcer. Here, we attempt to prove the use of this plant as gastroprotective agent. Study Design: This study was conducted to evaluate the antiulcer activity of methanol extract obtained from the leaves of Barleria prionitis Linn. Place and Duration of Study: The experiments were conducted at Pharmacology lab of Institute of Pharmaceutical Sciences, Kurukshetra University during the period of July 2012 to December 2012. Material and Methods: Antiulcer activity was performed using the protocols of ulcer induced by ethanol and indomethacin at two different doses (250 and 500mg/kg). Parameters like volume of gastric juice, pH, free acidity, total acidity, aspartate amino transferase (AST) and alanine amino transferase (ALT) were also determined in ethanol induced ulcer model. Results: The reduction in ulcer index in Barleria prionitis treated animals was found to be statistically significant (P=.05), when compared with control groups in both the models. Significant changes were observed in total acidity only at dose 500mg/kg only and changes were significant in AST, ALT levels at both the doses. Other parameters showed non-significant results. Conclusion: The results of the present study show that the methanolic extract of Barleria prionitis L. possess antiulcer activity. This work supports the traditional use of this plant in treating gastric ulcer.

The aim of the study was to determine anti ulcer activity of stem bark of Careya arbore Roxb. on the Wister strain albino rats. Dried stem bark of Careya arbore Roxb. was powdered and this coarse powder was extracted with 70% ethanol by soxhlet extraction method to yield a Ethanol extract of stem bark of Careya aborea Roxb. (EECA). The extract was subjected for preliminary phytochemical analysis and was evaluated for anti ulcer activity against various models such as Ethanol induced, cold restraint stress induced and Pylorus ligation induced models. In acute toxicity study, EECA was found to be safe till 3000mg/kg. So the doses of EECA at various concentration of 300 and 600mg/kg body weight was administered orally, twice daily for 5 days for prevention of ulcer from Pylorus ligation, Ethanol and cold restraint stress - induced ulcers. Analytical parameters like Percentage of Ulcer protection was calculated based on Ulcer index and Gastric juice volume, pH and acidity of gastric juice. Preliminary phytochemical analysis of EECA showed the presence of carbohydrates, glycosides, phytosterols, phenolic compounds, tannins and saponins. The EECA has shown significant activity at both 300mg/kg and 600mg/kg dose level in a dose dependent manner. Phytoconstituents like tannins and saponins may be responsible for anti ulcer activity of EECA.

ABSTRACT

Activit, a herbomineral formulation, containing extracts derived from Mucuna pruriens, Withania somnifera, Argyreia speciosa, Centella asiatica, Tribulus terrestris, Asparagus racemosus, Piper longum, Anacyclus pyrethrum, Nux vomica, Tinospora cordifolia and Shring bhasma, was studied for its protective effect against gastric lesions induced by ethanol and pylorus-ligation. The formulation was tested at the doses of 125, 250, 500 and 1000 mg/kg body weight (p.o.) in rats for its effect on various gastric and antioxidant parameters. The reduction in ulcer index in both the models; along with the reduction in volume and total acidity, and an increase in the pH of gastric fluid in pylorus-ligated rats proved the anti-ulcer activity of Activit. The increase in the levels of endogenous antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), reduced glutathione (GSH)] along with membrane bound enzymes (Ca2+ATPase, Mg2+ATPase and Na+K+ATPase) and a decrease in lipid peroxidation (MDA) in both the ulcer models by Activit demonstrated its antioxidant effect. Thus it can be concluded that Activit possesses anti-ulcer activity, which can be attributed to its antioxidant mechanism of action.

ABSTRACT

Garcinia indica Linn (Clusiaceae), a medicinal plant mentioned in Ayurveda has been used for treatment of liver disorders, dysentery, sunstroke, cancer and heart diseases. The present study was undertaken to investigate ulcer protective effect of aqueous and ethanolic extract of Garcinia indica Linn fruit rind. The aqueous and ethanol extract of Garcinia indica Linn were investigated for ulcer protective activity against indomethacin induced ulcerogenesis and HCl/ethanol induced gastric lesion. Oral administration of the aqueous and ethanol extracts of Garcinia indica fruit rind at the dose 500 mg/kg provided significant (p<0.001) reduction of ulcer index in the HCl/ethanol and indomethacin induced gastric lesion rat models.

ABSTRACT

Tabernaemontana divaricata (L.) R.Br belonging to Apocynaceae family is traditionally used by people in many parts of the world to treat various disorders. The present study was undertaken to investigate anti-ulcer property of Tabernaemontana divaricata flower methanolic extract (TDFME 500 mg/kg, p.o) by pyloric ligation induced gastric ulceration model using Omeprazole (8mg/kg, p.o) as a standard drug in wistar rats. Five parameters i.e., volume of gastric juice, pH, free & total acidities and ulcer index were assessed. The test extract significantly (p< 0.01) decreased volume of gastric juice, free & total acidities and ulcer index. Like standard, it also raised pH of gastric acid. The observed percentage protection for standard and test were 89.84% and 79.53%, respectively. Thus, TDFME 500 mg/kg had a positive effect on all the parameters under study and the results were similar to that of standard. From the above results, it can be concluded that TDFME exhibits remarkable gastroprotective effect.

ABSTRACT

To evaluate antiulcer effects of aqueous, chloroform and ethanol extracts prepared from the henna leaves in rats employing the pylorus ligation and aspirin induced models. Gastric ulcers induced in Swiss albino rats (200g, N=6) by oral administration of aspirin suspension and pylorus ligation. The antiulcer activity was assessed by determining and comparing the ulcer index in the test drug groups with that of the vehicle control and standard ranitidine. In case of aspirin induced ulcers, the chloroform extract showed significant reduction of ulcers in a dose dependent manner. The parameters taken to assess antiulcer activity were volume of gastric juice, free acidity, total acidity and ulcer index. The results indicated that aqueous, ethanol and chloroform extract significantly (p<0.001) decreased the volume of gastric acid secretions, free acidity and total acidity and ulcer index.

ABSTRACT

The effect of aqueous extract of Ficus bengalensis (FBE) was assessed in different acute and chronic gastric ulcer models in rats. Gastric ulcers induced in swiss albino rats (200g, N=6) by oral administration of aspirin suspension and pylorus ligation. The anti ulcer activity was assessed by determining and comparing the ulcer index in the test drug groups with that of the vehicle control and standard ranitidine & sucralfate. FBE, 250–500 mg/kg administered orally, twice daily for 5 days showed dose-dependent ulcer protective effect in pylorus ligation (51.28, 63.24% protection, P < 0.01 to P < 0.001), aspirin (28.94, 64.91 protection, P < 0.001). The parameters taken to assess antiulcer activity were pH of gastric juice, total acidity and ulcer index. The results indicated that aqueous extract significantly (p<0.05) Ph, total acidity and ulcer index. On the basis of histopathology analysis, The results indicate that FBE possesses antiulcer activity in a dose dependent manner.
