Abstract Anredera cordifolia (Ten.) Steenis is a vine species native to Brazil that is considered an unconventional food plant and a medicinal species whose phenolic compounds exert antioxidant activity. Since the production of metabolites is determined by environmental factors and leaf maturity, it is important to track these changes in order to determine the best time to harvest. This study aimed to verify whether leaf phenology and seasonality cause variations in the amount of phenolic compounds and in the antioxidant activity of this species. The leaves were collected in different seasons between September 2018 and April 2019, and separated according to maturity: young, mature, and senescent. Daily atmospheric temperature and rainfall data were used to characterize the collection period. The total phenolic content (TPC), determined by Folin-Ciocalteu method, was significantly higher in the young leaves collected in winter, a season of lower temperatures. These leaves showed 54.4 mg of gallic acid equivalents per 100 g of dry matter (mg GAE 100 g-1DM). Other results averaged 25.6 mg GAE 100 g-1DM. The highest antioxidant activity, assessed via the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method, regardless of leaf phenology, was observed in leaves harvested in autumn (73.7%) and winter (71.1%), seasons with lower rainfall. Leaves harvested in summer and spring had lower antioxidant action rates (54.3 e 37.5%, respectively). There was no significant correlation between the total phenolic content and antioxidant activity. Thus, the phenolic composition of A. cordifolia, and consequently its activity on free radicals, varies seasonally in response to temperature and rainfall, and may or may not interact with the age of the leaves.


Resumo Anredera cordifolia (Ten.) Steenis é uma trepadeira nativa do Brasil considerada uma planta alimentícia não convencional e uma espécie medicinal, cujos compostos fenólicos exercem ação antioxidante. Como a produção de metabólitos é determinada por fatores ambientais e pela maturidade das folhas, é importante acompanhar essas mudanças a fim de determinar a época de colheita. Neste estudo objetivou-se verificar se a fenologia foliar e a sazonalidade ocasionam variação na quantidade de compostos fenólicos e na ação antioxidante dessa espécie. As folhas foram coletadas em diferentes estações, entre setembro de 2018 e abril de 2019, e separadas conforme a maturidade: jovens, maduras e senescentes. Os dados diários de temperatura atmosférica e pluviosidade foram usados para caracterizar o período de coleta. O conteúdo de fenólicos totais (TPC), determinado pelo método Folin-Ciocalteu, foi significativamente maior nas folhas jovens coletadas no inverno, período caracterizado por temperaturas mais baixas. Estas folhas apresentaram 54,4 mg equivalentes de ácido gálico por 100 g de matéria seca (mg GAE 100 g-1DM) . A média dos outros resultados foi de 25,6 mg GAE 100 g-1DM. A maior ação antioxidante, avaliada pelo método do radical livre 2,2-difenil-1-picrilhidrazil (DPPH), foi observada em folhas colhidas no outono (73,7%) e inverno (71,1%), independentemente da fenologia foliar. Nestes períodos a pluviosidade foi menor. Folhas colhidas no verão e na primavera apresentaram menores percentuais de ação antioxidante (54,3 e 37,5%, respectivamente). Não houve correlação significativa entre o conteúdo de fenólicos totais e a atividade antioxidante. Conclui-se que a composição fenólica de A. cordifolia e, consequentemente, sua ação sobre radicais livres, varia sazonalmente em resposta à temperatura e pluviosidade, podendo ou não interagir com a idade das folhas.

Abstract This study explores the antioxidant activity, phytochemical screening, total phenolic and flavonoids contents in the extracts of four locally available weeds plants namely Convolvulus arvensis, Chenopodium murale, Avena fatua and Phalaris minor with different solvents. The antioxidant activities of these extracts were determined via various in-vitro methods such as total antioxidant activity (TAA), reducing power (RP), DPPH (2,2-Diphenyl-1-Picrylhydrazyl) free radical scavenging and hydrogen peroxide scavenging assays. Phytochemical screening was performed both qualitatively as well as quantitatively. Total phenolic content (TPC) and total flavonoid content (TFC) were determined through Folin- Ciocalteu reagent and aluminium chloride methods respectively. Methanol-chloroform solvent showed the presence of a high amount of TPC in milligram of gallic acid equivalent per gram of dry weight (mg of GAE/g of DW) in the extracts of all weeds. Their descending sequence was Avena fatua (74.09) Phalaris minor (65.66) Chenopodium murale (64.04) Convolvulus arvensis (61.905), while, chloroform solvent found to be best solvent for the extraction of TFC. Methanol-chloroform solvent was also found to be best solvent for TAA (Total antioxidant activity assay) which showed values in milligram of ascorbic acid equivalent per gram of dry weight (mg of AAE /g of DW), for DPPH scavenging activity, reducing power (antioxidant activity) and hydrogen peroxide scavenging activity. Phytochemical screening indicated the presence of polyphenols, flavonoids, tannins, saponins, alkaloids and glycosides in these weeds.


Resumo Este estudo investiga a atividade antioxidante, a triagem fitoquímica, os teores de fenólicos totais e de flavonoides nos extratos de quatro plantas daninhas disponíveis localmente, quais sejam, Convolvulus arvensis, Chenopodium murale, Avena fatua e Phalaris minor com diferentes solventes. As atividades antioxidantes desses extratos foram determinadas por meio de vários métodos in vitro, tais como atividade antioxidante total (TAA), poder redutor (RP), sequestro de radicais livres DPPH (2,2-Difenil-1-Picril-hidrazil) e ensaios de sequestro de peróxido de hidrogênio. A triagem fitoquímica foi realizada tanto qualitativamente quanto quantitativamente. O teor de fenólicos totais (TPC) e o teor de flavonoides totais (TFC) foram determinados pelos métodos do reagente de Folin-Ciocalteu e do cloreto de alumínio, respectivamente. O solvente metanol-clorofórmio mostrou a presença de elevada quantidade de TPC em miligramas de ácido gálico equivalente por grama de peso seco (mg de GAE/g de DW) nos extratos de todas as plantas daninhas. Sua sequência descendente foi Avena fatua (74,09) Phalaris minor (65,66) Chenopodium murale (64,04) Convolvulus arvensis (61,905), enquanto o solvente clorofórmio foi o melhor solvente para a extração de TFC. O solvente metanol-clorofórmio também foi considerado o melhor solvente para AAT (ensaio de atividade antioxidante total), que apresentou valores em miligramas de equivalente de ácido ascórbico por grama de peso seco (mg de AAE/g de DW), para atividade sequestrante de DPPH, RP (atividade antioxidante) e atividade de sequestro de peróxido de hidrogênio. A triagem fitoquímica indicou a presença de polifenóis, flavonoides, taninos, saponinas, alcaloides e glicosídeos nessas plantas daninhas.

Background: The edible green algae Caulerpa racemosa (Forsskal) J. Agardh (Caulerpaceae), also known as "sea grape", is an excellent source of phenolic compounds known for their activity to reduce free radicals. Objectives: The research aims to evaluate the total phenolic content and antioxidant activity of C. racemosa (70% ethanol extracts) obtained from different extraction methods, such as maceration, Soxhlet, and ultrasound. Methods: Total phenolics of the extracts were determined by the colorimetry method using the Folin-Ciocalteu reagent. Total phenol content was expressed as mg gallic acid equivalent (GAE) per g extract. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical was used to assess the antioxidant activity of the extracts. Results: the ethanol extract of C. racemosa obtained from the ultrasonic methods had the highest phenolic content (39.38 mg GAE/g) compared to other extraction methods (Soxhlet was 36.88 mg GAE/g and maceration was 22.05 mg GAE/g). The IC50 value against DPPH of the C. racemosa ethanol extracts using ultrasonic, Soxhlet, and maceration was 352.95, 365.73, and 375.84 µg/mL, respectively. Conclusions: the variation of the extraction methods affected the total phenolics content of C. racemosa ethanol extracts and their antioxidant activity. We reported here the potential of C. racemosa extracts as an antioxidant raw material from marine plants for medicinal, nutraceutical, cosmetics, and food products; however, more research is needed.


Antecedentes: El alga verde comestible Caulerpa racemosa (Forsskal) J. Agardh (Caulerpaceae), también conocida como "uva de mar", es una excelente fuente de compuestos fenólicos conocidos por su actividad para reducir los radicales libres. Objetivos: La investigación pretende evaluar el contenido fenólico total y la actividad antioxidante de C. racemosa (extractos de etanol al 70%) obtenidos a partir de diferentes métodos de extracción, como maceración, Soxhlet y ultrasonido. Métodos: Los fenoles totales de los extractos se determinaron por el método colorimétrico utilizando el reactivo de Folin-Ciocalteu. El contenido total de fenoles se expresó como mg de ácido gálico equivalente (GAE) por g de extracto. Para evaluar la actividad antioxidante de los extractos se utilizó el radical libre 2,2-difenil-1-picrilhidrazilo (DPPH). Resultados: el extracto etanólico de C. racemosa obtenido por ultrasonido presentó el mayor contenido fenólico (39,38 mg GAE/g) en comparación con otros métodos de extracción (Soxhlet fue de 36,88 mg GAE/g y maceración fue de 22,05 mg GAE/g). El valor IC50 frente a DPPH de los extractos etanólicos de C. racemosa mediante ultrasonido, Soxhlet y maceración fue de 352,95, 365,73 y 375,84 µg/mL, respectivamente. Conclusiones: la variación de los métodos de extracción afectó al contenido total de fenoles de los extractos etanólicos de C. racemosa y a su actividad antioxidante. Aquí reportamos el potencial de los extractos de C. racemosa como materia prima antioxidante a partir de plantas marinas para productos medicinales, nutracéuticos, cosméticos y alimenticios; sin embargo, se necesita más investigación.

ABSTRACT

Natural antioxidants have an important role in the prevention of many age-related diseases and promotion of health. Among natural antioxidants from plants, flavonoids and other phenolic compounds are potent antioxidants and chelating agents. Panchavalkala the barks of five trees i.e. Nyagrodha (Ficus benghalensis L.), Udumbara (Ficus racemosa L.), Ashwatha (Ficus religiosa L.), Plaksha (Ficus virens Aiton) and Parisha (Thespesia populnea (L.)Sol.ex Correa) are also known as Pancha Ksheeri Vrikshas in use since Vedic period. Barks of these trees are dried in shade and are used for different formulations (Pancha Kashaya Kalpanas), in different pathological conditions, especially as wound healing, gynecological disorders and etc. The plant samples were extracted using ethanol and water, and subjected for the phytochemical analysis. It was confirmed that samples contain many biologically active compounds like flavonoids, polyphenols, tannins, alkaloids, glycosides and terpinoids etc. The marker compound of each trial drug and the quantitative analysis has been carried out by high performance liquid chromatography. The antioxidant study was done by using in vitro method 1, 1­-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay. The marker compounds caffeic acid and gallic acid were quantified in each extract for their quality and efficacy. PVK barks showed high free radical scavenging activity as evidenced by the low IC50 values in DPPH (EE PVK- 20.46µg/ml, AE PVK-37.79µg/ml, EE T.poulenea-22µg/ml, AE T. poulenia- 23.31µg/ml AE F. benghalensis- 25.53µg/ml, EE F. benghalensis- 26.23µg/ml, EE F. religiosa - 34µg/ml). Quercetin- IC50 value 4.026µg/ml is used as standard. The results of the study demonstrated that PVK barks possess phyto-constituent’s viz. tannins, flavonoids, polyphenols etc. and has potential antioxidant activity. Thus these barks have good therapeutic potential as natural antioxidant and might be used in life style related conditions like hyperlipidemia, diabetes, obesity, cardiovascular disorders and etc.

In this work, we were interested in a medicinal plant named Chrysopogon nigritanus, belonging to the Poaceae family. In order to carry out this study, we were interested first in the phytochemical screening test, after having carried out the extraction with two solvents (water and isopropanol), to characterize the different families of metabolites present in the extracts. Then, the assays of polyphenols and flavonoids were carried out in order to quantify the contents of these two compounds in each extract. Finally, the antioxidant activity of the extracts was evaluated by three methods: the CUPRAC method, the trapping of the 2,2-diphenyl-1-picryl-hydrazyl radical (DPPH•) and the trapping of the ABTS•+ radical-cation (2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)). The best antioxidant activities were obtained with the ABTS•+ radical scavenging with IC50 of 0.13 mg/mL for the aqueous extract and 0.19 mg/mL for the isopropanol extract. Based on these first results, an in-depth study is underway for various biological tests and the isolation of bioactive molecules.

Background: Berberis commutata Eichler is a berry that grows in the Peruvian Andes and has been consumed in the Andes of South America since ancient times. The edible fruits have an intense purple color and are rich in anthocyanins and phenolic compounds that are available from February until May each year. The color of the fruits is a soft purple dye for natural fibers, and many birds use them as food. Objective: This study quantified the total phenolic, monomeric anthocyanin content and antioxidant activity of Berberis commutata Eichler berries. Methods: The total phenolic content was determined by the Folin-Ciocalteu colorimetric assay. Monomeric anthocyanin content was determined by the method is pH differential, and the antioxidant activity was measured using the Brand-Williams method. Results: The total phenolic content was 7,490 ± 0.85 mg GAE/100g, and the monomeric anthocyanin content was 70 ± 0.03 mg/100g. The antioxidant activity of the berries showed a tendency to increase with B. commutata extract concentration; an EC50 of 0.91 mg/mL was calculated, indicating a high antioxidant power. Conclusion: Our results showed that B. commutata E. has both high total phenolic content and monomeric anthocyanins comparable to other superfruits and high antioxidant activity, which means that it is possible to use this berberis species as a functional food.


Introducción: Berberis commutata Eichleres una baya que crece en los Andes peruanos. Los frutos comestibles tienen un intenso color púrpura rico en antocianinas y componentes fenólicos que están disponibles desde febrero hasta mayo de cada año. El color de sus frutos se utiliza como un suave colorante púrpura para las fibras naturales y muchas aves los utilizan como alimento. Sin embargo, desde la antigüedad los frutos de esta especie han sido consumidas en los Andes de Sudamérica. Objetivo: Este estudio cuantificó el contenido fenólico total, antocianinas monoméricas y la actividad antioxidante usando el método del radical DPPH de las bayas de Berberis commutata Eichler. Método: El contenido fenólico total se midió a través del ensayo colorimétrico de Folin-Ciocalteu, el contenido de antocianinas monoméricas se determinó mediante el método del pH diferencial y la actividad antioxidante se midió con el método de Brand-Williams. Resultados: El contenido fenólico total fue de 7,490 ± 0.85 mg GAE/100g y el contenido de antocianinas monoméricas fue de 70 ± 0.03 mg/100g. La actividad antioxidante de las bayas mostró una tendencia a aumentar con la concentración del extracto de B. commutata, se calculó un EC50 de 0.91 mg/mL que indica un alto poder antioxidante. Conclusión: Nuestros resultados mostraron que B. commutata E. tiene tanto un alto contenido fenólico total, así como antocianinas monoméricas comparables con otras superfrutas y una elevada actividad antioxidante, lo que significa que es posible utilizar esta especie de berberis como alimento funcional.

The study was aimed to evaluate the therapeutic effects of Zizyphus oxyphyla leaves methanolic (ZOX-LME), on serum liver, kidney and hematology along with other serum parameters in Carbon tetrachloride (CCl4) intoxicated rabbits. Experimental animals were divided into five groups, six rabbits in each. These were: group NC (normal control), group, TC (toxic control) and group ST i.e. silymarine administered group at dose rate (50) mg/kg body weight (BW). Group ET1 and group ET2 treated with (ZOX-LME) at dose 200 mg/kg BW and 400 mg/kg BW. CCl4 administration caused significant (P> 0.05) impairment in serum liver enzymes, blood factors and other serum indices. Treatment with (ZOX-LME) significantly (P<0.05) reduced and normalized the levels of serum alanine transaminase (ALT) aspartate transaminase (AST) and alkaline phosphatase (ALP) and hematological indices. Also significant (P< 0.05) reduction was observed in creatinine, urea, uric acid, blood urea nitrogen (BUN), and albumin and glucose concentrations. The altered levels of lipid profile and serum electrolytes (Ca, Mg, Cl, Na, K, and P) were significantly (P<0.05) change toward normal levels with (ZOX-LME) feeding. In addition (ZOX-LME) ingestion caused significant improvement in GSH, GST and CAT levels, while reducing the TBARS levels, exhibited antioxidant capacity. Also (ZOX-LME) showed increase inhibition against percent scavenging of 2, 2-diphenile-1-picrylehydrazyle (DPPH) free radical. Significant (P<0.05) normalizing effects were observed with high dose 400 mg/kg BW of (ZOX-LME and were equivalent to silymarine administered groups. The histological study of liver supported the hepatoprotective and renal curative activity of (ZOX-LME).


O estudo teve como objetivo avaliar os efeitos terapêuticos das folhas metanólicas de Zizyphus oxyphyla (ZOX-LME) no fígado, rim e hematologia séricos, juntamente com outros parâmetros séricos em coelhos intoxicados com tetracloreto de carbono (CCl4). Os animais experimentais foram divididos em cinco grupos, seis coelhos em cada. Estes foram: grupo NC (controle normal), grupo TC (controle tóxico) e grupo ST, isto é, grupo administrado com silimarina na taxa de dose (50) mg / kg de peso corporal (PC). Grupo ET1 e grupo ET2 tratado com (ZOX-LME) na dose de 200 mg / kg de peso corporal e 400 mg / kg de peso corporal. A administração de CCl4 causou prejuízo significativo (P > 0,05) nas enzimas hepáticas séricas, fatores sanguíneos e outros índices séricos. O tratamento com (ZOX-LME) reduziu significativamente (P < 0,05) e normalizou os níveis de alanina transaminase (ALT), aspartato transaminase (AST) e fosfatase alcalina (ALP) e os índices hematológicos. Também foi observada redução significativa (P < 0,05) nas concentrações de creatinina, ureia, ácido úrico, nitrogênio ureico no sangue (BUN), albumina e glicose. Os níveis alterados de perfil lipídico e eletrólitos séricos (Ca, Mg, Cl, Na, K e P) foram significativamente (P < 0,05) mudando em direção aos níveis normais com a alimentação (ZOX-LME). Além disso, a ingestão de (ZOX-LME) causou melhora significativa nos níveis de GSH, GST e CAT, enquanto reduzia os níveis de TBARS, exibindo capacidade antioxidante. Também (ZOX-LME) mostrou inibição aumentada contra a eliminação percentual do radical livre 2, 2-difenila-1-picrilehidrazila (DPPH). Efeitos de normalização significativos (P < 0,05) foram observados com altas doses de 400 mg / kg de peso corporal de (ZOX-LME) e foram equivalentes aos grupos administrados com silimarina. O estudo histológico do fígado confirmou a atividade hepatoprotetora e curativa renal de (ZOX-LME).

CCl4 administration caused significant (P> 0.05) impairment in serum liver enzymes, blood factors and other serum indices. Treatment with (ZOX-LME) significantly (P 0.05) reduced and normalized the levels of serum alanine transaminase (ALT) aspartate transaminase (AST) and alkaline phosphatase (ALP) and hematological indices. Also significant (P 0.05) reduction was observed in creatinine, urea, uric acid, blood urea nitrogen (BUN), and albumin and glucose concentrations. The altered levels of lipid profile and serum electrolytes (Ca, Mg, Cl, Na, K, and P) were significantly (P 0.05) change toward normal levels with (ZOX-LME) feeding. In addition (ZOX-LME) ingestion caused significant improvement in GSH, GST and CAT levels, while reducing the TBARS levels, exhibited antioxidant capacity. Also (ZOX-LME) showed increase inhibition against percent scavenging of 2, 2-diphenile-1-picrylehydrazyle (DPPH) free radical. Significant (P 0.05) normalizing effects were observed with high dose 400 mg/kg BW of (ZOX-LME and were equivalent to silymarine administered groups. The histological study of liver supported the hepatoprotective and renal curative activity of (ZOX-LME).


Resumo O estudo teve como objetivo avaliar os efeitos terapêuticos das folhas metanólicas de Zizyphus oxyphyla (ZOX-LME) no fígado, rim e hematologia séricos, juntamente com outros parâmetros séricos em coelhos intoxicados com tetracloreto de carbono (CCl4). Os animais experimentais foram divididos em cinco grupos, seis coelhos em cada. Estes foram: grupo NC (controle normal), grupo TC (controle tóxico) e grupo ST, isto é, grupo administrado com silimarina na taxa de dose (50) mg / kg de peso corporal (PC). Grupo ET1 e grupo ET2 tratado com (ZOX-LME) na dose de 200 mg / kg de peso corporal e 400 mg / kg de peso corporal. A administração de CCl4 causou prejuízo significativo (P > 0,05) nas enzimas hepáticas séricas, fatores sanguíneos e outros índices séricos. O tratamento com (ZOX-LME) reduziu significativamente (P 0,05) e normalizou os níveis de alanina transaminase (ALT), aspartato transaminase (AST) e fosfatase alcalina (ALP) e os índices hematológicos. Também foi observada redução significativa (P 0,05) nas concentrações de creatinina, ureia, ácido úrico, nitrogênio ureico no sangue (BUN), albumina e glicose. Os níveis alterados de perfil lipídico e eletrólitos séricos (Ca, Mg, Cl, Na, K e P) foram significativamente (P 0,05) mudando em direção aos níveis normais com a alimentação (ZOX-LME). Além disso, a ingestão de (ZOX-LME) causou melhora significativa nos níveis de GSH, GST e CAT, enquanto reduzia os níveis de TBARS, exibindo capacidade antioxidante. Também (ZOX-LME) mostrou inibição aumentada contra a eliminação percentual do radical livre 2, 2-difenila-1-picrilehidrazila (DPPH). Efeitos de normalização significativos (P 0,05) foram observados com altas doses de 400 mg / kg de peso corporal de (ZOX-LME) e foram equivalentes aos grupos administrados com silimarina. O estudo histológico do fígado confirmou a atividade hepatoprotetora e curativa renal de (ZOX-LME).

Abstract Present study analysed the therapeutic potential of traditionally acclaimed medicinal herb Nanorrhinum ramosissimum, using plant parts extracted with different solvents (10 mg/mL). Shoot extracts exhibited comparatively better antimicrobial properties, in comparison to root extracts. Total phenolic content was estimated, to ascertain its dependency on antioxidant properties of plant extracts. Antioxidant assay revealed promising results in comparison to IC50 value of standard ascorbic acid (52.2±0.07 µg/mL), for methanolic extracts of shoot (61.07±0.53 µg/mL and 64.33±0.33 µg/mL) and root (76.705±0.12 µg/mL and 89.73±0.28 µg/ mL) for in vivo and in vitro regenerants respectively. Correlation coefficient R2 values ranged between 0.90-0.95, indicating a positive correlation between phenolic contents and antioxidant activity. Plant extracts were also able to inhibit DNA oxidative damage again indicating their antioxidative potential. Antidiabetic potential was confirmed by alpha amylase inhibition assay where shoot methanolic extracts (invivo, in vitro) exhibited the best IC50 values (54.42±0.16 µg/mL, 66.09±0.12 µg/mL) in comparison to standard metformin (41.92±0.08 µg/mL). Ethanolic extracts of roots (in vitro, invivo) exhibited the relative IC50 values (88.97±0.32µg/mL,96.63±0.44 µg/mL) indicating that shoot parts had a better alpha amylase inhibition property; thus proving the herb's bioactive potential and its prospective therapeutic source for curing various ailments.

ABSTRACT

A panel of 2,3-disubstituted thiazolidin-4-ones 4a-n was synthesised from Schiff bases 3a-n derived from sulfanilamide, by reaction with thioglycolic acid. The compounds were characterised by means of IR, NMR, and Mass spectral data. Compounds 4a-n were screened for DPPH scavenging assay and compounds 4e, 4h, 4i, and 4n exhibited moderate activity. Compounds 4e, 4h, and 4i were tested at 200 mg/kg and 4e at 50 mg/kg b.w. orally for antidiabetic activity in fructose induced diabetic rats. They exhibited significant antidiabetic activity compared to the control group. Pioglitazone was used as a standard drug. The tested compounds exhibited better and ignificant serum cholesterol lowering activity when compared with the control and standard groups. They also reduced the triglyceride level after the 21st day; however, it was insignificant when compared to the control group. Compound 4n displayed the highest binding energy when docked with PPAR-γ followed by compounds 4e, 4h, and 4i when compared to pioglitazone. The physicochemical, drug likeness and ADME properties of the title compounds were found to be satisfactory.


Se sintetizó un panel de tiazolidinas-4-onas 2,3-disustituidas 4a-n a partir de las bases de Schiff 3a-n derivadas de la sulfanilamida por reacción con ácido tioglicólico. Los compuestos se caracterizaron por IR, RMN y datos espectrales de masa. Los compuestos 4a-n se analizaron para DPPH y los compuestos 4e, 4h, 4i y 4n mostraron una actividad moderada. Los compuestos 4e, 4h y 4i se probaron a 200 mg/kg y 4e a 50 mg/kg b.w. oralmente para la actividad antidiabética en ratas diabéticas, inducida por fructosa. Los compuestos mostraron una actividad antidiabética muy significativa en comparación con el grupo control. La pioglitazona se utilizó como fármaco estándar. Los compuestos ensayados mostraron una mejor y significativa actividad reductora del colesterol sérico en comparación con los grupos control y estándar. Estos compuestos también redujeron el nivel de triglicéridos después del 21° día, aunque fue insignificante en comparación con el grupo control. El compuesto 4n mostró la mayor afinidad de unión cuando se acopló a PPAR-γ, seguido de 4e, 4h y 4i en comparación con la pioglitazona. Las propiedades fisicoquímicas, la similitud con el fármaco y las propiedades ADME de los compuestos fueron satisfactorias, lo que los convierte en útiles agentes antidiabéticos.

Os compostos 4a-n foram rastreados para O ensaio DPPH de limpeza radical e os compostos 4e, 4h, 4i e 4n exibiram actividade moderada. Os compostos 4e, 4h e 4i foram testados a 200 mg/kg e 4e a 50 mg/kg de peso corporal por via oral para antidiabéticos. actividade em ratos diabéticos induzidos por frutose. Exibiram uma actividade antidiabética altamente significativa actividade em comparação com o controlo. A pioglitazona foi utilizada como droga padrão. Os compostos testados exibiu uma melhor e significativa actividade de redução do colesterol sérico quando comparado comde triglicéridos após o 21° dia; no entanto, foi insignificante quando comparado com o controlo. O composto 4n mostrou a maior afinidade de ligação quando acoplado com PPAR-γ seguido de 4e, 4h, 4i quando comparado com pioglitazona. O propriedades físico-químicas, de semelhança com drogas e ADME dos compostos do título de propriedade também foram encontrados paraser satisfatórios, tornando-os agentes antidiabéticos úteis.

ABSTRACT

The importance of Nigella sativa plant is increasing day by day in many medical and nutritional fields, and the demand for it is increasing in foreign markets. Therefore, we chose the nigella plant to study the effect of spraying with a natural growth stimulant (Ascobein) at concentrations of zero, 50, 100 and 150 ppm on the productivity of seed and oil, as well as its effect on the activity of antioxidants. The results showed that the concentration of 100 ppm gave the highest values of vegetative growth traits; (Plant height (86.3 and 89.5cm), Number of branches (21.7 and 25.1), Fresh and Dry weight per plant (118.43 and 143.85gm, 23.86 and 28.29gm/plant) respectively for both seasons. Also, results revealed that foliar application of 100 ppm (Ascobein) improved the seed yield/plant (16.95 and 19.74gm), Oil % (45.73 and 48.02%), DPPH% (99.04 and 102.39%), TPC (447.17 and 450.54 mg GAE/g) and TFC (110.19 and 114.43 mg QE/g) for both seasons 20/2021 and 21/2022. The percentage of major fatty acids in nigella seeds oil has been estimated (Linoleic, Oleic, Palmitic and Stearic).

ABSTRACT

Fennel plant (Foeniculum vulgare, Mill.) is one of most widely used plants in food and medicine. Our study aimed to show the response of fennel plants from Egypt and China planted in Egypt to foliar application of benzyladenine (BA) at various concentrations; control, 50, 100 and 150 ppm. The obtained results indicated that treated fennel plants with BA resulted in a significant increase in the vegetative growth, number of umbels/plant, fruit, yield and oil yield per plant and feddan as well as main components, total phenolic contents and antioxidant activity of essential oils compared to untreated plants. In general, the application of benzyladenine was more effective on Egyptian fennel plants than that of Chinese plants. Egyptian fennel plants gave higher values of growth, fruit yield, oil percentage and oil yield than Chinese plants. However, the major components, TPC and antioxidant activity (DPPH %) had higher values in Chinese essential oil than Egyptian essential oil. The highest values of all studied parameters were obtained by using BA at 100 ppm. GC–MS analysis of the volatile oils showed that the major components were trans-anethole followed by fenshone, ?- pinene and D-limonene, respectively for both essential oils. 100 ppm of BA gave the highest values of trans-anethole content and radical scavenging activities of oils. Therefore, we concluded that spraying fennel plants with 100 ppm BA can be used for obtaining higher vegetative growth, fruit and quantity and quality of oil.

ABSTRACT

The study assessed the phytochemical, nutritional and antioxidant properties of a functional tea made from mixture of Utazi leaf Gongronema latifolium), Nchanwu leaf (Ocimum gratissimum) and Turmeric rhizome (Curcuma longa). The three spices were individually processed by drying and mincing before combining them at different ratios that resulted to 16 samples. The samples were generated through D-optimal mixture design using statistical software (Design Expert version 12). The samples were evaluated for phytochemical, nutritional and antioxidant properties. The fresh raw materials were destalked (for the leaves), sorted, washed, withered, oxidized for 24 h, oven dried at 60oC for 30 min, cooled and milled, and packaged. Fresh turmeric rhizome were sorted, washed blanched at 90oC for 2 min, cooled, peeled, sliced, oven dried at 60oC for 30 min, cooled, milled and packaged in teabags and high density polyethylene. The alkaloids, saponin, phenol, cardiac glycoside, flavonoid, tannin, steroid and anthocyanin content ranged from 12.00-22.00, 0.42-1.15, 11.67-15.63, 0.06-0.67, 12.67-22.00, 0.56-0.89, 1.00-1.67, 46.00-54.00 mg/100g respectively. The vitamin C and E content ranged from 3.03-4.13, 0.31-0.64 mg/100g respectively. The minerals iron and zinc content of the tea samples ranged from 3.53-4.70, 0.06-0.78 ppm respectively. The sensory evaluation score of the tea infusion of the sample AB and DA were ‘liked very much’ with the rating of 8.60 and 8.20 respectively, these samples were more acceptable to the panelist. The antioxidant scavenging activity ranged from 42.00-47.00% with sample EC exhibiting the highest level of activity, and the proportion was 70g, 24.62g, 5.38g of Utazi leave, Nchanwu leave, and Turmeric rhizome.

Background: Ilex guayusa Loes. belongs to the family Aquifoliaceae. It is an evergreen tree native to the Amazon region. According to traditional uses, it is used as a diaphoretic, narcotic, purgative, among other uses. Objective: To evaluate the antioxidant capacity, quantify phenols and total flavonoids of extracts obtained from Ilex guayusa leaves. Methods: Total ethanolic extract was obtained using Soxhlet-type equipment, with subsequent liquid/liquid fractionation with solvents of different polarities (petroleum ether, dichloromethane, and ethyl acetate); were reacted with 2,2-diphenyl-1-picrylhydrazyl (DPPH•) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS•+), determining the inhibitory concentration 50 (IC50) and relative antioxidant activity (%AAR), with subsequent statistical analysis (ANOVA and Tukey's post-hoc). Total phenols and flavonoids were quantified using the Folin-Ciocalteu method and complex formation with AlCl3, respectively. Results: The ethanolic extract of leaves showed the highest antioxidant activity in both the DPPH• and ABTS•+ methods with IC50 values of 4.58 and 3.82 ppm, respectively. Conclusions: According to the results obtained in this study, it was possible to corroborate that the ethanolic extract (EE) obtained from the leaves of I. guayusa showed the highest antioxidant capacity, by the DPPH• and ABTS•+ methods. Additionally, it was possible to relate this capacity to the high flavonoids content present in this extract. Based on the above and accompanied by future studies, the Ilex guayusa species could be proposed as an important source of antioxidant compounds with possible application in medicine and the food industry


Antecedentes:Ilex guayusa Loes. pertenece a la familia Aquifoliaceae. Es un árbol perenne, nativo de la región amazónica. De acuerdo con los usos tradicionales es empleada como diaforético, narcótico, purgativo, entre otros usos. Objetivo: Evaluar la capacidad antioxidante, cuantificar fenoles y flavonoides totales de extractos obtenidos a partir de las hojas de Ilex guayusa. Métodos: El extracto etanólico total se obtuvo mediante un equipo tipo "Soxhlet", con posterior fraccionamiento líquido/líquido con solventes de diferentes polaridades (éter de petróleo, diclorometano y acetato de etilo); se hicieron reaccionar con 2,2-difenil-1-picrilhidracilo (DPPH•) y ácido 2,2'-azino-bis-3-etilbenzotiazolin-6-sulfónico (ABTS•+), determinando la concentración inhibitoria 50 (IC50) y la actividad antioxidante relativa (%AAR), con posterior análisis estadístico (ANOVA y post-hoc de Tukey). Los fenoles y flavonoides totales se cuantificaron con el método de Folin-Ciocalteu y el de formación de complejos con AlCl3, respectivamente. Resultados: El extracto etanólico de las hojas fue el que presentó mayor actividad antioxidante tanto en el método DPPH• como en el ABTS•+ con valores de IC50 de 4,58 y 3,82 ppm, respectivamente. Conclusiones: De acuerdo con los resultados obtenidos en este estudio, se pudo corroborar que el extracto etanólico obtenido de las hojas de I. guayusamostró la mayor capacidad antioxidante, por los métodos DPPH• y ABTS•+. Adicionalmente, se pudo relacionar esta capacidad con el alto contenido de flavonoides presentes en este extracto. Con base en lo anterior y acompañado de futuros estudios, la especie I. guayusa podría proponerse como una importante fuente de compuestos antioxidantes con posible aplicación en los campos de la medicina y la industria alimentaria

ABSTRACT

To study the chemical constituents from the the deep-sea fungus Alternaria sp. F49. Seven compounds were isolated from the EtOAc extract by using silica gel, Sephadex LH-20, ODS and HPLC methods. Based on the spectroscopic analysis, their structures were identified as (8R)-5-O-methyl-orcinotriol (1), orcinotriol (2), α-acetylorcinol (3), 3'-hydroxyalternariol 5-O-methyl ether (4), altenusiol (5), altenusin (6), and 5'-methoxy-6-methyl-biphenyl-3,4,3'-triol (7). (8R)-5-O-Methyl-orcinotriol (1) is a new phenolic compound which has never been reported in the literature. Compounds 4-7 showed strong DPPH free radical scavenging activity; whereas compounds 1-7 showed strong ABTS free radical scavenging activity.

By analyzing the chromatographic and biological activity fingerprints of 16 batches of Polygoni Multiflori Radix Praeparata, the dose-effect relationship was established and the total antioxidant activity was quantified by activity addition.The results suggested that the online HPLC-DPPH method can evaluate the antioxidant activity of different bathches of Polygoni Multiflori Radix Praeparata, with different processing methods, aiming to provide datasupport and scientific basis forquality evaluation of Polygoni Multiflori Radix Praeparata.

@# Objective: To evaluate the antioxidant potential and pancreatic lipase inhibitory action of optimized hydroethanolic extracts of Solanum nigrum. Methods: Optimized extraction for maximum recovery of metabolites was performed using a combination of freeze-drying and ultrasonication followed by determination of antioxidant and antiobesity properties. The ultra-high performance liquid chromatography equipped with mass spectrometry was used to analyze metabolite profiling of Solanum nigrum. Computational studies were performed using molecular docking and electrostatic potential analysis for individual compounds. The hypolipidemic potential of the most potent extract was assessed in the obese mice fed on fat rich diet. Results: The 80% hydroethanolic extract exhibited the highest extract yield, total phenolic contents, total flavonoid contents along with the strongest 2,2-diphenyl-1-picrylhydrazyl scavenging activity, total antioxidant power, and pancreatic lipase inhibitory properties. The 80% hydroethanolic extract not only regulated the lipid profile of obese mice but also restricted the weight gain in the liver, kidney, and heart. The 80% hydroethanolic extract also reduced alanine transaminase and aspartate transaminase concentrations in serum. The effects of plant extract at 300 mg/kg body weight were quite comparable with the standard drug orlistat. Conclusions: Solanum nigrum is proved as an excellent and potent source of secondary metabolites that might be responsible for obesity mitigation.

Abstract The total phenolic compound and fatty acid profiles of lipids from microalgae are unique. The present study was designed to investigate aqueous, ethanolic and acetone extracts of several algae (Spirogyra sp., Spirulina sp.,Chlorella sp and Chara sp.) for their antioxidant capacities of the crude extracts and fractions by radical scavenging activity against the stable radical 1,1-diphenyl-2-picrylhydrazyl DPPH as well; total phenolic content. The results showed that Spirulina sp. indicated significantly higher total phenolic compound and antioxidant activities compared to the other species (P 0.05) and acetone extracts showed higher quantity among three extracts. The fatty acids analysis using High performance liquid chromatography HPLC showed the presence of palmitic acid, stearic acid, oleic acid, and linoleic acid, palmitic acid showed high quantity than other fatty acid classes in all studied algae. This study concluded that high antioxidant capacity of microalgae could be inspected for different industrial applications.


Resumo O composto fenólico total e os perfis de ácidos graxos dos lipídios das microalgas são únicos. O presente estudo foi desenhado para investigar extratos aquosos, etanólicos e acetona de várias algas (Spirogyra sp., Spirulina sp., Chlorella sp. e Chara sp.) Quanto às suas capacidades antioxidantes dos extratos brutos e frações por atividade de eliminação de radicais contra o radical estável 1,1-difenil-2-picrilhidrazil DPPH também; fenólico total. Os resultados mostraram que a Spirulina sp. indicaram atividade antioxidante e compostos fenólicos totais significativamente maiores em relação às outras espécies (P 0,05), e os extratos de acetona apresentaram maior quantidade entre os três extratos. A análise de ácidos graxos por cromatografia líquida de alta eficiência - HPLC mostrou a presença de ácido palmítico, ácido esteárico, ácido oleico e ácido linoleico; o ácido palmítico apresentou maior quantidade do que outras classes de ácidos graxos em todas as algas estudadas. Este estudo concluiu que a alta capacidade antioxidante pode ser inspecionada para diferentes aplicações industriais.

ABSTRACT

Food handlers plays a primary role in the transmission of pathogenically important protozoans and helminth parasites. This study was aimed to evaluate the prevalence of intestinal pathogenic protozoans and helminth parasites among food handlers in and around University of Malakand, Lower Dir, Pakistan. Stool samples were collected from 642 food handlers (all of male) in a cross-sectional study from January to November, 2017. Wet Mount Techniques and concentration methods by using salt and formol­ether solutions. Three hundred and eighty four cases (59.8%) were found infected with one more parasites. Most of the individuals were found infected with helminth (47.6%) as compared to intestinal protozoans (0.93%). Seventy two cases (11.2%) of the cases presented mixed infection with both intestinal protozoan and helminth parasites. The order of prevalence for intestinal helminth was Ancylostoma duodenale (n = 258, 40.1%), followed by Taeniasa ginata (n=96, 14.9%) Ascaris lumbricoides (n = 54, 8.40%) and Trichuris trichura (n=30, 4.60%). For intestinal protozoa, Entamoeba histolytica/díspar (n = 36, 5.64%) was the only protozoan detected. Mono-parasitism was higher than poly-parasitism. Family size income and education level were the factors significantly (P<0.05) associated in the parasites prevalence. Current research showed that IPIs are primarily the foodborne pathogens still an important public health problem in Pakistan. Effective control programs on parasitic diseases transfer and their associated factors are recommended.


Os manipuladores de alimentos desempenham um papel fundamental na transmissão de protozoários e helmintos parasitas patogenicamente importantes. Este estudo teve como objetivo avaliar a prevalência de protozoários patogênicos intestinais e helmintos parasitas entre manipuladores de alimentos na Universidade de Malakand, Lower Dir, Paquistão. Amostras de fezes foram coletadas de 642 manipuladores de alimentos (todos do sexo masculino) em um estudo transversal de janeiro a novembro de 2017. Técnicas de montagem úmida e métodos de concentração usando soluções de sal e formol-éter. Trezentos e oitenta e quatro casos (59,8%) foram encontrados infectados com mais um parasita. A maioria dos indivíduos foi encontrada infectada por helmintos (47,6%) em comparação com protozoários intestinais (0,93%). Setenta e dois casos (11,2%) dos casos apresentavam infecção mista com protozoários intestinais e helmintos parasitas. A ordem de prevalência de helmintos intestinais foi Ancylostoma duodenale (n = 258, 40,1%), seguido por Taeniasa ginata (n = 96, 14,9%) Ascaris lumbricoides (n = 54, 8,40%) e Trichuris trichura (n = 30, 4,60 %). Para protozoários intestinais, Entamoeba histolytica / dispar (n = 36, 5,64%) foi o único protozoário detectado. Monoparasitismo foi maior do que poliparasitismo. A renda familiar e o nível de escolaridade foram os fatores significativamente (P <0,05) associados na prevalência de parasitos. A pesquisa atual mostrou que os IPIs são principalmente os patógenos de origem alimentar, ainda um importante problema de saúde pública no Paquistão. Programas eficazes de controle da transferência de doenças parasitárias e seus fatores associados são recomendados.
