ABSTRACT

Objective To investigate the clinical data,brain pathology and molecular genetic characteristics of retinal vascular disease families with leukoencephalopathy and systemic damage.Methods The clinical data of two families of RVCL-S(retinal vasculopathy with cerebral leukoencephalopathy and systemic manifestations,RVCL-S)were collected and family maps were drawn to analyze the clinical manifestations,imaging,brain pathology and molecular genetic characteristics.Results Family 1:there were 3 male cases and the age of onset was 10 years,29 years and 40 years,respectively.Family 2:there was one male case and the age of onset was 32 years.Both families presented with retinal vascular disease,leukoencephalopathy and multi-system damage,the latter including liver,kidney and digestive tract involvement.There were 4 asymptomatic carriers in two families.The cranial CT of family 1-Ⅱ2 showed lamellar low density near the posterior corner of the left ventricle with multiple intracranial high density calcification.Brain MRI plain scan showed lateral ventricle lesions.The brain MRI plain and enhancement scans of family 1-I5 showed frontotemporal cortex lesions with peripheral edema and space occupying effect,and the ring enhancement was remarkable.The brain MRI plain scan and enhancement scan of family 2-Ⅱ1 showed the right and left frontal lobe lesions,accompanied by peripheral edema and enhancement,and the occupying effect was obvious.The operative pathology of brain tissue from family 1-I5 showed endothelial cell hyperplasia.The pathological manifestations of family 2-Ⅱ1 encephalopathy were consistent with"cerebral infarction"after two operations.The small blood vessels in the small intestinal wall showed endothelial cell proliferation.Molecular genetics:TREX1 D272Rfs heterozygous mutation was present in family 1-Ⅱ2,and his offspring including two daughters and one son were asymptomatic mutation carriers.TREX1 S246Ifs heterozygous mutation was detected in the 2-Ⅱ1 gene of the family which was not found in either the father or the mother found the mutation,and the son was asymptomatic carrier of the mutation.Conclusion The main clinical manifestations of RVCL-S are retinal vascular disease,nervous system involvement and multi-system damage.Imaging findings showed that intracranial lesions were space-occupying,accompanied by peripheral edema and enhancement.The pathological features were small vessel endothelial cell proliferation and lumen stenosis.Genetic results confirmed the presence of TREX1 gene mutation.

ABSTRACT

Objective To observe the effect of penetrating electro-acupuncture at scalp-acupoints on the motor function and oxidative stress action of mice with Parkinson's disease (PD).Methods Mouse model of PD was established by 5-day intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP,30 mg/kg),and then PD mice were given electro-acupuncture set at different parameters.The effect of the electroacupuncture on the behavior and expression of tyrosine hydroxylase (TH) of mice were observed to screen out the appropriate treatment parameters.Healthy C57BL/6 mice were randomly divided into blank group,model group,positive group (madopar,6.25 mg/kg) and electro-acupuncture group.The mice except for blank group were given intraperitoneal injection of MPTP to induce PD,and then the PD mice of positive group and electroacupuncture group were treated with madopar(6.25 mg/kg) and electro-acupuncture respectively from the 6th day for 7 continuous days.Behavioral test was carried out on the 5~ modeling day and on the Th electro-acupuncture treatment day to evaluate the motor function of the mice.After the mice were killed and their brains were isolated,mitochondrial complex Ⅰ ~Ⅳ activities,reactive oxygen species(ROS) level,malondialdehyde (MDA) content,and total activities of superoxide dismutase (SOD) in the cerebral mitochondria were determined to evaluate the effect of electro-acupuncture at acupoints on anti-oxidative stress of PD mice.Results The motor function of PD mice was improved,the activity of mitochondrial complex Ⅰ was increased,the contents of ROS and MDA in cerebral mitochondria were decreased,and the activity of SOD was increased in the electro-acupuncture group,the differences being significant compared with those of the model group (P < 0.05 or P < 0.01).Conclusion Penetrating electro-acupuncture at scalp-acupoints can increase the anti-oxidative ability of the cerebral mitochondria and improve the motor function of PD mice.

ABSTRACT

Objective To observe the effect and mechanism of extracts of Liangxue Tongyu Recipe(LTR) on acute cerebral hemorrhage rats. Methods Twelve SD rats were randomly divided into sham operation group, model group, LTR water-extract group and LTR alcohol-extract group. Rats of sham operation group were injected with 2μL of normal saline into the left caudate nucleus, and rats of the other groups were injected with 2μL (0.25 U/ μL) of type Ⅶ collagenase into the left caudate nucleus to induce the model of acute cerebral hemorrhage. And then the rats of LTR groups were given intra-gastric gavage of LTR water-extract or LTR alcohol-extract. At the end of the experiment, we observed the macroscopic features of rat brain, pathological changes in rat brain under microscope after HE staining, awake time, rectal temperature, neurobehavioral scores and serum-specific enolase (NSE) level. Results Rats except for the sham operation group had various degrees of right hemiplegia symptoms, and the symptoms became severest on the third day of experiment. LTR alcohol-extract group and water-extract group had an effect on improving neurobehavioral scores, shortening awake time, and decreasing rectal temperature and serum NSE level of model rats. Conclusion LTR alcohol-extract and water-extract can relieve acute cerebral hemorrhage induced by type Ⅶcollagenase .

ABSTRACT

Objective To observe the effect of Da Jianzhong Decoction on cerebral cyclooxygenase-2 (COX-2) mRNA and protein and calmodulin-dependent protein kinase II(CaMKⅡ) mRNA in spleen-yang deficiency pain rats, so as to investigate the therapeutic mechanism of Da Jianzhong Decoction for spleen-yang deficiency pain. Methods Forty SD rats were randomly divided into normal group, model group, and high-and low-dose Da Jianzhong Decoction groups(3, 0.75 g·kg-1·d-1), 10 in each group. Spleen deficiency rat model was established by comprehensive modeling methods. After the rats in various groups were given gastric administration of Chinese medicine for 15 continuous days, rat cervical dislocation was performed for the sampling of the brain tissue. The expression levels of COX-2 mRNA and CaMKⅡmRNA in brain tissue were detected by real time polymerase chain reaction (RT-PCR). The COX-2 protein expression was determined by immunohistochemistry. Results Compared with the normal group, mRNA and protein expression levels of COX-2 in rat brain tissue of spleen-yang deficiency model group were increased(P﹤0.01), and CaMKⅡ mRNA expression was decreased (P<0.01). Compared with the model group, COX-2 mRNA and protein expression levels were decreased(P<0.01) and CaMKⅡexpression was increased(P<0.01) in high-and low-dose Da Jianzhong Decoction groups(P<0.01). Conclusion Da Jianzhong Decoction can suppress cerebral COX-2 mRNA and protein expression, and up-regulate CaMKⅡmRNA expression, which probably contribute to its therapeutic mechanism for relieving the pain.

Objective To observe the protective effect of Huoxue Jiannao Capsule(HJC) on neuronal cells in rats after focal cerebral ischemia-reperfusion injury.Methods SD rats were randomized into nimodine group,mode1 group,pseudo-operation group(at the dose of 10.8mg?kg-1?d-1) and HJC group(at the dose of 1.8g?kg-1?d-1).The rats in the latter 3 groups received 2-hour ischemia,and 6,12 and 24 hours of reperfusion respectively.Focal cerebral ischemia-reperfusion model was induced by intra-luminal thread occlusion.Cytochrome C(Cyt C)of neuronal cells was measured by immunohistochemistry.In-situ hybridization(ISH) was used to determine the expression of Caspase-3 mRNA and Caspase-9 mRNA.Results Weak positive expression of Cyt C was presented in HJC group after cerebral ischemia-reperfusion for 6 hours,and the difference was significant as compared with the model group(P0.05).After ischemia-reperfusion for 24 hours,the expression of Caspase-3 mRNA and Caspase-9 mRNA in hippocampus arrived to the peak in the model group,while was decreased in HJC group,and the difference between the model group and HJC group was significant(P

ABSTRACT

Objective To investigate the effect of electroacupuncture(EA)in improving the reconstruction of synapse after cerebral ischemia.Methods Forty-five Wistar rats were divided into 3 groups:sham-operated group,ischemia group and EA group.The number density(Nv)of synapses and the surface density(Sv)of synaptic joint band were observed 1 hour,1 day,3 days,1 week and 3 weeks after ischemia and EA, respectively.Heat-coagulation-induced occlusion of the middle cerebral artery was performed to establish the model of focal cerebral ischemia.EA group received EA on the acupoints of Baihui and Dazhui.The changes of synaptie structure,Nv and Sv as well as the influence of EA on the above indexes were observed in different time.Results There existed significant decrease of Nv and Sv 1 hour,1 day and 3 days after establishment of models in the isehemia group(P0.05).Conclusion EA can reduce the cerebral ischemic injury by increasing the synaptic amount and area in the cortex of rat model of cerebral isehemia at early stage,promoting the cerebral recovery after ischemia,which will provide substantial morphological basis for the improvement of synaptic reconstruction and recovery from cerebral isehemia by EA at different time after ischemic cerebral injury.

Objective To observe the effect of Fujian Tablets(FT),a prescription with the actions of nourishing liver and kidney,on the expression of microtubule-associated protein 2(MAP-2) in cerebral infarction rats at different time points,and to explore its mechanism on promoting nerve cell differentiation.Methods A total of 240 male Sprague Dawley rats were divided into normal control group,sham-operation group,model group and FT group.Focal cerebral ischemia models were established by occlusion of the middle cerebral artery according to the reformed Longa method in rats,and were subdivided into 3-,7-,14-,28-,and 42-day cerebral infarction subgroups.FT group was given FT in the dose of 9 g?kg-1?d-1,and the other groups were given the same dose of distilled water.Results In the model group,MAP-2 expression in the hippocampus increased and reached a peak on day 3,which were notably different from that in the normal control group(P0.05).In FT group,MAP-2 expression increased on day 3,reached a peak on day 7,remained high level till day 14,and then decreased,the difference being significant compared with the model group(P
