ABSTRACT
Cancer still has elevated morbidity and mortality, which undoubtedly impacts the life quality of affected individuals. Remarkable advances have been made in cancer therapy, although the toxicities of traditional therapies remain an obvious challenge. Dahuang Zhechong Pill (DHZCP), developed by Zhongjing Zhang in the Synopsis of the Golden Chamber, represents an effective anticancer traditional Chinese medicine (TCM). In this review, it was found that DHZCP is therapeutically utilized in liver, lung, gastric, pancreatic and other cancers in clinic. Pharmacological evidence showed that its anti-tumor mechanisms mainly involve induced cell cycle arrest, apoptosis and autophagy, as well as suppressed tumor cell proliferation, obstructed angiogenesis and metastasis, enhanced immunity, and reversal of multidrug resistance. The present review provides a solid basis for the clinical application of DHZCP and may promote the wide use of TCM in clinical antitumor application.

The infiltration of immune cells into the hepatocellular carcinoma microenvironment is the main reason why hepatocellular carcinoma patients are prone to carcinoma recurrence and the disease are incurable. Notably, the infiltration of Treg cells is the main trigger. Dahuang Zhechong pill (DHZCP) is a traditional Chinese herbal compound successful in the treatment of hepatitis and hepatocellular carcinoma. DHZCP can heal and nourish while slowing the onset of the disease, thereby strengthening the body's immune function. It can localize tumors and ultimately achieve the goal of eliminating tumors. In this study, an orthotopic liver cancer model of mice was used to explore the mechanism of DHZCP enhancing anti-tumor immunity, which showed more Th1 cells in the peripheral blood and spleen after DHZCP treatment, while more IFN-γ was secreted to activate CD8+ T cells and Treg cell production was inhibited, thereby suppressing the growth of HCC. Finally, we also analyzed the potential components of DHZCP from the perspective of modern targets using network pharmacology methods and experimental results.

ObjectiveTo investigate the clinical effect of Dahuang Zhechong capsules combined with entecavir in the treatment of chronic hepatitis B (CHB) patients with liver fibrosis. MethodsA total of 100 CHB patients with liver fibrosis who visited or were hospitalized in Shenzhen Hospital of Peking University from October 2014 to January 2016 were enrolled and randomly divided into Western medicine group and combined treatment group, with 50 patients in each group. The patients in the Western medicine group were given entecavir, and those in the combined treatment group were given Dahuang Zhechong capsules in addition to entecavir. The course of the treatment was 48 weeks. The changes in liver function, HBV DNA load, four serum parameters of liver fibrosis, liver stiffness, and aspartate aminotransferase-to-platelet ratio index (APRI)/FibroIndex were observed in both groups. The t-test was used for comparison of continuous data between groups, and the chi-square test was used for comparison of categorical data between groups. ResultsBoth groups showed significant reductions in serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and HBV DNA load after 48 weeks of treatment (all P<0.05). After treatment, the four serum parameters of liver fibrosis all returned to normal after treatment, and the serum levels of hyaluronic acid, type Ⅲ precollagen, and type IV collagen showed significant differences between the two groups (all P<005), and the portal vein diameter, thickness of the spleen, liver stiffness and APRI/FibroIndex also showed significant differences between the two groups (both P<0.05). The combined treatment group had a significantly higher overall response rate than the Western medicine group (920% vs 720%, χ2=6.775, P=0009). ConclusionDahuang Zhechong capsules combined with entecavir have a better effect in the treatment of CHB patients with liver fibrosis compared with entecavir alone.

Objective To explore effect of small dose of diuretics combined with Dahuangzhechong pill on testosterone, Cystatin C(CysC), leptin and β2 microglobulin(β2-MG) in serum in patients with chronic renal failure.Methods 69 cases of kidney disease patients were collected and divided into two groups.Testosterone, Cystatin C, leptin and β2 microglobulin in serum were compared before and after the treatment.The control group was given hydrochlorothiazide tablets, treatment group was given Dahuangzhechong pill based on the control group.All patients were treated for 3 months. Results After treatment, levels of testosterone elevated (P <0.05), CysC, leptin and β2 microglobulin decreased (P <0.05) in the two groups compared with before treatment;Compared with the control group , level of testosterone was higher (P<0.05), levels of CysC ,leptin and β2-MG were lower (P<0.05).Conclusion Low dose diuretics combined with Dahuang Zhechong pill has good effect in treatment of chronic renal failure curative.The mechanism may be related to elevate serum testosterone levels and reduce serum CysC , leptin and β2-MG level in patients.

OBJECTIVE:To evaluate the clinical effect of Dahuang zhechong pill on chronic hepatitis with hepatic fibrosis.METHODS:A total of 128 patients with chronic hepatitis with hepatic fibrosis were randomly divided into treatment group(68 cases) and control group(60 cases).The control group received routine hepatic protective therapy,while the treatment group received routine hepatic protective therapy and Dahuang zhechong pills(6 g?d-1) concomitantly.The course of treatment lasted for 6 months.The clinical symptoms,liver function,hyaluronic acid(HA),pro-collagen Ⅲ(PC Ⅲ),cholyglycine(CG) and collagen type Ⅳ(ⅣC),and ultrasound image were observed in both groups.RESULTS:Levels of serum HA,PCⅢ,CG,and ⅣC decreased significantly in treatment group,and the difference between two groups was significant(P

objective Microbial limit test(MLT)for Dahuang Zhechong pill was established and verified to supply scientific foundation for the establishment of MLT for antibacterial and anti-inflammatory Chinese patent medicine,and to supply principle guidance for the verification of the method issued in Chinese pharmacopoeia(2005 edition);meanwhile the antibiogram of Dahuang Zhechong pill was probed,which can provide evidence for its clinical administration.Methods With centrifugal sedimentation method and culture medium diluted method,the recovery test was done on each tested bacterial strain to validate the accuracy and feasibility of MLT.Results Dahuang Zhechong pill had a stronger inhibitory effect on staphylococcus aureus and bacillus subtilis,had a weak inhibitory effect on escherichia coli,and had no effect on candida albicans and aspergillus niger,indicating that the antimicrobial spectrum of Dahuang Zhechong pill is Gram-positive bacteria.Conclusion Centrifugal sedimentation method combined with culture medium diluted method can eliminate the anti-bacterial effect of Dahuang Zhechong pill during the experiment,thus the microorganisms in the polluted pill can be detected successfully.

AIM: To observe the anti-atherosclerotic mechanism of Dahuang Zhechong Pill(Radix et Rhizoma Rhei,Eupolyphaga seu steleophaga,Hirudo,Radix Scutellariae,etc) in the aspect of VSMC proliferation and apoptosis,collagen synthesis in the vessel wall. METHODS: The atherosclerotic model was established by hypercholesterol feeding together with immune-injured endothelium in rabbits.The level of hydroxyproline in vessel wall was determined.Histological changes were examined by electronic microscopes.The proliferation of VSMC was determined by immunohistochemistry and apoptosis of VSMC by TUNEL. RESULTS: Dahuang Zhechong Pill improved the histological changes of vessel wall of rabbit with atherosclerosis and reversed vascular remodelling.The level of hydroxyproline in the vessel wall and the number of proliferation of VSMC decreased,and the number of apoptosis of VSMC increased in Dahuang Zhechong Pill-administered groups compared with the model control group. CONCLUSION: Dahuang Zhechong Pill can inhibit the development of atherosclerosis by inhibiting the synthesis of collagen fiber in vessel wall and the proliferation of VSMC as well as inducing the apoptosis of VSMC.
