ABSTRACT

Objective To establish a new dialysis washing method for human frozen platelets and to evaluate a new type of washing machine based on dialysis. Methods Twenty bags of platelets(1U)were divided into two groups:centrifugation group and dialysis group. The effect of the washing machine was evaluated by measuring platelet function in vitro. Results After the platelets were treated by the two methods, the recovery rate of dialysis group[(73.57 ± 12.49)%]was significantly better than that of centrifugal group[(62.21 ±7.07)%].The osmotic pressure of dialysis group was 305.8 ±5.27mOsm/kg, compared to 383.2 ±22.00 mOsm/kg in centrifugation group. There was significant differences in the recovery rate and osmotic pressure(P<0.05), but there was no significant difference between dialysis group and centrifugation group in the mean platelet volume[(9.44 ±0.93)vs(9.23 ±0.50)fl], ADP-induced aggregation rate[(20.7 ±8.00)%vs(27.7 ±11.83)%],thrombin-induced platelet aggregation rate[(84.1 ±7.88)%vs(92.9 ±5.28)%], hypotonic shock responses[(19.02 ±4.42)% vs(23.19 ±9.08)%], CD62p activation rate [(16.00 ±9.40)%vs(21.86 ±8.14)%]or apoptosis rate[(46.31 ±10.28)% vs(54.59 ±16.76)%](P>0.05). Conclusion Compared to centrifugation group, the frozen platelet washing machine can not only improve platelet recovery, but also solve the problem of excessive DMSO residues.

ABSTRACT

Objective To investigate the status of microbial contamination in domestic washing machines, and pro-vide basis for strengthening hygiene and health management. Methods On-the-spot sampling, laboratory microbial detection, and questionnaire survey were performed to investigate and analyze the hygiene treatment and microbial contamination of domestic washing machines of Beijing residents. Results Source water of 115 washing machines were collected,no bacteria were detected. The total number of bacteria and fungi in washing water from 115 washing machines was 0-135 000 CFU/mL (13 206 CFU/mL on average)and 0-690 CFU/mL (229 CFU/mL on average) respectively. There was significant difference in bacterial colony forming unit in washing water from washing ma-chines of different length of service (P <0.05).15 specimens of washing water from 15 washing machines were per-formed bacteriological detection,17 kinds of bacteria were detected, most of which were opportunistic pathogenic bacteria. The major detected bacteria was Pseudomonas aeruginosa (66.67%),followed by Staphylococcus aureus (60.00%)and Escherichia coli (53.33%).Survey showed that 32.17% of residents reflected the washing ma-chines had peculiar smell and mildew spots,48.69% of users chose to open the covers for ventilation,38.26%adopted local wiping and cleaning of filter bags, only 10.43% chose to soak with disinfectant, while 24.35% didn’t know how to deal with. Conclusion Microbial contamination of domestic washing machines is serious, improper use and maintenance of washing machine is not conducive to health.

ABSTRACT

Objective To improve the processing protocol of the frozen erythrocyte ( RBC) washing machine of the dialysis type to enhance its efficiency of treating the 2 U frozen RBCs .Methods The frozen RBC washing machine for dialysis was used to deglycerolize the 2 U frozen RBCs after thawing .Measures including adjustment of the Pump speed , washing buffer adding and solution osmolarity were taken to decrease the processing time .Hemoglobin(Hb) content, Hb recovery, residual glycerol, free Hb content, deforming ability, and apoptosis of the deglycerolized RBCs were assessed to evaluate the efficiency of the improved protocol .Results As for the cells processed respectively by the previous and improved protocols, contents of Hb were (35.03 ±4.20) g and (53.82 ±2.08) g, respectively; Hb recoveries were (61.10 ±8.46)% and (80.22 ±3.73)%,respectively.Osmolarities representing residual glycerol were (307.00 ± 14.50)mOsm and (322.00 ±29.00)mOsm.Free Hb values were (0.74 ±0.04) and (0.80 ±0.08)g/L.The processing time was (1.64 ±0.31) and (1.04 ±0.16) h,respectively. There were significant differences in Hb content , Hb recovery, and deforming ability between the cells processed by the two protocols respectively .However , there were no significant changes in glycerol contents , free Hb, and apoptosis extent between the cells processed by the two protocols .Conclusion The innovated protocol can improve the quality of washed frozen RBCs , decrease the processing time and enhance the efficiency of the machine.
