ABSTRACT

For sustainable agriculture to succeed, especially in the production of organic food, the use of biofertilizers and biopesticides is essential. Arbuscular mycorrhizal symbiosis is important for improving plant development through increased nutrient intake, soil stabilization, and carbon sequestration through the creation of glomalin, a distinct and specialized protein. It was not determined whether glomalin affects soil-borne fungi infections. Therefore, using glomalin isolated from the soil used to raise maize plants inoculated with various arbuscular mycorrhizal fungi (AMF), the inhibition of soil-borne plant diseases Sclerotium rolfsii and Fusarium oxysporum was examined. The outcomes demonstrated that there was variation in the quantity of glomalin generated by various AMF species. Glomalin isolated from soil inoculated with Glomus coronatum suppressed Sclerotium rolfsii and Fusarium oxysporum more effectively in in-vitro tests than soil inoculated with Glomus intraradices and Glomus mosseae.

To optimize nursery practices for efficient plant production procedures and to keep up to the ever growing demand of seedlings, identification of the most suitable species of arbuscular mycorrhizal fungi (AMF), specific for a given tree species, is clearly a necessary task. Sixty days old seedlings of Neem (Azadirachta indica A. Juss) raised in root trainers were inoculated with six species of AMF and a mixed inoculum (consortia) and kept in green house. Performances of the treatments on this tree species were evaluated in terms of growth parameters like plant height shoot collar diameter, biomass and phosphorous uptake capabilities. Significant and varied increase in the growth parameters and phosphorous uptake was observed for most of the AMF species against control. Consortia culture was found to be the best suited AMF treatment for A.indica, while Glomus intraradices and Glomus mosseae were the best performing single species cultures. It is the first time in the state of Gujarat that a wide variety of AMF species, isolated from the typical semi-arid region of western India, were tested for the best growth performance with one of the most important tree species for the concerned region.

ABSTRACT

Ferns, which are usually colonizing different environments and their roots frequently present mycorrhization, have two adult stages in their life cycle, the sporophytic and the gametophytic phase. This paper describes the experimental mycorrhizal association between Pteris vittata leptosporangiate fern and a strain of Glomus intraradices during the life cycle of the fern, from spore germination to the development of a mature sporophyte. The aim of this study was to compare the colonization pattern of in vitro cultures of G. intraradices along the fern life cycle with those found in nature. For this, mature spores were obtained from fertile P. vittata fronds growing in walls of Buenos Aires city, Argentina. Roots were stained and observed under the light microscope for arbuscular mycorrhizal colonization. Approximately, 75 fern spores were cultured in each pot filled with a sterile substrate and G. intraradices (BAFC N° 51.331) as inoculum on the surface. After germination took place, samples were taken every 15 days until the fern cycle was completed. In order to determine colonization dynamics each sample was observed under optical and confocal microscope after staining. Gametophyte was classified as Adiantum type. Male and female gametangia were limited to the lower face, mycorrhizal colonization started when they were differentiated and took place through the rhizoids. Spores and vesicles were not found in this cycle stage. Paris-type mycorrhizal colonization was established in the midrib and in the embrionary foot. It was colonized by external mycelium. When the first root was developed soil inoculum colonized de novo this structure and Arum-type colonization was observed. This study proves that the type of colonization is determined by the structure of the host, not by the fungus. Both the gametophyte and embryo foot have determined growth and Paris-type colonization, while, sporophyte roots have undetermined growth and Arum-type colonization. Los helechos presentan dos etapas en su ciclo de vida, una fase esporofítica y una gametofítica. Estos por lo general pueden colonizar diferentes ambientes y frecuentemente presentan raíces micorrizadas. Este estudio describe la asociación experimental entre Pteris vittata, un helecho leptosporangiado y una cepa de Glomus intraradices durante el ciclo de vida del helecho, desde la germinación de las esporas hasta el desarrollo del esporofito maduro. El objetivo de este estudio fue comparar los patrones de colonización de G. intraradices a lo largo de todo el ciclo de vida del helecho con los tipos encontrados en la naturaleza. Las esporas maduras fueron obtenidas de frondes fértiles de P. vittata que crecen sobre las paredes de la ciudad de Buenos Aires, Argentina. Las raíces se tiñeron y fueron observadas bajo microscopio óptico para el estudio de la colonización micorrízica. Aproximadamente 75 esporas de helecho se cultivaron en macetas con un sustrato estéril y con un inóculo de G. intraradices (N° 51.331 BAFC) en la superficie. Después de la germinación, se tomaron muestras cada 15 días hasta que se completó el ciclo de vida del helecho. Con el fin de determinar la dinámica de la colonización, cada muestra se observó con el microscopio óptico y el microscopio de confocal luego de la tinción correspondiente. El gametofito fue clasificado como del tipo “Adiantum”. Los gametangios femeninos y masculinos se desarrollaron en la cara inferior del mismo. La micorrización comenzó cuando los gametangios estaban ya diferenciados y la colonización se produjo a través de los rizoides. Las esporas y las vesículas no se encontraron en esta fase del ciclo. La micorrizacion tipo Paris se observó sobre la línea de la nervadura central. El pie del esporofito fue colonizado por el micelio externo. Cuando la raíz se desarrolló, se colonizó “de novo”, y se observó una colonización de tipo Arum. Este estudio demuestra que el tipo de colonización está determinado por la estructura del helecho y no por el hongo. Tanto el gametofito como el pie del embrión tienen crecimiento definido y colonización tipo Paris, mientras que las raíces del esporofito presentan un crecimiento indeterminado y una colonización tipo Arum. Las estructuras que se encontraron bajo cultivo coinciden con las que se encontraron en condiciones naturales.

ABSTRACT

This study evaluated the effects of inoculation with the arbuscular mycorrhizal fungi Glomus mosseae, Glomus intraradices A4 and Glomus intraradices B1 and two phosphorus levels (10 and 40 mg kg-1) on root colonization, plant growth, nutrient uptake and essential oil content in Mentha piperita L. The experiment was carried out in a greenhouse, in 4x2 factorial arrangement, in completely randomized design. At sixty days after transplanting, the mycorrhizal plants had significantly higher fresh matter, dry matter and leaf area compared to non-mycorrhizal plants. The inoculation increased P, K and Ca levels in the shoot which were higher under 40 mg P kg-1 of soil. Plants grown with 40 mg P kg-1 soil increased the essential oil yield per plant by about 40-50% compared to those cultivated with 10 mg P kg-1, regardless of the mycorrhizal treatment. Among the studied fungal species, inoculation with G. intraradices A4 and a high level of P significantly increased plant growth and essential oil yield, compared to the other studied mycorrhizal fungal species. In conclusion, inoculation of arbuscular mycorrhizal fungi into peppermint plants is a feasible alternative to increase the essential oil production and reduce the use of fertilizers required to obtain economic production of peppermint under phosphorus-deficient soil condition.


Este estudo avaliou os efeitos da inoculação de fungos micorrízicos arbusculares Glomus mosseae, Glomus intraradices A4 e Glomus intraradices B1 e duas doses de fósforo (10 e 40 mg kg-1) sobre a colonização radicular, crescimento, absorção de nutrientes e óleos essenciais em Mentha piperita L. O estudo foi conduzido em casa de vegetação no delineamento inteiramente casualizado em esquema fatorial 4x2. Sessenta dias após o transplantio, as plantas micorrizadas apresentaram massa fresca, massa seca, e área foliar significativamente maior em comparação as não-micorrizadas. A inoculação aumentou o teor de P, K e Ca na parte aérea sendo superiores em 40 mg P kg-1 de solo. As plantas cultivadas com 40 mg P kg-1 de solo aumentaram a produção de óleo essencial por planta cerca de 40-50% em relação às cultivadas com 10 mg de P kg-1, independentemente da micorrização. Dentre as espécies fúngicas estudadas, a inoculação com G. Intraradices A4 e com um elevado nível de P, aumentou significativamente o crescimento e rendimento de óleos essenciais em comparação com outras espécies de fungos micorrízicos estudados. Em conclusão, a inoculação dos fungos micorrízicos arbusculares em plantas de hortelã é uma alternativa viável para aumentar a produção de óleos essenciais e reduzir o uso de fertilizantes necessários para a produção econômica de hortelã-pimenta com deficiência de fósforo no solo.

ABSTRACT

Con el objetivo de incrementar y acelerar el proceso de germinación de las semillas y obtener una alta producción y homogeneidad de plántulas de Carica papaya variedad Maradol en vivero, se evaluó el efecto de tres biofertilizantes aplicados solos o en combinación (Azotobacter chroococcum, Azospirillum brasilense y Glomus intraradices), y un biorregulador del crecimiento vegetal, el ácido giberélico (AG3), en la germinación y el crecimiento vegetal. Se realizó un experimento bajo un diseño completamente al azar con ocho tratamientos y tres repeticiones. A las semillas se les aplicó un pretratamiento germinativo con alternancia de temperatura para superar la dormancia. Los tratamientos simples con A. chroococcum y A. brasilense, incrementaron el porcentaje de germinación a 90,28 y 88,89% respectivamente. Además, con la aplicación de los biofertilizantes y el AG3, la velocidad de germinación se incrementó y el tiempo medio de germinación se redujo. La doble aplicación en semillas y foliar de los biofertilizantes y el AG3 en plántulas mejoró el crecimiento vegetal. La población de A. chroococcum fue mayor cuando se inoculó en combinación con G. intraradices. La prevalencia de colonización de las plántulas inoculadas con G. intraradices varió de 18,53 a 26,67%, con el mayor valor registrado para el tratamiento combinado con A. brasilense. Finalmente, aplicando esta metodología se logró acelerar la germinación, obteniéndose una mayor homogeneidad en la emergencia de las plántulas, disminuyendo así el tiempo de permanencia en el vivero.


In order to increase and accelerate the process of seed germination and obtain a high yield and homogeneity of papaya seedlings cv. Maradol in nurseries, we evaluated the effect of three biofertilizers applied single or in combination (Azotobacter chroococcum, Azospirillum brasilense and Glomus intraradices) and a plant growth bioregulator, the gibberellic acid 3 (AG3), on the germination and subsequent growth of papaya seedlings. An experimental design completely random with eight treatments and three replications were used. The application of a pre-germinal treatment with alternating temperature had to be applied to seeds to overcome dormancy. Single biofertilization with A. chroococcum and A. brasilense, promoted the germination percentage 90.28 y 88.89% respectively. Germination rate could be enhanced and the mean germination time was reduced with the application of biofertilizer and AG3. Both applications on seeds and leaves of biofertilizers and AG3, had a positive effect on plant growth. The population of A. chroococcum was higher in the combined inoculation with G. intraradices. The prevalence of colonization of plants inoculated with G. intraradices ranged from 18.53 to 26.67%, with the greatest values recorded for the treatment involving combined inoculation with A. brasilense.
