ABSTRACT
The simple, precise and economic UV methods have been developed for estimation of Cefixime in single component. Cefixime has the absorbance maxima in zero order spectra in 230 nm (method A). Method B applied was first order derivative for the analysis of Cefixime at 238.5 nm .Method C applied was area under curve in the wavelength range of 234-228 nm. Drug followed Beer-Lamberts law in the concentration range of 503.5 μg/ml for zero order, 10-60 μg for area under curve methods and first order derivative spectrum. The percentage recovery of Cefixim ranged from 98.05 to 101.075 in pharmaceutical dosage from result of analysis was validated statistically and by recovered study.
ABSTRACT
AIM: To study the methodology of hepatic cytochromes P450 in Wistar rats. METHODS: After the Wistar rats were administered with gatifloxacin and ciprofloxacin once daily for 7 days,and liver microsome was distilled by different velocity centrifugation. The Lowry method was used to measure the concentration of protein in liver microsome,and the activities of gatifloxacin in hepatic microsomal enzymes (CYP450) of rats were studied by spectrophotometer. RESULTS: The linearity range of ox blood serum albumin standard curve was 25-250 mg?L -1 with the detection limits 25 mg?L -1 and correlation coefficient 0.9988 . The contents of cytochromes P450 and cytochromes b5 and the activity of NADPH-cytochrome C reductase indicated that ultraviolet spectrophotometer method was sensitive. The linearity range of formaldehyde standard curve,which was used to measure the activities of aminopyrine-N-demethylase and erytheomycin-N-demethylase was 0.05 - 0.5 mmol?L -1 with the detection limits 0.05 mmol?L -1 and correlation coefficient 0.9998 . The linearity range of resorufin standard curve,which was used to measure the activity of Coumarin 7-hydroxylation was 1-8 ?mol?L -1 with the detection limits 1 ?mol?L -1 and correlation coefficient 0.9998 . CONCLUSION: The methods of measuring the contents and activities of the six hepatic microsomal enzymes are sensitive and credible in Wistar rats by ultraviolet and fluorescence spectrophotometer.

AIM: To screen and optimize the preparation conditions of irisquinone-hydroxypropyl-?-cyclodextrin inclusion complex (Irisquinone-HP-?-CD) with stiring method. METHODS: The load-drug and recovery of irisquinone were adopted as the index, host-guest molar ratio, alcohol concentration and stiring rate as factors, have been investigated in the techniques of irisquinone-HP-?-CD. RESULTS: The load-drug and recovery of irisquinone-HP-?-CD were respective ( 15.79 ?0.22)% and ( 92.03 ?6.26)% by optimizing conditions which were host-guest molar ratio 2∶1, 90% EtOH and stiring rate 800r?min -1. At the same time, the dissolution test showed that irisquinone was faster released from inclusion complex than that from capsules and mixture dosage form. CONCLUSION: The optimizing techniques fit to prepare irisquinoe-HP-?-CD in industry.
